Abstract

Penelitian bertujuan untuk mengoptimasi aktivitas bakteriosin yang dihasilkan Lactobacillus brevis dengan variasi pH dan suhu serta uji konfirmasi enzim proteolitik.  Kultur bakteri berumur 24 jam yang telah diisolasi dari es pisang ijo pada penelitian sebelumnya ditumbuhkan pada media De Man Rogosa Sharp Agar (MRSA) dengan metode striking dan diinkubasi pada suhu 32°C selama 24 jam. Uji aktivitas antibakteri terhadap bakteri indikator menggunakan metode difusi agar.  Hasil penelitian menunjukkan Lactobacillus brevis berpotensi digunakan sebagai agen biopreservatif pada bahan pangan yang dapat menghambat Gram positif (Staphylococcus aureus) dengan aktivitas tertinggi pada pH 6 dan suhu 40°C dengan zona hambat secara berturut turut yaitu 12,10 mm dan 12,40 mm dan Gram negatif (Escherichia coli) dengan aktivitas tertinggi pada pH 6 dan suhu 40°C dengan zona hambat secara berturut turut yaitu 9,30 mm dan 8,5 mm.  Konfirmasi menggunakan enzim zona hambat ketika ditambahkan enzim katalase yaitu sebesar 7 mm dan pada bakteri Escherichia coli dan 10 mm pada bakteri Staphylococcus aureus. Brevicin diinaktifkan oleh enzim tripsin sehingga tidak terbentuk zona hambat pada koloni.Kata Kunci: Bakteriosin, bakteri asam laktat, pisang ijo

Abstract

Pada masa pandemi Covid-19 kebanyakan para pendidik memanfaatkan media sosial untuk menyampaikan file materi pembelajaran tanpa disertai penjelasan secara lisan, tidak seperti pada belajar tatap muka dikelas. Melalui sarana yang terbatas tersebut memberikan efek antara guru dan siswa mengalami kendala interaksi yang dapat dijadikan sebagai tolak ukur penyerapan materi yang diberikan guru dan pemahaman materi bagi siswa. Berdasarkan kondisi masalah ini maka kami berinisiatif membuat kegiatan pengabdian kepada masyarakat dengan memberikan pelatihan pemanfaatan aplikasi Powerpoint Presenter-View-Recorder dan Interactive Mentimeter sebagai media interaksi dalam pembelajaran daring, yang masih jarang digunakan oleh para pengajar khususnya di SMPN 264 Jakarta. Pelaksanaan program pelatihan ini bertujuan untuk memberikan terobosan dalam mengajar. Siswa diharapkan tidak bosan saat mengikuti kegiatan belajar. Inovasi yang digunakan adalah dengan mengoptimalkan aplikasi Powerpoint dengan bentuk Presenter-View-Recorder sekaligus penggunaan aplikasi Interactive Mentimeter dan langsung mengupload video pembelajaran yang telah dibuat ke aplikasi Google Classroom dan media sosial Youtube.

Abstract

Infeksi yang paling umum tersebar didunia salah satunya yaitu infeksi cacing. Prevalensi infeksi cacing di Indonesia berdasarkan Departemen Kesehatan Republik Indonesia (DepKes RI) masih tergolong cukup tinggi yaitu sebesar 24,1%. Ekstrak etanol daun alamanda mengandung metabolit sekunder yang diduga mempunyai aktivitas anthelmintik. Penelitian ini bertujuan untuk mengetahui aktivitas anthelmintik, pengaruh peningkatan konsentrasi, nilai LC50 dan nilai LT50 ekstrak etanol daun alamanda terhadap cacing Ascaridia galli dan Raillietina tetragona. Uji anthelmintik terbagi dalam 5 kelompok perlakuan yaitu 3 kelompok ekstrak etanol daun alamanda (konsentrasi 5 mg/ml, 25 mg/ml dan 50 mg/ml), kelompok kontrol normal (NaCl 0,9%) dan kelompok kontrol positif (mebendazol 5 mg/ml). Waktu dan jumlah kematian cacing dicatat dan dianalisis secara statistik menggunakan program komputer SPSS. Hasil penelitian menunjukkan waktu kematian masing-masing cacing pada tiap kelompok perlakuan berbeda bermakna dengan kontrol normal. Peningkatan konsentrasi ekstrak meningkatkan efek anthelmintik yang dihasilkan. Nilai LC50 ekstrak etanol daun alamanda (Allamanda cathartica L.) pada Ascaridia galli dan Raillietina tetragona yaitu 2,658 mg/ml dan 2,975 mg/ml sedangkan nilai LT50 yaitu 19,8 jam dan 5,1 jam. Hal ini menunjukkan bahwa ekstrak tersebut memiliki aktivitas anthelmintik.Kata Kunci: anthelmintik, Allamanda cathartica, Ascaridia galli, Raillietina tetragona.   One of the most common infection spread in the world is worm infection. The prevalence of worm infections in Indonesia based on the data form Ministry of Health of the Republic of Indonesia (DepKes Ri) is still quite high which is 24.1 %. The ethanolic extract of alamanda leaves contains secondary metabolites that are suspected of having anthelmintic activity. This study aims to determine the anthelmintic activity, the effect of increased concentration, LC50 and LT50 values of ethanolic extract of  alamanda leaves to Ascaridia galli and Raillietina tetragona worms. The anthelmintic test was divided into 5 treatment groups, i.e 3 groups treated with ethanolic extract of alamanda leaves (concentration of 5 mg/ml, 25 mg/ml and 50 mg/ml), normal control group (NaCl 0,9%) and positive control group (mebendazol 5 mg/ml). The time and amount of worm’s mortality were recorded and statistically analyzed using the SPSS computer program. The results showed that the mortality period of each worm in every treatment groups was significantly different with the normal control group. Increased concentration of the extract will increase the anthelmintic  effect. The LC50 values of ethanolic extract of alamanda leaves (Allamanda cathartica L.) in Ascaridia galli and Raillietina tetragona was 2,658 mg/ml and 2,975 mg/ml respectively, while LT50 values was 19,8 hours and 5,1 hours respectively. AbstrakKemitraan Perusahaan Grab dengan driver dalam Undang-undang nomor 20 tahun 2008 tentang Usaha Mikro, Kecil, dan Menengah adalah bentuk kemitraan waralaba. Sedangkan dalam hukum Islam sistem kemitraan perusahaan Grab dengan driver masuk dalam kategori syirkah ‘inan. Upah yang diterima driver dipotong 20% sebagai profit bagi perusahaan. Beberapa syarat tentang ma’qud ‘alaih (aset, usaha, dan profit) belum sepenuhnya sesuai dengan syarat musyarakah dan dalam beberapa praktik masih ditemukan unsur-unsur kemitraan yang dilarang dalam Islam seperti adanya gharar dan kecurangan.Kata Kunci: Sistem Kemitraan, Transportasi Online, Hukum Islam. AbstractThe Grab Company partnership with drivers in Law number 20 of 2008 concerning Micro, Small and Medium Enterprises is a form of franchise partnership. Whereas in Islamic law the Grab company partnership system with drivers is included in the syirkah ‘inan category. Wages received by drivers are cut by 20% as profit for the company. Some conditions regarding ma'qud ‘alaih (assets, business, and profit) aren’t yet fully in accordance with the requirements of musharaka and in some practices elements of partnership are still prohibited in Islam such as gharar and cheating.Keywords: Partnership Systems, Online Transportation, Islamic Law.

Abstract

ABSTRAKTanaman sirsak (Annona muricata L) merupakan salah satu tanaman berkhasiat yang dapat digunakan sebagai obat  kanker  dan  pengganti  kemoterapi.  Kanker  adalah  penyakit  berbahaya  yang menyerang DNA manusia. Annonaceous acetogenis atau yang lebih sering disebut dengan acetogenin yang terkandung dalam  daun  sirsak  dapat  membunuh  sel-sel  kanker  dengan  cara  menghambat  ATP  yang  menjadi sumber energi bagi pertumbuhan kanker. Senyawa fi tokimia yang menjadi anggota acetogenin seperti muricereacin  dan murihexocin C memiliki  kekuatan  yang melebihi  kefektifan dari adreamycin  (obat kemoterapi). Hasil  penelitian  menunjukkan  bahwa  senyawa  acetogenin  berperan  untuk  membunuh sel  kanker  dan derivat  senyawa acetoginin dapat  digunakan  sebagai pengganti  kemoterapi. Adanya acetoginin mempunyai aktivitas untuk membunuh sel kanker. Cancer is an insidious disease that attacks the human DNA. Annonaceous acetogenis or more often called acetogenin contained in soursop  leaves can kill cancer cells by  inhibiting ATP  the energy source  for  the growth of cancer. Phytochemical compounds  that are members acetogenin like muricereacin and murihexocin C has a power that exceeds the effectiveness of adreamycin (chemotherapy drugs). The result of research of soursop leaf to kill cancer cells and replacement of chemotherapy. That compounds may contribute acetogenin to kill cancer cells and derivate of acetoginin compounds can be used as a substitute for chemotherapy and have activity to kill cancer cells.Keywords: acetogenin, cancer, soursop leaves, chemotherapy

Abstract

Bacteriocin is a protein compound with a small molecular weight that has an antibacterial activity. Bacteriocin has been widely applied as a natural food preservative, as it effectively prevents the growth of pathogenic bacteria in foods and beverages. The aim of this research was to know the activity of bacteriocin produced by lactic acid bacteria Lactobacillus plantarum from Ce hun tiau beverages against Escherichia coli and Staphylococcus aureuswith different pH and temperature treatments. In this study, Lactobacillus plantarum bacteria isolated from Ce hun tiau beverages were characterized by Gram staining test. Bacteriocin was extracted from Lactobacillus plantarum bacteria which had been incubated in deMan Rogose Sharpe Broth media. Confirmation test using proteolytic enzyme and bacteriocin activity test with different pH (2, 4, 6, 8, and 10) and temperature (40°, 60°, 80°, 100°, and 121°C) treatments were conducted. The results showed that bacteriocin hadantibacterial activity against Escherichia coli but had no activity against Staphyloccus aureus. The inhibition zone was formed at pH 2 to 6 and at temperature range of 40°C to 121°C.

Abstract

Chronic complication of diabetes mellitus is diabetic foot ulcers. Diabetic foot ulcer can be defined as an open wound in the feet which will become infected as the result of high blood sugar levels that develops and become place of bacteria. One of the bacteria in diabetic foot ulcer is Escherichia coli. Imprecise use of antibiotics in the treatment of diabetic foot ulcers can cause antibiotic resistance to bacteria. This study aims to determine antibiotic sensitivity of Escherichia coli bacteria in diabetic foot ulcer Wagner grade III and IV. Samples of diabetic foot ulcer swab’s with Wagner grade III and IV were taken as Consecutive sampling. Identification of Escherichia coli bacteria is done by using a biochemical test and Gram stain test. Antibiotic sensitivity test is conducted by using Kirby Bauer’s disc of diffusion method. Escherichia coli bacteria that were tested are sensitive to amikacine, gentamicine, ciprofloxacin, levofloxacine, ceftriaxone, cefotaxime, imipenem and meropenem antibiotics but showed resistant to cefadroxil.

Abstract

Aloe vera is one of the typical plants found in Pontianak, West Kalimantan. This plant’s leaf peel is nontoxic and can be used as an antibacterial and anti-inflammatory agent. Nowadays, Aloe vera is one among 10 most popular plants in the world that have a potency to be developed as medicinal plant. This study aimed to determine antibacterial activity of the extract of Aloe vera leaf peel in liquid soap formulation against several pathogenic bacteria. Aloe vera leaf peel extract was prepared with maceration using ethanol. Then, it was formulated into a liquid soap. The soap was evaluated for its organoleptic, pH value, specific gravity, and foam height. Antibacterial activity assay of the liquid soap was carried out using diffusion method. The results showed that the characteristic of the liquid soap was viscous, yellowish white colored, and had distinctive smell of Aloe vera. The pH values on the day 0, 7 and 14 were 8; 8,9; and 9,4; respectively. The specific gravity was 1,033 g/ml. The foam height on minute 5, 10 and 15 were 76.92%, 19.23% and 19.23%, respectively. The liquid soap from the extract of Aloe vera leaf peel was found to have antibacterial activity against Gram positive bacteria (Staphylococcus aureus, Staphylococcus epidermidis, Bacillus subtilis, and Bacillus cereus) and Gram negative bacteria (Salmonella typhimurium, Proteus mirabilis, Pseudomonas aeruginosa, and Escherichia coli).

Abstract

The combined use of natural materials and antibiotic compound is one of treatment that can be done against infections caused by bacteria. This combination is expected to be more potent to inhibit the bacteria with lower side effects. FICI (Fractional Inhibitory Concentration Index) is an index that can indicate the activity of a combination of natural ingredients and antibiotic compounds. FICI would indicate that the combination has a synergistic effect, additive, indifferent or antagonistic. The aim of this study is to determine the FICI to the combination of Aloe vera leaves skin (Aloe vera (L.) Burm.f.) ethanol extract and gentamicin sulphate againts Escherichia coli. FICI was determined by disc diffusion method. Combination of Aloe vera leaves skin (Aloe vera(L.) Burm.f.) ethanol extract and gentamicin sulphate with concentration of 1.25 mg/mL + 2,5 µg/mL was not form zone of inhibition; while the concentration of 2.5 mg/ml + 5 µg/ mL formed zone of inhibition of 6.95 mm; 6.75 mm; and 6.65 mm. The results showed that the combination of Aloe vera leaves skin (Aloe vera (L.) Burm.f.) ethanol extract and gentamicin sulphate against the Escherichia coli has the indifference effect on Escherichia coli, and FICI to this combination is 2.

Abstract

Leaves agarwood (Aquilaria microcarpa Bail) is one of the natural materials that can be used as an antibacterial. This study aims to determine the content type of secondary metabolites and antibacterial activity against Staphylococcus aureus and Proteus mirabilis by the method of Kirby-Bauer disc diffusion. Simplicia leaves agarwood macerated with ethanol 96%. Results of phytochemical screening of ethanol extract of leaves agarwood contain flavonoids, phenols, tannins, saponins and steroids. Furthermore, the TLC for the assertion of their chemical compounds in extracts of leaves agarwood using a mobile phase of butanol: acetic acid: water (4: 1: 5) with the stationary phase silica gel GF254 with spotting FeCl3 5% for phenolic and AlCl3 5% for flavonoids later in UV detection at 254nm and 366nm. Control positive used is siprofloksasin 50 μg while control negative used is DMSO 20 % . Based on the results of the average diameter zone obstruent extract ethanol leaves agarwood in staphylococcus aureus by concentration of the 300 mg / ml, 400 mg / ml, 500 mg / ml is 12.50 mm, 13,51 mm, 15,80 mm. while in proteus mirabilis by concentration of the 300 mg / ml, 400 mg / ml, 500 mg / ml is 12,10 mm, 13,26 mm, 15.19 mm. This shows that extracts ethanol leaves agarwood having activity on Gram positive and Gram negative.
