Abstract

This study aimed to determine the addition of duck egg yolk in skim milk – fructose extender on sperm motility and viability of Muscovy duck (Cairina moschata) at 5°C storage. Ejaculates of Muscovy duck were divided into four portions equally. The first portion was for the control group (diluted in skim milk – fructose extender), and the second, third, and fourth portions were for the treatment groups of T1, T2, and T3 (diluted in skim milk – fructose extender added with 2.5, 5.0, and 7.5% v/v duck egg yolk). Sperm motility and viability were evaluated two hourly until the motility percentage reached 40%. The data were analyzed using ANOVA and Duncan test. The result showed that the highest percentage of spermatozoa motility and viability (p<0.05) was in the T2 group compared to the other groups. It could be concluded that the Muscovy duck semen extended in skim milk - fructose with 2.5 to 7.5% egg yolk of duck qualified for artificial insemination at storage up to 10 hours in 5°C storage.

Abstract

This study aims to determine the effect of administration of ciplukan (Physalis angulate Linn.) leaf extract on the variables of testicular weight, seminiferous tubules diameter, and spermatozoa plasma membrane integrity of rats (Rattus norvegicus) exposed to heat as model animals. Rats were divided randomly into five groups. In the NC group, rats were not exposed to heat and were only given 1% Na-CMC. In the PC, T1, T2, and T3 rats were exposed to heat followed by administration of 1% Na-CMC, 100, 200, and 400 mg/kg bw of ciplukan leaf extract (CLE) in 1% Na-CMC. Heat exposure at 40°C was conducted for 60 minutes every day for 21 days. On day-22, all rats were sacrificed for testicular evaluation. The results showed that all variables in the PC group were lower (p <0.05) than in the NC group. All variables in the T3 group were higher (p <0.05) than in the PC group. Testicular weight and spermatozoa plasma membrane integrity in the T3 group were not significantly different (p >0.05) from the NC group. However, the diameter of the seminiferous tubules in the T3 group were lower (p <0.05) than in the NC group. This study revealed that the administration of ciplukan leaf extract as an antioxidant increased testicular weight, seminiferous tubules diameter, and spermatozoa with intact plasma membrane in rats exposed to heat.

Abstract

One of the many efforts to increase the quality of livestock genetics is through artificial insemination (AI). Other than increasing it, AI can be conducted to preserve semen. A successful AI is determined by seminal quality, therefore, a method to preserve semen for a longer storage time is needed. The method used is adding an extender that fulfills prerequisites for a semen extender such as coconut water combined with egg yolk citrate extender. Coconut water is rich in carbohydrates, protein, vitamins, and antioxidants while egg yolk contains lecithin. This study aims to find out the Pote buck spermatozoa quality stored in coconut water and egg yolk extender. This study uses three groups of treatments (T0: 0.1 ml semen + 0.9 ml egg yolk citrate, T1: 0.1 ml semen + 0.9 ml coconut water, and T2: 0.1 ml semen + egg yolk citrate (20%) + coconut water). All three of these are stored at 5oC and evaluated every day until day 5 of their motility, viability, intact plasma membrane, abnormality, and MDA level. Data analysis used is ANOVA and a further test called BNT is conducted if a significant difference is determined. No significant difference was found between T0 and T1 (p>0.05). The highest progressive motility, viability, and intact plasma membrane (%) among the three groups of treatments happened to be from T2. Meanwhile, a low percentage of spermatozoa abnormality and MDA level were also found in T2 with its extender being coconut water combined with egg yolk citrate. To conclude, the best extender for storing Pote buck semen is stored at 5oC is coconut water combined with egg yolk citrate extender.

Abstract

This study aimed to evaluate the spermatozoa motility and viability of turkey semen extended with egg yolk nanoparticles-PBS extender at various concentrations, stored at 3-5°C. Semen was collected from two turkeys and divided into four treatment groups. Group T0 consisted of fresh turkey semen extended with 20% (v/v) fresh egg yolk in phosphate-buffered saline (PBS). Groups T1, T2, and T3 consisted of fresh turkey semen extended with 5, 10, and 20% (v/v) egg yolk nanoparticles in PBS, respectively. Extended semen was stored in a refrigerator (3-5°C), and progressive spermatozoa motility and viability were evaluated every two hours until spermatozoa motility decreased to 40%. The results showed that turkey semen in T3 extender containing 20% egg yolk nanoparticles in PBS was able to maintain spermatozoa motility for up to eight hours, better than T0 and T2 extenders which supported spermatozoa motility for six hours. In contrast, T1 extenders only support spermatozoa motility for four hours. T3 extenders were also able to maintain spermatozoa viability for up to eight hours, while T0, T1 and T2 extenders could only support viability for 6 hours. The extender with 20% egg yolk nanoparticles in PBS was significantly better (p <0.05) in maintaining spermatozoa quality compared to those with 5 and 10% egg yolk nanoparticles, as well as the extender with 20% fresh egg yolk. In conclusion, 20% egg yolk nanoparticles in PBS semen extender was effective in maintaining the motility and viability of turkey (Meleagris gallopavo) spermatozoa stored at 3-5°C for up to eight hours.

Abstract

Monosodium glutamate (MSG) could cause increased production of reactive oxygen species (ROS), causing oxidative stress in the testicles, which adversely affected sperm quality. Dayak onion extract which is known for its high antioxidant content, could help alleviate oxidative stress caused by MSG. This research examined the effect of Dayak onion (Eleutherine palmifolia) on sperm abnormalities and plasma membrane integrity in mice (Mus musculus) exposed to MSG. Twenty-five male mice, 8 weeks old with a body weight approximately 20g, were divided into five groups, C– (received 0.5% Na-CMC), C+ (received 4 mg/g bw MSG), and three treatment groups (T1, T2, T3) which were given 4 mg/g bw MSG accompanied by Dayak onion extract at doses of 30, 60, and 120 mg/kg bw respectively. All treatments lasted for 52 days. Significant differences (p <0.05) in sperm abnormalities and plasma membrane integrity were observed among the groups. Sperm abnormalities found were (12.3 ± 1.92), (61.1 ± 3.10), (41.6 ± 2.87), (30.4± 1.91), and (18.2 ± 2.10) % respectively for C–, C+, T1, T2 and T3. Meanwhile, sperm plasma membrane integrity found were (33.0 ± 3.24), (69.3 ± 2.32), (41.8 ± 2.42), (55.4 ± 3.11), and (64.2 ± 1.27) % respectively for C–, C+, T1, T2 and T3. These results indicate that Dayak onion extract could help reduce sperm abnormalities and maintain plasma membrane integrity in mice exposed to MSG.
