Abstract

DNA barcoding based on partial Cytochrome Oxidase subunit I (COI) gene in the mitochondrial has widely used in species identification and biodiversity studies. COI gene application is expected to obtain genetic characteristic, genetic variations and phylogeny of giant featherback. The aim of this research was to analyze genetic diversity of giant featherback Chitala lopis in Lampung and Kalimantan. To analyse genetic distance, Kimura two parameter (K2P) model was performed where to determine nucleotide variation & polymorphism and also reconstructed of phylogenetic tree was used MEGA 7.0 software. Total nine individuals were obtained from three populations, i.e. Lampung, West Kalimantan and South Kalimantan. The results showed that giant featherback has 689 bp conserve, 18 bp variation, 13 bp parsimony-informative, and 2 bp singleton sites from 707 bp COI partial gene. The average within-species, in-group, and out-group based on K2P distances were 1.24%, 1.43% & 1.58% (AP008922.1; KM213054.1), and 13.00% respectively. The Single Nucleotide Polymorphism (SNP) was obtaining from 13 SNP sites. West Kalimantan samples have two SNP (471 and 528 site). The South Kalimantan samples showed more specific nucleotides with nine SNP (120, 129, 144, 201, 306, 324, 474, 615 and 644). Based on genetic distance, the biggest difference was in the South Kalimantan sample (1.58%) compared with Lampung and West Kalimantan. The results of the K2P neighbour-joining phylogenetic tree reconstruction show that the South Kalimantan samples are in a different group. The West Kalimantan sample shows that it is closely related to the Lampung. Abstrak Barkoding DNA berdasarkan gen parsial Cytochrome Oxidase subunit I (COI) mitokondria telah banyak digunakan pada identifikasi spesies dan studi biodiversitas. Penggunaan gen COI mampu memperoleh karakteristik genetik, variasi genetik, dan filogeni. Tujuan penelitian ini adalah menganalisis keragaman genetik spesies ikan belida Chitala lopis asal Lampung dan Kalimantan. Analisis yang digunakan yaitu menghitung jarak genetik dengan model Kimura two parameter (K2P), melihat variasi nukleotida, polimorfisme dan merekonstruksi pohon filogenetik menggunakan software MEGA 7.0. Sebanyak sembilan individu ikan belida dikoleksi dari tiga populasi yaitu: Lampung, Kalimantan Barat dan Kalimantan Selatan. Hasil analisis urutan nukleotida yang didapat menunjukkan 689 pb situs konservatif, 18 pb situs variasi, 13 pb situs parsimoni, dan 2 pb situs singleton dari 707 pb gen parsial COI. Jarak rata-rata intraspesies, ingroup, outgroup berdasarkan K2P adalah 1,24%, 1,43% & 1,58% (Kode akses Genbank: AP008922.1; KM213054.1), 13,00%. Single Nucleotide Polymorphism (SNP) yang diperoleh sebanyak 13 situs. Sampel Kalimantan Barat memiliki dua SNP pada situs (471 dan 528). Kalimantan Selatan memiliki sembilan SNP (situs ke 120, 129, 144, 201, 306, 324, 474, 615 dan 644). Berdasarkan jarak genetik, perbedaan terbesar terletak pada sampel Kalimantan Selatan (1,58%) dibandingkan dengan Lampung dan Kalimantan Barat. Hasil rekonstruksi pohon filogenetik K2P neighbor-joining, menunjukkan bahwa sampel Kalimantan Selatan berada pada grup yang berbeda. Sampel Kalimantan Barat menunjukkan kekerabatan lebih dekat dengan grup Lampung.

Abstract

Penelitian ini bertujuan untuk mengkaji perubahan struktur histologis organ insang, hati dan ginjal ikan nila akibat dari paparan merkuri. Penelitian dilakukan dari bulan Februari hingga Juni 2013. Ikan nila berukuran panjang 11-13 cm dengan bobot rata-rata 20 gram dipaparkan pada konsentrasi sub kronik merkuri klorida (0,164 mgL-1) selama 56 hari. Pembuatan preparat histologis hati dan dilakukan dengan metode histoteknik menggunakan pewarnaan Haemotoxylin dan Eosin. Hasil penelitian menunjukkan adanya pengaruh negatif pada hati dan ginjal ikan nila akibat dari paparan merkuri. Paparan merkuri pada organ insang menyebabkan terjadinya perubahan struktur histologis berupa perbesaran sel organ (hypertrophy), penambahan jumlah sel (hyperplasia) pembengkokan lamela sekunder (curling of secondary lamella), penghimpitan lamella sekunder (fusion in secondary lamella)  dan kematian sel  (neukrosis). Perubahan histologis pada organ hati berupa perbesaran sel organ (hypertrophy), penambahan jumlah sel (hyperplasia), penciutan inti  sel (shrinkage of hepatocytes), pendarahan (hemorage), dan kematian sel (neukrosis).Kata kunci: merkuri klorida, histologis, hypertrophy, hyperplasia, neukrosis.

Abstract

The rearing technology of green catfish larvae has not yet established. Hight mortality occurred in the early larval stages. This experiment was conducted to study the effect of temperature and dissolved oxygen on metabolic rate and survival rate of green catfish larvae. 3600 green catfish larvae with initial individual mean weights of 1.24 ±0.19 mg and lengths 5.85 ±0.71 mm (one day after hatching) were reared in the aquarium of 30 cm x 30 cm x 40 cm dimension. Treatments were done at 3 levels of temperature 27oC, 30oC, and 33oC; and 3 levels of dissolved oxygen 6.47 ml/L, 1.05 ml/L, and 0.78 ml/L and 3 replications. The result showed that temperature and dissolved oxygen was significant to the oxygen consumption. Oxygen consumption of 0.67 ±0.25 mg O2/h/g and 0.86 ±0.005 mg O2/h/g was highest in larval reared at 33oC and dissolved oxygen 6.47 ml/L for 3 weeks. The higher water temperature the greater the oxygen consumption. The oxygen consumption decline with decreasing dissolved oxygen in the water, indicating an axyconformer type. The temperature was not significant to the survival rate and growth of the larvae. However the effect of dissolved oxygen was significant to the growth rate (P<0.05). Growth rate of 25.69 ±0.55% was highest in larval reared at 6.47 mg/L for 3 weeks.

Abstract

Alkaline proteases, amylase and cellulase activities of digestive organ of yellow rasbora, Rasbora lateristriata Blkr., was evaluated with four different feeding levels of 0.34g protein+0.03g fiber, 1.01g protein+0.10g fiber, 1.69g protein+0.16g fiber and 2.36g protein+0.23g fiber/day/100 g fish biomass. A total of 280 fish with average body weight of 0.71±0.06g were used in this study. The results showed that the difference in the feeding levels resulted in a significant difference in trypsin and chymotrypsin  activities (P <. 05), but not for amylase and cellulase activities (P > .05). In conclusion, protein digestion capacity increased, but not to the digestion of starch and fiber in response to different feeding levels and the optimal feeding level for yellow rasbora was 1,01g protein+0,10g fiber/day/100 g fish biomass.

Abstract

This study was to evaluate the digestive enzyme activity  included a total protease, trypsin, chymotrypsin, lipase, amylase, cellulase and alkaline phosphatase in Rasbora. This research was carried out using three different ages (2, 4 and 6 months) and in each age consisted of six groups (replicates). In this study 150 fish (±2 months age), 120 fish (± 4 months age) and 90 fish (± 6 months age) were used. All digestion enzyme activity was measured by the spectrophotometric method, except the lipase activity was by the titration method. The results showed that the distinctness of age resulted in a significant difference on total protease, trypsin, lipase, cellulase and alkaline phosphatase (P <0.05), but no significant difference in amylase activity (P> 0.05). Total protease and trypsin activities were higher in fish of age two months than fish age four and six months, but the activity of lipase, cellulase and alkaline phosphatase were higher in fish age of four months compared to two months age fish. Fish, with distinct age has the different nutrient digestion capacity as expressed by differences in the activity of the enzyme digestion, except amylase. These results contribute to the future development of digestive physiology, especially in Rasbora.Key Words: alkaline phosphatase, carbohydrase, lipase, protease, Rasbora

Abstract

Climbing perch, Anabas testudineus (Bloch) is a potential species for aquaculture in Kalimantan, Indonesia and belongs to Anabantidae family. The development of its digestive tract was evaluated on larvae reared under culture conditions of 28oC-30oC, from hatching to 30 days after hatching using histological and morphological methods. The larvae were kept in six 100-L tanks. They were fed with rotifers and microalgae from day 2nd after hatching to day 10th; Artemia nauplii from day 7th to day 15th; Artemia meta-nauplii from day 15th to day 20th; and Tubifex worm from day 20th onwards. The development of digestive tract in climbing perch followed the general pattern described for other species. Shortly after hatching, its digestive system was found to be consisted of an undifferentiated straight tube laying dorsally to the yolk sac. At first feeding (day 2nd), both mouth and anus had opened and the yolk sac was partially absorbed. On day 3, the digestive tract was fully differentiated into buccopharynx, esophagus, intestine and rectum. The two pyloric caeca appeared on day 25th after hatching, indicating the transition from larval to juvenile stage and acquisition of an adult type of digestion.

Abstract

Waduk Koto Panjang dibangun dengan membendung Sungai Batang Mahat dan Sungai Kampar Kanan. Waduk ini selesai dibangun pada tahun 1996 dan terdapat di Kabupaten Kampar Provinsi Riau. Banyak biodiversitas jenis nekton ditemui baik di perairan mengalir yang menjadi inlet maupun waduknya sendiri. Salah satu nekton yang ditemui di Perairan Inlet Waduk Koto Panjang adalah Ikan Bunga Air. Penelitian ini bertujuan untuk mengidentifikasi dan mengetaui sebaran ukuran Ikan Bunga Air. Pengambilan sampel ikan dilakukan selama empat bulan yaitu dari April – Juli 2010 dengan mengambil 5 lokasi penelitian. Stasiun I Muara Takus, Stasiun II Gunung Bungsu I, Stasiun III Gunung Bungsu II, Stasiun IV Tanjung I dan Stasiun V Tanjung II. Dari hasil penelitian didapatkan bahwa Ikan Bunga Air kedalam Kelas Actinopterygii Ordo Clupeiformes Famili Clupeidae Genus Clupeichtyhs dan Spesies Clupeichthys goniognathus Bleeker, 1855. Sedangkan distribusi ukuran yang dominan secara keseluruhan, ikan jantan dan betina adalah 30 – 47 mm dan 30 – 48 mm.Koto Panjang reservoir built with dam of Batang Mahat and Kampar Kanan river. This reservoir finishing built at 1996 and location in Kampar regency province Riau. Many biodiversity species of necton that finded in this aquatic, in river stream and in reservoir. One of necton that finded in Inlet Koto Panjang reservoir is Sumatran River Sprat. This research aims are for identified and to know distributed size of Bunga Air fish. Take the fish sample during 4 months, from April until July 2010 with five location research, namely Muara Takus, Gunung Bungsu I, Gunung Bungsu II, Tanjung I and the Tanjung II. The result research show Bunga Air fish include Class Actinopterigii Order Clupeiformes Family Clupeidae Genus Clupeichthys and Species Clupeichthys goniognathus Bleeker, 1855. While for the size distributed that dominant for all of it, male fish and female fish are 30 – 47 mm and 30 – 48 mm.

Abstract

Penelitian ini dilakukan di perairan ekosistem mangrove Suaka Margasatwa Muara Angke, Penjaringan Jakarta Utara, pada bulan Februari - April 2012 dengan menggunakan jala dan jaring insang berbagai uku-ran. Selama penelitian berlangsung tertangkap sebanyak 1.535 individu ikan yang terdiri dari 32 jenis yang mewakili 29 marga dan 26 suku. Keanekaragaman ikan-ikan di perairan ekosistem mangrove ini bervariasi antara 1,939-2,673 dengan keanekaragaman tertinggi di danau angke sedangkan terkecil di suaka, dengan dominasi jenis hasil tangkapan Pepetek (Leiognathus equulus).

Abstract

Serayu termasuk sungai besar yang alirannya melewati lima Kabupaten di Jawa Tengah dan terfragmentasi oleh waduk di wilayah Banjarnegara. Informasi mengenai biodiversitas ikan di sungai ini masih sedikit. Penelitian ini bertujuan untuk mengungkap keanekaragaman jenis ikan, potensi, status jenis, dan upaya konservasinya. Penelitian menggunakan metode survei yang dilakukan di tiga zona (di bawah waduk, kawasan waduk, dan di atas waduk). Hasil penelitian ditemukan 22 spesies yang tergolong ke dalam 13 famili, sebagian besar merupakan ikan konsumsi (54,55%), status jenisnya sebagian besar bersifat umum atau mudah ditemukan (81,82%) dan introduksi (18,18%). Ancaman terhadap kelestarian sumber daya ikan di wilayah ini beragam dan diperlukan upaya konservasinya.

Abstract

Ikan pepija merupakan ikan demersal dengan penyebaran di perairan estuaria dan laut dangkal. Penelitian ini bertujuan untuk mengkaji distribusi spasial dan temporal ikan pepija di perairan P. Tarakan. Penelitian dilakukan dari Februari 2013 sampai dengan Februari 2014. Penangkapan ikan dilakukan dengan menggunakan pukat hela (trawl) dengan ukuran panjang sayap 7 meter dengan besar mata jaring pada sayap, badan dan kantong masing-masing berukuran 2,2 dan 1 inch. Pengoperasian jaring trawl pada masing-masing lokasi stasiun dilakukan “zig zag” dengan 2 kali masa penarikan pukat hela (towing) selama 30 menit. Ikan yang tertangkap ditimbang seluruhnya. Hasil tangkapan bervariasi saat waktu pengamatan dan antara satu stasiun dengan stasiun lainnya. Laju tangkap tertinggi pada bulan Desember dan Januari sebesar 75.56 dan 77.37 kg/jam dan terendah pada bulan April sebesar 7.41 kg/jam. Ikan pepija melakukan migrasi harian dari Tanjung Simaya (tanggal 7 kalender Hijriah), tanggal 8 di perairan Tanjung Selayu, tanggal 9 antara perairan Tanjung Selayu dan Tanjung Juata, dan tanggal 10 pada penanggalan Hijriah di perairan Tanjung Juata. Berdasarkan data tangkapan tersebut terungkap bahwa distribusi ikan pepija di perairan Pulau Tarakan berkaitan dengan pasang surut, ikan ini hanya ditemukan pada saat pasang perbani pada tanggal 7, 8, 9 dan 10 bulan Hijriah.
