Abstract

Avian Influenza (AI) is a strategic communicable and zoonotic disease. The cause is a virus with Highly Pathogenic Avian Influenza (HPAI) subtype H5N1. The poultry market has important roles in the preservation, propagation, and spreads of the Avian Influenza (AI) virus from poultry to other species and humans. The purpose of this study was to determine the level of seroprevalence of Avian Influenza (H5N1) in free-range chickens at the Beringkit and Galiran market. A total of 120 free-range chickens were used as a sample. They have taken 60 serum samples per market. Serum removal is made from 5 merchants under the provisions of 3 samples from merchants who sell 6 to 10 free-range chickens. The sampling period is carried out for 2 months every 2 weeks 4 times. Sample testing was performed at the Denpasar Veterinary Centre with Haemagglutination (HA) and Haemagglutination Inhibition (HI) as barriers. The data titer of the antibodies obtained was analyzed by Non-Parametric Statistic Test Chi-Square (χ2) using IBM SPSS for windows. The results of the study showed that the AI subtype of the H5N1 subtype in both Beringkit Markets is 3.3% and Galiran Market is 6.7%, with seroprevalence in the two markets of 5.0% which is statistically not dissimilar (P<0.05). To prevent the transmissions of AI disease at the Beringkit Market and Galiran is recommended for vaccination and more attention to the market management and the free-range chicken maintenance system.

Abstract

Avian Influenza subtype H5N1 (AI-H5N1) is a malignant virus that is very detrimental to laying chickens because it is highly contagious and mutates easily. Prevention of AI-H5N1 disease in laying chickens is carried out by vaccination, therefore to maintain the quality of the vaccine, continuous research is needed. This study aims to determine the potential of AI-H5N1 vaccine isolates from Bali as measured based on cellular immune response based on total and differential leukocyte cells. Formation of antibodies is influenced by the nonspecific and specific immune system involving leukocytes, especially lymphocytes. Total of 40 layers of Novogen Brown strain were used for the research sample, kept since the age of one day on a commercial farm in Perean Village, Tabanan Regency, Bali. Laying chickens are vaccinated at 5 weeks of age by intramuscular injection. Total of 20 chikens were taken randomly and used for the sample. Blood draws were performed once pre-vaccination and five times each week after vaccination with anticoagulants. Total leukocytes were examined by an auto hematology analyzer, while differential leucocytes with thin blood smear stained with Giemsa. Total and differential leukocyte data were analyzed by means of the variance test followed by the Duncan test. Results showed that AI-H5N1 vaccination from Bali isolates could increase total and differential leucocytes of laying chickens and had significant effect on the mean total leukocytes, the absolute values of heterophyll cells, eosinophils, lymphocytes, and monocytes, but had no significant effect on post-vaccination basophil cells.

Abstract

ABSTRAK : Telah dilakukan penelitian isolasi dan identifikasi senyawa aktif yang dapat menurunkan kadar glukosa darah dari ekstrak etanol buah pare terhadap tikus putih jantan (Rattus novergicus) yang diinduksi aloksan. Ekstrak etanol buah pare dipartisi dengan n-heksana, kloroform, dan n-butanol. Berdasarkan hasil uji, fraksi n-heksana paling cepat menurunkan kadar glukosa darah pada hari ke-14. Fraksi n-heksana selanjutnya dipisahkan dengan kromatografi kolom menggunakan eluen n-heksana : etil asetat (6 : 4,5) diperoleh 5 fraksi (FA - FE). Berdasarkan hasil uji, fraksi A merupakan fraksi yang paling aktif menurunkan kadar glukosa darah dibandingkan fraksi lainnya (fraksi B, C, D, E). Fraksi A dapat menurunkan kadar glukosa darah pada hari ke-7. Hasil identifikasi isolat aktif (FA) menggunakan GC-MS menunjukkan adanya 10 senyawa yaitu  n-tetradekana, metil dodekanoat, metil heksadekanoat, etil heksadekanoat, metil-9,12oktadekadienoat, metil-9-oktadekenoat, metil oktadekanoat, etil oktadekanoat, metil-9cis-11trans-13trans oktadekatrienoat, dan mono (2-etilheksil)-1,2-benzenadikarboksilat.ABSTRACT : The aim of this research was to isolate and identify the active compounds from ethanol extract of bitter melon fruit to reduce the blood glucose levels of male mice (Rattus novergicus) which was alloxan induced. Partition of the ethanol extract using n-hexane, chloroform, and n-butanol was conducted and we found that the n-hexane fractions was the fastest to reduce blood glucose levels on day 14. The n-hexane fractions were then separated by coloumn chromatography using n-hexane: ethyl acetate (6: 4,5) as eluent and five fractions (FA - FE) were obtained. Based on the test result, fraction A was the most active fraction to reduce the blood glucose levels than other fractions (fractions B, C, D, E). The fraction A could reduce blood glucose levels on day 7. Identification of the active isolates (FA) was conducted by using GC-MS showed 10 compounds which were n-tetradecane, methyl dodecanoate, methyl hexadecanoate, ethyl hexadecanoate, methyl-9,12octadecadienoate, methyl-9-octadecenoate, octadecanoic acid methyl ester, octadecanoic acid ethyl ester, methyl-9cis-11trans-13trans octadecatrienoate, and 1,2-benzenadikarboxylic acid mono (2-ethylhexyl) ester.

Abstract

Chitosan is the modification of chitin which found on the outer skin of Crustacea species such as shrimps and crabs. This study aims to isolate chitin, syntesize and charactetize chitosan from shrimp shells. Chitin isolation stage included demineralization using HCL 1.5M and deproteination step with NaOH 3.5%. Transformation of chitin into chitosan was done through the deacetylation reaction by using NaOH 60%. The result showed that characteristic of the chitosan obtained in this research was as follows: the yield of transformation chitin into chitosan was 67.08%; it had a white color powder texture with had no smell; water content 1.55%; It solubled in 2 % acetic acid with 84.85 % deacetylation.

Abstract

High rate of population growth causes population problems to be huge problems that should be solved; ifdon’t’ there will be “Population Bomb”( Paul R. Ehrlich,1968). Factors affecting the rate of population growth afterconducting analysis with quantitative and qualitative descriptive research approach and statistical analysis and publicregulation theory by Edward III; in which the research object is in all Regencies/City in Bali. The rate of populationgrowth determined by the change of population dynamics as well as: fertility, mortality, and migration in BaliProvince. The rate of population growth is much higher than the national rate of population growth. Although theacquired bonus demography, in which Total Fertility Rate (TFR) has been decreasing from 2,5 to be 2,3 per womenhowever the Mortality Rate is still above one percent and migration rate level in the area of Sarbagita is very higheven reaches more than 4 percent. The effort of controlling the population through Keluarga Berencana (FamilyPlanning) is still stagnant, as the consequences of the weakness of the government regulation implementation.

Abstract

Live Bird Market (LBM) has a high potential for spreading Avian Influenza Virus (AIV) between fowls or from fowl to human. Up to now, a dinamic of avian flue incidents at many LBMs in Bali has not been reported. This research aimed to reveal a dynamic of seroprevalences of avian influenza in ducks at Beringkit (Badung) and Galiran (Kelungkung) LBMs. A total of 35 duck blood samples was collected from each of LBMs. Sampling was conducted monthly from March to August, 2012 . AIV antibody of duck serum was measured using Rapid Hemagglutination Inhibition (Rapid HI) test. Seroprevalence differences were analyzes with Chi-square (?2) Nonparametric statistical test. The results showed that seroprevalences of AIV H5 in ducks at Beringkit and Galiran LBMs were very high, ranged from 68.6% to 100% and 65.7% to 97.1% respectively. A Dynamic of AIV H5 seroprevalences in ducks at Beringkit and Galiran LBM had a similar pattern, except in July 2012. This indicates that VAI H5 has been circulating for a long time and has been to be an endemic virus infection in ducks at LBMs in Bali. It can be suggested that an Avian Influenza Virus monitoring should be done continuously over a long period.

Abstract

Newcastle Disease (ND) is one of the poultry diseases that can infect domestic chickens. It is an infectious disease that has a detrimental impact on chicken farmers. ND infection can be acute to chronic, and it can also affect all types of poultry, especially chickens, both broiler and domestic chickens. Tabanan Subdistrict is a location where poultry trade activities are often carried out between regions and/ or provinces, so domestic chickens have the potential to be exposed to ND. This study aims to determine the disease seroprevalence in domestic chickens in Tabanan District, Tabanan Regency, Bali. The sample of this study was serum from chicken’s blood samples that were taken from six villages in Tabanan Sub-district. Samples were taken from villages where each village consisted of three banjars. Banjar is a division of Bali administrative areas, and eight samples of domestic chickens were collected from each sampled banjar. A total sample of 144 serums of non-vaccinated domestic chicken. Serum was tested serologically with hemagglutination (HI) test at the Virology Laboratory of the Faculty of Veterinary Medicine, Udayana University. The results showed that ND seroprevalence in domestic chickens in Tabanan Subdistrict was 3.17%. Positive samples were gathered from Gubug, Bongan, and Wanasari villages with antibody titers ranged from 24 to 25. The conclusions of seropositive results showed that domestic chicken in the Tabanan district has currently been infected with the Newcastle Disease virus.

Abstract

This study was conducted to determine the primary immune response post vaccination using EDS inactivated vaccine polyvalent. The sample used was a commercial layer farm in the village of Tiga, regency of Bangli, Bali. A total of 25 layer which were14 weeks old vaccinated using EDS-76 inactivated vaccine containing polyvalent Newcastle disease antigen virus, infectious bronchitis and egg drop syndrome by intramuscularly injection. Examination of EDS antibody titer using serologic test by Hemagglutination Inhibition (HI) test. Egg drops syndrome antibody titer checked four times, once before vaccination, and every week for three weeks post-vaccination to see the immune responses. The average antibody titer then analyzed using an univariate of variance test followed by a test of Least Significant Difference, Duncan test and regression analysis. The result showed an increase antibody of EDS was significantly every week post vaccination. The average antibody titers of EDS are 22,6 HI unit at one weeks post vaccination, about 25,04 HI unit at two weeks post vaccination and 26,4 HI unit at three weeks post vaccination.

Abstract

This study aims were to inventory the profile of bali calf praesen status. Praesen status examination includes body temperature, respiration, heartbeat, and arterial pulse. This study used 15 bali calf with the age group of four months as much as five, aged five months as much as five,and aged six months as much as five. Praesen status measurement performed in the morning,afternoon, and evening. The results of the research praesen status of bali calf held in Bali Cattle Breeding Center, Sobangan, Mengwi, Badung is the average body temperature of calves aged fourmonths 38.80 ± 0.20°C, aged five months of 38.60 ± 0.53°C, aged six months 38.47 ± 0.42°C. The average frequency of respiration calf aged four months 45.60 ± 3.27 times/min, aged five months31.67 ± 5.31 times/min, aged six months 37.93 ± 10.25 times/min. The average frequency of arterial pulse calf aged four months 93.40 ± 5.55 times/min, aged five months 82.80 ± 2.55 times/min, aged six months 82.40±8.49 times/min. The average frequency of the heartbeat calfaged four months 91.73 ± 4.39 times/min, aged five months 83.20 ± 3.50 times/min, aged sixmonths 83.40 ± 8.86 times/min. Based on the research results bali calf (4-6 months) it can be concluded that: body temperature was 38.62±0.17°C; respiration frequency was 38.40 ± 6.98 times/min; arterial pulse frequency was 86.20±6.24 times/min; and heart beat frequency was 86.11± 4.87 times/min. Praesen status is expected to be used as a reference for bali calf health examination.

Abstract

Conjunctivitis is inflammation of the conjunctiva. The case of a dog named Ve is the breed Pug dogs 10 months of age, having an eye disorder. Ve has conjunctivitis in the eye of the dexter and sinister part. Ve was reported to have conjunctivitis at 3 months of age. Based on the results of the physical examination of the red eye mucosa, in the eyes sinister the eyeball is not visible (atrophy), which is accompanied by mucopurulent exudate discharge, the part eye dexter eyes looks cloudy, accompanied by mucopurulent exudate and hyperpigmentation of the skin due to Demodex sp. Based on identification and examination, conjunctivitis suffered Ve is caused by trauma accompanied by secondary bacterial infection. From the results of routine hematology examination, it shows lymphocytosis, eosinophilia and thrombocytopenia. Eye therapy was given in the form of Erlamycetin® eye ointment antibiotics and Calvidog® vitamin, while the skin was given dipenhidramine HCl injection and invermectin injection.
