Abstract

In everyday life, queues often occur. Waiting at the counter to get train or movie tickets, at the toll gate, at the bank, at the supermarket, and in other situations that we often encounter Queues occur when the need for services exceeds the capacity or capacity of the service facility. As a result, users of the facility cannot get immediate service due to the busyness of the service. The Amplas Toll Gate queue is the object of this research. The Amplas Toll Gate is one of the densest toll gates that is heavily traveled by vehicles both entering and exiting. This makes it often seen a fairly long queue, especially during peak hours in the late afternoon to evening. The Medan City Government built an off ramp at the Amplas flyover in 2016. This off ramp leads directly to the Amplas toll gate. The vehicle arrival rate increases along with the queue length because vehicles can arrive faster to the toll gate. This study aims to calculate the queue length at the Amplas toll gate before and after the construction of the ramp off. Data is obtained by recording the volume of vehicles at the research location. With an average service time of 7 seconds, the queuing method produces a queue length of 11.98 meters, while the results using Pro Model software are 11.98 meters. In addition, the queue length after the construction of the ramp off decreased to 6.67 meters from before the construction of the ramp off. Promodel is a windows-based simulation software used to simulate and analyze a system.

Abstract

Studies in transgenic sugarcane (Saccharum officinarum L.) demonstrated that sucrose transporter (SUT) genes were essential in sucrose translocations. Sucrose transporter gene isolated from sugarcane were designated as SoSUT1 and SoSOSUT2 respectively as encode protein of 518 and 747 amino acids. The genes were constructed into plasmid pYES2 for SoSUT1, and pYX112 for SoSOSUT2. cDNA SoSOSUT2 had also constructed into plasmid pBIN-At-GFP which was possible to trace the gene inserted. The constructed plasmid was transformed into yeast (Saccharomyces cerevisiae) and grown in minimal medium (SD–urasil) as selection medium. The transformed colony was confirmed using PCR. The functional expression was studied by growing yeast in YPD medium with 2% Sucrose, then the sucrose uptake was measured in number interval time using resorcinol method. The result showed that yeast INVSc1-pYES2-SoSUT1 and BF264- pYX112-SoSOSUT2 had higher ability in sucrose transport compared to the control-INVSc1 for SoSUT1 and control-BF264 for SoUT2. Moreover, the result showed that SoSUT1 had higher ability to transport sucrose than SoSOSUT2. Confocal microscope observation showed that transformation gene SoSOSUT2 was successful, which was indicated by green exposure of GFP protein.Keyword: sugarcane, sucrose transporter, yeast

Abstract

This study explores the impact of visual marketing elements on consumer purchasing behavior within the fashion e-commerce industry, focusing on visual aesthetics, user-generated content (UGC), and influencer endorsements. Using a qualitative approach, semi-structured interviews were conducted with 25 participants to capture in-depth insights into how visual cues shape consumer perceptions, build trust, and influence purchase intentions. Thematic analysis of the data revealed three main findings: high-quality visuals evoke emotional responses that increase product appeal, UGC serves as a crucial factor in establishing trust and reducing purchase hesitation, and influencer endorsements significantly impact consumer decision-making by adding perceived value and trend relevance. These findings contribute to existing theories, such as the Stimulus-Organism-Response (S-O-R) model, by illustrating the diverse ways in which visual stimuli influence consumer behavior on emotional, social, and practical levels. Managerial implications include the strategic use of high-quality visuals, encouragement of UGC, and selection of authentic influencers to enhance consumer engagement and conversion. This study provides a comprehensive understanding of how visual marketing shapes consumer behavior in fashion e-commerce, offering valuable insights for both academic research and practical applications in the digital marketplace.

Abstract

Identifying pathogenic fungi at the species level based on morphology, especially for leaf rust, is quite difficult. Further investigation, such as molecular techniques, is required to identify leaf rust fungi precisely. One way that can be employed is DNA barcoding. This study seeks to detect leaf rust fungi (Puccinia spp.) on sugarcane. Accurate species identification allows early detection of Puccinia kuehnii, helping farmers and extension agencies to apply targeted control strategies. A fungal sample was collected from sugarcane fields in Bondowoso, East Java, Indonesia. Morphological identification was done with an SEM, whereas molecular identification was done through DNA extraction, amplification, visualization, sequencing, and phylogenetic tree creation. The primers employed are obtained from the ITS region of ribosomal DNA, which contains enough diversity to differentiate fungi at the species level. The amplification findings revealed a DNA band of 500 bp at both loci. Sequencing results indicate that samples The sequencing results indicatethat the samples from Bondowoso are closely related to the Puccinia kuehnii sequence from NCBI. The findings of the phylogenetic tree construction revealed that samples from both locations are still associated with P. kuehnii sequences from other nations. This is the first study documenting P. kuehnii in Indonesia, especially East Java, that integrates morphological and molecular characterization.

Abstract

The article seeks to elicit some current tendencies that have made science, philosophy and religion in the third millennium change their self-perception, hence a paradigm shift. The hermeneutic point of view, the bias of gender, the negative impacts, the critique of the ideological presupposition behind each field, along with their respective internal evolution, have all put science, philosophy and religion into a redefinition and recontextualization. By now science has recognized the limits of its own logic and language, and seems to see the possibility of different kinds of ‘logic’ used by other types of knowledge. Philosophy, after Heideggerian phenomenology, has seemed to lose the possibility of creating an ‘all encompassing’ system of thought, and instead has to be more modest in simply identifying hermeneutically various modes of knowledge and their interconnection with pre-reflective modes of being, as well as in uncovering the root of today’s problems of life and humanity. As for religion, to revitalize itself today it has to shift its basic orientation, from doctrine and its propositional truth-claim to the complexity of religious experience, from ‘what one believes’ to ‘what one does’. Keywords :*Flux, *Web, *Relasional, *Ideologis, *Logika, *Lebenswelt, *hermeneutik, *narasi besar, *pra-reflektif, *doktrin, *pengalaman, *religion without religion

Abstract

Intercultural encounter  is something inevitable and crucial today. Its significance for religions depends on how religions conceive of intercultural translatability  and the meaning of ‘the other’. Concerning the former, there are three possibilities : different cultures can be seen as radically untranslatable, mutually translatable in terms of universal economic medium, or  mutually translatable in terms of  universal doctrinal message. Each brings its own consequences.  Concerning the latter, the other may be viewed  as the outer-other or the inner-other of which both require some kind of self-relativization on the part of religion. If  Christianity is consistent with its ‘logic of love’, it would be governed by heteronomous reason in which the self lives from out of itself , whereas its dwelling place is not the privileged centre.

Abstract

For the Javanese the whole life is a manifestation of the Absolute. Therefore it is not possible to separate the sacred from the profane, neither morality from religiosity, nor epistemology from ethics.Knowledge, as the pursuit of the truth, is not viewed as in empiricaltradition in which the mind perceives objects out there through thecreation of representations of them by the nervous systems andregistered by the brain (Representationalism); neither as inCartesian intellectualism in which knowledge is founded onindubitable certainties (Foundationalism). It is, instead, a matter ofdoing and venturing (Laku) in which one is to go deeper: fromgross physical body, through subtle body, to the sublime soul. Thecentre of gravity of this pursuit is rasa ( inner feeling, ultimatesecret, the bearer of the divine life, the vehicle of life). Rasa is thequintessence of all the three parts of human structure : the head, the chest and the genital ( the scrotum).
