Abstract

This research aimed to characterize VirB4 protein of local isolate Brucella abortus with Western blotting method. The result showed that there were four protein bands with molecular weights of 64.61, 59.25, 21.63, and 16.70 kDa by triggering a reaction between the whole Brucella abortus and anti-Brucella abortus serum. The results also revealed that there was only one protein band with a molecular weight of 59.25 kDa triggering a reaction between the whole Brucella abortus and anti-VirB Brucella abortus serum. Finally, it can be concluded that VirB4 protein can affect the virulence factor of Brucella abortus, successfully characterized with the appearance of one band with a molecular weight of 59.25 kDa by using Western blotting method.

Abstract

The aim of this research was to determined the prevalence and infection degrees of endoparasite on layer chicken in Sub-district Udanawu, Blitar. Ninety-six samples were taken from layer chicken in 3 different villages namely Bakung Village, Tunjung Village, and Slemanan Village. The examinations taken in this study are fecal examination using native, sediment, and floating methods and blood examination using blood smear method. Result showed that 81.25% samples are positive for helminthiasis infection consisting of Ascaridia galli (66.67%), Heterakis gallinarum (45.83%), Raillietina sp. (31.25%), and Strongyloides avium (7.29%). Blood examination result shown there is no positive sample that infect layer chicken in Sub-district Udanawu, Blitar. Chi-Square test result showed there are significant difference (P<0.05) of Ascaridia galli and Heterakis gallinarum in Bakung Village, Tunjung Village, and Slemanan Village in Sub-district Udanawu, meanwhile there are no significant difference (P>0.05) of Raillietina sp. and Strongyloides avium. Range of infection degrees of helminthiasis in Bakung village, Tunjung village, and Slemanan Village are 608.75 ± 588.53, 223.12 ± 359.21, 156.25 ± 332.39. There are significant difference (P<0.05) on helminthiasis infection degree of layer chicken in Udanawu District, Blitar.

Abstract

Fasciolosis is one of the endemic parasitic diseases in Indonesia and causes significant economic losses. Fasciola gigantica is the main species found to infect livestock. This research aimed to describe the morphology of F. gigantica histologically using hematoxylin and eosin staining as well as ultrastructure morphology using scanning electron microscopy (SEM). Besides, the study was also carried out to determine the prevalence and estimation of economic losses due to fasciolosis in Berau Regency. The histological identification of flukes morphology found several organs including oral sucker (OS), ventral sucker (VS), cirrus (C), genital pore (GP), uterus (U), ovary (O), tegument (Te), testis (T), and caecum (C). In this study, ultrastructure morphology was divided into anterior and midbody region. In the anterior region, there are an oral sucker (OS) with a diameter of 304.5 µm and a ventral sucker (VS) with a diameter of 571 µm. It was also found the anterior spine with a width of 25.4 µm and a height of 22.8 µm. In the midbody, the spine has a width of ± 26.7 µm and a length of ± 38.9 µm leading posteriorly. Postmortem examination results showed that the prevalence of fasciolosis in Berau regency was 21.65% or 34 of 157 cows which were positive with an estimated loss of 228,656,475.00 IDR/16,450 USD (1 USD=13,900 IDR). The amount of loss due to fasciolosis must be a particular concern in carrying out control and prevention measures. Histology and ultrastructure morphology can become a reference in the development of anthelmintic testing that has been used so far in Indonesia.

Abstract

Abstract The development of fish consumption in Indonesia was promising increasingly and growing very rapidly. One of the priority commodities in the development of aquaculture subsector was the gourami (Osphronemus gouramy). Many factors must be faced in the cultivation of gouramy, including the problem of fish diseases caused by Lernaea cyprinacea. The aim of this research is to find out the description of anatomic pathology and histopathology of gouramy which infested by L. cyprinacea at different degrees of infestation. This research uses descriptive method. The research showed that the gouramy integument which infested by L. cyprinacea at low, moderate and heavy infestation cause moderate until heavy defect with 2.48; 2.42 and 2.32 in reaverage scoring. Hence, accordance with pathological anatomy, L. cyprinacea infestation caused necrotic nodules, lesion which covered by exudate and haemorrhage. While the histopathology of the integument changes found were inflammatory cell infiltration, congestion and haemorrhage in the low, moderate and heavy infestation.

Abstract

Antibiotic resistance now days become a main issue to the medical researches as found many positive result of antibiotic resistance test. One of the causes of antibiotic resistance is using antibiotic as a feed additive to animal. Bacteria that are resistant to antibiotics can be a danger to humans, in this case the resistant bacteria as a result of treatment errors animals, especially chickens that uses low-dose antibiotics as growth promoters. This study aimed to determine the contamination of Salmonella sp and its antibiotics resistance patterns of Salmonella sp isolated from broiler meat sold at wet market in the Center of Surabaya: (a) Pasar Kembang, (b) Pasar Kupang, (c) Pasar Dukuh Kupang, (d) Pasar Kedungsari, (e) Pasar Kedungdoro and (f) Pasar Keputran. The method that used in this study was bacteriological isolation and identification method. The method started with pre-enrichment using Buffered Pepton Water, selective enrichment using Tetrathionate Broth and Selenite Cysteine broth, selective media using Salmonella-Shigella Agar, Biochemical test using Triple Sugar Iron Agar, Simon Citrate, Methyl Red – Voges Proskauer, and Sulfide Indol Motility, and followed with susceptibility test according to Kirby-Bauer method using Mueller-Hinton Agar. The antibiotics that used in susceptibility test were: (a) Meropenem, (b) Ampicillin Sulbactam, (c) Amikacin, (d) Ofloxacin and (e) Nalidixic Acid. The results of this study were found 90% or 27 of 30 samples positive contaminated with Salmonella sp. The results of antibiotics resistance from 27 isolates 0% were resistant to Meropenem, 0% were resistant to Amikacin; 3.7% were resistant to Ampicillin-Sulbactam; 11.1% were resistant to Ofloxacin and 44.4% were resistant to Nalidixic Acid.

Abstract

One of the plants with immunostimulant activity is Tenggulun leaves which contain of flavonoid, like terpenoid. The aim of this research is to find the potential of Tenggulun's leaves extract to have the immunostimulant activities. The potential of immunostimulant activity is identified by the increasing the amount of T-cell CD4+ expression and IFNγ secretion. The research method is conducted through cultured chicken PBMC which is infected by ND virus; it is then treated with Tenggulun's leaves extract with immunostimulant. The result of immunocitochemistry examination CD4+ secretion on PBMC cultures shows how tenggulun is significantly different from the control in the secretion CD4+. The 10μg of tenggulun extract can modulate the T cell CD4+ secretion 68.8±0.83. It is significantly different from K (control) (p<0,05) and treatment group K+, P0, and P1. The examination of IFNγ level using ELISA from tenggulun leaves extract of 10μg doses were inoculated after being infected by ND virus contained immonostimulant potential in increasing the secretion of IFNγ 120.91±6.44. It is significantly different from K-, K+, and P1, yet not significantly different from P0. The content of terpenoid can increase IFNγ secretion on the macrofag cells culture and limfosit cells.

Abstract

Interaksi yang dekat antara monyet ekor panjang dengan manusia dapat meningkatkan risiko penularan penyakit zoonosis. Blastocystis sp. adalah protozoa gastrointestinal pada manusia dan hewan yang yang bersifat zoonosis. Penelitian ini bertujuan untuk mengidentifikasi Blastocystis sp. yang menginfeksi monyet ekor panjang melalui identifikasi molekuler. Identifikasi Blastocystis sp. pada penelitian ini menggunakan metode morfologis dan molekuler. Sebanyak 90 feses individu monyet ekor panjang Taman Nasional Baluran dilakukan pemeriksaan secara mikroskopis setelah dilakukan kultur pada Jones Medium. 28 dari sampel yang positif secara mikroskopis dilanjutkan dengan uji PCR dengan target primer barcode region yang mempunyai visualisasi 600bp. Tiga sampel dengan band positif 600bp dilanjutkan dengan squencing. Hasil sekuens diproses dalam BLAST dan MLST. Satu sampel yang terkonfirmasi sebagai Blastocystis sp. dengan infeksi campuran dari subtipe 1 alel 2 dan subtipe 3 alel 34. Hasil menunjukkan bahwa Blastocystis sp. terdapat pada monyet ekor panjang di Taman Nasional Baluran dengan prevalensi rendah.

Abstract

Ptyas korros or Indo-China Rat Snake is one of the snakes that are commonly hunted in Indonesia. This study aimed to identify Spirometra sp. on P. korros in Sidoarjo. A total of 18 snakes were collected within 3 months. Identification of Spirometra sp. was done using a native method, Semichen Acetic Carmine, and HE staining. As a results, 8 (44.4%) of 18 samples showed positive infection with Spirometra sp. The results of this study indicate the presence of spargana in P. korros in Sidoarjo and no adult Spirometra sp. was found.

Abstract

The problem most often encountered in fish farming is disease which can cause a decrease in fish production levels. Parasites cause one of the infectious diseases found in fish farming. The presence of parasites can cause a decrease in the quality of the growth of fish production. The purpose of this study was to identify the prevalence of ectoparasites on Nile tilapia in Sidoarjo, Indonesia. Ninety-six samples from two ponds were examined under the microscope to find out the presence of ectoparasite that exists in the outside body part of the fish. Samples were collected from skin, fins, tail, and gills using native methods and observed under a microscope in the Division of Parasitology Laboratory, Faculty of Veterinary Medicine, Universitas Airlangga. The results of this study showed 86 samples positively infected by ectoparasite and 10 showed negative. The ectoparasites found in this study were Trichodina sp., Chilodonella sp., Ichthyophthirius multifillis, Gyrodactylus sp., and Dactylogyrus sp. The highest prevalence is Trichodina sp. and Dactylogyrus sp. The number of fish infested by ectoparasite is 89.56%, this is classified in the usual category. The highest prevalence occurs in infestations of Trichodina sp. i.e.72.91% which were contained in the 70 samples. While the lowest prevalence was in Gyrodactylus sp. by 8.32% in 8 fish samples. Double infections from two different ponds were 70% within 33 samples which were included in the usual category.

Abstract

Antibiotic resistance in animals and humans has become a global problem that needs attention. The use of antibiotics in inappropriate on food-producing animals can lead to resistance many of the pathogenic bacteria to the various types of antibiotics, one of which is the Escherichia coli (E. coli) which produces extended spectrum β-lactamase (ESBL). The aim of this study was to isolate and identified ESBL- E. coli isolate from dairy cow rectal swabs in Sendang, Tulungagung district using the Vitek-2 method. The number of rectal swab samples used in the present study was 50. The result of the study showed that from all of the samples could be isolated and indentified E. coli, based on the colony characteristics on EMBA and biochemical test. Based on the double disc synergy test method using antibiotic disc amoxicylyn-clavulanate, ceftriaxone, aztreonam, ceftazidime and cefotaxime, 10 isolates could be identify els ESBL- E. coli. furthermore 3 out of 10 isolates DDST positives were confirmed ESBL- E. coli using Vitek-2 method.
