Abstract

As the prevalence of type 2 diabetes mellitus continues to increase over the years, research into antidiabetic drugs needs to continue. Probiotics are non-pathogenic microorganisms that have potential as α-glucosidase inhibitors. This study aimed to characterize and determine the α-glucosidase inhibitor activity of LBSU9 isolate, a probiotic isolated from "trites," a traditional food of North Sumatra. The results showed that LBSU9 had bacillus morphology, Gram positive, negative catalase test, TSIA (Triple Sugar Iron Agar) A/A test and non-hemolysis. LBSU9 had good tolerance to simulated gastric acid and bile salts, with growth percentages of 66.54% and 64.74%. LBSU9 also has potent antimicrobial activity against S.aureus and E.coli, with 20 mm and 17 mm inhibition zones, respectively. The alpha-glucosidase inhibitor activity of LBSU9 was 98.4% greater than that of acarbose, which was 97%. Based on the results of this study, it is concluded that LBSU9 isolate has the potential to be a complementary therapy to prevent or treat type 2 diabetes mellitus

Abstract

Risiko penyakit kardiovaskular dan obesitas dikaitkan dengan kadar kolesterol yang tinggi. Bakteri Asam Laktat sangat bermanfaat bagi fungsional tubuh manusia sebagai bakteri probiotik. Probiotik memiliki manfaat terapeutik seperti membantu pengobatan laktosa intoleran, mencegah kanker usus besar, dan menurunkan kadar kolesterol dalam darah. Tujuan dari penelitian ini untuk menentukan potensi probiotik isolat DNH 16 yang di isolasi dari makanan tradisional Sumatera Utara, Dali ni Horbo. Penelitian ini merupakan penelitian eksperimental. Rancangan penelitian yang digunakan adalah Post Test Only Control Group Design. Metode penelitian aktivitas antimikroba dilakukan dengan metode kertas cakram, uji ketahanan terhadap pH asam, garam empedu dan penurunan kolesterol di lakukan dengan metode spektrofotometer. Uji aktivitas antimikroba Isolat DNH 16 menunjukan aktivitas zona hambat terhadap bakteri S. aureus sebesar 71,5 mm dan bakteri E. coli sebesar 14,5 mm. Uji ketahanan terhadap pH asam kurang baik yaitu sebesar 20,05%. Uji ketahanan terhadap garam empedu cukup tinggi yaitu 95,4%. Penurunan kadar kolesterol secara In vitro cukup baik yaitu 65,85%. Kontrol positif penurunan kolesterol yaitu 14,63%. Kesimpulan yang di dapat dari penelitian ini adalah probiotik isolat DNH 16 dapat menurunkan kadar kolesterol.

Abstract

BACKGROUND: Type 2 diabetes mellitus (T2DM) cases tend to rise throughout the year in all parts of the world. The α-glucosidase inhibitors are frequently used to both prevent and treat T2DM. This α-glucosidase inhibitor activity is seen in some probiotic species, and a certain strain of Pediococcus acidilactici exhibits promising characteristics as an α-glucosidase inhibitor. This study was conducted to assess the hypoglycemic activity and safety of P. acidilactici strain DNH16 (PADNH16) in experimental rats with T2DM induced by streptozotocin.METHODS: The experiment was conducted in vivo using Wistar rats. Acarbose was employed as a positive control, and Lactobacillus casei was used as a comparative. For 24 days, post-prandial blood glucose levels were assessed every 3 days, followed by serum biochemical levels measurement of liver, kidney, and lipid profiles. The pancreas was histopathologically examined utilizing the Hematoxyline-Eosin staining procedure.RESULTS: Administration of PADNH16 to T2DM rats lowered post-prandial blood glucose levels and gave hypoglycemic benefits comparable to acarbose and commercial probiotics. PADNH16 dosing did not affect serum glutamic oxaloacetic transaminase (SGOT), serum glutamic pyruvic transaminase (SGPT), urea, or creatinine levels, showing that PADNH16 was not hazardous to liver or kidney function. The lipid profile assessment revealed that the values, including high-density lipoprotein (HDL), low-density lipoprotein (LDL), and triglycerides, were comparable to the control group. The pancreatic histopathology revealed that injection of PADNH16 caused no alterations to pancreatic β cells.CONCLUSION: P. acidilactici isolate DNH16 has a hypoglycemic effect on T2DM rats, but does not affect liver function enzymes, kidneys, lipid profiles and does not provide significant changes in pancreatic β cells.KEYWORDS: diabetes mellitus, inhibitor α-glucosidase, Pediococcus acidilactici strain DNH16

Abstract

Seorang dokter merupakan tenaga medis yang mempunyai kemampuan dalam hal mengobati dan menyembuhkan penyakit pasien. Untuk mendapatkan lulusan dokter yang berkualitas, maka perlu adanya pendukung kurikulum pembelajaran yang menghubungkan antara pembelajaran teori dan praktik yang disebut dengan ECE (Early Clinical Exposure), pembelajaran berbasis ECE memiliki berbagai manfaat bagi mahasiswa. Saat ini Fakultas kedokteran UNPRI menerapkan ECE sebagai pendukung kurikulum pembelajaran terhadap Mahasiswa Kedokteran Semester V. Agar manfaat pelaksanaan ECE dapat berjalan dengan baik dan lancar. Oleh karena itu, perlunya Persepsi Mahasiswa Semester V Fakultas Kedokteran UNPRI terhadap Pelaksanaan ECE di RSU Royal Prima Marelan. Penelitian menggunakan deskriptif kualitatif dengan desain cross sectional. Data yang digunakan adalah data primer yang berupa kuesioner dengan skala likert 1-5. Sampel penelitian ini diambil menggunakan teknik Cluster Random Sampling, yaitu mahasiswa semester V yang berjumlah 103 orang yang berpartisipasi dari 124 orang. Analisis statistik dilakukan dengan analisis univariat. Hasil analisis menunjukan bahwa rata-rata persepsi mahasiswa semester V terhadap pelaksanaan ECE sebesar 3,17 (63%), Struktur dan Organisasi ECE sebesar 3,11 (62%), dan Manfaat Implementasi ECE sebesar 3,37 (67%). Bahwa mayoritas Persepsi Mahasiswa Semester V Fakultas kedokteran UNPRI menjawab netral terhadap pelaksanaan ECE (cukup), Struktur dan Organisasi ECE (cukup), manfaat implementasi ECE (Baik) di RSU Royal Prima Marelan.

Abstract

Hiperlipidemia adalah sebuah kondisi abnormal metabolik lipid dengan indikasi kadar lipid di dalam darah mengalami kenaikan. Secara umum, hiperlipidemia sering diikuti dengan adanya penyakit lain, sehingga sering digabungkan dengan obat jenis lain dan memicu terjadinya interaksi obat. Tujuan dari pelaksanaan penelitian ini yaitu untuk  mengetahui apakah ada kejadian interaksi obat antihiperlipidemia di rumah sakit umum royal prima medan dan untuk mengidentifikasi tingkat keparahan interaksi obat antihiperlipidemia pada pasien di rumah sakit umum royal prima medan. Penelitian ini termasuk penelitian observasi(survey), jenis penelitian yang diterapkan yaitu survey analitik dan pengambilan data dilaksanakan dengan retrospeksi. Jenis analisis yang digunakan yaitu analisis data univariat dan pengolahan dilakukan secara teoritis menggunakan stokley drug interaction dan review aplikasi drugs.com. Dari 120 lembar resep yang dikaji, dapat disimpulkan bahwa karakteristik sesuai dengan jenis kelamin menunjukkan persentase tertinggi dari peresepan yaitu pasien yang berjenis kelamin perempuan sebanyak 64 pasien (53,3%). Karakteristik berdasarkan usia tertinggi 50-59 tahun (29,2%). Berdasarkan ada tidaknya interaksi yang menunjukkan persentasi tertinggi yang menggunakan lebih dari 5 obat yaitu sebanyak 56 lembar resep (58,3%). Berdasarkan penggunaan antihiperlipidemia terbanyak yaitu golongan atorvastatin sebanyak 114 resep (70%). Berdasarkan dengan tingkat keparahan interaksi obat terbanyak terjadi pada tingkat keparahan moderat yaitu sebanyak 189 kasus (84,4%).

Abstract

Phytochemical screening of ethanolic extract of cabbage (Brassica oleracea) has been successfully conducted. A phytochemical test was performed using FeCl3 to know the content of the phenolic compound from Cabbage. Cabbage is extracted until the ethanolic extract was obtained. It contained secondary metabolic compounds. mengandung senyawa metabolic sekunder. The largest secondary metabolic in cabbage extract was the flavonoid compound. The flavonoids total contained in the ethanolic extract of cabbage was 1.68 mg QE/g extract and the level of total phenol contained in the ethanolic extract was 45.090 mg GAE/g extract.

Abstract

Introduction: Diabetes mellitus is a chronic metabolic disorder with a high global prevalence and serious complications. The prediabetes stage is a critical target for prevention, often managed with α-glucosidase inhibitors. However, synthetic drugs such as acarbose are frequently associated with gastrointestinal side effects, highlighting the need for safer alternatives. Endophytic microbes from medicinal plants, including areca nut (Areca catechu), have the potential to produce bioactive compounds with antioxidant and antidiabetic activities. Objective: This study aimed to explore endophytic bacteria from fresh areca nut seeds as a source of metabolites with antioxidant and α-glucosidase inhibitory activity. Methods: Fresh areca nut seeds were sterilized using the surface sterilization method, followed by endophytic bacterial isolation and Gram staining. Cultures were fermented in Nutrient Broth, and supernatants were obtained by centrifugation. Antioxidant activity was assessed using the DPPH method, while α-glucosidase inhibition was tested in vitro with p-nitrophenyl α-D-glucopyranoside as substrate. Acarbose served as the positive control. Results: Seven Gram-negative, rod-shaped endophytic isolates (BP1-BP7) were obtained. Isolate BP1 exhibited the highest antioxidant activity with 45.7% inhibition. Furthermore, the BP1 supernatant demonstrated potent α-glucosidase inhibitory activity up to 93,6%, comparable to that of acarbose (91%). Conclusion: Endophytic bacteria from areca nut seeds possess dual bioactivity as antioxidants and α-glucosidase inhibitors, suggesting their potential as sustainable alternative antidiabetic agents worthy of further investigation.

Abstract

Kualitas tidur yang buruk dapat memengaruhi keseimbangan metabolik tubuh dan berdampak pada perubahan Indeks Massa Tubuh (IMT). Mahasiswa kedokteran merupakan kelompok yang rentan mengalami gangguan tidur akibat beban akademik yang tinggi dan pola hidup tidak teratur. Penelitian ini bertujuan untuk mengetahui hubungan antara kualitas tidur dengan Indeks Massa Tubuh (IMT) pada mahasiswa Fakultas Kedokteran Universitas Prima Indonesia angkatan tahun 2022. Jenis penelitian ini adalah kuantitatif dengan metode analitik cross-sectional. Data dikumpulkan melalui kuesioner Pittsburgh Sleep Quality Index (PSQI) untuk menilai kualitas tidur serta pengukuran berat dan tinggi badan untuk menentukan IMT. Jumlah sampel sebanyak 178 mahasiswa yang dipilih dengan metode total sampling. Analisis data dilakukan menggunakan uji Chi-Square dan uji Cramer’s V dengan bantuan program SPSS. Hasil penelitian menunjukkan bahwa 94 responden (52,8%) memiliki kualitas tidur baik dan 84 responden (47,2%) memiliki kualitas tidur buruk. Mayoritas responden memiliki IMT normal sebanyak 113 orang (63,5%), sedangkan 29 orang (16,3%) tergolong obesitas. Uji Chi-Square menghasilkan nilai p < 0,001 yang menunjukkan adanya hubungan signifikan antara kualitas tidur dengan IMT, dengan kekuatan hubungan sedang (Cramer’s V = 0,492). Kesimpulan penelitian ini adalah semakin buruk kualitas tidur seseorang, semakin tinggi kecenderungan mengalami peningkatan IMT.

Abstract

This study aims to test the administration of ethanol extract of cat's whisker leaves (Orthosiphon stamineus) to reduce cholesterol levels in female Sprague Dawley rats. This type of research is a quasi-experiment design using a one group pre-test and post-test design. The population in this study was 30 female Sprague Dawley rats aged ± 3 months with a body weight of around 150-250 grams. Data were analyzed using the t test statistic at = 5%. The results of the research showed that the highest decrease in cholesterol levels was in female Sprague Dawley rats at a test dose of 1000 mg/kgBW and there was an effect of ethanol extract of cat's whisker leaves (Orthosiphon stamineus) on reducing cholesterol levels in female Sprague Dawley rats

Abstract

Diabetes, a disorder of hemostasis of carbohydrate and lipid metabolism, is one of today's leading killers. The most prevalent form of diabetes is type 2 diabetes mellitus (T2DM). Rapid hydrolysis of starch by pancreatic α-amylase and α-glucosidase, followed by intestinal absorption of glucose, causes a sudden increase in blood glucose. Available therapies for T2DM are oral insulin secretagogues, sulfonylureas, repaglinide, nateglinide, biguanides, thiazolidinediones, α-glucosidase, inhibitors and insulin. However, several hypoglycemic agents have limitations, such as side effects and increased diabetes complications. α-glucosidase inhibitors are structurally similar to natural oligosaccharides with a higher affinity for α-glucosidases, and they produce a reversible inhibition of membrane-bound intestinal α-glucoside hydrolase enzymes. This causes delayed carbohydrate absorption and digestion and results in a reduction in postprandial hyperglycemia. Natural α-glucosidase inhibitor drugs from natural sources can be used as a therapeutic approach to treat postprandial hyperglycemia for their assumed lower side effect and more affordable price than synthetic drugs. In this article, the author summarizes the potential of α-glucosidase inhibitors from microorganisms, namely fungi and bacteria, along with several active compounds with better activity than commercial α-glucosidase inhibitors.
