Abstract

One of the potentials in Teluk Rhu Village is beach tourism. The development of coastal tourism in Teluk Rhu Village needed a principal role from the Women's Fishermen, specifically to make processed products from fishermen's catch. The community service activity aims to improve the Women Fishermen's knowledge and skills in making processed fishery products that can be marketed at coastal tourist locations. This program implementation used the PALS (Participatory Action and Learning System) method with three stages. There was socialization, increasing knowledge, and direct practice. The community service activities showed that there was potential to develop coastal tourism in Teluk Rhu Village. The several potentials were the Women Fishermen can get the raw material easily, they have a lot of time used to produce and sell the products, and they have skills to make some processed food with diversification techniques such as nugget, meatball, and otak-otak. Thus, Increasing the knowledge and skills of the Women's fishermen through diversification technology can be used to increase the number of tourists at the coastal tourist location of Teluk Rhu Village, Rupat Utara District.

Abstract

Research on the population structure of bamboo shells (Solen lamarckii) in the intertidal zone of Sri Tanjung Village, Rupat District, Bengkalis Regency was carried out in October 2021-March 2022. This study aimed to determine the population structure of bamboo shells and the relationship between the size of bamboo shells and the depth of bamboo shells in the sediment. The intertidal zone is divided into 5 subzones. Each subzone is placed in 3 plots measuring 1 x 1 m2 along the transect line as a replication by drawing a straight line from the boundary of the highest tide area to the lowest low tide area. The results showed that the average population density of bamboo shells was 1.20 ind/m2 or 12,000 ind/ha with the lowest density located in the highest tide area and the highest density located in the lowest low tide area. The distribution pattern of bamboo shells is uniform. The size of the most common bamboo shells found is 5.14-5.70 cm. The relationship between the size of the bamboo shells and the depth of the bamboo shells in the sediment is very weak

Abstract

The Media is a material consisting of a mixture of nutrients used by microorganisms to grow and breed. Molasses and liquid waste know one of the alternative media which has the least inexpensive carbon source and is easily obtained from the waste food industry that is rich in nutrients and minerals, so it has the potential for bacterial growth of Bacillus cereus. The consetration of molasses used for growth medium is 1%; 1.5%; and 2% while liquid waste knows 8%, 10%, and 12%. Cell growth is measured by the method of Spectrophotometry, total plate count (TPC) and biomass while the incubation period of cell growth is measured every 0, 6, 12, 18, and 24 hours. The results showed from the molasses + skim milk and the tofu liquid waste + skim milk, the best medium of bacterial cell growth of B. cereus is a liquid waste know isolates K 12%. While the best growth concentrations molasses Medium bacterial B.cereus was N2% and K2%.

Abstract

Pro coffee is a fermented beverage using Bacillus cereus bacterial consortium with single-cell protein. This study aims to determine the best composition of the B. cereus consortium bacteria mixture in organoleptic aspects that panelists most prefer and the effect of adding B. cereus consortium bacteria on proximate analysis and pro coffee flavor. Organoleptic testing of probiotic coffee was conducted on 20 panelists, then continued with proximate analysis of pro coffee. The composition with the best organoleptic flavor is obtained in code D, with the treatment of adding 5% bacterial consortium B. cereus for 15 days. The analysis of taste obtained has a value of 0.000 <0.05, and the study results show that the significance value is smaller than the significance level. Hence, the treatment results give the effect of adding bacterial consortium to pro-coffee. The addition of bacterial consortium B. cereus to probiotic coffee, in the form of moisture, ash, protein, fat, and carbohydrate content, is 0.000 <0.05. The analysis results show that the significance value is greater than the significance level, so the treatment results do not give the effect of adding bacterial consortium to pro coffee

Abstract

Vaname shrimp is one of Indonesia's leading export species and is widely cultivated. The main problem in vaname shrimp pond cultivation is the attack of shrimp diseases caused by bacteria or viruses. This study aims to isolate and identify lactic acid bacteria (BAL) from vaname shrimp (Litopenaeus vannamei) and test their antagonism against pathogenic bacteria. The research was conducted from February to April 2023. The method used in this study is a survey method. Sampling was conducted at the vaname shrimp pond of the Fisheries Service Task Unit (UPT), Tanjung Punak Village, Rupat Utara District, Bengkalis Regency, Riau. The method used in this research was a survey method. BAL was isolated using MRS Agar + CaCO3 media. The Kirby-Bauer method was used to test bacterial antagonism against pathogenic bacteria. Based on the identification of seven isolates, it was found that only four isolates were positively identified as LAB bacteria, namely 47D, 54T1, 73DB1, and 73BD2. The results of the antagonism test against pathogenic bacteria showed that all BAL isolates could not suppress the growth of E. coli bacteria. However, these isolates suppressed the growth of A. hydrophila bacteria, with the widest inhibition zone diameter on isolate 54T1 (4.2 mm). On Vibrio sp. bacteria, the widest inhibition zone of BAL was on isolate 73DB1 (6 mm). These results indicate that the antibacterial activity produced by BAL is still relatively weak.

Abstract

Bacillus cereus SN7 is a bacterium with potential applications in producing Single-Cell Protein (SCP). SCP can be an alternative to protein derived from traditional sources like agriculture, fisheries, and animal husbandry. Encapsulation is a process used to stabilize cells, aiming to potentially enhance probiotics' viability and stability throughout production, storage, and handling. This study aims to make capsules from B. cereus SN7 single-cell protein to protect them from contaminants. This study used a survey method, which carried out in-situ processing of bacteria, as well as conducting experiments on the formation of bacterial encapsulation, testing the effectiveness of encapsulation, and observing the viability of B. cereus SN7 bacteria by counting the number of colonies. B. cereus produced biomass with a total dry weight of 1.3708 g/mL. The alginate encapsulation experiments can be formed into capsule beads that protect bacteria. The best encapsulation process is an alginate-based coating, which is indicated by the ability of the alginate to withstand temperature, pH, and salinity.

Abstract

Bacillus cereus is a probiotic bacteria that requires media as a source of nutrients for its growth. This study aims to the media used were tofu liquid waste, sago liquid waste, and molasses. The method used is an experimental method, namely testing two groups of bacterial isolates; B. cereus SN7 and the consortium (combination of five isolates from B.cereus with different strains). Each group of isolates was grown with three replications on each waste medium. bacterial growth every six hours for twenty four hours using two methods; Total Plate Count (TPC) and bacterial cell biomass. There was no significant difference in growth between the two groups of bacterial isolation. The optimal growth of each isolate was found in molasses waste. The growth between two bacterial isolates in the liquid waste media was B. cereus SN7 bacterial isolate.

Abstract

This study aims to find out that Bacillus paramycoides and Enterobacter cloacae bacteria can produce amylase enzymes and have the ability to degrade organic matter, especially mangrove litter. From this study it was found that the optimal growth of B.paramycoides and E. cloacae bacteria occurred at 12th hour. The results of measurements and calculations of absorbance values ​​at 630 10.238 x 108 cells/mL (B. paramycoides) and 12.030 x 108 cells/mL (E. cloacae) using the spectrophotometric method. Meanwhile, with the TPC method at 12 hours, the number of bacterial cells was 2.08 x 108 CFU's/mL (B.paramycoides) and 2.44 x 108 CFU's/mL (E. cloacae). The ability to produce the largest amylolytic bacterial amylase enzyme also occurred at 12 hours as much as 0.306 mg/mL (B.paramycoides) with an increase of 0.046 mg/mL and 0.243 mg/mL (E. cloacae) with an increase of 0.028 mg/mL. The bacteria that have the highest amylase enzyme ability is E.cloacae as evidenced by the diameter of the clear zone of 10.10 mm. Testing the ability of amylolytic bacteria in degrading mangrove litter was carried out by adding amylase enzyme as much as 0%, 50% and 100%. Amylolytic bacteria can degrade organic matter by hydrolyzing starch contained in mangrove litter. The most degraded starch content was in the 100% enzyme treatment, which was 1.021 mg/mL (B. paramycoides) and 1.189 mg/mL (E.cloacae).

Abstract

This research was conducted from March-June 2019. The purpose of this study was to determine the characteristics of secondary metabolites prduced by of heterotrophic bacterial from sea water Sungai Kayu Ara Village, Siak Regency as an antimicrobial and to determine the storage time of these bacteria by measuring at the optimal growth time. Five bacterial secondary metabolite extracts used were B, C, D, and H (B. cereus) and J (V. fluvialis) obtained from the collection of Marine Microbiology Laboratory, Department of Marine Sciences, Faculty of Fisheries and Marine sciences, University of Riau. phytochemical test showed that extracts of isolates B, D, and H contained saponin compounds, while isolate J contained flavonoid compounds, however, all extracts contained alkaloid compounds. Antimicrobial test indicated that J extract inhibited A. Hydrophila at concentration 500 µg/ml but the extract could not inhibit V. algynolyticus and Pseudomonas sp concentrations determined. In the bacterial storage time test, the optimal growth of each bacterial concentration at was 7th day incubation and decreased on the 14th day.

Abstract

Proteolytic bacteria have important role in the degradation of complex compounds to simple compound by the enzyme proteases. Sediment in mangrove ecosystem of Dumai Marine Station may contain the bacteria. This research aims to identify the proteolytic bacteria of mangrove sediments molecularly and to examine the antagonism of pathogenic bacteria. The method used in this research was survey. The results obtained 10 bacterial isolates (AZ1, AZ2, AZ3, AZ4, AZ6, AZ10, AZ11, AZ15, AZ18, and AZ20). Identification using 16S rRNA analysis revealed that 3 isolates showed different results, namely AZ2 had a similarity to Bacillus proteolyticus with strain MCCC 1A00365, AZ6 had a similarity to the bacteria B. monlinensis with strain BL4-6 and AZ20 had similarity to the bacterium B. toyonensis with strain MCCC 1A00365 BCT-7112. Three of the 10 isolates that had the highest inhibition against pathogens. Isolates AZ2 shovued inhibition zone against Pseudomonas aeruginosa with a diameter of 9.65 mm, AZ6 shovued inhibition zone against Escherichia coli bacteria with a diameter of 5.18 mm, and AZ20 shovued inhibition zone against Vibrio alginolyticus bacteria with a diameter of 4, 01 mm.
