Abstract

The aim of this study was to test whether high intensity swimming exercise on young white mice affect memory retention. A Randomized Post-Test Only Control Group Design was used. Subjects were 20 male rats (Rattus norvegicus wistar strain), age 1 to 1.5 months, body weight 60-100 grams, which were divided randomly into 2 groups (each consist 10 rats). The treatment group got the high intensity swimming exercise (plus 9% of body weight added load). Each treatment was conducted once a day during 1 minute and 45 seconds, three sets with rest period three times the exercise time, for four weeks. Memory retention was measured through a Y- maze test in two sessions, each with six replications. Results showed that memory retention between groups on day 4, was (65.00 ± 31.38)% in the control group, and (30.56 ± 26.68)% in the treatment group. Independent t-test results revealed significant differences between the control group and the treated group (p = .020). The conclusion is that high intensity exercise decline memory retention in young white rats. Perlakuan diberikan satu kali per hari selama 1 menit 45 detik, tiga set dengan waktu istirahat tiga kali lama waktu latihan, selama empat minggu. Retensi memori diukur melalui test Y-maze selama dua sesi, masing-masing enam kali pengulangan. Abstract

Staphylococcus aureus (S. aureus) is the most common pathogen in humans that causes various infections. A variety of infections due to S. aureus are related to virulence factors, one of which is Toxic Shock Syndrome Toxin-1 (TSST-1) as the main cause of Toxic Shock Syndrome (TSS). This study aimed to explore the genetic pattern of the tsst-1 genes among MSSA and MRSA from clinical isolates of patients at Dr. Soetomo General Academic Hospital, Surabaya-Indonesia. This study performed a PCR test on 106 clinical samples of S. aureus isolated to determine the presence of the tsst-1 genes; subsequently, this study also performed phylogenetic analysis. The positive tsst-1 genes were found in 3 (5.7%) MSSA isolates and 2 (3.8%) MRSA isolates from 106 samples that were divided into 53 MSSA isolated and 53 MRSA isolates. There was no significant difference between the tsst-1 genes on both the MSSA and MRSA bacteria (p = 0.648). The positive tsst-1 genes were found in 1 (4.5%) of pus specimens and 2 (16.7%) of blood specimens from MSSA isolates. However, the positive tsst-1 genes were found in neither sputum nor urine specimens. The genes were found in 2 (16.7%) of blood specimens from MRSA isolates. Based on the phylogenetic tree, Indonesian tsst-1 isolated belonged to the same clade as Japan, Iran, Iraq, India, Egypt, and the United Kingdom. The prevalence of tsst-1 genes of both MSSA and MRSA from clinical isolates in Dr. Soetomo General Academic Hospital Surabaya was 5.7% and 3.8%, respectively. The tsst-1 genes of Surabaya-Indonesia, Japan, Iran, Iraq, India, Egypt, and the United Kingdom belonged to the same clade.

Abstract

Cases of overweight and obesity in the world increased continuously. In 2016, obesity increased by 3% in men and 6% in women compared to 1975. Overweight cases also increased by 20% in men and 23% in women compared to 1975. Overweight and obesity have been linked to an increase in adipose tissue in the body. Increased adipose tissue associated with inflammation, which one of its characteristics is an increase levels of C - Reactive Protein (CRP). This article aim is to describe the mechanism of physical exercise to diminish CRP level in overweight and obesity. Adipose tissue produces and releases various pro-inflammatory and anti-inflammatory factors such as leptin, adiponectin, resistin, TNF-α, IL-6, MCP-1 and CRP. One of the prevention and treatment of inflammatory for overweight and obesity cases is to do physical exercise. In cases of overweight and obesity, the physical exercise aims to increase energy expenditure. Physical exercise decreases the volume and amount of adipose and pre-adipose tissue as well as the number of endothelial cells and macrophages in adipose that contain pro-inflammation such as IL-1, TNF-α, CRP, serum amyloid protein (SAA), and cytokines. Physical exercise rises anti-inflammatory properties such as IL-10, IL-1ra which play a role in inhibiting the transduction of IL-1β signals and inhibiting TNF-α synthesis. Physical exercise also amplifies antioxidant enzymes such as SOD and GPX. The antioxidants play a role in fighting free radicals to reduce inflammation.

Abstract

Highlights: • This article reports the use of the Y-maze as a simple yet effective method to measure spatial working memory in mice. • The Y-maze method can be used safely without exposing the animals to additional stressors, as evidenced by the absence of mortality following the test. Abstract: Spatial working memory (SWM) in humans and animal models with impaired cognitive functions has been analyzed through a number of methods. However, this is still understudied in animal models treated with a high-calorie diet (HCD) and moderate-intensity exercise (MIE). The Y-maze was utilized as the assessment method in this study. A 40 x 9 x 9 cm3 Y-maze was employed to observe the animal models' spontaneous alternation (SA) as the representation of their SWM. This was done by calculating the total alternation percentage divided by total entry minus two. A total of 17 female Mus musculus mice aged 8 weeks were tested in the Y-maze to investigate their SWM using SA calculations. Each mouse was analyzed for eight minutes and recorded in a dark and quiet room to minimize bias due to environmental noise and lighting. Comparing the treatment group's (HCD+MIE) SA to the control group's SA revealed no statistically significant difference (p=0.451). Seven mice in the treatment group performed similarly to the mice in the control group in the Y-maze test, with no significant difference in their ability to complete the task. The mice in the treatment group exhibited no motor impairment, as indicated by complete movements of all their extremities while exploring the Y-maze within the allotted time. In conclusion, the Y-maze can be used as a reliable method to analyze SWM in overweight/obese Mus musculus animal models treated with moderate-intensity physical exercise.

Abstract

Obesity becomes a global epidemic nowadays. The high-fat diet is used as an alternative therapy for obesity. The optimal composition of a high-fat diet to reduce body weight is still unknown. This study aimed to determine which components of a high-fat diet can decrease body weight, visceral fat, and PPARG expression of visceral fat. This study was conducted at the Faculty of Veterinary Medicine, Universitas Airlangga, for three months by using a randomized post-test only control group design. Fifty male mice, 2-3 months old, 18-30 grams were adapted for one week given standard diet AIN93-M, then mice were divided into five groups, namely K1 (control group, 12% fat, 20% protein, 62% carbs); K2 (30% fat, 60% proteins, 0% carbs); K3 (45% fat, 45% protein, 0% carbs); K4 (60% fat, 30% protein, 0% carbs); and K5 (75% fat, 15% protein, 0% carbs). Bodyweight was measured before and after treatment, then the visceral fat and PPARG expressions were evaluated. Statistical comparisons were performed using Statistical Package for the Social Sciences (SPSS) software. After treatment, there were forty-three mice. The body weight and visceral fat weight of the mice with a high-fat diet were decreased. The most significant changes in body weight were in K4 with -9,60 ± 3,806 grams reduction. The bodyweight of mice in K5, slightly increased than K2-K4. This could be caused by the hormesis phenomenon. PPARG expressions decreased in groups with a high-fat diet but increased in K5. The composition of a high-fat diet in group K4 was the most optimal to decrease the body weight, visceral fat, and PPARG expressions in mice
