Abstract

Abstract

Mucositis is a common complication in cancer patients undergoing 5-fluorouracil (5-FU) chemotherapy. Roselle (Hibiscus sabdariffa) extract in synbiotic yogurt could be a beneficial alternative because it might change the inflammatory response and oral microbiota. The study aimed to investigate the effects of synbiotic roselle extract yogurt on inflammatory responses and oral microbiota in oral mucositis caused by 5-FU. An experimental study with twenty-four Sprague-Dawley rats divided into four groups (n=6): healthy control (NC), disease control (PC), synbiotic roselle extract yogurt therapy (P1), and standard therapy (P2) groups. This study evaluated the number of bacterial colonies, expression of COX-2 and caspase-1, and levels of IL-1β and VEGF (days 4 and 7). The P1 group had a significantly increased beneficial lactic acid bacteria (6.91±0.87) and a decrease in pathogenic bacteria, including Staphylococcus aureus (3.89±0.05), Escherichia coli (0), and Enterobacter aerogenes (1.78±0.29), compared to the PC group. Additionally, there was a statistically significant increase in VEGF levels in the tissue (0.07±0.03 ng/mL), a decrease in serum IL-1β levels (48.02±10.29 pg/mL), and an expression of caspase-1 and COX-2 compared to the PC group (p<0.05). Synbiotic roselle extract yogurt shows promise as a therapeutic strategy for managing mucositis by restoring microbial balance and mitigating inflammation.

Abstract

Preeclampsia (PE) stands as a prominent cause of maternal mortality in developing nations, yet a standardized therapy for PE has not been established. Some pregnant women in Lampung have consumed the water of young kopyor coconut fruit from the Kalianda variety (WKK), although its therapeutic effects remain unexplored. This study aimed to investigate WKK phytochemicals through in vitro and silico analyses. Liquid Chromatography-Mass Spectrometry (LCMS) was employed for phytochemical analysis, while an in silico study involved Autodock, Autodock Tools, Autodock Vina, Biovia Discovery Studio 2020, and Open Babel GUI, alongside pharmacokinetics prediction using the pkCSM strategy. The study assessed the inhibitory potential of WKK on Gentiatibetine and curcumenolacto C, targeting the PE ACE2 receptor (1R24) through molecular docking with the 3D structure. Post-docking analysis, including binding affinities, hydrophobic interactions, and pharmacokinetic predictions, was conducted. WKK exhibited relatively low binding affinities for Gentiatibetine (-4.86 kcal/mol), curcumenolactone C (-2.96 kcal/mol), and aspirin (-5.12 kcal/mol). Multiple hydrophobic interactions were observed, such as Van der Waals, Salt Bridge, Conventional Hydrogen Bond, Alkyl, 162, and Lys 129. The receptor IR displayed a high bond-free energy, like aspirin docked with the same gene receptor. Pharmacokinetics predictions indicated that WKK possesses a favorable profile. In conclusion, WKK phytochemicals demonstrated a notable docking score comparable to aspirin, suggesting its potential for preventive therapy use.

Abstract

Iron deficiency anemia (IDA) in female adolescents is one of the health problems, which occupied 8th position as a cause of global disability and is the main cause of anemia in reproductive women. Iron tablet supplementation is the government program to reduce IDA prevalence in Indonesia but often results in gastrointestinal side effects. Jamaican cherries fruits contain vitamin C, iron and flavonoids, which play essential roles in iron metabolism. This study aimed to investigate the effects of Jamaican cherries fruits ethanol extract on clinical manifestations and behavior changes in female Wistar rats. This randomized controlled trial with pre-posttest control group design used 24 female Wistar rats, aged 10 -12 weeks and were randomly divided into 6 groups: N, positive control (PC: AIN-93M low iron food + elemental iron) and treatments (T1-4: AIN-93M low iron food + 0.26, 0.35, 0,5 or 0.75g/ 100g Body Weight (BW) Jamaican cherries fruits extract respectively for 28 days). Clinical manifestations, reflexes and behavior changes were observed at day 0, 14 and 28 interventions. BW data were analyzed using one way ANOVA and repeated measures ANOVA tests. All rats had normal reflexes and did not show clinical manifestations and behavior changes. The average of rat BW in T1-4 groups did not significantly differ before (p=0.896), during (p=0.884) and after treatment (p=0.775) but the average of rat BW in the T1 group increased significantly (p=0.012). In conclusion, administering Jamaican cherries fruits ethanol extracts up to 0.75g/100g BW has no toxic effects in female Wistar rats. Abstract

The imbalance between food intake and energy expenditure can lead to obesity. Obesity is a major risk factor for chronic diseases such as cardiovascular disease and can cause premature death worldwide. This study is aimed to investigate the effect of Star Fruit Leaves Extract (SLE) and T. accumulate Leaves Extract (TLE) combination on Body Weight (BW), Body Mass Index (BMI), and Fat Content (FC) of obese rats. Male Wistar rats aged 5 weeks, with BW of 100-150 g were induced with a High Fat Diet (HFD) and 10% fructose solution for 30 days. A total of 18 rats were randomly divided into 3 groups, namely Negative Control (NC), Therapy (T), and Normal (N). After 35 days of treatment compared to the NC group, the mean BW of the T group was significantly lower (p=0.014) and the mean decrease was significantly higher (p less than 0.001).  Similarly, the mean BMI and FC of the T group were significantly lower and the decrease in the mean of both groups was significantly higher than that of the NC group (p less than 0.001). Therefore, the combination of SLE and TLE can be used as a treatment for obesity because it reduces BW, BMI, and FC better than only a low-calorie diet. Abstract

Anemia is a serious maternal health problem that can result in long-term negative impacts such as preeclampsia, hemorrhage before and after delivery, stunting, reduced baby immunity, fetal retardation growth, and fetal death. Moringa oleifera is a miracle plant that has been used for anemia treatment. This study aimed to analyze systematically the effect of M. oleifera leaves on hemoglobin levels and dietary intake of iron and zinc in pregnant women with anemia. Our study used the Cochrane tool to select all randomized control trial (RCT) articles and met the requirements of the PRISMA diagram, filtered using some keywords: pregnant women, anemia, M.oleifera powder, and Fe supplements. The selected articles were further screened using the Population, Intervention, Comparator, Outcomes, and Study Type (PICOS) protocol and assessed for their quality using the Cochrane tool bias risks. Five full-text articles used M.oleifera leaves for anemia treatment in women with their first and or third trimester pregnancies. However, increased Hb levels in pregnant women who consumed 100g of fresh M.oleifera leaves with iron supplementation were higher than that of pregnant women who consumed M oleifera powder and extract. In addition, only one full-text article reported dietary intake of Fe and Zn as secondary outcomes after treating M. oleifera leaves. In conclusion, the administration of M. oleifera leaves has a minor effect on Hb levels and no effect on dietary intake of Fe and Zn in pregnant women with anemia.

Abstract

Latar Belakang.
