Effects of Radiation on the Histology of the Tongue and Lip Mucosa in Rats : Preliminary Research Sagita Yudha; Aisyah Elliyanti; Eti Yerizel; Sukri Rahman

Abstract

The tongue and lip mucosa, as tissues with high cell proliferation rates, are highly susceptible to injury caused by ionizing radiation. This study aimed to evaluate histological changes in the tongue and lip mucosa of rats after exposure to ionizing radiation at various doses. Nine rats were used, each consisting of three rats, divided into a negative control group (K) and three treatment groups receiving radiation doses of 13 Gy (30 seconds of radiation), 15 Gy (66 seconds of radiation), and 17 Gy (110 seconds of radiation). Samples of tongue and lip mucosa tissue were harvested after 7 days and processed for histological examination using Hematoxylin and Eosin (HE) staining at 100x objective magnification. The results of this preliminary study showed that radiation at 13 Gy caused epithelial erosion, loss of filiform papillae, subepithelial bullae formation, and acute inflammatory cell infiltration on the tongue. At 15 Gy, more extensive ulceration with subepithelial edema was observed. At 17 Gy, total epithelial loss with massive subepithelial edema and vascular dilatation occurs. This preliminary study concludes that the tongue mucosa exhibits greater radiation sensitivity than the lip mucosa, with a dose-dependent progression of damage ranging from erosion to total epithelial loss.

Abstract

Pelayanan kesehatan terhadap lanjut usia (lansia) perlu mendapatkan perhatian khusus, karena lansia rentan terhadap penyakit Tujuan: Pengabdian masyarakat ini dilakukan dengan tujuan untuk mengidentifikasi faktor risiko penyakit dan memberikan penyuluhan pada kelompok lanjut usia di Kenagarian Sumanik. Metode: Kegiatan yang dilakukan adalah pemeriksaan kesehatan lanjut usia dan pengkajian psikologis dengan menggunakan kuisioner DASS 42. Selanjutnya dilakukan penyuluhan kesehatan dengan harapan meningkatkan pengetahuan para lansia sehingga mampu mengidentifikasi tanda bahaya dan mendapat pertolongan lebih awal. Hasil: Kegiatan dihadiri oleh 63 orang warga, 31 orang diantaranya di atas usia 60 tahun. Hasil pemeriksaan ditemukan 22 orang mengalami hipertensi stadium 2, dan 5 orang menderita diabetes yang sebelumnya sudah diketahui akan tetapi kontrol tidak teratur. Pada pemeriksaan mata terdapat 25 orang menderita gangguan refraksi, 5 orang ekstensi lensa dan dirujuk ke RSUD melalui Puskesmas. Dari kajian psikologis didapatkan 5 orang memiliki gejala depresi, 15 orang dengan ansietas dan 48 orang mengalami stress-psikologis. Kesimpulan: Penyuluhan kesehatan sangat diperlukan terutama kelompok lanjut usia supaya lansia dapat mengenal tanda-tanda penyakit sehingga dapat pelayanan kesehatan lebih dini. Aktivitas fisik seperti berolahraga diperlukan namun lansia perlu mengetahui jenis olah raga yang tepat sesuai dengan usia.

Abstract

Introduction: Pelvic organ prolapse (POP) is a gynaecological disorder with a rising prevalence, significantly affecting quality of life. While the pathogenesis of POP is multifactorial, alterations in the extracellular matrix (ECM) have been implicated, particularly the role of transforming growth factor-beta (TGF-β) in ECM remodelling. However, the precise relationship between TGF-β and POP remains unclear. Methods: A systematic review was conducted following PRISMA guidelines. Relevant studies were identified from PubMed, Scopus, Web of Science, and the Cochrane Library. Inclusion criteria focused on studies investigating TGF-β expression in women with POP. Data extraction included study design, patient demographics, TGF-β detection methods, and key findings. Results: Analysis of nine studies revealed mixed findings. Four studies reported a significant reduction in TGF-β1 expression in POP patients, suggesting impaired ECM regulation via the TGF-β1/Smad3 pathway. Three studies found no significant differences in TGF-β levels between POP and control groups, while one study linked increased TGF-β1 levels to severe prolapse, possibly as a compensatory response. Variations in methodology, sample size, and patient characteristics contributed to inconsistencies across studies. Conclusion: Current evidence suggests that TGF-β1 plays a role in POP pathogenesis, with lower levels potentially weakening ECM integrity and higher levels in advanced cases reflecting a failed reparative response. Further research is needed to explore TGF-β interactions with other biomarkers and assess its potential as a therapeutic target.

Abstract

Thyroid cancer poses a persistent clinical challenge, particularly when conventional therapies fall short. Growing evidence suggests that molecular iodine (I₂) may exert direct antitumor effects beyond its classical role in thyroid hormone synthesis. This study evaluated the antiproliferative and apoptosis-inducing effects of I₂ on primary cell cultures derived from differentiated thyroid cancer tissue and established dose- and time-response relationships. Primary cultures were exposed to I₂ at concentrations of 5, 10, 20, 40, 80, and 100 µM for 24, 48, and 72 hours. Cell viability was assessed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, with IC₅₀ calculated accordingly. Apoptosis was characterized by acridine orange/propidium iodide (AO/PI) double-staining, analyzed by fluorescence microscopy and ImageJ software. I₂ demonstrated clear antiproliferative and apoptotic activity in a dose- and time-dependent manner. The strongest effect appeared at 100 µM after 72 hours, reducing viability to 1.39% (p < 0.001). At the IC₅₀ of 2.09 µM, iodine significantly suppressed proliferation and induced apoptosis, with an apoptosis rate of 81.77% ± 12.24% (p = 0.004). These findings confirm the meaningful antineoplastic potential of I₂ against differentiated thyroid cancer cells. This study supports SDG 3 (Good Health and Well-Being), contributing to efforts to develop more equitable, effective cancer treatment options and to reduce cancer-related morbidity globally.
