Abstract

Dendrobium is great interest because of its high value as a floricultural commodity and a kind of elite herbage plant in Indonesia. An efficient in vitro propagation system via protocorm-like body (PLBs) induction has been developed using one-year-old axillary node explants. The potential of different media and supplements triptone for reliable Dendrobium orchid plantlet regeneration from in vitro PLBs explants were studied. In all two media under investigation, shoot regeneration was achieved when the PLBs were transferred onto solidified MS half strenght and Growmore medium with or without 2 g L-1 triptone supplemented with 15% (v/v) coconut water, 20 g L-1 sucrose, and 2 g L-1 charcoal. The shoots development investigation after three month of culture. 1/2 MS and Growmore media significantly influenced the plantlet regeneration of Dendrobium orchid. Among the media, Growmore showed better performance on shoots and leaves per plantlet, but triptone showed no significant effect on number shoots and leaves. The longest shoots were found in Growmore medium without triptone. Key words: Dendrobium, in vitro, PLBs, planlet, triptone.

Abstract

Two consecutive experiments that aimed to investigate effects of basal media and activated charcoal (AC) on (1) in vitro seed germination and in vitro seedling growth of Phalaenopsis hybrids were conducted. Phalaenopsis seeds taken from sterilized ripe pods were sown aseptically on MS or Growmore medium with or without 2 g/l AC, then the protocorms produced were counted after two months of culture. The same combinations of basal media and AC were used to grow protocorms produced from previous experiment for another two months, then the growth of seedlings were measured. The two experiments were arranged in a completely randomized design with three replicates. Each experimental unit consisted of four culture bottles each of which contained the same amount of seeds (in the first experiment) or five protocorms (in the second experiment). Data were subjected to analysis of vari-ance and if there was any significant F result (s), the means were separated using least significant difference at P? 0.05. Results of the first experiment showed that for in vitro germination of Phalaenopsis seeds, Grow-more medium produced significantly higher number of protocorms campared to MS, and addition of AC in both media significantly decreased the number of protocorms. Results of the second experiment showed that for in vitro growth of Phalaenopsis seedlings, either MS or Growmore medium resulted in the same seedling growth. However, addition of 2 g/l AC in the media significantly increased seedling growth as shown by higher values in plant height, number and length of roots as well as seedling fresh weight. So, it was concluded that Growmore medium was better than MS for in vitro seed germination and the same as MS for seedling growth of Phalaenopsis. The addition of AC in the medium was inhibitory for seed germination, but stimulatory for seedling growth. Keywords: activated charcoal, basal media, Phalaenopsis, seed germination, seedling growth.

Abstract

The increasing number of Gladiol Flower demand every year in Indonesia is not comparable with the low productivity of Gladiol Edible root. The technique of growing stimulation by using growth controller essence becomes one alternative to increase the number of sprout and edible root of Gladiol. This research is being purposeful to know the concentrate, the long term of submersion, and the combination between the concentrate and the best long term kinetin submersion which could increase the number of sprout and edible root of Gladiol (Gladiolus hybridus L.). The treatment is done by factorial (3 x 2) in Perfect Disordered Group Arrangement with four times repetition. The first factor is kinetin concentrate, those are 30, 40, and 50 ppm, the second factor is the long term of kinetin submersion, and those are 24 and 48 hours. The result of the research shows that there is no real influence between the treatments of concentrate and the long term kinetin submersion. The percentage of germinated edible root, the number of edible root, the height of the plants, the number of leaves, the number of beet, the weight of beet, the diameter of beet, the number of beet offspring, the weight of the offspring, and the dry weight of one root of beets.Key words: kinetin, multiplication, Gladiolus

Abstract

Indonesia is one of the largest cassava producing countries in the world with various types and advantages. Waxy clones have a high starch content and therefore they can be used as a food substitute for rice. Tissue culture through somatic embryogenesis is an effective technique for rapid and mass seedling propagation. This research aimed to determine the effect of the type and concentration of auxin 2,4-D or picloram and the addition of NAA on the formation of Waxy cassava clone somatic embryos. This research used a completely randomized design (CRD) which was arranged in a non-factorial. Auxin 2,4-D and picloram were used at four concentration levels: 0 mg.l-1, 8 mg.l-1, 10 mg.l-1, 12 mg.l-1, and additional NAA treatment of 6 mg.l-1 each. Results showed that MS + 6 mg.l-1 NAA treatment produced the highest callus weight. Meanwhile, the percentage of embryonic callus and number of embryo was higher in the treatment of MS + picloram 12 mg.l-1 and NAA 6 mg.l-1 (19,05 ± 5,82 %) and MS + 8 mg.l-1 2,4-D + 6 mg.l-1 NAA (11,11 ± 3,17 %) with 5,48 ± 3,56 and 4,93 ± 1,95 embryos, respectively. Thus, auxin picloram 12 mg.l-1 + NAA 6 mg.l-1 were the most effective in inducing the formation of somatic embryos on waxy cassava clone.

Abstract

This experiment aimed to study the formation of scalps and  banana shoots 'Tanduk' in vitro in response to thidiazuron (TDZ). Shoot explants measuring ± (1 x 1 x 1) cm3 were cultured in precondition media (MS+ 5 mg/l benzyladenine) for 4 weeks, then transferred to treatment media, namely MS + TDZ. This experiment was carried out using a completely randomized design with 3 replications and 7 treatments of TDZ concentrations (0.5; 1.0; 1.5; 2; 2.5; 3; 3.5; 4.0 mg/l).  Each experimental unit consisted of 5 bottles, each containing one explant. Observations of scalp number were carried out at 8 weeks of age. The scalps were subcultured on MS+5mg/l BA media to induce shoots. After 8 weeks in the shoot induction medium, the number of shoots was recorded.  The results showed that at 8 weeks on the culture all of the explants formed scalps, with the highest number of scalps (6 scalps/explant) was at 1 mg/l TDZ.  The scalp number decreased to 4.4 to 3.5/explant as the TDZ concentration increased from 1.5 to 4 mg/l. After 8 weeks on shoot induction media containing 5 mg/l BA, the highest number of shoots (3.8 shoots/explant) was obtained on the scalp from the 0.5 mg/l TDZ treatment. The shoot number decreased to (3.2-1.9/explant) with increasing TDZ concentrations from 1.0 to 4.0 mg/l.Keywords: Tanduk, plantain, thidiazuron, in vitro, scalp.

Abstract

The growth and production of cassava can be increased through various efforts, including the use of superior varieties, fertilization, the addition of organic materials, and the application of growth regulators (PGR). Naphthaleneacetic acid (NAA) and indole-3-butyric acid (IBA) are PGRs that have been widely documented to stimulate root initiation and development in cuttings. The application of aauxin mixtures on cassava cuttings followed by fertilization and the addition of organic materials is expected to improve the growth and yield of cassava. The aim of this experiment is to study the effects of the application of an NAA and IBA mixture (1:1) and the type of fertilizer on the growth and yield of the Garuda variety of cassava. This research is designed using a clustered randomized block method with three replications. The design used is factorial 2x3, consisting of two main treatment factors. The first factor is the combination of growth regulators NAA and IBA in a 1:1 ratio at three concentration levels, namely 0 ppm, 500 ppm, and 1,000 ppm. The second factor includes the type of fertilizer used, which is a single inorganic fertilizer and a combination of inorganic fertilizer with cow manure. Each experimental unit in this study consists of 10 cassava cuttings planted in an orderly manner with a planting distance of 1 meter x 1 meter between plants. The results of the experiment showed that the application of inorganic fertilizer produced plant growth and the number of productive roots equivalent to the use of inorganic fertilizer + cow manure. The application of a mixture of auxins NAA and IBA (1:1) at concentrations of 500 ppm and 1,000 ppm significantly increased the growth of shoots, the number and weight of productive roots of the Garuda variety cassava, but both treatments resulted in cassava growth and yield that were not different. Abstract

Rice growth and yield can be increased through various efforts, including the use of "high-yielding varieties, fertilization, soil conditioners with application of plant growth regulators (PGRs). Soil conditioners namely as biochar are carbon products that can enhance plant productivity through enhancing the physical, chemical, and biological characteristics of the soil, while cytokinine benzyladenine (BA) is a PGR that has been widely documented to stimulate cell growth, cell division, and differentiation in rice plants. The purpose of this study was to determine the effect of biochar and benzyladenine (BA) on the growth and yield of the IR Nutrizinc rice variety. Data collection was carried out in October 2024 - February 2025 in Kampung Payung Rejo, Pubian District, Central Lampung Regency. The treatments were arranged factorially (2x3) using a completely randomized design with 3 replications. Factor I was without biochar and given 20 tons/ha of biochar. The second factor was 3 benzladenin (BA) concentrations, namely 0 ppm, 50 ppm, and 100 ppm applied by spraying on the surface of the plant at 20, 30, and 40-days after transplanting, while the biochar application was implemented 2 weeks before transplanting. Observed variables included plant height, maximum tillers per clump, count of productive tillers, length of panicles, and grains per panicle, percentage of empty grains per panicle, dry harvested grain yield, dry harvest grain yield, 1000 grain weight and dry weight of the crownThe Bartlett test was employed to assess the homogeneity of variance among treatments then analysis of variance was carried outIf a notable difference existed between treatments, the BNT test was carried out at the 5% significance level. The findings indicated that applying 20 tons/ha of biochar enhanced the yield components in terms of dry harvested grain, dry milled grain, and dry shoot weight The application of 50 ppm BA was able to increase the growth and yield of grain compared to without benzyladenine (BA). The combination of biochar and BA enhanced the growth and yield of grain in terms of the number of grains per panicle, dry harvested grain, and dry milled grain.
