Abstract

Moderate and high aerobic exercises are the activities that can induce high oxidant. Demandof endogen and exogen antioxidant for neutralizing oxidant induced by exercise will playimportant role in maintaining body performance. Microtrace elements, such as selenium,may able to balance high oxidant formation. However, limited research and data have beencollected with regards to the exercise and selenium. Therefore, this research was conductedto investigate the effect of aerobic exercise on selenium level in plasma. An analytic studywith experimental method of data collection was performed from October -November 2016.17 male students with the age between 19-25 years old and without medically compromiseddisease from Universitas Padjadjaran, Jatinangor, were selected as subjects. Blood sampleswere taken pre- and post-aerobic exercise, i.e., brisk walking with the intensity of 4.5 mi/h.Selenium level in plasma was then examined by enzyme linked immunosorbent assay (ELISA)kit as selenium binding protein (SELENBP1). This study found that 11 (65%) subjectshad a decrease in their selenium plasma after exercise (3.1 ± 2.58 ng/ml), 5 (29%) subjectshad an increase in their selenium plasma after exercise (2.08 ± 2.8 ng/ml) and 1 (6%) subjecthad unchanged selenium (SELENBP1) plasma (1.4 ± 1.4 ng/ml) level. Abstract

Background: Chronic Ultraviolet B (UVB) exposure causes oxidative stress that may induce damages to the collagen matrix and thus plays a role in the wrinkle formation. Lactobacillus reuteri is a probiotic that may exerts antioxidant effects, thus helping to reduce damages caused by UVB-induced oxidative stress in the skin.Materials and Methods: Twenty-eight male Balb/c mice were divided equally into control group, UVB radiation only group, oral L. reuteri supplementation only group, and UVB radiation with oral L. reuteri supplementation group. UVB irradiation was given 3 times a week (100 seconds/exposure, within 3 cm distance) for 10 weeks, with a total dose of 166 mJ/cm2. Oral L. reuteri supplementation (0.2 mL, 108 CFU) was provided every morning after meal via orogastric feeding tube for 10 weeks. Wrinkle formation on the dorsal skin of the mice was evaluated in accordance with the Bissett method and type I procollagen levels was evaluated by western blotting.Results: In comparison with the group receiving only UVB irradiation, the group receiving probiotic and UVB irradiation showed significantly lower wrinkle scores (Group 1 vs. Group 3, 2.50±0.55 vs. 1.00±0,00; p<0.05) and significantly higher type I procollagen levels (Group 1 vs. Group 3, 0.88±0.36 vs. 1.92±0.46; p<0.05).Conclusion: Results of the current study showed that L. reuteri supplementation may reduce wrinkle formation and increase type I procollagen production in UVB-exposed dorsal skin of male Balb/c mice.Keywords: Lactobacillus reuteri, type I procollagen, photoaging, wrinkles, ultraviolet B

Abstract

Autofagi merupakan proses adaptasi yang dilakukan sebagai pertahanan dalam respon seluler, seperti kekurangan nutrisi atau stress metabolik lain. Mekanisme autofagi diregulasi oleh protein yang dinamakan Autophagy-Related Genes (ATG). Autofagi juga telah banyak dikaitkan dengan berbagai penyakit pada manusia, misalnya kanker atau penyakit degeneratif lainnya. Ricin merupakan protein toksik yang berasal dari biji jarak Ricinus communis L. dan banyak dieksplorasi untuk aktivitas antikanker melalui jalur pensinyalan caspase (apoptosis), namun belum ada penelitian pada jalur autofagi. Penelitian ini dilakukan untuk menelaah mode ikatan yang terjadi antara ricin-A dan protein-protein yang berperan pada setiap tahap proses autofagi (Beclin-1, LC3 atau Light Chain 3, dan p62/Sequistrosome1). Metode yang digunakan adalah simulasi penambatan protein-protein menggunakan server online ClusPro (https://cluspro.org). Hasil penelitian menunjukkan bahwa ricin-A dapat berinteraksi dengan Beclin-1. LC3, dan p62 melalui pembentukan ikatan hidrogen dengan afinitas baik. Dapat disimpulkan bahwa ricin-A berperan penting dalam proses autofagi dan dapat dikembangkan menjadi fitofarmaka terapi kanker.

Abstract

Jumlah kepemilikan hewan di Indonesia terus tumbuh dan berasal dari berbagai kalangan, termasuk dari kalangan mahasiswa jenjang sarjana yang memutuskan untuk memelihara kucing. Hal ini dilakukan untuk meredakan stres dan menjadi pengalih perhatian semua masalah yang sedang dialami pada usia tersebut. Naiknya jumlah kepemilikan hewan ini, harus diikuti dengan pemahaman dan pengetahuan mengenai kesejahteraan hewan untuk menghindari terjadinya penelantaran hewan dan kekerasan fisik serta psikis yang dilakukan terhadap hewan peliharaan. Penelitian ini bertujuan mengetahui tingkat pengetahuan dan penerapan kesejahteraan oleh mahasiswa Universitas Padjadjaran, serta menganalisis hubungan karakteristik mahasiswa (jenis kelamin, fakultas, tingkat pendidikan, jumlah hewan peliharaan, dan tujuan pemeliharaan) terhadap pengetahuan dan penerapan kesejahteraan hewan. Data yang terkumpul kemudian dianalisis menggunakan beberapa variabel, antara lain: jenis kelamin, fakultas, tingkat pendidikan, jumlah hewan peliharaan, dan tujuan pemeliharaan. Penelitian ini bersifat deskriptif kuantitatif dengan melakukan survei kuesioner dan wawancara terhadap 100 responden yang merupakan mahasiswa aktif Universitas Padjadjaran dan memiliki hewan peliharaan kucing. Hasil penelitian menunjukkan bahwa terdapat hubungan yang signifikan antara tingkat pengetahuan kesejahteraan hewan dengan penerapan konsep kesejahteraan hewan dengan hasil uji chi square <0,05. Di sisi lain, tidak terdapat hubungan yang signifikan antara variabel yang diteliti terhadap tingkat pengetahuan kesejahteraan hewan dan penerapan kesejahteraan hewan.

Abstract

Abstract: The athlete's physical condition can affect the athlete's appearance during training or competition, so that they cannot perform optimally. This activity aims to provide knowledge and skills for implementing the Aquarobic Training for Fitness (ATF) program. The method of this activity is carried out through two activities, namely the presentation and discussion method as well as the practical method of aquarobics training for fitness, which was held on August 13 and 14, 2022, at Club House Paramount Village, Semarang. Physical fitness checks include measurements of weight, height, blood pressure, body fat percentage, and heart rate. The results of community service through Aquarobics Training for Fitness for athletes and coaches went well; as many as 4 [38 %] participants had very good fitness and 8 [62%] had good fitness, which shows good reciprocity in the implementation of community service activities  Increased understanding of body fitness is indicated by changes in blood pressure and heart rate that are monitored during activities.  Abstrak, kondisi fisik atlet dapat mempengaruhi penampilan atlet pada saat latihan atau pertandingan, sehingga tidak dapat tampil secara maksimal. Kegiatan ini bertujuan untuk memberikan pengetahuan dan keterampilan dalam menerapkan program Aquarobic Training For Fitness (ATF). Metode kegiatan ini dilakukan melalui dua kegiatan yaitu metode presentasi dan diskusi, serta metode  praktik Aquarobics Training for Fitness yang dilaksanakan pada tanggal 13dan 14 Agustus 2022 di Club House Paramount Village, Semarang. Pemeriksaan kebugaran tubuh mencakup pengukuran berat badan, tinggi badan, tekanan darah, presentase lemak tubuh, dan denyut jantung. Hasil pengabdian masyarakat melalui Aquarobics Training for Fitness pada atlet dan pelatih berjalan dengan baik,  sebanyak 4 [38 %] peserta memiliki kebugaran sangat baik dan 8 [62 %]  memiliki kebugaran baik, hal ini menunjukkan adanya timbal balik yang baik dalam pelaksanaan kegiatan pengabdian masyarakat. Peningkatan pemahaman kebugaran tubuh  ditandai dengan perubahan  tekanan darah dan denyut jantung yang dipantau selama kegiatan berlangsung.

Abstract

Metabolic syndrome (MS) is currently emerging globally and increases the risk of dangerous chronic diseases. Obesity involving inflammatory mediators secreted by adipocytes appears to be the major cause. While white adipose tissue (WAT) functions as energy storage, brown adipose tissue (BAT) involves in thermogenesis and is known to have the ability to dissipate energy. Currently, researchers are developing BAT through the browning mechanism of adipose tissue to prevent and cure MS. The extract derived from nutmeg, a tropical seed of Myristica fragrans, has a peroxisome proliferator-activated receptor γ (PPARγ) agonistic effect which is known to stimulate brown adipocytes development. PPARγ activation stimulates the expression of uncoupling protein 1 (UCP1), uncoupling protein 2 (UCP2), and uncoupling protein 3 (UCP3). In the present study, we explore nutmeg-induced stimulation of UCP1, UCP2, and UCP3 protein level characterization shift in 3T3- L1 cell lines which indicates the browning mechanism of adipose tissue exerted by nutmeg. Nutmeg was first extracted and 3T3-L1 cell lines taken from ATCC were used which were grown in Dulbecco’s Modified Eagle Medium (DMEM). Cells were treated with nutmeg extract (NuSE) with dose: 0.01; 0.1; 1; and 10 ppm. Western blot and ImageJ software were used for protein analysis and all experimental data were analyzed using SPSS. Results showed that NuSE stimulates the increase of UCP1 and UCP2 protein levels significantly at a 10-ppm dose. This suggests that polyphenol- containing malignant in NuSE has the probability to regulate PPARγ which potentially alters characteristics of white adipose tissue in 3T3-L1 cell lines.

Abstract

A. pauciflorum is known to have various pharmacological activities, including antimicrobial, antioxidant, antidiabetic, antidiarrheal, anticancer, and anti-gastric activity. In-vivo studies were conducted to determine the therapeutic and metabolic effects of A. pauciflorum. This research aimed to study the Effect of A. pauciflorum added to the diet in rats. The treatment group was divided into 4 to six mice each. The treatment group consisted of normal control, a negative control, and two test groups. The normal control group was given a normal diet (NCD), and the negative control group was given a high-fat diet (HFD) diet containing palmitic fatty acid to be overweight. Both test groups were given a normal diet and preparations containing A. pauciflorum (NCD-JK), and high-fat diets were fed with A. pauciflorum (HFD-JK). The treatment was carried out for nine weeks. Adding A. pauciflorum to feed can increase hemoglobin, RBC, WBC, glucose, and body weight caused by the content of Se in A. pauciflorum, which can protect cells from free radicals and affect the composition of gut microbiota. However, the administration of A. pauciflorum did not affect the platelets, ALT, and AST levels, which might be due to the lack of Se content in A. pauciflorum.

Abstract

Aging caused such overwhelming effects on the mass of skeletal muscle. The increasing period of rest and lack of physical activity that indicate the reduced movement most likely explain much in reducing the phenotype of muscle and bone in older individuals. Sarcopenia, the age-related loss of muscle mass and strength, is the term to explain this condition. Metabolic abnormalities such as alterations in insulin sensitivity, greater deposition of fat, myosteatosis, low level of hormones, systemic inflammation, and impaired oxidative defenses caused by decreased mitochondrial activity. Accumulated evidence showed that resistance training (RT) had an abundant effect on sarcopenia, functional status, bone density, and hip fracture. Although numerous studies showed resistance training had a significant effect to improve muscular strength in the elderly. This study aims to explore the role of bodyweight exercise in improving skeletal muscle strength in the elderly. From 19 original articles, we analyzed the effects of bodyweight exercise in the population who underwent aging. Several articles were also added to explain the muscular condition in aging and its improvement after exercise. Bodyweight training has a significant role in improving skeletal muscle strength in the elderly. It induced satellite cell activity, preventing excessive proteasomal degradation, and type II motor-unit fibers proliferation which has a significant role in muscular strength.

Abstract

Nutmeg as one of Indonesia native plants has already been used widely in herbal treatment. It has been demonstrated that nutmeg presents activities related to mitochondria functions. Nutmeg potentially has the effect of peroxisome proliferator activated receptors Υ (PPAR Υ) which contributes to myogenesis, and may alters mitochondrial homeostasis in respiration and biogenesis. In this study, how nutmeg extract alters oxidative process in skeletal muscle of aging rats was explored. Twenty aging Wistar male rats aged 80 weeks old were divided into two groups (control and nutmeg treatment). Nutmeg extract was administered for 12 weeks using a gavage. After that, the soleus muscles were collected, weighted, frozen, and stored at -800C until used. Nutmeg was observed to increase the COX1 (1.9 folds, p<0.01), COX2 (1.6 folds, p<0.05), and COXIV (2.8 folds, p<0.01) gene expressions, in parallel with increased protein carbonyl levels (1.5 folds, p<0,01). Nutmeg treatment also significantly increased the expressions of antioxidant endogen COQ7 (2 folds, p<0,01) and PDSS2 (2 folds, p<0,01) in the soleus muscle. Taken together, nutmeg extract may improve mitochondrial respiratory oxidative activities in type I aging skeletal muscle.

Abstract

Komunitas Duta Kampus Merdeka (DKM) 2022 memiliki sifat (nature) go-to-person dalam melaksanakan tugasnya untuk mengakselerasi implementasi kebijakan Merdeka Belajar Kampus Merdeka (MBKM) sesuai dengan spirit transformasi pendidikan tinggi. Oleh karena itu, DKM diharapkan mampu berperan sebagai problem shooter yaitu mampu menjawab dan membantu memecahkan masalah tentang implementasi program Kampus Merdeka baik di lingkungan Perguruan Tinggi (PT) maupun di PT sekitar. Salah satu upaya untuk mengoptimalkan peran dan nature DKM serta membantu penyelesaian masalah dan mengakselerasi implementasi Kampus Merdeka di perguruan tinggi, maka DKM Leads menjalin kerjasama dengan Lembaga Layanan Pendidikan Tinggi (LLDIKTI) untuk melaksanakan kegiatan kolaborasi. Kolaborasi dilaksanakan dalam bentuk Pendampingan Akselerasi Implementasi MBKM yang bertujuan meningkatkan kapasitas DKM sebagai problem shooter dengan solusi yang disampaikan secara on the spot. Selain itu, kegiatan tersebut juga bertujuan untuk meningkatkan sinergi dalam memetakan dan memberikan rekomendasi tematik bagi perguruan tinggi dan program studi yang masih memiliki kendala dalam implementasi Kampus Merdeka. Melalui Pendampingan Akselerasi Implementasi MBKM, DKM dan LLDIKTI dapat membantu mengakselerasi implementasi MBKM secara sistemik dan komprehensif.
