Abstract

Shrimp disease caused by Vibrio sp is one of the main limiting factors in the increasing production in shrimp farming. This disease may kill the shrimp and cause high loss in shrimp culture in South East and East Asia. Samples of 10 individuals of a ten months cultured giant tiger prawn as well as 1 liter pond water and sea waters were collected from shrimp pond in Bengkalis Island, Sumatra. Samples of shrimps were also collected from Jepara shrimp pond in Central Java and they were selected by looking at their behavior and unhealthy physical characteristics. Amplification, rDNA 16s sequencing and bioinformatics analysis to identify Vibrio species were conducted in Biotech Center, BPPT Serpong Banten. The results of DNA sequencing of each bacteria isolate were compared to DNA sequence from international DNA bank database. Tracing were made by BLAST (Basic Local Alignment Search Tool) system accessed through the internet at http://www./ncbi.nlm.nih.gov/blast. The result of this study found seven strains of Vibrio sp bacteria where five of which (V. alginolyticus, V. parahaemolyticus, V. harveyi, V. shilonii and V. vulnificus) were already registered in the world gen bank with homolog level above 97%. Meanwhile, another two strain found in this study were not found in the list of world gen bank and therefore are considered as native Vibrio sp bacteria from Indonesia. Keywords: Vibrio sp, bacteria, shrimp, aquaculture, fish disease

Abstract

One of the ways to reduce energy consumption on the air plane and the other bluff bodies are by decreasing the drag. Drag isclosely related to the flow separation. The earlier separation, then the drag will increase more. Based of the fact the effort todecrease drag is conducted by manipulating the field of fluid flow. Stream manipulation was be done by installing Triangleobstacle in front of cylinder. The purpose of this research is to analyze the effect of various distance triangle obstacle in front ofcylinder on drag. The present experiment was done by placing triangle rod in front of the cylinder. In the present research, theexperiment was conducted in the wind tunnel, which consisted of blower, pitot pipe, manometer, cylinder pipe, and triangle rod.The triangle was positioned at L/D = 1.19, L/D = 1.43, L/D = 1.67, L/D = 1.9, L/D = 2.14, L/D = 2.38, L/D = 2.62, and L/D =2.86 by upstream from the cylinder. The triangle was 8 mm uniform side. The Reynolds number based on the cylinder diameter (D= 42 mm) was Re = 1.81 x 104. The research results showed that the triangle rod could decrease the drag of cylinder. Coefficientdrag for cylinder without triangle rod was 0.1276 while the biggest decrease of coefficient of drag with triangle rod washappened at L/D = 1.43 which was 0.0188. It means that the drag of cylinder with triangle rod was 85.25% lower than thecylinder alone.

Abstract

One of the interesting and environmentally friendly microbiology strategies and approaches to control the impact of microplastics is to approach bioremediation technology by harnessing the potential of microbes or indigenous bacteria (local bacteria).  Dumai sea waters currently show a high enough of microplastic pollution which allows the potential of indigenous bacteria to adapt to a plastic environment. The purpose of this study is to isolate and identify indigenous bacteria to degrade plastics from the sea waters of Dumai and to know whether or not there is a difference in the number of bacteria found between stations in this study. This research was conducted in October-December 2020 with experimental methods at the Marine Microbiology Laboratory Faculty of Fisheries and Marine Science Universitas Riau. Based on the results of the study, 12 bacterial isolates were isolated from the research stations.  Isolates of these bacteria have diamaters ranging from 0.2-1.1 cm. Microplastic degradation test results by bacteria found that ISL 10 is an isolate that shows the highest PET degradation activity, which is 17.27% and the diameter of biofilm formation 0.8 cm. Based on biochemical and morphological tests, similar results were obtained that ISL 10 bacteria are a bacterium of the genus Bacillus. The most bacterial colonies were seen in statiun IV (TPI) with an average number of bacteria of 214.9 x 104 CFU/ml.

Abstract

The pattern of antibiotic consumption by the community which is carried out independently without medical guidance causes antibiotic pollution has reached sea waters, one of its impacts on the nature of bacterial resistance. This study aims to look at the pattern of antibiotic sensitivity to E coli that isolated from Dumai sea waters. E coli isolates that grown on MHA media were tested based on the Kirby Bauer disk diffusion method. Seventeen isolates were identified, among which a high rate of resistance to penicillin, isoniazid and streptomycin (17), a high rate of resistance to chloramphenicol (5), intermediate susceptibility to chloramphenicol (11) and only 1 sensitive to chloramphenicol. The multiple antibiotic resistance (MAR) index of E coli isolates was 0.82. These show that antibiotics like penicillin, isoniazid, streptomycin and chloramphenicol were not suitable antibiotic for E coli infection treatment. The community is expected to be more disciplined in using antibiotics and to be careful in using the Dumai sea for fishery activities
