Abstract

High levels of fat in the blood are a risk factor for nonalcoholic steatohepatitis liver disease. Indonesian medicinal plants that can decrease blood fat levels are turmeric and mangosteen peel. One of the mechanisms of blood fat-lowering drugs is to increase the expression of the PPARα gene. The purpose of this study was to assess the effect of turmeric and mangosteen peel on the expression of the PPARα gene in the rat liver induced by a high-fat diet. This research was conducted at Maranatha Biomedical Research Laboratory in February–October 2021, using male Wistar rats that were divided into 5 groups (n=5): negative control groups (no treatment), positive control groups (high-fat diet), turmeric, mangosteen, and fenofibrate groups, that was given high-fat diet continued by ethanol extract of turmeric, ethanol extract of mangosteen peel, and fenofibrate. At the end of the study, the animals were terminated, and the liver was extracted for RNA extraction and semi-quantitative PCR. The results showed that there was an increase in PPARα gene expression in the turmeric group and fenofibrate group, which were significantly different from the positive control group that received a high-fat diet (p<0.05) and between the fenofibrate group compared to negative controls that received standard chow diet (p<0.05). In conclusion, turmeric and fenofibrate are suggested to increase the expression of the PPARα gene in the liver induced by a high-fat diet.

Abstract

Vitamin D has a broad impact on the human body, including affecting the bones and the immune system. Vitamin D deficiency in pregnant women is a risk factor in several circumstances, such as preeclampsia, gestational diabetes, premature birth, and low birth weight babies. This study aimed to determine the vitamin D levels and their relationship to predisposing factors and estimated fetal weight in the third trimester of pregnancy. Eighteen pregnant women in their third trimester of pregnancy who received antenatal care in the Obstetric Clinic of Immanuel Hospital Bandung from January to December 2022 were used as subjects of this study. The inclusion criteria are third-trimester pregnant women, healthy, while the exclusion criteria are currently taking drugs that have side effects in pregnancy or having pregnancy disorders. Blood was taken to measure vitamin D levels, fetal weight was estimated with obstetric ultrasound, and the subjects filled in questionnaires about predisposing factors. The results showed ten pregnant women (56%) had vitamin D deficiency below 20 ng/mL in their third-trimester pregnancy. We found non-significant relationships (p>0.05) between predisposing factors (daily consumption of vitamin D, sun exposure, maternal age), estimated fetal weight, and vitamin D levels. This study concludes that 56% of pregnant women in their third trimester of pregnancy have vitamin D deficiency with no significant relationship with its predisposing factors and estimated fetal weight.

Abstract

Changing learning patterns from senior high school student to college student status is difficult, and each individual's response varies, some of which cause stress. Factors that cause stress are internal and external, and external factors consist of physical, conflict, emotional, and behavioral conditions. In comparison, the external factors consist of the physical environment, work environment, community environment, family environment, economic and legal problems. The burden of life stress is heavy and impacts the physical, such as feeling dizzy, nauseous, ulcers, and difficulty sleeping. Sometimes causes a person to take self-medication to overcome these uncomfortable symptoms. This study aims to determine the characteristics of early-level students and the stress level of early-level students at one of the State Universities of East Kalimantan Province. This study used a qualitative descriptive design on 121 students. Using a questionnaire through a cross-sectional approach with a purposive sampling technique. The results showed that the stress level of students showed mild stress as much as 30.58%, moderate stress at 56.20%, and severe stress as much as 13.22%. The number of students who did self-medication was more than those who did not provide treatment for the symptoms, 79.34%.

Abstract

Dysoxylum alliaceum belongs to the Dysoxylum genus (Meliaceae) and there are few reports concerning the phytochemical components of this plant. To examine the chemical constituents of Dysoxylum alliaceum stem bark, a phytochemical study has been conducted and identified five known steroids, 3β,16β-dihydroxy-24(S)-methyl cholestenol (1), ergosta-5,22-dien-3β-ol (2), ergosta-7,24(28)-dien-3β-ol (3), 22(E)-ergosta-6,22-dien-3β,5α,8α-triol (4), and 20α-dihydroprogesterone (5) from its ethanolic extract. Spectroscopic data such as FT-IR, HR-ESI-MS, 1D, and 2D NMR as well as comparison with previously published spectral data were used to identify the chemical structures of compounds 1–5. Furthermore, these steroids 1-5 were assessed in vitro regarding their cytotoxic effect against A549 lung cancer cell lines and revealed weak to inactive with IC50 values ranging from 68.52 to >150 µM. Abstract

ABSTRACTIntroduction: Periodontitis treatment includes scaling and root planing, which can be supplemented with additional therapy, such as the administration of propolis. Propolis, a potential local natural resource, contains active metabolites that act as anti-inflammatory agents in periodontal healing. This study aims to analyze the effectiveness of propolis extract gel from Kelulut (stingless bees) as an anti-inflammatory agent in periodontal treatment. Methods: This laboratory-based experimental research employed a post-test-only control group design, divided into five groups: 50%; 60%; 70% propolis extract gel; Metronidazole Gel and Placebo Gel. The anti-inflammatory test was conducted using 50 male Wistar rats induced with periodontitis through silk ligature and P.gingivalis bacteria. The rats were sacrificed on the third and seventh days after treatment, and macrophage cells count were performed under a microscope at 400x magnification using Haematoxylin-Eosin staining. Results: The ANOVA test results showed no significant differences in the macrophage cells counts among the treatment groups on day 3 and day 7 in the periodontitis-induced rat model (p>0.05). The anti-inflammatory test results indicated a decrease in the mean macrophage cells count in each group by day 7. Conclusion: The use of Kelulut propolis extract gel from Kabanjahe was effective in reducing the number of macrophages in periodontitis-induced rats, with the 70% propolis extract gel demonstrating the best anti-inflammatory effectiveness in periodontal treatment. Potensi gel ekstrak propolis lebah kelulut kabanjahe sebagai agen anti inflamasi pada perawatan periodontal: eksperimental laboratoris Pendahuluan: Perawatan periodontitis meliputi scaling dan root planing serta dapat didukung dengan terapi tambahan berupa pemberian propolis. Propolis merupakan salah satu sumber daya alam lokal yang potensial, mengandung metabolit aktif yang berperan sebagai zat anti-inflamasi dalam penyembuhan periodontal. Penelitian ini bertujuan untuk menganalisis efektivitas gel ekstrak propolis dari lebah Kelulut (stingless bees) sebagai anti-inflamasi pada perawatan periodontal. Metode: Penelitian eksperimental laboratoris ini memiliki desain post-test-only control group design yang dibagi ke dalam lima kelompok yaitu kelompok gel ekstrak propolis 50%; 60%; 70%, gel Metronidazole dan gel Placebo. Pengujian anti-inflamasi menggunakan 50 ekor tikus wistar jantan yang diinduksi periodontitis dengan ligatur Silk 3/0 dan bakteri P.gingivalis. Tikus dikorbankan pada hari ketiga dan hari ketujuh setelah perlakuan, kemudian dilakukan penghitungan sel makrofag dengan mikroskop pada perbesaran 400x dengan pewarnaan Haematoxylin-Eosin. Hasil: Hasil uji ANOVA, tidak terdapat perbedaan bermakna pada setiap kelompok perlakuan terhadap jumlah makrofag. sel pada hari ke-3 dan hari ke-7 pada model tikus periodontitis (p<0.05). Hasil uji anti-inflamasi menunjukkan adanya penurunan rerata jumlah sel makrofag pada masing-masing kelompok pada hari ke-7. Simpulan: Terdapat efektivitas penggunaan gel ekstrak propolis lebah Kelulut Kabanjahe terhadap jumlah makrofag pada tikus yang diinduksi periodontitis dan gel ekstrak propolis 70% memiliki efektivitas terbaik sebagai anti-inflamasi pada perawatan periodontal.

Abstract

Selenium is a trace element that has antioxidant properties and is a component of selenoproteins. In certain circumstances, such as injury, selenium supplementation is required to regulate the inflammatory response. Contusion injury increases the inflammatory response to a muscle or tissue that begins with vasodilation, increases blood flow, increases vascular permeability, and leads to edema formation at the site of injury. Objectives: To determine the effect of selenium supplementation on changes in blood markers in rats treated with contusion injury. Methods: Experimental design, using experimental male rats (Rattus norwegius), eight weeks old, weighing 200-220 grams, divided into three groups, namely: The Control group, the contusion group, the contusion + Selenium group. Selenium supplementation was given orally on days 1-3 after injury at a dose of 0.0153 mg into 2% PGA. Examination of markers such as White Blood cells (WBC), lymphocytes, monocytes, and neutrophils was carried out on day three after treatment. Results: This study found that the administration of post-muscular selenium supplementation based on examination of blood markers such as WBC, Lymphocytes, Monocytes was higher in the contusion group and lower in the selenium group. Neutrophils were higher in the contusion and selenium groups. This was not significantly different from the control group. The result of blood marker examination tended to change, but it was not statistically significant. Conclusion: Selenium supplementation in post-contusive injury mice did not provide significant changes in blood markers such as WBC, lymphocytes, monocytes, and neutrophils.Keywords:  antioxidant, blood marker, contusion, selenium

Abstract

The immunosurveillance system in the human body can detect and eradicate abnormal cells. However, cancer cells can avoid immune destruction by exploiting the immunosuppression system. Extensive research has been done to find therapeutic agents that could reverse this condition. Novel drugs called checkpoint inhibitors have been developed and showed a promising result. Besides, “living’’ drug, an engineered T-cells which can accurately target cancer cells, has increased public attention in immunotherapy. These immunotherapies might bring a new era of cancer management, a highly effective and specific treatment with fewer adverse effects. Abstract

A long-term high-fat diet (HFD) was proven to induce metabolic dysfunction and causes various organ inflammation, including the retina. As the regulator of lipid metabolism, PPARa plays a role in retinal lipid metabolism and served as one of the targets for decreasing lipid deposition in the retina. Turmeric and mangosteen peel are Indonesian medicinal herbs with enormous health effects, including antiinflammation and hypolipidemic properties. This study aims to determine the effect of ethanol extract from turmeric and mangosteen peel on PPARa  gene expression in the retina of an HFD-induced rat model. Twenty male Wistar rats were divided into 5 groups: negative control, positive control, turmeric, mangosteen, and fibrate. At the end of the study, total cholesterol and triglyceride levels from the blood were measured. The retina was extracted to conduct Realtime PCR for PPARα gene expression. The result showed a significant difference in triglyceride levels between positive control and turmeric groups, and PPARa gene expression in the retina between the control negative, positive, and turmeric groups, but no significant difference was found in other groups. This study concludes that the extract of turmeric increases the expression of the PPARa gene expression in the retina in an HFD-induced rat model.

Abstract

Abstract: Pregnancy is a critical period involving significant physiological and psychological changes, particularly in primigravida women who are more susceptible to anxiety and stress. Elevated stress levels during pregnancy may increase the risk of complications like high blood pressure, preeclampsia, and premature delivery. Non-pharmacological interventions like Hatha Yoga and gymnastic prenatalhave been widely recommended to improve maternal well-being. The purpose of this research was to evaluate and compare the impacts of two different interventions on maternal psychological and physiological health through a Systematic Literature Review (SLR) conducted in accordance with PRISMA guidelines. Relevant literature was retrieved from databases including Pubmed, Scopus, ScinceDirect, Google Scholar, focusing on research published between 2014 and 2024. The findings indicate that Hatha Yoga is more effective in reducing anxiety, cortisol levels, and systolic blood pressure, while prenatal gymnastic better supported diastolic blood pressure regulation and physical endurance for labor. Both interventions contribute to better cardiovascular function, increased oxygen saturation, and a smoother labor process. Additionally, yoga is associated with improved neonatal outcomes, while studies on prenatal exercise’s effects on newborn health remain limited. Future research should explore the long-term effects and key differences between these interventions to determine the most effective approach for maternal and neonatal health. These findings support integrating yoga and gymnastic prenatalinto antenatal care programs to enhance pregnancy outcomes.

Abstract

Background Protein is essential for skin integrity, while exercise modulates cellular and molecular processes that regulate autophagy, mitochondrial biogenesis, and collagen synthesis. This study evaluates the combined effects of dietary protein restriction and varying exercise intensities on epidermal thickness, autophagy markers (LC3, p62), mitochondrial biogenesis (PGC1α), and collagen synthesis (Col1a1, Col3a1) in male Wistar rats.    Methods An experimental laboratory study was conducted involving 32 male Wistar rats. They were randomized into 8 groups: control, low protein, low intensity, group IV, moderate intensity, moderate intensity low protein, high intensity, and high intensity low protein. Low protein diet (5%) compared to the control (24%) was given for 60 days, while the exercise regimen was eight weeks of treadmill exercise (low/10 m per minute, moderate/20 m per minute, and high intensity/30 m per minute) for 30 minutes a day, five days a week. Epidermal thickness, LC3, p62, PGC1α, Col1a1, and Col3a1 mRNA expression were measured at the end of the study. One-way ANOVA/Kruskal-Wallis followed by Tukey HSD/Mann–Whitney test was used to analyze the data.   Results A low-protein diet significantly reduced epidermal thickness (p=0.048), increased LC3 (p=0.043) and p62 (p=0.011), indicating impaired skin regeneration and autophagy. However, exercise, especially at high intensities with low protein, mitigated this effect by increasing both LC3 (p=0.011) and PGC1α (p=0.011).   Conclusions These findings highlight the compensatory role of exercise in maintaining tissue integrity under protein-deprived conditions in male rats. High-intensity exercise potentially enhances autophagy and mitochondrial biogenesis.
