Abstract

Alginate is known as an immunostimulant. The comprehensive study of Artemia on its co-activity with Lactobacillus bulgaricus resolves the relationship of feed digestibility, hematological parameters, gene expression, feed digestibility, and disease-resistant have not been covered. This study aimed to determine the effect of various doses alginate with Lactobacillus bulgaricus bio-encapsulated Artemia in Vibrio vulnificus, V. parahaemolyticus, and V. harveyi challenged. Alginate from Sargassum sp. L. bulgaricus were cultured and grown with de Man, Rogosa and Shape media. Nauplii Artemia was encapsulated for one hour with 400, 600, 800 ppm alginate doses, with and without probiotics. The Artemia then was challenged with three Vibrio spp. The Artemia mortality, immune parameters (Phenol Oxidase, Super-oxide Dismutase) were evaluated. Pro Phenol Oxidase, b-1,3-glucan-binding proteins (LGBP), and Lectin gene expression as well as gut evacuation time, fullness of gut were recorded.  Compared to the non-probiotic alginate, co- probiotic activated and improved the mortality rate of 400 ppm alginate from 100% to 30-70% (36 h), CMI of 276-702, and mortality reduction (41.21-74.59%). The co-activity of 400 ppm alginate and L. bulgaricus resulted in higher PO and SOD activity. LGBP and proPO gene expression were also upregulated 233.44% and 185.17%. The gut evacuation time and fullness of alginate 400 ppm and L. bulgaricus treatment have also resulted in better performance than those of 800 ppm alginate without probiotics. Alginate and L. bulgaricus probiotics cooperated synergically through pre and probiotic mechanisms. This Artemia bio model defines that this combination will improve the survival rate, immune system, and gene expression. In the future this will be beneficial in terms of shrimp production in ponds.

Abstract

The lack of consumer disloyalty in the shopping process, especially amid Shopee's dominance as a market leader, is the focal point of this research. Shopee managed to achieve top rankings in shaping customer loyalty in the online marketplace. This study aimed to identify several factors that affect customer loyalty on the Shopee platform through trust, good service at Shopee, and satisfaction. Considers related to the important role e-satisfaction plays as a mediating variable in understanding the impact of e-trust and e-service quality on Shopee customer loyalty. A quantitative research design was used, with 100 respondents receiving a questionnaire via Google Form, using simple random sampling techniques in data collection. Data analysis was performed through Structural Equation Modeling (SEM) using the SMART PLS application. Research findings show that the impact of e-trust on Shopee consumer loyalty is quite significant and that the quality of e-service also significantly impacts Shopee customer e-loyalty. In contrast, e-satisfaction is a mediator that significantly affects customers’ e-loyalty levels. Managers must focus on improving the quality of e-services for customer satisfaction. Abstract

A molecular docking study demonstrates the interaction between naringin, a citrus flavonoid, with SARS-CoV-2 spike RBD. Nevertheless, in vitro investigation of the inhibitory effect of naringin on SARS-CoV-2 entry and syncytia models has yet to be carried out. We synthesized VSV∆G-GFP/Spike* pseudovirus (PSV) as a SARS-CoV-2 model by pseudotyping VSV∆G-GFP/S* in BHK-21 cells overexpressing the SARS-CoV-2 spike glycoprotein. In the SARS-CoV-2 PSV entry assay, we utilized CHO-K1 cells transfected with hACE2 plasmid, which were then treated with naringin and SARS-CoV-2 PSV/naringin. After 16-18 h incubation, PSV internalization represented by the GFP signal was observed under a fluorescence microscope. Immunofluorescence staining was also performed to probe the SARS-CoV-2 spike and confirm the PSV entry. We performed a syncytia assay using 293T cells co-transfected with SARS-CoV-2 spike/hACE2. Six hours after transfection, the cells were treated with naringin and incubated for another 16-18 hours. Then, we observed syncytia using a phase contrast microscope. Based on fluorescence foci quantification, the results indicated that naringin might inhibit SARS-CoV-2 PSV entry at a concentration of 100 µM (P<0.05). However, naringin did not prevent syncytia formation compared to solvent control. These PSV entry and syncytia assay results suggested that naringin potentially inhibited SARS-CoV-2 viral infection but not cell-to-cell viral transmission.
