Abstract

Secang adalah tanaman berguna dalam bidang farmakologis, namun budidayanya belum banyak dilakukan. Teknik budidaya yang dapat dilakukan yaitu dengan pengaturan jarak tanam dan aplikasi fitohormon berupa asam salisilat (SA) dan metil jasmonat (MeJA). Tujuan penelitian ini yaitu untuk mengetahui interaksi antara jarak tanam dengan fitohormon SA dan MeJA terhadap respons fisiologis tanaman secang. Penelitian dilaksanakan di Kebun Percobaan dan Laboratorium Pangan dan Perkebunan, Fakultas Pertanian, Universitas Padjadjaran pada bulan Juli hingga Desember 2024. Percobaan menggunakan Rancangan Petak Terbagi, dengan jarak tanam (J) sebagai petak utama dan aplikasi hormon eksogen (H) sebagai anak petak. Penelitian ini dilakukan dengan 3 ulangan. Petak terdiri dari tiga taraf, yaitu jarak tanam: j1 = 1 m x 1 m, j2 = 2 m x 2 m, dan j3 = 3 m x 3 m. Adapun anak petak, yaitu aplikasi hormon eksogen (H) yang terdiri dari 3 taraf, yaitu: h0 = tanpa aplikasi hormon eksogen, h1 = SA 200 ppm, dan h2 = MeJA 100 ppm. Pengamatan fisiologis yang dilakukan meliputi indeks klorofil daun, nilai konduktansi stomata, dan kerapatan stomata. Hasil penelitian menunjukkan bahwa indeks klorofil daun dipengaruhi secara mandiri oleh h1, dengan interaksi terbaik pada j2 × h2. Konduktansi stomata tertinggi diperoleh pada jarak tanam lebar (j3), baik secara mandiri maupun dalam interaksi dengan AS. Sementara itu, kerapatan stomata dipengaruhi secara mandiri oleh jarak tanam sempit (j1) serta MeJA yang tidak berbeda nyata dengan kontrol, dan interaksi terbaik ditunjukkan oleh j1 × h2. Secara keseluruhan, pengaturan jarak tanam dan aplikasi fitohormon SA serta MeJA berpotensi dioptimalkan sebagai strategi budidaya untuk meningkatkan respons fisiologis tanaman secang secara lebih efektif.

Abstract

Groundcherry (Physalis angulata L.) is a plant with many medicinal potentials due to its rich secondary metabolites such as phenolic and flavonoid. However, conventional agriculture practices are still limited, especially in Indonesia. This study was divided into two stages. The first stage was conducted to determine optimal 6-BAP and 2,4-D combination for callus induction, while the second stage was conducted to determine callus phenolic, flavonoid, and antioxidant response to salinity stress. The first stage was arranged in factorial completely randomized design with two factors: 6-BAP (0, 2, and 4 mg/L) and 2,4-D (0, 0.5, 1, and 1.5 mg/L). The second stage was arranged in simple completely randomized design with different NaCl concentration (0, 25, 50, 75, 100 mM) as treatments. The results showed significant interaction (p < 0.05) between 6-BAP and 2,4-D on callus induction. Combination of 2 mg/L 6-BAP and 1 mg/L 2,4-D showed the highest callus formation percentage (46% increase), callus size (60.12% increase), and fresh weight (179.69% increase), and greener compact callus. Application of NaCl as salinity stress at second stage experiment served as an elicitor to enhance callus antioxidant capacity. Salinity level at 100 mM NaCl showed the most accumulation of phenolic content (17.8% increase), flavonoid content (25.17% increase), and antioxidant activities (6.84% IC50 decrease). This study demonstrates plant growth regulator optimization with salinity stress elicitation integration as an effective strategy to enhance antioxidant production in P. angulata callus, providing a practical approach for controlled secondary metabolite production.

Abstract

This study analyzes the influence of farmer group dynamics on the application of Good Agricultural Practices (GAP) in the cultivation of red chili in Sukajaya District, Bogor Regency. Red chili is an important commodity that maintains the stability of the national economy which is vulnerable to fluctuations in price and production. Data shows that there has been a decrease in the productivity of red chili in Sukajaya District despite an increase at the regency level, indicating that the application of GAP is not yet optimal. Using a descriptive quantitative approach and multiple linear regression analysis, this study involved 101 respondents from farmer group members. The results showed that the factors that significantly influenced the application of GAP were group task function, group objectives, guidance, group structure, effectiveness, and pressure, with a contribution of 72.2%. Characteristically, the majority of farmers are men (94%), of productive age 36-45 years (34.7%), with elementary school education (38.6%), very experienced in farming (40.6%), and have sufficiently productive land (41.6%). The strategy to increase the dynamics of farmer groups to optimize the implementation of Good Agricultural Practices (GAP) needs to pay attention to these factors, especially the function of group tasks that have the greatest influence, through training, technical guidance, counseling, and routine monitoring.

Abstract

Rising temperatures associated with climate change threaten tomato productivity, yet the contribution of auxin signaling components to heat-stress adaptation remains incompletely understood. The IAA9 gene, encoding an Aux/IAA transcriptional repressor, is well known for its role in auxin-regulated development, but its role in heat responses is still unclear. This study aims to elucidate the function of IAA9 in modulating tomato responses under heat stress conditions. Researchers utilized tomato iaa9-3 and iaa9-5 mutants and exposed them to prolonged elevated temperatures of 40 to 45 °C for 6 weeks to assess morphophysiological traits, and to 38 to 40 °C for 6 days to evaluate molecular responses through SlDREBA4 gene expression analysis. Under prolonged heat stress, all genotypes exhibited reduced leaf area, leaf number, and total chlorophyll content, accompanied by increased plant height compared to plants grown under normal conditions. Specifically, wild-type Micro-Tom (WT-MT) showed the lowest values in leaf area (165.89 cm²), leaf number (23 leaves), and total chlorophyll content (115.7 µg g-1). In contrast, the iaa9-3 and iaa9-5 mutants recorded the highest plant heights at 11.98 and 12.13 cm, respectively, indicating a differential growth response under stress. Gene expression analysis revealed that SlDREBA4 expression was upregulated in both iaa9-3 and iaa9-5 mutants compared to normal temperature conditions, with increases of 0.45-fold and 1.78-fold, respectively. These results indicate that IAA9 mutations confer enhanced thermotolerance in tomato, as reflected by altered morphology and increased heat-responsive gene expression. This study highlights IAA9 as a potential genetic target for improving heat stress resilience in tomato breeding programs.
