Abstract

BACKGROUND: Connective tissue growth factor (CTGF) plays a central role in fibrotic processes affecting both renal and retinal tissues in diabetes. Although sodium-glucose cotransporter 2 (SGLT2) inhibitors have been shown to exert renoprotective and antifibrotic effects, their impact on CTGF expression in renal and retinal tissues has not been clearly established. This preliminary study was conducted to evaluate whether SGLT2 inhibitors (SGLT2i) could influence CTGF expression in the kidneys and eyes of diabetes-induced rats.METHODS: After two weeks of adaptation, 24 rats were randomized and distributed equally into four groups (n=6 each): 1) Healthy Control, healthy rats without diabetic induction; 2) Negative Control, diabetic rats induced with streptozotocin (STZ) without treatment; 3) Metformin Group, diabetic rats treated with metformin; and 4) SGLT2i Group, diabetic rats treated with empagliflozin. Following eight weeks of intervention, CTGF expression was analyzed by Western blot in renal tissue (right kidney) and chorioretinal tissue (right eye). Four samples per group yielded analyzable bands and were included in the final quantification.RESULTS: In renal tissue, CTGF levels (mean±SD) were highest in Negative Control Group (0.81±0.06). Both the Metformin Group (0.58±0.14) and SGLT2i Group (0.57±0.33) demonstrated a trend toward reduced CTGF expression. In chorioretinal tissue, CTGF values were relatively similar across groups (Healthy Control: 0.67±0.05; Negative Control: 0.63±0.12), with Metformin Group (0.61±0.12) and SGLT2i Group (0.64±0.22) showing a modest reduction trend.CONCLUSION: In diabetic rats, CTGF expression levels are markedly increased. Following treatment with metformin and SGLT2i, CTGF expression demonstrates a noticeable reduction trend.KEYWORDS: SGLT2 inhibitor, metformin, connective tissue growth factor, cellular communication network factor 2, renal, retina

Abstract

Dysmenorrhea and anemia are common health problems among adolescent girls, while pharmacological therapy has limitations, highlighting the need for safe and comprehensive alternative interventions. This study aimed to analyze the effects of Fe supplementation and the combination of Fe supplementation with yoga on hemoglobin levels and dysmenorrhea intensity in adolescent girls. A quasi-experimental design with a two-group pre-test post-test approach was employed. Participants were adolescent girls from SMAN 03 Rejang Lebong who received either Fe supplementation or a combination of Fe supplementation and yoga for four weeks. Data were analyzed using the Wilcoxon and Mann-Whitney tests. The results showed that the combination of Fe supplementation and yoga significantly improved hemoglobin levels and reduced dysmenorrhea intensity (p = 0.000). Moreover, the combination was more effective than Fe supplementation alone. These findings suggest that combining yoga with Fe supplementation can serve as an effective non-pharmacological strategy to manage anemia and dysmenorrhea among adolescent girls. Abstract

Skabies merupakan salah satu infeksi kulit yang sering dijumpai pada kucing yang mana disebabkan oleh tungau Sarcoptes scabiei ataupun Notoedres cati. Gejala penyakit ini termasuk rasa gatal (pruritus) yang menyebabkan aktifitas menggaruk secara berlebih dan adanya penebalan kulit. Jika tidak ditangani, pertumbuhan tungau akan tidak terkontrol dan dapat menyebabkan imunosupresi. Ivermectin 1% merupakan obat yang umum digunakan untuk menangani penyakit ini. Disamping itu, hypochlorous acid (HOCl) 0,01% yang telah distabilkan terbukti memiliki efek antiinflamasi, anti-pruritus. Penelitian ini bertujuan untuk melihat gambaran HOCl terhadap infeksi kulit skabies pada kucing. Penelitian ini menggunakan 3 kucing yang dibagi menjadi 3 perlakuan, yaitu kontrol negatif (tidak diberi perlakuan), kontrol positif (pemberian ivermectin 1% 0,05 mL/kgBB secara subkutan), dan perlakuan HOCl 0,01% (pH 5,5-6,5) yang berdurasi selama 7 hari. Pengamatan secara makroskopis dilihat dari adanya eritema, pinnal-pedal reflex, alopecia, hiperkeratosis, pus, warna, dan bentuk permukaan lesi. Dokumentasi dilakukan dengan menggunakan penanda 2x2 cm untuk perbandingan ukuran lesi. Hasil yang didapatkan, HOCl terbukti mengurangi hiperkeratosis dan mempercepat penyembuhan luka akibat garukan. Perbandingan waktu kesembuhan antara kontrol positif dan perlakuan teramati sama, yaitu dimulai pada hari ke-3. Asam hipoklorit (HOCl) secara topikal dapat membantu mempercepat fase inflamasi dan proliferasi penyembuhan luka.

Abstract

Background: The limited research on the determinants of performance in short-distance trail running in tropical environments is a major problem in understanding the influence of environmental conditions on runners' performance and pacing strategies.Objectives: This study aimed to examine the influence of race distance, sex, and age on trail running performance in the 5K and 10K categories of the Merapi Merbabu de Trail 2025.Methods: A retrospective observational race result analysis was conducted using official race data from the Merapi Merbabu de Trail 2025. The study included 429 runners (5K: n = 144; 10K: n = 285), consisting of 275 male and 154 female participants. The primary outcome variable was finish time (in minutes), while the independent variables included race, distance, sex, and age. Statistical analyses were performed using SPSS version 26, including descriptive statistics, independent-samples t-tests, Cohen’s d effect sizes, and multiple linear regression to evaluate predictors of performance.Results: Results showed a significant difference in performance between race distances. The mean finish time for the 5K category was 152.8 ± 44.3 minutes, while the 10K category averaged 306.1 ± 74.3 minutes (t = -26.68, p < 0.001, Cohen’s d = 2.50). Male runners completed the 10K race faster than female runners (293 vs. 345 minutes). Regression analysis indicated that finish time increased by approximately 2–3 minutes per year of age, indicating a progressive decline in performance with increasing age.Conclusions: These findings indicate that race distance, sex, and age are significant determinants of trail running performance in tropical environments.

Abstract

Background: Cardiac aging is closely associated with mitochondrial dysfunction and impaired quality control mechanism. Exercise has been shown to modulate mitochondrial homeostasis, however, its effects on cardiac mitophagy and mitochondrial dynamics during aging remain unclear. This study investigated the effects of moderate-intensity treadmill training on mitochondrial maintenance-related gene expression in the hearts of young and aged Wistar rats.Methods: Young and aged rats were divided into four groups: young control, young exercise, aged control, and aged exercise (n=6 per group). Exercise groups performed treadmill running at 20 m/min for 30 minutes/day, 5 days/week for 8 weeks. Cardiac gene expression levels of Pink1, Parkin, Mfn1, Mfn2, Opa1, Drp1, and Fis1 were analyzed using semi-quantitative polymerase chain reaction (PCR). Data were analyzed using one-way ANOVA or Kruskal–Wallis test followed by appropriate post hoc analyses.Results: Exercise significantly increased Mfn2 expression in aged exercise rats compared with aged controls (p=0.029), suggesting partial restoration of age-related decline. Expression of Mfn1 and Drp1 varied among groups but showed no significant pairwise differences. Expression levels of Opa1, Fis1, Pink1, and Parkin remained unchanged. These results indicate that moderate exercise selectively enhances mitochondrial fusion capacity while maintaining balanced fission and basal mitophagy activity.Conclusion: Moderate-intensity treadmill training promotes mitochondrial  fusion-related adaptation in aging cardiac tissue. Regular moderate exercise may represent a potential non-pharmacological strategy to support mitochondrial function and mitigate cardiovascular aging.

Abstract

Background: The increasing use of tobacco and electronic nicotine delivery systems (ENDS) among youth represents a significant public health concern. In Indonesia, nicotine use among university students may contribute to long-term health risks. This study evaluated the correlation between nicotine dependence, measured using the Hooked-on Nicotine Checklist (HONC), and cardiorespiratory fitness assessed through maximal oxygen uptake (VO₂max) within the Wellness Matrix Framework.Methods: A cross-sectional study was conducted among 40 male undergraduate students aged 18–24 years at Universitas Padjadjaran who actively used cigarettes, e-cigarettes, or vape products. Participants were selected using purposive sampling. Nicotine dependence was assessed using a modified HONC questionnaire, whereas VO₂max was estimated using the YMCA 3-Minute Step Test. Descriptive statistics, the Shapiro-Wilk tests, and Pearson correlations analysis were applied (α=0.10).Results: Participants showed moderate nicotine dependence (mean HONC score: 5.6 ± 2.37) and moderate fitness (mean VO₂max: 44.31±3.86 mL/kg/min). A weak inverse correlation was found between HONC scores and VO₂Max (r=-0.275, p=0.085). Body mass index showed a strong negative correlation with VO₂max (r=-0.824, p<0.001). Subgroup analysis showed consistent negative trends between nicotine dependence and VO₂max across single, dual, and triple product users. Duration of nicotine use showed negligible correlation with VO₂max (r=0.041, p=0.802). Conclusion: Higher nicotine dependence is associated with lower cardiorespiratory fitness and affects multiple wellness dimensions. These findings underscore the importance of holistic interventions targeting physical and psychosocial wellness among young adults.

Abstract

Sedentary lifestyle can cause the accumulation of energy reserves in the form of lipids in white adipose cells. Lifestyle modification by means of exercise is done to reduce fat reserves in white adipose tissue (WAT). This study aims to determine the effect of exercise in various intensities on the histological changes of white adipose cells. Research method exploratory with Federer formula to determine the number of samples carried out. Using 28 Wistar rats located at the Central Laboratory of Padjadjran University, Sumedang from May to June 2022. The data taken were the average diameter ± standard error (SEM) of the inguinal white adipose cells of the experimental rats. Data analysis using Microsoft Excel 2016 and SPSS v.28 software. Of the 28 samples studied, only 25 samples met the inclusion criteria. Group K is 0.15% (p = 0.024) greater than P2, and 0.25% (p 0.001) is greater than P3. The P1 group had a value of 0.17% (p = 0.025) greater than P3. Moderate and vigorous exercise can be used to reduce the size of white adipose cells. While low-intensity exercise is considered ineffective. Further research can be carried out to assess changes in white adipose cells based on exercise intensity from other sides besides their morphology. Abstract

Acute inflammatory conditions such as uveitis in pets can lead to discomfort and unnecessary pain. The treatment for uveitis involves the administration of anti-inflammatory drugs, including NSAIDs or herbal remedies generally containing flavonoids. Physalis angulata Linn., known as Ciplukan, encompasses compounds exhibiting anticoagulant, antimicrobial, anti-inflammatory, and anticancer activities. Ciplukan is composed of withanolids, terpenoids, carotenoids, and flavonoids. This study was conducted to investigate the anti-inflammatory effect of the ciplukan ethyl acetate fraction (CEAF) on Wistar rats as egg white-induced inflammation models and the most effective dose. The test was carried out on 6 test groups, each consisting of 5 rats. The test groups consisted of a normal control group, a 4,5 mg/kgBW diclofenac induced group, a negative control (NC) group which were induced inflammation using egg whites, and CEAF administration at respective doses of 5, 10, and 15 mg/kgBW groups. The parameters used in this research were plant determination, phytochemical tests, measurement of edema volume, and analysis of anti-inflammatory activity. The volume of edema on the rats’ paws was measured using a plethysmometer periodically, with a total period of 6 hours. All the data in the study were analysed statistically using the One-Way Analysis of Variance (ANOVA) method and the Tukey test with 95% CI. The results indicated that the CEAF with dose of 15 mg/kgBW exhibited a percentage of edema inhibition (47,8%) closest to that of the diclofenac sodium group (61,6%). The study findings suggest that the CEAF possesses anti-inflammatory activity with the most effective dose of 15 mg/kgBW.

Abstract

Background: UCP-1 deficiency can be addressed through the activation of thermogenesis using various herbs, including Mitragyna speciosa (M. speciosa ). Purpose: This study aims to evaluate the efficacy of M. speciosa extracts, encapsulated in nanoparticles, in stimulating the activity of UCP-1 . Methods: The research utilized an in silico approach to test the docking of Mitragynine and epinephrine ligands against β1 and β3 adrenergic receptors. The nanoparticles were prepared using chitosan, alginate, and polyethylene glycol (PEG). An in vivo study was performed utilizing Wistar rats subjected to a high-fat diet (HFD) and assigned to six groups: Normal, HFD, HFD + MG50, HFD + Npm5, HFD + Npm10, and HFD + Npm20. The induction period for all groups was set at four weeks. Results: The docking tests yielded binding scores of -189.55 for Mitragynine against the β1 adrenergic receptor and -179.76 for the β3 receptor. The interactions between Mitragynine and the adrenergic receptors involved amino residues similar to those found in the native ligand epinephrine, specifically Asn1006A, Ser1007A, and Asn1022A for β1, and Asn332R for β3. Nanoparticles at doses of 5, 10, and 20 mg/kg BW demonstrated significant differences (p < 0.05) in body temperature and UCP-1 levels. Notably, the 10 and 20 mg/kg BW dosages did not lead to weight gain in the rats (p > 0.05). Conclusion: Mitragynine shows potential as an agonist for β1 and β3 adrenergic receptors. Furthermore, the nanoparticles at 5, 10, and 20 mg/kg BW were effective in stimulating UCP-1 activity and elevating body temperature in rats subjected to a high-fat diet.
