Abstract

Plant regeneration post-genetic transformation play an important an role in genome editing activities that should be optimized via several factors such as the composition of the medium and the concentration of plant growth regulators. This study aimed to optimize the kinetin concentrations and medium compositions for shoot multiplication originating from cotyledon node explants of several local citrus cultivars. The cotyledonary nodes from three citrus cultivars (Batu 55, Siam Madu, and Proksi-1 Agrihorti) were incubated in MS medium with Morel and Wetmore vitamins (VMW) supplemented with several kinetin concentrations (0; 0.2; 0.4; 0.6; 0.8; and 1 mg/L). The best kinetin concentrations for number of shoots variable were then combined with MT medium. The results showed that kinetin concentration at 0.8 mg/L gave the best number of shoot in Batu 55 cultivar as well as 1 mg/L concentration in Siam Madu and Proksi-1 Agrihorti cultivars. The combination 1 mg/L kinetin with Murashige and Tucker (MT) medium showed the best number of shoots, percentage of shoot formation, number of leaves, number of nodes, and shoot length in this study. This medium composition could be further used for shoot multiplication in genetic transformation in those three citrus cultivars, including genome editing activities in development of new improved citrus varieties.

Abstract

Tetraploid citrus are important for interploidal hybridization to create triploid seedless citrus. Colchicine is the most commonly used as antimitotic agent to induce polyploid plants. Tetraploid induction by colchicine in Pummelo ‘Nambangan’ was conducted in vitro using different types of explants. The aim of this research was to induce tetraploid pummelo ‘Nambangan’ by colchicine treatment using germinated seed, shoot tip and cotyledonary node as explants. Tetraploid shoot induction was conducted by soaking germinated seeds, shoot tips and cotyledonary nodes in 0.1 % colchicine for 1, 3 and 5 hours. Regenerant shoots were grown on MS medium and their growth was observed after four weeks in culture. Ploidy level was determined using flow cytometry analysis. Stomata density, length and width of stomatal guard cell were also recorded. The results showed that shoot elongation was inhibited by colchicine treatment. Soaking of shoot tip explants in 0.1 % colchicine for 1 hour resulted in 66.66 % of putative tetraploid shoots. Compared to diploid shoots, tetraploids had lower stomata density but bigger in guard cell size.

Abstract

Abstract: Phalaenopsis orchids are one of the ornamental plants that are very popular with the public because they have attractive flower colors and shapes. One of the obstacles in the success of orchid cultivation is the attack of soft rot disease caused by Dickeya dadantii. Therefore, efforts are needed to find Phalaenopsis clones that are resistant to soft rot disease. This study aims to identify the resistance response of the hybrid orchid genotype Phal. Salu Spot Phal. bellina no. 1102-44. The plant material used was the hybrid orchid Phal. Salu Spot Phal. bellina no. 1102-44 inoculated with Dickeya dadantii bacteria. Identification of the resistance response was carried out through the infection method using the leaf disk technique. The results showed that the clone Phal. Salu Spot Phal. bellina no. 1102-44 was successfully grouped into five resistance classes at 24 JSI (hours after inoculation). A total of three accessions were identified as having resistance to soft rot disease, with two accessions originating from the 22.19 M media treatment (T2-15 and T2-17) and one other accession originating from the 44.39 M media treatment (T4-23).Keywords: orchid, Dickeya dadantii, plant resistance, leaf disk technique Abstrak: Anggrek Phalaenopsis merupakan salah satu tanaman hias yang sangat diminati masyarakat karena memiliki warna dan bentuk bunga yang menarik. Salah satu hambatan dalam keberhasilan budidaya tanaman anggrek adalah serangan penyakit busuk lunak yang disebabkan oleh Dickeya dadantii. Oleh karena itu, diperlukan upaya untuk menemukan klon Phalaenopsis yang memiliki ketahanan terhadap penyakit busuk lunak. Penelitian ini bertujuan mengidentifikasi respons ketahanan genotipe anggrek hibrida Phal. Salu Spot Phal. bellina no. 1102-44. Bahan tanaman yang digunakan berupa anggrek hibrida Phal. Salu Spot Phal. bellina no. 1102-44 yang diinokulasi dengan bakteri Dickeya dadantii. Identifikasi respons ketahanan dilakukan melalui metode infeksi menggunakan teknik leaf disk. Hasil penelitian menunjukkan bahwa klon Phal. Salu Spot Phal. bellina no. 1102-44 berhasil dikelompokkan ke dalam lima kelas ketahanan pada 24 JSI (jam setelah inokulasi). Sebanyak tiga aksesi teridentifikasi memiliki ketahanan terhadap penyakit busuk lunak, dengan dua aksesi berasal dari perlakuan media 22,19 M (T2-15 dan T2-17) dan satu aksesi lainnya berasal dari perlakuan media 44,39 M (T4-23).Kata kunci: Anggrek, Dickeya dadantii, ketahanan tanaman, tekhnik leaf disk

Abstract

The evaluation of citrus resistance to Huanglongbing (HLB) disease is still challenging due to the incapability of the bacteria to be cultured purely in artificial medium, the complexity of inoculation methods, and the long duration required for phenotypic observation. Thus, the use of molecular markers is one of the alternatives to solve this problem. The focuses of this study were to perform in silico analysis of the nucleotide variations in NONEXPRESSOR OF PATHOGENESIS-RELATED GENES 1 (NPR1) gene sequence among several citrus genotypes whose resistance information to HLB have been known previously and to employ phylogenetic analysis among them. The NPR1 gene sequences from 20 genotypes which consisted of 14 citrus and six relative genotypes were collected in silico from Citrus Genome Database and analyzed using multiple sequence alignment program. A total of six interesting SNPs that could distinguish between susceptible and resistant citrus genotypes were detected in this study. As many as five SNPs were non-synonymous, while only one synonymous SNP that did not cause the amino acid change was identified in this study. The phylogenetic analysis also revealed the separation between susceptible and tolerant/resistant citrus genotypes in two main clusters. The SNPs found in this study are expected to be useful for designing new functional markers as a selection tool in future studies.
