Abstract

Somatic embryogenesis, a crucial plant regeneration method, has become indispensable for crop improvement, particularly for species reliant on somatic cell manipulation techniques. Optimization of this process necessitates an understanding of the developmental stages involved. This study investigates the histological aspects of somatic embryogenesis in Musa acuminata ssp. malaccensis derived from immature zygotic embryos. Through detailed histological analysis, we aimed to elucidate the morphological changes and cellular organization occurring during the various stages of somatic embryogenesis, from induction, culture proliferation, and somatic embryo development to plantlet conversion. The initial stages of embryogenesis, characterized by nodules, were primarily composed of meristematic cells with high cell division activity. These cells contained tetrad-like structures that could develop into distinct two- and four-celled proembryoids or proembryogenic aggregates. Our histo-anatomical analysis revealed that embryogenic cultures proliferated through multiple pathways simultaneously: somatic embryo budding, proembryo formation, and pro-embryonic mass formation from both internal and peripheral cells. At the stage of somatic embryo development, embryos with a well-defined protoderm layer, containing cells with prominent nuclei and dense cytoplasm, potentially regenerate into plantlets. Furthermore, histological examination revealed the presence of procambium within mature somatic embryos, which subsequently developed into the vascular system of the complete plantlet

Abstract

Establishing a regeneration media of Indonesian local garlic is necessary for several purposes, including plant breeding and large-scale propagation. This study was aimed to evaluate media formulation on callus induction and proliferation of four local garlic varieties (Geol, Lumbu Hijau, Lumbu Kuning, and Lumbu Putih) using root cuttings as the explants. MS media supplemented with different concentration of picloram (4 and 6 mg L-1) without and in combination with glutamine (100 mg L-1) alone and casein hydrolysate (3 g L-1) were evaluated. The results showed that the responses of induction and proliferation of embryogenic callus were genotype-dependent because there was no significant interaction between varieties and media formulations. Still, the varieties had a significant interaction with the observed variables. The fastest initiation time of callus induction was obtained from Lumbu Putih, less than 2 weeks after culture. Geol showed the highest percentage of callus formation and fresh weight of callus, 59% and 0,92 g respectively. There were three different types of the callus: (1) friable, glossy, clear white, (2) friable, glossy, transparent yellow, and (3) semi compact, glossy, yellowish to milky white. ABSTRAK Pemantapan media regenerasi bawang putih lokal Indonesia penting dilakukan untuk berbagai tujuan, termasuk pemuliaan tanaman dan perbanyakan skala besar. Penelitian ini bertujuan untuk mengetahui respon in vitro empat varietas bawang putih lokal (Geol, Lumbu Hijau, Lumbu Kuning, dan Lumbu Putih) terhadap komposisi media induksi dan proliferasi kalus dengan menggunakan akar sebagai eksplan. Komposisi media yang diujikan meliputi media dasar MS yang mengandung pikloram (4 dan 6 mg L-1), baik tanpa atau dengan penambahan glutamin (100 mg L-1) dan kasein hidrolisat (3 g L-1). Hasil penelitian menunjukkan respon induksi dan proliferasi kalus embriogenik bersifat genotype dependent, sebab tidak terdapat interaksi yang nyata antara faktor varietas dan formulasi media, namun faktor varietas berpengaruh nyata terhadap variabel amatan. Waktu inisiasi kalus tercepat diperoleh dari Lumbu Putih, yaitu kurang dari 2 minggu setelah kultur. Varietas Geol memiliki persentase pembentukan kalus dan bobot segar kalus tertinggi, berturut-turut sebesar 59% dan 0,92 g. Terdapat tiga tipe kalus yang terbentuk, yaitu (1) remah, mengkilap, putih bening, (2) remah, mengkilap, bening kekuningan, dan (3) kompak, mengkilap, kekuningan-putih susu.

Abstract

Durio testudinarius, known as Durian Kura-Kura, is a native Bornean durian species in the Malvaceae family with fruit at the base of the main stem. Recent advances in genomic-based next-generation sequencing are being used to conserve germplasm and enhance plant breeding. This method rapidly and cost-effectively sequences plant genomes, allowing for simple sequence repeat markers to be constructed in silico. This study aimed to design and evaluate novel, simple sequence repeat markers utilizing next-generation sequencing microsatellite data of D. testudinarius. A total of 20 sequences containing di-, tri-, tetra-, and penta-nucleotide motif repeats were selected and designed to generate primers using Primer3 online software. The PCR results of the twenty primers tested on D. testudinarius and 19 genomic DNA samples of species from the Malvales order showed successful amplification. These newly developed simple sequence repeat markers are crucial for genetic population analyses, determining population structure, and enhancing plant breeding programs and conservation strategies for D. testudinarius and its relative's germplasm resources.

Abstract

Wild banana Musa acuminata ssp. malaccensis, an ancestor of cultivated bananas, possesses valuable genetic diversity, including resistance genes to fusarium wilt, and demonstrates high environmental adaptability. These traits are important for pre-breeding programs, whether by conventional breeding, which is a lengthy process, or by taking advantage of somatic cell manipulation techniques such as somatic hybridization, which requires an efficient plant regeneration system like somatic embryogenesis. We have established an efficient and comprehensive protocol for somatic embryogenesis of this wild Musa using immature zygotic embryo explants covering culture induction and proliferation, somatic embryo development, and subsequent plant conversion. Embryogenic culture was induced on a complex-modified MS medium supplemented with 1 mg.L-1 2,4-D auxin or 5 mg.L-1 picloram. The embryogenic cultures proliferated in the form of granular or nodular structures, which was best obtained by reducing the picloram concentration to 1 mg.L-1 and combining it with the same concentration of 2,4-D at a half-strength macro-nutrient of basal medium. Embryo development from embryogenic cultures and regeneration into shoots. Proembryos as granular structures spontaneously matured into early-stage somatic embryos upon withdrawal of the strong auxin inducer. Increasing the sucrose and gelling agent concentrations in the growth medium improved somatic embryo formation from embryogenic cultures. The frequency of shoot formation from developed somatic embryos was increased by incorporating 0.5 mg.L-1 BA and 0.5 mg.L-1 GA3 into the regeneration medium.

Abstract

Plant growth-promoting rhizobacteria (PGPR) is a promising technology for controlling viral diseases, including pepper yellow leaf curl disease (PYLCD) of chili pepper caused by Begomovirus infection. The objectives of this research were to investigate the effectiveness of PGPR containing Pseudomonas fluorescens PF1 and Bacillus polymyxa BG25, as well as their combination with other protective agents, to control PYLCD under field conditions in an endemic region. The treatments consisted of a single application of PGPR (a mixture of P. fluorescens PF1 and B. polymyxa BG25), guano tea, endophytic fungus H5, and neem oil; combination of PGPR with guano tea, endophytic fungus H5, and neem oil; conventional pesticide that relies on synthetic chemical insecticide sprayed weekly; and untreated plots. The experiment was arranged in a randomized block design with four replications. Treatment with PGPR alone was able to delay disease onset by 2.25 weeks, but it caused only a slight reduction in disease incidence. The combination of PGPR + guano tea and PGPR + endophyte H5 provided the best results in controlling PYLCD. The combination of PGPR + guano tea and PGPR + endophyte H5 delayed disease onset by 2.75 weeks and 3.25 weeks, respectively, and reduced disease incidence with effectiveness rates of 52.72% and 52.08%, respectively. These two treatment combinations gave the best performance for plant growth and yield.

Abstract

Pepper yellow leaf curl disease (PYLCD) is the primary disease that affects chili plants, resulting in a loss of quality and quantity. The identification of chili plants resistant to PYLCD is a solution to support optimal chili cultivation. The purpose of this study was to estimate genetic parameters to obtain information on the genetic control of chili resistance to PYLCD. The genotypes used in this study consisted of four parental, four hybrid genotypes, and 180 individuals of the F2 population. The results showed that the genotypes "F6074" and "IPBC12" were categorized as resistant, while the genotypes “IPBC5” and “YUNI” were categorized as susceptible. The distribution of resistance level in chili peppers to yellow curly leaf disease tends to follow a ratio of 13:3, with a dominance of susceptible traits. The assumption for this phenomenon is that resistance to PYLCD is controlled by at least two genes. Both genes have dominant and recessive epistatic interactions. Abstract

The sterilization is a primary step in in-vitro culture. plant breeding through in-vitro culture hinges on the successful effectiveness of the sterilization method in producing sterile plants. This research aims to optimize sterilization methods using a completely randomized factorial design. The first factor is the size of the material (small and large), and the second factor is the graded concentration of the bleach solution. The study revealed that Material sizes do not have a significant effect on contamination level, all the same, large size materials increase the survival rate of sterilized rodent tuber plants. The application of graded bleach solution concentrations at 25%, 20%, and 15% is able to reduce contamination to 0% on small size materials and 5 % on large size materials. Material sizes do not have a significant effect on contamination level, all the same, large size materials increase the survival rate of sterilized rodent tuber plants.

Abstract

Excessive use of pesticides can have an impact on the environment, including the loss of pesticide residues in soil and agricultural products. The purpose of this study was to determine the spreading of organophosphate pesticide residues in the shallot production area of  Brebes Regency, Central Java. Research locations were two districts selected by purposive sampling based on production area, namely Kersana and Wanasari Districts. Organophosphate compounds analyzed include chlorpyrifos, paration, profenofos, diazinon, fenitrotion, metidation and malation. Organophosphate residue analysis was carried out on seven soil samples and seven shallot products. Pesticide residue analysis was carried out using the standard method by using a gas chromatography device with an electron capture detector. Based on analysis, seven residues of organophosphate pesticides were chlorpyrifos, paration, profenofos, diazinon, fenitrotion, metidation and malation, spread in Kersana District, and six  pesticides residues (except profenofos) spread in Wanasari District. In soil samples, the highest residues obtained in two districts were chlorpyrifos compounds, chlorpyrifos compounds in Kersana District was higher than Wanasari District. In the shallot sample, the highest residues in two districts are diazinon compounds, diazinon compounds in the sample of Kersana District was  higher than Wanasari District. There were  several samples of shallot product contained organophosphate residue  exceed the maximum concentration of residual limit (RML).

Abstract

The development of new superior varieties will be more efficient if it is based on accurate information about the genetic diversity and population structure of available breeding resources, so that the combination of the most potential parents in the breeding program that will be carried out can be identified. Indonesia still lacks information of strawberry diversity. The aim of this research is to study strawberry genetics based on morphological and molecular markers in order to develop strawberry varieties efficiently. Genetic relationship analysis was carried out on 20 genotypes of strawberries collected by Indonesian Instruments Standardization Testing Center for Citrus and Subtropical Fruits (BPSI Jestro), based on morphological characteristic and molecular Random Amplified Polymorphic DNA (RAPD). Morphologycal markers analysis showed that there were two groups of strawberry genotypes using Gower dissimilarity coefficient value of 0.0936–0.5977. The result of molecular marker analysis was also separated into two groups by dissimilarity coefficient value of 0,1321-0,5566. Combination of morphologycal and molecular analysis was carried out and also showed two groups by disimilarity coefficient of 0,1657–0,5264. The information obtained on morphological characteristics and genetic distances can be used as a basic consideration in parental selection for strawberry breeding program. Pengembangan varietas unggul baru akan lebih efisien bila didasarkan pada informasi yang akurat tentang keragaman genetik dan struktur populasi sumber genetik yang tersedia, sehingga dapat diketahui kombinasi tetua yang paling potensial dalam program pemuliaan yang akan dilakukan. Namun, informasi tentang keragaman genotipe-genotipe stroberi di Indonesia masih terbatas. Penelitian ini bertujuan untuk mempelajari keragaman genetik stroberi berdasarkan marka morfologi dan molekuler dalam rangka pengembangan varietas stoberi yang efisien. Analisis kekerabatan genetik dilakukan pada 20 genotipe stroberi koleksi Balai Pengujian Standar Instrumen Tanaman Jeruk dan Buah Subtropika (BPSI Jestro), berdasarkan karakter morfologi dan molekuler dengan metode Random Amplified Polymorhpic DNA (RAPD). Pengamatan pada 41 karakter morfologi menunjukkan terdapat 34 karakter yang beragam dan 7 karakter yang seragam. Hasil analisis menggunakan marka morfologi menunjukkan bahwa 20 genotipe stroberi terbagi menjadi dua kelompok dengan nilai koefisien ketidakmiripan Gower 0,0936–0,5977. Hasil analisis menggunakan marka molekuler juga terbagi menjadi dua kelompok dengan nilai koefisien ketidakmiripan sebesar 0,1321-0,5566. Analisis gabungan morfologi dan molekuler yang dilakukan juga membagi ke 20 genotipe menjadi dua kelompok dengan koefisien ketidakmiripan 0,1657–0,5264. Informasi karakter morfologi dan jarak genetik yang diperoleh dapat dijadikan sebagai dasar pertimbangan dalam pemilihan tetua pada kegiatan pemuliaan tanaman stroberi.
