Abstract

The primary and secondary somatic embryogenesis can be used to propagate Coffea arabica L clonally.  However, the success of this propagation was depended on plant growth regulator and varieties. This study aimed to examine the possibility of 2,4-D and thidiazuron application to form primary and secondary somatic embryo to support Arabica coffee clonal propagation. The study consisted of two activities (1) 2,4-D and thidiazuron Application to Induce Primary Somatic Embryogenesis of Arabica Coffee and (2) The Application of thidiazuron in Solid and Semi-Solid Media to Induce Secondary Somatic Embryos.  The results indicated significant effect of varieties and plant growth regulator on fresh weight, number of torpedo and germinated embryo.  However, it showed no significant effect on callus formation percentage. The best medium to induce primary somatic embryogenesis depending on variety, on the treatment of 4.52 μM 2,4 -D +18.16 μM thidiazuron was the best for AS2K and Sigarar Utang varieties, S 795 at 4.52 μM 2,4-D + 9.08 μM thidiazuron, whereas Kartika at 4.52 μM 2.4-D + 13.62 μM thidiazuron.  The morphology of coffee somatic embryo was normal.  Primary somatic embryo was developed indirectly, whereas the secondary somatic embryo was directly.  The application of 9.08 μM thidiazuron  increased the percentage and number of secondary somatic embryos, hence enhancing number of Arabica coffee planlet. Keywords : Coffea arabica L, 2,4-D, thidiazuron, semi-solid media, Indirect somatic embryogenesis

Abstract

Dalam memilih kontraktor yang berkualifikasi diperlukan proses pemilihan yang disebut pelelangan (tender). Tender adalah kegiatan yang bertujuan untuk menyeleksi, mendapatkan, menetapkan dan menunjuk perusahaan yang paling layak untuk mengerjakan suatu paket pekerjaan Proses tender memiliki beberapa tahapan, mulai dari tahap pengumuman, pendaftaran, pemberian penjelasan (aanwijzing), penyampaian, pembukaan, hingga evaluasi dokumen penawaran. Dalam pelaksanaan proses tender proyek konstruksi terdapat berbagai faktor. Faktor-faktor yang diteliti berdasarkan kombinasi beberapa literature dan hasil penelitian terdahulu meliputi empat faktor yaitu; karakteristik administratif (X1), karakteristik teknis (X2), karakteristik harga (X3), karakteristik klarifikasi (X4). Berbagai faktor tersebut perlu diteliti oleh para pemangku kepentingan agar dapat mengantisipasi kerugian yang terjadi sedini mungkin. Penelitian ini bertujuan untuk mengindentifikasi faktor-faktor yang dapat mempengaruhi kegagalan kontraktor dalam proses tender dengan sistem elektronik. Penelitian ini dilakukan di Kota Surabaya dengan mata anggaran proyek Tahun 2018. Responden penelitian ini adalah para penyedia jasa konstruksi (kontraktor) proyek Kota Surabaya yang tergabung dalam anggota GAPENSI 2018. Teknik pengumpulan data menggunakan kuisioner dan wawancara dengan memakai instrument Skala linkert 1-5. Analisis data menggunakan analisis kuantitatif dan kualitatif. Hasil penelitian menyimpulkan bahwa ketidaksesuaian sisa kemampuan paket atau disingkat SKP (X1.1) dan kurangnya dukungan supplier (X2.1) adalah faktor yang paling dominan menyebabkan kegagalan, sedangkan faktor-faktor lainnya merupakan faktor yang perlu diperhatikan juga untuk mengantisipasi atau meminimalisir terjadinya kegagalan. Kata kunci : faktor, kegagalan, proyek konstruksi, tender

Abstract

The study on growth respons of Lansium domesticum  poliembryoni seed on MS and WPM media with added by coconut water had been carried out from Maret up to Mei 2006. This experiment had been done in Tissue Culture Laboratory, Agronomy-Holticulture Depatment, Agronomy Faculty, Institute Pertanian Bogor, using  two factorial of Completely Randomized Design with  nine treatments, and ten replications. The first factor treatment were  MS, ½ MS and WPM media, and the 2nd  factor were 0, 15 and 20% coconut water. The result showed that the emergence time of the first adventitious shoot were beginning on 26,5 days after planting, with one until four numerous of shoots, on MS, ½ MS, WPM media with added  until 20% coconut water. There were only 50-70% shoot formed on 8 weeks after planting. With 20% coconut water suplemented on MS culture media showed that the height of shoots were significant than controle.Key words: Lansium domesticum, coconut water, polyembryonic seed, culture media.

Abstract

Low productivity of shallot in Indonesia can be improved through development of high yielding variety. The availability of genetic information related to the character being improved is inevitable for effective breeding program. In this study, seven shallot genotypes were evaluated for their combining ability for yield improvement using half diallel cross.  Heterosis effect and heritability estimate was also investigated for yield-related characters.  The results showed that there were significant differences in general combining ability (GCA) and specific combining ability (SCA) among the evaluated genotypes. Tiron and Timor had the greatest GCA.  The greatest SCA and heterosis value was found in crosses of Kuning/Tiron, Timor/Bima Juna, Tiron/Timor and Kuning/Sibolangit. Heterosis effect varied from low to high. Broad sense heritability estimate for all characters was high, but narrow sense heritability was low for most charaters.  The dominant gene action observed on all yield-related characters suggests that the evaluated genotypes are potential to be used in breeding for high yielding hybrid varieties.   Keywords: GCA, SCA, heterosis, heritability, shallot

Abstract

Improvement of selected induction culture media on callus induction in anther culture of anthurium and a histologicalstudy on its callus formation were studied at the tissue culture laboratory of the Indonesian Ornamental CropsResearch Institute from February to October 2008. The objectives of the study were to optimize selected media forcallus formation, reveal cell origin of callus derived from anther culture and shoot formation process. Selectedmedia improved in the study were 1) MMS-TBN containing 0,5 mg/l TDZ, 1,0 mg/l BAP and 0,01 mg/l NAA (Winartomedium, WM) and 2) MMS III supplemented with 1,5 mg/l TDZ, 0,75 mg/l BAP and 0,02 mg/l NAA (Winarto andRachmawati medium, WRM). Improvement treatments were carried out by omission and application of 2,4-D in 0.5mg/l and reduction of medium strength of full, half, quarter, one eighth, one sixteenth, and zero strength. Afactorial experiment was arranged using a randomized complete block design with four replications. Results ofthis study indicated that the highest callus induction was clearly established in WRM. The medium stimulatedpotential growth of anther (PGA) up to 81% with 49% of percentage of anther regeneration (PAR) and 2.7 number ofcallus formed per replication (NCF). Significant improvement in callus formation was also recorded by reduction ofmedium strength of WRM to one eighth compared to others. The reduction induced PGA up to 58% with 29% of PARand 1.8 NCF. From histological studies it was well recognized that regenerated callus on half anthers cultured wasoriginated from middle layer cells of anther wall. The morphogenic response of anther wall cells caused primarilyon no androgenesis effect in microspore cells.

Abstract

Perbanyakan klonal anggrek melalui kultur in vitro dibutuhkan untuk memperoleh sejumlah besar propagul tanaman yang berkualitas. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh konsentrasi BAP terhadap kemampuan multiplikasi tunas dalam propagasi klonal beberapa anggrek hibrida Phalaenopsis dan menganalisis keragaman genetik menggunakan marka Pto-SNAP pada planlet klonal. Percobaan dilakukan secara terpisah pada 3 populasi tunas anggrek hibrida Phalaenopsis yaitu, tunas hasil perkecambahan biji dari Phal. amabilis x Phal. ‘KHM 421’ dengan 5 taraf perlakuan BAP (0.00, 11.09, 22.19, 33.29, 44.39 µM), tunas klon Phal. ‘Salu Spot’ x Phal. bellina ‘1102-38’ dan Phal. ‘Salu Spot’ x Phal. bellina ‘1102-44’ dengan 4 taraf perlakuan BAP (11.09, 22.19, 33.29, 44.39 µM). Hasil penelitian menunjukkan bahwa konsentrasi BAP terbaik untuk multiplikasi tunas pada tiga populasi yang diuji adalah 22.19 µM dengan jumlah tunas masing-masing 3.7, 4.7 dan 6.0 tunas pada 12 minggu setelah tanam (MST). Protocorm like bodies (PLBs) hanya dihasilkan pada populasi Phal. amabilis x Phal. ‘KHM 421’ dengan jumlah PLBs terbanyak perlakuan BAP 11.09 µM. Evaluasi keragaman genetik menggunakan 11 marker Pto-SNAP (single nucleotide amplified polymorphism) dilakukan pada planlet klonal Phal. ‘Salu Spot’ x Phal. bellina ‘1102-44’. Hasil analisis menunjukkan adanya variasi somaklonal dari planlet yang diuji perlakuan BAP 22.19 µM dengan keragaman 7.7% pada lokus Pto-241 dan perlakuan BAP 44.39 µM dengan persen keragaman 11.1% pada lokus Pto-424. Abstract

ABSTRACTPreliminary study: Gene transfer from transgenic potato Katahdin RB to non transgenicpotato. One of the concerns associated with the release of transgenic crops, is the possibilityof the gene flow from transgenic crops to neighboring crops of the same species or to relatedspecies. In plants, gene flow is a routine process occur through the natural hybridization. Theopportunity for gene flow occur depends principally on two factors, the degree of sexualcompatibility between donor and recipient species, and the physical distance between thetwo. The experiment was conducted to determine whether the gene flow from transgenicpotato Katahdin RB to non transgenic was occurred, based on selection using a 50 mg/lkanamycin, and to estimate gene flow mediated by natural hybridization at different isolationdistances. Preliminary result indicated that a rapid and simple method using MS0 liquid mediawith kanamycin 50 mg/l was effective for screening the seeds. There was a gene flow fromtransgenic potato Katahdin RB to non transgenic, based on a rapid and simple selectionmethod using 50 mg/l of kanamycin as selectable marker. The isolation distance used in thestudy were 0.8, 1.6, 2.4, 3.2, 4.0, 4.8, 5.6, 6.4, 7.2, 8.0, 8.8, 9.6, 10.4, and 11.2 m from the row oftransgenic potato Katahdin RB. The gene flow through natural hybridization at a isolationdistances of (0.8 - 1.6 m), (2.4 ? 4 m), and (4.8 ? 6.4 m) from transgenic to non transgenic plantswere 13.78, 10.92, and 3.82%, respectively. At a distance of 7.2 ? 8 m, the frequency of gene flowwas declined to 0%. The frequency of gene flow from transgenic potatoes to non transgenicpotatoes markedly decreased by increasing the isolation distance, and was negligible at 7.2 m.Key words : natural hybridization, transgenic potato RB, kanamycin selection

Abstract

In Vitro Multiplication of  Black Orchid (Coelogyne pandurata Lindl.) Using the Combination of NAA and BAPABSTRACTBlack orchid is an indigenous plant from Kalimantan, Indonesia. It becomes endangered because of forest over-exploitation and its low natural reproduction rate. Tissue culture is considered to offer a solution to conserve and propagate this species. The aim of this research was to evaluate the effect of Naphtalene Acetic Acid (NAA) and 6-Benzile Amino Purine (BAP) on shoots multiplication of black orchid. The basic medium used was a half of Murashige & Skoog (MS) composition supplemented with 150 mLL-1 coconut water. Initial explants used were 6-month-old shoots of germinating seeds. The shoot cultures were incubated for 23 weeks. Results showed that the best combination for shoot multiplication was NAA 0.0 mgL-1 with BAP 0.2 mgL-1. Shoot grew better on medium with BAP and without NAA while roots growth was better on medium without the two plant growth regulators. The addition of BAP up to 0.3 mgL-1 increased the leaf number, which however decreased at higher BAP concentration.Keywords: BAP, black orchid, Coelogyne pandurata, multiplication, NAA ABSTRAKAnggrek hitam merupakan flora langka asli Kalimantan, Indonesia. Keberadaa anggrek ini di alam semakin langka akibat eksploitasi berlebihan dan sulitnya perbanyakan secara alami. Kultur jaringan merupakan metode untuk mengatasi kelangkaan anggrek ini. Penelitian ini bertujuan untuk mengetahui pengaruh kombinasi NAA dan BAP terhadap multiplikasi anggrek hitam. Media dasar yang digunakan adalah ½ MS dengan penambahan air kelapa 150 mLL-1. Eksplan yang digunakan adalah tunas hasil semai biji umur 6 bulan. Kultur tunas diinkubasi selama 23 minggu. Hasil penelitian menunjukkan bahwa kombinasi terbaik untuk multiplikasi tunas adalah NAA 0 mgL-1 dengan BAP 0,2 mgL-1. Tunas tumbuh lebih baik dalam media dengan penambahan BAP tanpa NAA, sedangkan akar pada media tanpa NAA dan BAP. Penambahan BAP sampai 0.3 mgL-1 mampu meningkatkan jumlah daun, namun menurun dengan penambahan di atas konsentrasi tersebut.Kata Kunci: anggrek hitam, BAP, Coelogyne pandurata, multiplikasi, NAA

Abstract

INDUCTION MUTATION AND SELECTION OF IN VITRO PLANT OF WHEAT (Triticum aestivum L.)The goal of this research was to produce wheat crop which is tolerant to lowland condition.Six varieties were used, Dewata, Selayar, Alibey, Oasis, Rabe and HP1744. This research consisted of 4 stages: production of the best callus on MS medium containing 3 g/L 2.4-D, induced mutation of embryogenic callus using EMS, in vitro selection of callus at temperature of 27–35°C, and callus regeneration. The best result for callus production was 76% for Dewata and 70% for Selayar varieties. Higher concentration of EMS and longer soaking time decreased the percentage of callus growth. LC50 for Dewata was 0.3% EMS at 30 minutes and that for Selayar was 0.1% EMS at 60 minutes. The higher the temperature was, the lower was the adaptation tolerant of the plants, and callus growth was inhibited. At the highest temperature (35°C) the callus did not grow at all.Keywords: Induced mutation, Triticum aestivum, EMS, in vitro selection, callusABSTRAKTujuan penelitian ini adalah untuk merakit tanaman gandum yang toleran pada dataran rendah. Varietas yang digunakan ada 6 yaitu Dewata, Selayar, Alibey, Oasis, Rabe dan HP-1744. Penelitian terdiri atas empat tahap yaitu induksi pembentukan kalus terbaik menggunakan media MS + 3 g/L 2,4-D (dipilih dua varietas yang terbaik), induksi mutasi kalus embriogenik menggunakan EMS, seleksi kalus in vitro pada suhu 27–35°C, dan regenerasi. Hasil induksi kalus terbaik terdapat pada varietas Dewata sebesar 76% dan Selayar sebesar 70%. Semakin tinggi konsentrasi EMS dan semakin lama waktu perendaman yang digunakan maka semakin menurun persentase pertumbuhan kalus. LC50 varietas Dewata adalah EMS 0,3% waktu 30 menit sedangkan LC50 varietas Selayar adalah EMS 0,1% waktu 60 menit.Semakin tinggi suhunya maka semakin berkurang toleran adaptasi tanaman tersebut, dan pertumbuhan kalus semakin sedikit. Abstract

Potato late blight, caused by Phytophthora infestans is one of the most devastating plant diseases. Potato yield losses due to this disease ranged from 47-100%. Frequent intervals and high rates of fungicide spray, currently practiced by potato growers to control the disease are expensive. Host resistance is an alternative control measure that is more economically and environmentally sustainable. Development of late blight resistant plants was conducted by crossing RB transgenic Katahdin SP904 andSP951 as male and two susceptible (Atlantic, Granola) varieties as female parents. F1 progenies were molecularly characterized for the integration of the RB transgene and evaluated for their segregations. Crossing data of Atlantic x transgenic Katahdin SP904 and SP951 produced 71 (57.72%) berries with average number of seeds per berry 139.58 and 83 (41.29%) berries with average number of seeds/berry 85.23, respectively. Granola x transgenic Katahdin SP904 and SP951 crosses gave higher results in terms of berry set (79.55 and 84.44%, respectively) than Atlantic x transgenic Katahdin crosses. A total of 554 F1 progenies were analyzed for the presence of the RB PCR marker. An expected 619-bp and 840-bp band were amplified in the progenies that contain the RB gene. The RB gene was integrated in 65 (45.45%), 77 (47.83%), 47 (45.63%), and 71 (48.30%) F1 progenies of Atlantic x transgenic Katahdin SP904, Atlantic x transgenic Katahdin SP951, Granola x transgenic Katahdin SP904, andGranola x transgenic Katahdin SP951, respectively. Chisquare tests showed that all the four cross combinations followed a 1 : 1 segregation ratio.
