Abstract

The identification of Vibrio sp. bacteria in the water of intensive and traditional ponds for vannamei shrimp in Teluk Pambang Village was conducted from March to June 2023. The aim of this research was to calculate the quantity of Vibrio sp. bacteria, understand the morphological characteristics, and identify the various types of Vibrio sp. bacteria in the water of intensive and traditional ponds for vannamei shrimp. The sampling involved collecting water from 2 bottles each from intensive and traditional ponds. Subsequently, Vibrio isolation from the water was cultured on selective TCBS media through three dilution steps. The bacterial count was then determined. Further, bacterial cultures from different colonies were streaked on TSA media, followed by identification through a series of morphological observations and biochemical tests. The research results indicate that the presence of Vibrio bacteria in intensive ponds complies with the maximum threshold and quality standards for a favorable environment for both the pond and shrimp. However, for traditional ponds, there is an abundance of Vibrio bacteria exceeding the threshold, where the maximum limit or good quality standard for Vibrio in the environment and shrimp is 103 CFU/ml. The morphological characteristics found include a round shape, green and yellow color, and a flat edge with emerging elevations. Furthermore, infections of Vibrio hollisae, Vibrio anguillarum, and Vibrio damsela were identified in the water of intensive and traditional ponds for vannamei shrimp (Litopenaeus vannamei) in Teluk Pambang Village, Bengkalis Regency.

Abstract

Vaname shrimp is one of Indonesia's leading export species and is widely cultivated. The main problem in vaname shrimp pond cultivation is the attack of shrimp diseases caused by bacteria or viruses. This study aims to isolate and identify lactic acid bacteria (BAL) from vaname shrimp (Litopenaeus vannamei) and test their antagonism against pathogenic bacteria. The research was conducted from February to April 2023. The method used in this study is a survey method. Sampling was conducted at the vaname shrimp pond of the Fisheries Service Task Unit (UPT), Tanjung Punak Village, Rupat Utara District, Bengkalis Regency, Riau. The method used in this research was a survey method. BAL was isolated using MRS Agar + CaCO3 media. The Kirby-Bauer method was used to test bacterial antagonism against pathogenic bacteria. Based on the identification of seven isolates, it was found that only four isolates were positively identified as LAB bacteria, namely 47D, 54T1, 73DB1, and 73BD2. The results of the antagonism test against pathogenic bacteria showed that all BAL isolates could not suppress the growth of E. coli bacteria. However, these isolates suppressed the growth of A. hydrophila bacteria, with the widest inhibition zone diameter on isolate 54T1 (4.2 mm). On Vibrio sp. bacteria, the widest inhibition zone of BAL was on isolate 73DB1 (6 mm). These results indicate that the antibacterial activity produced by BAL is still relatively weak.

Abstract

Bacillus cereus SN7 is a bacterium with potential applications in producing Single-Cell Protein (SCP). SCP can be an alternative to protein derived from traditional sources like agriculture, fisheries, and animal husbandry. Encapsulation is a process used to stabilize cells, aiming to potentially enhance probiotics' viability and stability throughout production, storage, and handling. This study aims to make capsules from B. cereus SN7 single-cell protein to protect them from contaminants. This study used a survey method, which carried out in-situ processing of bacteria, as well as conducting experiments on the formation of bacterial encapsulation, testing the effectiveness of encapsulation, and observing the viability of B. cereus SN7 bacteria by counting the number of colonies. B. cereus produced biomass with a total dry weight of 1.3708 g/mL. The alginate encapsulation experiments can be formed into capsule beads that protect bacteria. The best encapsulation process is an alginate-based coating, which is indicated by the ability of the alginate to withstand temperature, pH, and salinity.

Abstract

This study aims to find out that Bacillus paramycoides and Enterobacter cloacae bacteria can produce amylase enzymes and have the ability to degrade organic matter, especially mangrove litter. From this study it was found that the optimal growth of B.paramycoides and E. cloacae bacteria occurred at 12th hour. The results of measurements and calculations of absorbance values at 630 10.238 x 108 cells/mL (B. paramycoides) and 12.030 x 108 cells/mL (E. cloacae) using the spectrophotometric method. Meanwhile, with the TPC method at 12 hours, the number of bacterial cells was 2.08 x 108 CFU's/mL (B.paramycoides) and 2.44 x 108 CFU's/mL (E. cloacae). The ability to produce the largest amylolytic bacterial amylase enzyme also occurred at 12 hours as much as 0.306 mg/mL (B.paramycoides) with an increase of 0.046 mg/mL and 0.243 mg/mL (E. cloacae) with an increase of 0.028 mg/mL. The bacteria that have the highest amylase enzyme ability is E.cloacae as evidenced by the diameter of the clear zone of 10.10 mm. Testing the ability of amylolytic bacteria in degrading mangrove litter was carried out by adding amylase enzyme as much as 0%, 50% and 100%. Amylolytic bacteria can degrade organic matter by hydrolyzing starch contained in mangrove litter. The most degraded starch content was in the 100% enzyme treatment, which was 1.021 mg/mL (B. paramycoides) and 1.189 mg/mL (E.cloacae).

Abstract

The purpose of this study is to determine the rate of shoreline change and the level of abrasion in Bukit Batu District, Bengkalis Regency, Riau Province based on several physical variables in the coastal area. End Point Rate (EPR) is a method that can be used to calculate the rate of shoreline change by displaying transects according to their category (high abrasion, medium abrasion, low abrasion, stable, and accretion). From 1991-2021 there was an abrasion of ±139.71 m equivalent to ±4.65 m/year which mostly occurred in Sukajadi Village and Pangkalan Jambi Village, while accretion of ±27.51 m equivalent to ±1.58 m/year mostly occurred in Pakning Asal Village and Sejangat Village. Moderate abrasion is the predominant type. Sukajadi and Pangkalan Jambi are villages that have experienced high levels of abrasion. The area affected by abrasion has moderate and fast currents as well as coastal protection in the form of mangroves, most of which have been damaged and require repairs. Therefore, the main factors causing the abrasion are thought to be the lack of coastal protection and high-speed currents and waves flowing from the Malacca Strait.

Abstract

Blood clams are soft-bodied animals that live sessile and obtain food by filtering (filter feeders). This results in many microorganisms accumulating in the body of the blood clams, one of which is the coliform bacteria group. Coliform bacteria are divided into two types, namely fecal coliform and non-fecal coliform. Fecal coliform comes from animal and human feces, such as Escherichia coli. According to SNI 01-7388-2009, the MPN value of E. coli bacteria for fresh Mollusca, Crustaceans, and Echinodermata is <3/g. This study aimed to calculate the density of E. coli bacteria in blood clams. This study was conducted from December 2024 to February 2025. Sampling was conducted at the lowest ebb with one transect consisting of 3 plots measuring 1 × 1m². The MPN method is a method for analyzing coliform bacteria and E. coli. The results of the estimation test obtained at station 1, namely the Tebing Dian Beach Area, ranged from 7-10 MPN/g with an average of 12.6 MPN/g and station 2, namely the "Cinta Mangrove" Tourism Area, ranged from 4-15 MPN/g with an average of 10 MPN/g. While the results of the estimation test obtained at station 1 ranged from 3-4 MPN/g with an average of 2.3 MPN/g, stations 2 ranged from 3 MPN/g with an average of 1.0 MPN/g. These results indicate that the abundance of E. coli bacteria in blood clams does not exceed the quality standards set based on SNI 01-7388-2009
