Abstract

Red alga Acanthophora spicifera is a marine species with potential as a source of natural antihistamines. This study evaluated the bioactivity of its secondary metabolites through in vivo, in vitro, and in silico approaches. Extracts were obtained by successive maceration using n-hexane, ethyl acetate, and methanol. In vivo testing on BALB/c mice showed that extract administration did not cause significant weight loss. However, its effect on allergic symptoms was significant (p < 0.05), with the methanol extract identified as the most effective by the Mann–Whitney test. In vitro assays revealed a significant reduction in histamine levels (p < 0.05) during sensitization, treatment, and post-challenge phases, particularly in the methanol extract group. In silico docking demonstrated that two compounds from the methanol extract exhibited strong binding affinity to the H1 receptor (PDB ID: 3RZE), with binding energies of −6.27 and −5.06 kcal/mol. These findings suggest that A. spicifera, particularly its methanol extract, is a promising source of natural antihistamines with potential applications in the development of safer allergy treatments.

Abstract

Tuberculosis remains a global health issue, with treatment hindered by drug resistance. This study aims to identify the potential of Quercetin as an inhibitor of Enoyl-Acyl Carrier Protein Reductase (M. tuberculosis), a key enzyme in the fatty acid biosynthesis of the bacteria. Using molecular docking, Quercetin is evaluated based on its binding affinity and stability with the target protein. The docking results show that Quercetin binds to the active site of Enoyl-Acyl Carrier Protein Reductase through hydrogen bonds, π-alkyl, and π-sigma interactions, with an RMSD value of 1.6466 Å, indicating valid results. Comparison with the control compound, 5-hexyl-2-(2-methylphenoxy)phenol, reveals that Quercetin has a stronger binding affinity, supporting its potential as a more effective enzyme inhibitor. These findings open new opportunities for the development of natural compound-based anti-Tuberculosis therapies that can address the growing problem of drug resistance. Further research is needed for experimental validation in biological models and the development of more efficient Quercetin formulations.

Abstract

This review synthesizes research on transition metal layered double hydroxide nanosheets in breast cancerdrug encapsulation to address challenges in targeted delivery, drug resistance, and controlled release. Thereview aimed to evaluate synthesis and functionalization strategies, benchmark drug loading and releaseprofiles, elucidate metal doping effects on targeting and uptake, compare therapeutic outcomes, and identifyclinical translation barriers. Literature selection focused on experimental in vitro and in vivo studies publishedup to mid-2024, emphasizing physicochemical characterization, cellular internalization, and therapeuticefficacy. Findings reveal that transition metal doping and surface modifications enable ultrahigh drug loadingcapacities with pH-responsive, tumor-specific release, enhancing intracellular delivery via receptor-mediatedand endocytic pathways. Multifunctional nanosheets demonstrate synergistic anticancer effects throughcombined chemo-, photothermal, and immunotherapies, with favorable biocompatibility and low systemictoxicity in preclinical models. However, variability in synthesis reproducibility, incomplete mechanisticunderstanding of metal ion interactions, and limited long-term safety data impede clinical translation.Integrating these findings underscores the potential of transition metal-doped LDH nanosheets as versatileplatforms for breast cancer nanotherapy while highlighting the need for standardized evaluation andcomprehensive in vivo assessments. These insights inform future research directions toward optimizingnanosheet design and advancing clinical applications in precision oncology.

Abstract

The presence of lead ions (Pb²⁺) in fishery products poses potential risks to human health. Therefore, simple, rapid, and easily applicable detection methods are highly required. This study aimed to develop a simple colorimetric sensor for the visual detection of Pb²⁺ based on color change as an analytical signal. The sensor was prepared through the interaction of a complexing indicator with the target metal ion, resulting in a distinct and visually observable color response. The novelty of this work lies in the application of a simple colorimetric sensor with a low visual detection limit and high selectivity toward Pb²⁺, as well as its direct applicability to simulated fishery product samples without the need for complex analytical instruments. The results demonstrate that the developed sensor is capable of selectively detecting Pb²⁺ with a clear visual response, indicating its potential use as a low-cost and practical method for preliminary screening of lead contamination.

Abstract

Metal-Organic Frameworks (MOFs) have gained increasing attention as promising nanocarriers for targeted cancer drug delivery due to their high surface area, tunable porosity, adjustable chemical structure, and capacity for controlled drug release. Conventional chemotherapy often faces limitations such as systemic toxicity, poor tumor selectivity, low bioavailability, and damage to healthy tissues. Stimuli-responsive MOFs enable drug release in response to specific tumor microenvironment conditions, including acidic pH, redox imbalance, enzyme overexpression, hypoxia, or external triggers such as light and temperature. This systematic review synthesizes 30 selected studies, focusing on MOF types, therapeutic cargo, stimuli-responsive mechanisms, cancer models, and reported therapeutic outcomes. Findings show that ZIF-8, UiO-66, MIL-series, and related MOF platforms are widely investigated, with doxorubicin and other anticancer drugs frequently used. pH-responsive and redox-responsive systems dominate, while multifunctional MOFs combining chemotherapy with photothermal or photodynamic therapy show strong potential. Clinical translation remains limited due to unresolved concerns regarding long-term biosafety, biodegradation, pharmacokinetics, immune response, reproducibility, and scalability. Stimuli-responsive MOFs represent a promising direction for precision cancer therapy, but further in vivo validation and translational studies are required

Abstract

Metal-peptide complexes represent a promising avenue in medicinal chemistry due to their unique coordination and biological properties. This study aimed to design and evaluate a Fe(II)-proline-arginine-dipeptide complex as a potential therapeutic agent. Computational modeling constructed the ligand and optimized the coordination geometry with Fe(II) and two chloride ligands, forming a stable distorted tetrahedral structure. 1H- and 13C-NMR spectra confirmed preservation of the dipeptide framework. Physicochemical assessment revealed a molecular weight of 396.056 g/mol, low LogP of 0.21947, balanced hydrogen-bond donors and acceptors, and a polar surface area of 139.047 Ų, supporting oral drug-likeness. ADMET analysis predicted moderate intestinal absorption (38.444%), low CNS penetration, minimal CYP enzyme interaction, and moderate clearance (1.11 log mL/min/kg). Toxicity prediction indicated no AMES mutagenicity, hepatotoxicity, or hERG inhibition, with an oral rat LD50 of 2.174 mol/kg. These results demonstrate that the Fe(II)-proline-arginine-dipeptide complex possesses structural stability, favorable pharmacokinetic behavior, and a broad safety profile, highlighting its potential as a therapeutic candidate. The study provides a framework for integrating peptide ligand design with metal coordination and in silico pharmacokinetic evaluation, suggesting avenues for experimental validation and future drug development.
