Abstract

The COVID-19 pandemic caused by SARS-CoV-2 requires effective vaccines to be developed. This study aimed to assess the impact of a SARS-CoV-2 inactivated vaccine candidate in k18-hACE2 mice by monitoring their body weight, immune activation, and inflammatory cytokines including IL-4, IL-6, TNF-α, and IFN-γ. The study utilized k18-hACE2 mice expressing the human angiotensin-converting enzyme 2 (hACE2) receptor. The mice were administered the inactivated vaccine candidate compared with sham and vehicle. Body weight was monitored, and serum samples were collected to measure IL-4, IL-6, TNF-α, and IFN-γ levels using ELISA. Data were evaluated using SPSS statistical analysis software. The administration of the SARS-CoV-2 inactivated vaccine candidate in k18-hACE2 mice did not result in significant changes in body weight compared to the control group. Furthermore, the levels of IL-4, IL-6, TNF-α, and IFN-γ were significantly reduced in the vaccinated mice compared to the control group, suggesting a dampening effect on the inflammatory response. This study demonstrates that the SARS-CoV-2 inactivated vaccine candidate has a minimal impact on the body weight of k18-hACE2 mice. Nevertheless, it successfully regulates the levels of IL-4, IL-6, TNF-α, and IFN-γ, suggesting its safety and beneficial impact. These findings contribute to understanding the vaccine's efficacy and safety profile in vaccine development.

Abstract

The frequency of canary imports continues to increase every year. Antibiotic resistance is a global problem that threatens human and animal health worldwide. Human interaction with birds as pets is a public health concern because it has the potential to increase zoonotic diseases. This study was conducted to identify the antibiotic resistance of E. coli using fecal swab samples of imported canaries from Malaysia. Samples were grown on an EMBA medium for the isolation test, Gram staining test, and IMViC test were performed to continue the identification test -Kirby-Bauer diffusion test - to determine antibiotic sensitivity. Based on morphological culture, Gram staining, and biochemical tests, the sample examination results showed 18% (27/150) were positive for E. coli, 16% (24/150) showed the highest resistance to tetracycline, 15,33% (23/150) amoxicillin, 12,66% (19/150) trimethoprim-sulfamethoxazole, 6% (9/150) ciprofloxacin, and 14,66% (22/150) isolates were confirmed MDR because they were resistant to three to four antibiotics. Further efforts are needed to understand and address the factors that lead to antibiotic resistance in the context of animal and public health. Prudent management of antibiotic use and monitoring of antibiotic resistance needs to be improved to maintain animal health and prevent the risk of transmission of resistant bacteria to humans.

Abstract

This study aimed to determine the sex of Bangkok Zebra doves (Geopelia striata) based on external body morphometric measurements. A total of 32 doves were used, consisting of 16 females (8 bird each at 3 and 6 months of age) and 16 males (8 birds each at 3 and 6 months of age). Four morphometric parameters were measured: wing chord, antebrachium (forearm) length, longest primary feather, and longest tail feather. Wing and forearm lengths were measured using digital calipers with a precision of 0.01 mm, while the longest primary and tail feathers were measured using a ruler with 0.05 mm precision. The results showed that the average wing chord, longest primary feather, and longest tail feather lengths were significantly greater (p <0.05) in males compared to females. At six months of age, males also had a significantly longer forearm than females (p <0.05). However, no significant difference (p >0.05) was found in forearm length between sexes at three months of age. It could be concluded that wing and tail feather morphometrics can be used to distinguish between male and female Bangkok Zebra doves at both three and six months of age, except for forearm length at three months.

Abstract

Sex identification of Sun Conures (Aratinga solstitialis) is crucial for breeding and preservation, as well as increasing sun conure populations. These birds are sexually monomorphic. Therefore, Determination between male and female carried out by their morphology examination. The Polymerase Chain Reaction (PCR) method, utilizing molecular-based technology, was employed to determine the sex of Aratinga solstitialis in this study. The P2 and P8 primers were utilized in this method, which has been deemed suitable and accurate for sex identification through calamus samples. The research focused on two 28-month-old Aratinga solstitialis birds. Calamus samples were collected and subjected to PCR amplification using the extracted calamus. The resulting PCR products were then visualized using electrophoresis with a 1% agarose gel. In the electrophoresis photo, the presence of two bands indicated a female specimen, whereas a single band indicated a male specimen. The result of the gel electrophoresis research showed that both of the Aratinga solstitialis were male with one band of each bird on ranged from 300-400 base pairs. The result show that the Polymerase Chain Reaction method in terms for sex identification on monomorphic birds, especially Aratinga solstitialis birds is very effective to differentiate the sex of young birds and the adults.

Abstract

Cryptosporidiosis is a waterborne zoonosis caused by Cryptosporidium, which is a parasite that causes infection of the small intestine and leads to acute diarrhea in both humans and animals. Wild rats (Rattus spp.) are known to carry many zoonotic pathogens including Cryptosporidium. Given their close proximity to humans in urban environments, the likelihood of disease transmission is high. This study aims to detect the Cryptosporidium spp. infection in wild rats in Surabaya City, East Java Province. Intestinal fecal samples were collected using necropsy. The flotation test and Ziehl-Neelsen stain were used to observe the presence of Cryptosporidium under the microscope. The results showed that 69 samples were positive for Cryptosporidium spp. in the oocyst stage, which appeared dark pink with a clear cavity and the size of the oocysts ranged from 2-6 μm. The high incidence of cryptosporidiosis in wild rats is a potential health threat to both animals and humans.

Abstract

This study was conducted to determine the relationship between the estrous detector's reading value (Draminski®, DramiÅ„ski S.A, Poland) and serum progesterone levels on Etawa crossbred does. Eighteen healthynon-prehnant Etawa crossbreds does weighing 20-25 kg, in their first parity were used in this study. The does were divided equally into two groups. In the two groups, the does were synchronized their estrus by means of intramuscular and intravulvar injection of 7.5 mg prostaglandin FÉ‘ (PGF2É‘) twice in 11-day intervals. After the second injection, the estrous detector value measurement, visual observation of the estrus sign, and blood sampling were conducted for five days. The blood sample was processed into the serum to examine for progesterone levels by ELISA. The data analysis was using the simple correlation method on SPSS type 23 for windows. The result showed that a positive relationship (p <0.05) with coefficient correlation (r) was 0.609 between the values of the estrous detector measurement with serum progesterone levels. It could be concluded that the estrous detector's low value indicates that the Etawa crossbred was in the estrus phase and followed by the decreasing of progesterone level but not reaching the basal level.

Abstract

Predicting the turtle's reproductive status and sexual maturity is necessary for ex-situ breeding and conservation program. The histology of the seminiferous tubules of the Indonesian box turtle (Cuora amboinensis couro) at various ages has not been studied much. This study aims to develop equations to predict the sexual maturity of Indonesian box turtles based on straight carapace length (SCL). Six turtles with SCL sizes 10-21 cm were sacrificed and their testes were taken for morphometric measurements and histological preparations for Hematoxylin Eosin staining. Pearson's correlation of SCL with testicular maturity parameters was analyzed, followed by Anova Regression. The results showed that there were differences in the morphometry and topographic anatomy of the Indonesian box turtle testes between SCL below 15 cm and above 15 cm. Six turtles with SCL size of 10-21 cm were sacrificed and their testes were taken for morphometric measurements and histological preparations for Hematoxylin Eosin staining. Pearson's correlation of SCL with testicular maturity parameters was analyzed, followed by Regression Anova. Abstract

Second-degree burns are complex traumatic conditions requiring optimal tissue healing interventions. This experimental study investigated the impact of Gnetum gnemon L. leaf extract cream on collagen density in burn wounds. The research was conducted using 20 male Wistar rats (Rattus norvegicus), divided into five groups: negative control, positive control (silver sulfadiazine), and three treatment groups with varying leaf extract concentrations (2.5%, 5%, and 10%). Burn induction was performed on the right gluteal area using a thermostat at 85°C for 5 seconds. Collagen density was evaluated through histopathological analysis using ImageJ with density scores ranging from 0 to 4. Research results demonstrated significant collagen density increases in treatment groups, with the 10% concentration displaying the highest density (score +4). Significant differences between groups were confirmed by statistical analysis (p<0.05). Findings indicate the potential of Gnetum gnemon L. extract as an alternative therapy for improving burn wound healing through collagen density optimization.
