Abstract

Neurodegenerative diseases are mostly experienced by postmenopausal women and are often caused by estrogen deficiency, so it is necessary to replace the hormone estrogen in the form of phytoestrogens. One of the plants that contain phytoestrogens is Marsilea crenata. The objective of this study is to see if giving rotenone-induced zebrafish a 96% ethanol extract of M. crenata leaves increases locomotor activity. This research was conducted by inducing 5 µg/L rotenone as a compound that will interfere with the locomotor activity of zebrafish. Next, treatment was given with 96% ethanol extract of clover in each group with a dose of 2.5; 5; 10; and 20 mg/mL to determine the effect of the extract on increasing locomotor activity of rotenone-induced zebrafish. Observations were made by looking at the quantity of zebrafish swimming every five minutes until day 28. The treatment of 96% ethanol extract of M. crenata leaves could significantly increase zebrafish motility at the optimum dose of 2.5 mg/mL, because every week the zebrafish locomotor activity increased compared to the negative control. M. crenata leaves extract is proven to prevent neurodegenerative diseases. However, further research needs to be done on the degenerative effects of rotenone every week.

Abstract

Sexual arousal is an important aspect to achieve a harmonious sex life, but often disorders in normal sexual function disrupt the harmony of household relationships. One alternative natural ingredient that has the potential as an aphrodisiac agent is Lelak (Uvaria rufa) which is traditionally used by the people of East Nusa Tenggara (NTT), Indonesia, to overcome sexual dysfunction in men. This study aims to evaluate the safety and effectiveness of 70% ethanol extract of Uvaria rufa roots in vivo. This study is an experimental study to test the effectiveness of 70% ethanol extract of Uvaria rufa roots in vivo. This study was conducted with an initial stage, namely 70% ethanol extract of Uvaria rufa roots obtained through maceration, then the aphrodisiac activity was measured through the parameters of the number of leydig cells and the weight of the testes of male mice (Mus musculus). The results showed that the administration of the extract with an LD50 value of 2108 μg/ml which is included in the Relatively Harmless category, while group P2 with a dose of 0.85 mg/20g BB/day was stated as the optimal dose (p<0.05 against the negative control) for aphrodisiac activity in measuring testicular weight and the number of Leydig cells. Therefore, administration of 70% ethanol extract of Uvaria rufa roots at a dose of 0.85 mg/20g BW/day is the optimal dose for aphrodisiac activity.

Abstract

Carrageenan is widely used in various industries, such as food and beverages, cosmetics, and pharmaceuticals, due to its important role as a viscosity regulator, stabilizer, thickener, developer, gelling agent, and excellent stabilizing agent. Temperature and drying duration are important factors that affect the quality of the final product. This study aims to examine the quality characteristics of carrageenan from Eucheuma spinosum dried using the oven method, considering variations in temperature and drying time. This study used a completely randomized design (CRD) with two factors: temperature and drying time, each of which was repeated three times. Eucheuma spinosum was extracted using the microwave-assisted extraction (MAE) method with 10% KOH solvent in a ratio of 1:10 for 8 minutes. The first factor is the drying temperature, which consists of three levels: 40°C, 50°C, and 60°C. The second factor is the drying time, which also consists of three levels: 24 hours, 27 hours, and 30 hours. The results of the study showed that (1) temperature and drying time affect the quality characteristics of carrageenan from Eucheuma spinosum dried using the oven method, (2) the parameters affected include water content, ash content, sulfate content, viscosity, and gel strength of carrageenan, and (3) the higher the temperature and duration of drying, the more the water content, ash, protein, and crude fiber in the product tend to decrease. This decrease is in line with the viscosity and gel strength of the resulting carrageenan.

Abstract

This study aims to identify potential antimalarial compounds targeting the Plasmodium falciparum lactate dehydrogenase enzyme (PfLDH), given the parasite's dependency on glycolysis for energy production. Considering the high similarity of LDH enzymes across Plasmodium species, developing novel PfLDH inhibitors may offer therapeutic benefits, particularly against P. vivax, P. malariae, and P. ovale. Computational approaches were employed to screen natural compounds from the Cratoxylum genus (Cratoxylum glaucum and Cratoxylum arborescens). Molecular docking was performed using Molegro Virtual Docking (MVD) to assess binding affinity. SwissADME was utilized to evaluate pharmacokinetic, physicochemical, synthetic accessibility properties, and ProTox II was used for toxicity prediction. Molecular docking results indicated that fuscaxanthone C and 3-geranyloxy-6-methyl-1,8-dihydroxyanthraquinone exhibited strong inhibitory activity against PfLDH, with rerank scores of -103.068 kcal/mol and -111.141 kcal/mol, respectively surpassing the reference ligand chloroquine (-94.307 kcal/mol). Pharmacokinetic analysis revealed that all compounds, except stigmasterol and fuscaxanthone C, met Lipinski’s Rule of Five. Toxicity prediction categorized fuscaxanthone C as Class III (toxic). Additionally, synthetic accessibility predictions indicated that all compounds, except stigmasterol, are easy to synthesize. Natural compounds from the Cratoxylum genus, particularly 3-geranyloxy-6-methyl-1,8-dihydroxyanthraquinone, show promise as PfLDH inhibitors. Despite its potency, fuscaxanthone C's toxicity profile warrants caution. Further studies are needed to validate these findings through in vitro and in vivo testing.

Abstract

Skin dehydration is a common issue affecting 40–50% of the adult population, resulting in dry, rough, and dull skin, as well as itching, irritation, and premature aging. The "back to nature" trend has increased interest in natural skincare products that are more environmentally friendly and gentler on the skin. Virgin coconut oil (VCO) is a popular natural ingredient known for its ability to maintain skin moisture and provide protection. This study aims to develop a liquid soap formulation using VCO to address skin dehydration. The research methods included formulation and evaluation of liquid soap through organoleptic testing, foam stability, specific gravity, pH, as well as hedonic and irritation tests. The results showed that: (1) Formula III met all physical evaluation parameters, including organoleptic characteristics, foam stability and height, specific gravity, and pH, in accordance with SNI standards; (2) Formula III was the most preferred by panelists in the hedonic test and caused no skin irritation based on irritation test results. Therefore, Formula III can be concluded as the optimal formulation for VCO-based liquid bath soap.

Abstract

Diarrhea is one of the diseases caused by symptoms of intestinal tract infections due to various microorganisms. Goosegrass (Eleusine indica (L.) Gaertn) is a wild weed that has long been used as a traditional medicine, attributed to its phytochemical compounds such as tannins, alkaloids, flavonoids. This study aims to evaluate the antidiarrheal effects of Eleusine indica extract by comparing six treatment groups: a healthy group, a negative control group (Na-CMC), a positive control group (Loperamide 2 mg), and three variations of the extract dosage. Observations were made over 360 minutes at 30-minute intervals, using a fecal consistency scale from 1 (solid) to 5 (liquid). LC-MS/QToF analysis identified 25 secondary metabolites in the extract, including flavonoids (Schaftoside, Orientin, Vitexin, Tricin), phenolic compounds, and terpenoids. In vivo testing showed that the third dose of Eleusine indica grass extract had the most optimal antidiarrheal effect among the three doses, as evidenced by its ability to reduce diarrhea frequency, decrease fecal weight, shorten diarrhea duration, and improve fecal consistency, approaching the effectiveness of Loperamide 2 mg as a positive control. These findings suggest the potential of Eleusine indica extract as a promising natural antidiarrheal agent.
