Abstract

ABSTRACTSugarcane cultivars tolerant to drought stress are needed for specific location. In vitro selection can be used to obtain plants tolerant to drought stress through regeneration of callus in the culture medium. The purpose of this study was to obtain regenerants from irradiated callus on the stress medium. The experimental design was a completely randomized design with two factors. The first factor was the regeneration medium (RG) added with PEG 6000, i.e., 0%, 5%, 10% and 15%, while the second factor was the dose of gamma ray irradiation i.e. 0 krad, 5 and 10 krad, and 15 krad. There were 12 treatments, each treatment was repeated 18 times and each repetition was a culture bottles planted with three clums of callus 10 mm diameter. RG medium was the MS medium added with 0.5 mg L-1 BAP, Kinetin 0.1 mg L-1 and IBA 1.0 mg L-1. The higher the concentration of PEG, the less regenerants were produced. Regenerants could be generated from the selection medium PEG up to 15%. Several shoots still produced from callus irradiated with 10 krad. Selection medium with PEG up to 10% decreased the precentage of albino regenerants. The gamma ray irradiation dose and the concentration of PEG significanly affected the number of  roots produced.Keywords: drought tolerant, in vitro, polyethylene glycol, plantlets, albino

Abstract

Several bacterial pathogens can cause diseases of potato. Ralstonia solanacearum and Erwinia carotovora are those of the world’s most important diseases of potato, especially in tropical climates. This experiment was aimed to obtain putative potato cultivars having good tolerance to bacterial wilt caused by R. solanacearum and soft rot caused by E. carotovora. Two adopted potato cultivars in Indonesia, cv. Atlantic (2n=4X=48) and cv. Granola (2n=4X=48), were used as parents. The experiment was arranged in a Randomized Complete Design. Test for tolerance was perfomed in vitro using BF15 as susceptible control and Solanum stenotonum as tolerance control. The result showed that there was high diversity of tolerance level to both R. solanacearum and E. carotovora. Some clones showed good tolerance level as compared to cv. Atlantic and cv. Granola, but none showed that as compared to S. stenotonum.

Abstract

Endosperm quantity in kopyor coconut determines the quality of kopyor coconut. However, data of the best parental based on their progeny test is unavailable yet. In this study, we employed polycross approach and SSR molecular marker to estimate general combining ability of the best pollen contributor for the breeding purpose of this trait. Totally 151 kopyor coconuts were collected from the naturally open-pollinated population of kopyor Tall coconut Agom Lampung. The harvested nuts were rated for their endosperm quantity (scored 1-9). The analysis identified 31 paternal trees of the 53 progenies. The TJ-185 with the highest GCA has been considered as a promising paternal tree for development of high endosperm quantity of kopyor coconut.Keywords: combining ability, molecular marker, paternal trees, polycross, tall coconut

Abstract

Benih padi yang baru dipanen umumnya tidak berkecambah walaupun dikecambahkan pada kondisi optimum yang disebut after-ripening, dan secara alamiah akan hilang setelah waktu tertentu. Penelitian ini bertujuan melakukan karakterisasi agro-morfologi dan menetapkan periode after-ripening benih 5 lanras potensial di Timor-Leste. Penelitian dilaksanakan pada bulan April 2018-November 2019. Penanaman dan pengamatan karakter agro-morfologi 5 lanras potensial Timor-Leste, yaitu Hare R-oitu, Fos Mean, Hare Belit, Hare Nona Portu, dan Ale Mamea Ula Lesa, dilaksanakan di Desa Caibada Distrik Baucau, Timor-Leste. Pengamatan periode after ripening dan mutu benih ke lima lanras tersebut dilaksanakan di Laboratorium Pengujian dan Penyimpanan Benih, Departemen Agronomi dan Hortikultura, IPB University, dan dirancang dalam rancangan acak lengkap (RAL) dua faktor dengan empat ulangan. Faktor pertama adalah 5 lanras tersebut di atas dan faktor kedua adalah periode simpan yang terdiri atas 2, 3, 4, 5, 6, 7, dan 8 minggu setelah panen (MSP). Hasil penelitian menunjukkan bahwa 5 lanras potensial di Tinmor-Leste mempunyai keragaan tanaman yang pendek (76.3-105.50 cm), tergolong berumur sedang (129-141 HSS), dan jumlah anakan produktif tinggi (17.5-53.50 anakan), kecuali Ale Mamea Ula Lesa. Lima lanras tersebut mempunyai potensi produktivitas yang tinggi berkisar 8.1-9.8 ton ha-1. Periode after-ripening lanras Hare Belit dan Hare Nona Portu berakhir pada 2 MSP, Hare R-oitu pada 6 MSP, sedangkan Ale Mamea Ula Lesa dan Fos Mean pada 8 MSP. Semakin lama benih disimpan rasio kandungan ABA/GA3 semakin menurun dari 0.53-1.22 ppm pada 2 MSP menjadi 0.45-0.93 ppm pada 4 MSP. Lanras Hare Belit dan Hare Nona Portu merupakan lanras yang potensial dikembangkan menjadi varietas nasional di Timor-Leste, masing-masing untuk pengembangan di dataran rendah dan dataran sedang. Abstract

Shallot anthracnose, caused by Colletotrichum gloeosporioides, is a devastating disease in a tropical country with high humidity and rainfall. Chemical control of anthracnose is neither economical nor eco-friendly, and genetic resistance is considered an efficient management method. This study aimed to determine the resistance groups of several shallot varieties and predict resistance heritability characteristics. In this study, a total of 13 Indonesian shallot varieties were evaluated for anthracnose resistance and separated into two groups, resistance and susceptible, based on K-means clustering developed by using disease resistance/susceptibility characteristics such as incubation period, disease incidence, disease severity, and spot diameter. The results indicate that the Agrihorti, Maja Cipanas, Batu Ijo, and Rubaru varieties were identified as resistant groups based on lower disease severity and incidence, smaller spot diameter, and longer incubation period. Maja Cipanas and Rubaru were more consistent in all variables, which is recommended as a source of genetic resistance genotypes. On the other hand, the Biru Lancor, Bima Brebes, Srikayang, Violetta, Slupu Merah, Pancasona, Sakato, Katumi, and Kuning varieties were identified as susceptible groups based on high disease severity and incidence, large spot diameter, and short incubation period. Abstract

Rapid multiplication of potato seed using cuttings is a pivotal strategy for ensuring seed availability. This study aimed to elucidate the interactive effects of node number, paclobutrazol concentration, and planting density on seed potato and cutting productions. The study consisted of two experiments. The first experiment evaluated two genotypes (Granola and PKHT-6) with one or two nodes per cutting. The second experiment investigated planting density (one, three, or five plants per polybag) and paclobutrazol concentration (0, 15, 30, and 45 mg L-1). Notably, node number did not significantly influence cutting success, despite genotype-specific and interactive effects on vegetative parameters like plant height, stem diameter, leaf number, and root length. Planting density had no statistically significant impact on all vegetative parameters but demonstrably affected all production parameters. Conversely, paclobutrazol concentration significantly affected all vegetative parameters and influenced all production parameters except tuber weight per plant. From the results of this research, the use of single cutting is more recommended because it will obtain more plant material compared to two-node cuttings. Thus, in a more global context of potato development, this has the potential to increase the production of potato seeds from cuttings twice as much as the current seed production. The use of three plants/polybags is more recommended since it will produce more tuber but not different with five plants, and the use of paclobutrazol 15 mg L-1 is also recommended to increase the number of tubers. In this way, the production costs of G0 potato seeds can be reduced so that the availability of G0 potato seeds will be more guaranteed and affordable. Abstract

The gene that regulates callose could be modified by CRISPR/Cas9 technology. This research aimed to insert the CRISPR/Cas9-CsCS gene into Ponkan orange genome using Agrobacterium tumefaciens. The explants were soaked in a bacterial suspension for 20 minutes and incubated for 2-3 days. In vitro acceleration growth was conducted with a two-factor completely randomized design. The first factor is the type of explant with three levels (nucellar embryo, zygotic embryo, cotyledon node), and the second factor is the type of media with 6 levels (VMW, MT, MSK0, MSK1, MSK2, MSK3). The results showed of all explant types, the highest plant height average and number of leaves were obtained in a media combination of MS + Kinetin 2 mg L-1, MT, and MS + Kinetin 3 mg L-1. The highest transformation efficiency was in the nucellar embryo explant, while the highest regeneration efficiency was in the zygotic embryo explant. The highest shoot tip grafting percentage was achieved in the cotyledon node explant at 100%. In the grafting phase, the putative transformants before and after artificial bacterial inoculation showed that Ponkan 606 and Ponkan 597 had the highest plant heights, respectively. The intensity of Huanglongbing attacks after bacterial inoculation showed that three genotypes did not show HLB symptoms in the 24th week of observation, namely genotypes 598, 606, and 607. This study concluded that gene transformation in citrus plants produced three genotypes that did not show HLB symptoms. Abstract

Penurunan mutu benih atau deteriorasi merupakan salah satu penghambat dalam produksi benih jagung dan umumnya terjadi selama penyimpanan benih yang ditandai dengan turunnya viabilitas dan vigor benih sehingga mengakibatkan jeleknya pertanaman serta menurunnya hasil. Pengusangan cepat secara kimia merupakan salah satu metode yang digunakan untuk mengetahui percepatan penurunan kualitas benih dengan memanfaatkan kinerja uap etanol. Pengusangan ini dilakukan oleh APC IPB 77-1 MM yang berfungsi melakukan devigorasi benih baik secara fisik dengan uap panas maupun secara kimia dengan etanol. Hasil dari pengujian ini akan digunakan untuk menduga vigor daya simpan (VDS) benih jagung sehingga pemanfaatan dan pengolahan benih jagung dapat dilakukan secara efektif dan efisien. Penelitian dilaksanakan di Laboratorium Biologi dan Biofisika Benih, Laboratorium Penyimpanan dan Pengujian Mutu Benih, Departemen Agronomi dan Hortikultura, Fakultas Pertanian, Institut Pertanian Bogor dari bulan Maret 2019 hingga Agustus 2019. Penelitian ini terdiri dari dua percobaan. Percobaan pertama yaitu metode pengusangan dengan menyimpan benih pada kelembaban 98% dan suhu 40 oC selama 10 hari. Rancangan yang digunakan adalah Rancangan Kelompok Lengkap Teracak (RKLT) dengan satu faktor yaitu varietas jagung. Percobaan kedua yaitu pengusangan cepat benih jagung secara kimia dengan etanol 96% menggunakan APC IPB 77-1 MM. Rancangan yang digunakan adalah RKLT satu faktor varietas jagung. Hasil percobaan menunjukkan alat pengusangan cepat IPB 77-1 MM dapat digunakan untuk menduga penurunan vigor daya simpan pada benih jagung. Pengujian pengusangan cepat secara kimiawi lebih efektif dan efisien dalam menentukan nilai vigor daya simpan karena membutuhkan waktu yang relatif lebih singkat. Hasil perbandingan secara deskriptif menunjukkan bahwa APC IPB 77-1 MM dapat menunjukkan urutan nilai vigor daya simpan benih kelima varietas jagung yang sama dengan percobaan pengusangan dengan metode penyimpanan pada suhu dan kelembaban tinggi. Abstract

Pisang merupakan buah tropis yang sangat diminati oleh masyarakat Indonesia, sehingga dibutuhkan peningkatan produksi buah pisang yang tinggi dengan kualitas yang baik melalui manajemen pemupukan. Kegiatan penelitian bertujuan mempelajari pengelolaan pemupukan pisang Cavendish klon DM2 di Lampung Timur. Penelitian dilaksanakan pada Januari hingga Mei 2019. Pengamatan pengelolaan pemupukan meliputi konsep 5T (tepat jenis, tepat dosis, tepat waktu, tepat cara, dan tepat tempat) dan evaluasi produksi. Analisis data hasil penelitian menggunakan Uji t-student. Hasil pengamatan di areal dibandingkan dengan standar dari Departemen Research and Development. Pemupukan di kebun telah memenuhi kriteria tepat kerja. Ketepatan realisasi dosis pupuk Urea, Kiesrite, dan ZnSO4 telah tepat dosis, sedangkan pupuk KCl dan Petro cas kurang tepat dosis. Realisasi waktu pemupukan belum memenuhi standar kebun. Ketepatan cara pemupukan Urea, KCl, dan Petro cas telah memenuhi kriteria, namun Kiestrite dan ZnSO4 belum memenuhi standar kebun. Semua jenis pupuk belum tepat tempat saat aplikasi kecuali pupuk ZnSO4. Umur buah rata-rata yang dipanen pada sampel blok pengamatan belum maksimal dengan lebar buah yang masih di bawah standar kebun. Jumlah sisir dan hasil bobot tandan juga masih lebih rendah dibandingkan standar kebun. Kata kunci: pisang, pupuk, produksi, tandan

Abstract

Pembibitan tanaman hias biasanya menghasilkan banyak varietas anyelir. Keragaman morfologi antar varietas (dalam satu spesies) memberikan keleluasaan bagi konsumen dalam memilih produk yang akan dibeli. Penelitian bertujuan untuk memperoleh informasi mengenai produksi, kriteria panen komoditas anyelir serta mengevaluasi kelayakan usaha tani dari komoditas tersebut. Penelitian dilakukan di Cianjur, Jawa Barat, Indonesia, pada bulan Januari hingga April 2020. Terdapat 2 percobaan, yaitu (1) mengamati perbedaan 2 varietas anyelir yang diproduksi sebagai tanaman hias pot; varietas anyelir yang diamati memiliki kode DNT 417 dan DNT 917; dan (2) mengamati perbedaan 2 varietas anyelir yang diproduksi dalam polibag untuk digunakan sebagai tanaman hamparan, varietas anyelir yang diamati memiliki kode DNT 103 dan DNT 123. Secara umum keempat varietas yang diamati (untuk anyelir pot dan polibag) yang diproduksi dengan menggunakan prosedur standar perusahaan dapat memenuhi standar mutu yang ditetapkan perusahaan. Nilai R/C ratio pada tanaman anyelir pot dan anyelir polibag yaitu 3.47 dan 2.53. Nilai R/C ratio pada komoditas anyelir pot dan anyelir polibag menunjukkan hasil >1 yang berarti usahatani layak dijalankan dan bersifat menguntungkan.
