Abstract

Background Mesenchymal stem cells (MSCs) are valued in regenerative medicine for their multipotency, proliferative capacity, and immunomodulatory properties. Wharton’s jelly-derived MSCs (WJ-MSCs) from the umbilical cord offer a non-invasive, promising source for clinical applications, because easy isolation, lack of ethical concerns, and the presence of both embryonic and adult stem cells have made them a valuable source for use in therapeutic applications and regenerative medicine. This study aimed to optimize WJ-MSC isolation and characterization methods. Methods Human umbilical cords from three healthy donors were collected post-cesarean under strict inclusion criteria. WJ-MSCs were isolated using the explant culture method, with cells adhering to T75 flasks pre-coated with 2% gelatin. Cultures were maintained in Dulbecco’s Modified Eagle Medium (DMEM) supplemented with 10% freshly prepared Human AB serum and monitored for 21 days. Flow cytometry (BD FACSAria) was performed at passages 1 and 5 to assess MSC markers CD105, CD73, CD90, and CD44, alongside the exclusion marker CD45. Results WJ-MSCs exhibited fibroblast-like morphology by passage 1 and showed robust proliferation. Flow cytometry revealed high CD44 expression (~60%) at passage 1, while CD105, CD73, and CD90 became prominent by passage 5. CD45 remained low, suggesting minimal hematopoietic contamination. Conclusion This study confirms the feasibility of isolating and expanding WJ-MSCs using DMEM with 10% human AB serum. While consistent cell growth was achieved, the 21-day culture period may require optimization for scalability, including serum concentration, substrate coatings, and oxygen levels. CPJ-MSCs may be preferable for applications demanding rapid expansion and early marker expression.

Abstract

Food security, stunting issues, hunger, and malnutrition are all interconnected with the theme of extreme poverty. This phenomenon has become a leading discourse in developing countries. On one hand, it is imperative that a country consciously designs agricultural ecosystems and food security frameworks. Communities living in aquatic environments are naturally connected to the tools and resources available within the broader ecosystem that has sustained and supported them for years. Their ability to adapt and even go beyond the “natural-normal” state, in processing raw materials found in their surroundings, reflects their effort to survive through the development of derivative food products. This community service program was conducted in the administrative region of Kampung Dayun, Siak Regency. The main objectives and functions include promoting the use of local natural resources as alternative processed food ingredients, ensuring the availability of abundant local food sources, and preparing food reserves. The activities carried out involved delivering public lectures, question-and-answer sessions, and practical training. These aimed to transfer knowledge and innovation to local communities and food product innovators. The focus was on processing non-rice food sources, such as cassava, corn, sweet potatoes, sago, taro, breadfruit, and other similar types. All of these food sources require extensive knowledge and creativity to be transformed into diverse food products.

Abstract

Terrorism action exerts adverse impact on a state, in either defense or economic sector; therefore, it is always threatened with severe punishment by the criminal law in the state jurisdiction. The release of Law Number 5 of 2018 gives new color to the management of terrorism crime, in which the government holds deradicalization program for terrorism convict. This research aims to study the Republic of Indonesia Ministry of Law and Human Rights’ Decree Number PAS-1931.PK.05.09 of 2023 about Parole for Terrorism Convict in realizing the deradicalization attempt. The existence of deradicalization in the attempt of reducing radical activity and neutralizing radicalism for those involved in terrorism crime. This program is held as an attempt of paroling the terrorism crime. The weakness found in the implementation of deradicalization program is that there is no parameter concerning the successfulness level so that there is no parameter of achievement and the subject or the target of deradicalization is not obvious. Some contraints are found in the implementation of deradicalization: the convicts still hold on their ideology tightly, do not have fund, and their life is endangered by their group when they are free later. This research recommends the improvement of coordination between National Counter-Terrorism Agency (BNPT), Resort Police, Military District Command (Kodim), District Court, Religion Court, and Penitentiary to smooth the parole process and the increase in the number of Officers dealing with Terrorism Convicts

Abstract

Running has become a global phenomenon, including in Indonesia, with the number of runners tripling over the past few years. The Borobudur Marathon, which combines natural beauty and local culture, successfully attracts up to 10,000 runners annually. This study aims to analyze the impact of local culture and tourism attractions on purchase decisions among participants of the 2023 Borobudur Marathon, with brand image as a mediating variable. Adopting a quantitative approach, data were collected from 400 respondents using structured questionnaires and analyzed with SmartPLS 3.0 software. This study uses the Stimulus-Organism-Response (SOR) framework, considering tourism attraction and local culture as the stimuli, brand image as the organism, and purchase decision as a response. The results show that local culture and tourism attractions significantly enhance brand image, positively impacting purchase decisions. However, the direct relationship between local culture and purchase decisions was found to be insignificant, highlighting the critical role of brand image in shaping participant choices. These findings underscore the importance of leveraging local culture and tourism attractions to build a compelling brand image, fostering participant loyalty and engagement. For event organizers, these insights guide the development of strategic marketing approaches that enhance the overall participant experience. The study contributes to the theoretical understanding of the SOR and the dynamics between local culture, tourism attractions, brand image, and purchase decisions within the event object of sports tourism in the context of marketing communication. By applying these findings, organizers can create more memorable events.

Abstract
