Abstract

This study aims to determine the effect of administration of ciplukan (Physalis angulate Linn.) leaf extract on the variables of testicular weight, seminiferous tubules diameter, and spermatozoa plasma membrane integrity of rats (Rattus norvegicus) exposed to heat as model animals. Rats were divided randomly into five groups. In the NC group, rats were not exposed to heat and were only given 1% Na-CMC. In the PC, T1, T2, and T3 rats were exposed to heat followed by administration of 1% Na-CMC, 100, 200, and 400 mg/kg bw of ciplukan leaf extract (CLE) in 1% Na-CMC. Heat exposure at 40°C was conducted for 60 minutes every day for 21 days. On day-22, all rats were sacrificed for testicular evaluation. The results showed that all variables in the PC group were lower (p <0.05) than in the NC group. All variables in the T3 group were higher (p <0.05) than in the PC group. Testicular weight and spermatozoa plasma membrane integrity in the T3 group were not significantly different (p >0.05) from the NC group. However, the diameter of the seminiferous tubules in the T3 group were lower (p <0.05) than in the NC group. This study revealed that the administration of ciplukan leaf extract as an antioxidant increased testicular weight, seminiferous tubules diameter, and spermatozoa with intact plasma membrane in rats exposed to heat.

Abstract

The purpose of this study was to determine the effect of administering a dose of gonadotropin (PG-600) in cows with persistent corpus luteum (PCL) which were treated with prostaglandin F2 α (PGF2α) and human chorionic gonadotropin (hCG). This study used 20, three-year-old cows diagnosed with PCL, had recently given birth, and were not pregnant. Cows were randomly allocated into groups T0, T1, T2 and T3.  Cows in all groups were injected with PGF2α (25 mg/cow). Simultaneously, cows in groups T1, T2 and T3 were also injected with PG-600 at doses of 100, 150 and 200 IU/cow respectively. PGF2α and PG-600 were injected when PCL was identified. After that, cows showing signs of estrus were injected with hCG (100 IU/cow) and artificially inseminated 12-18 hours after the appearance of signs of estrus. AI was conducted using frozen thawed semen. Sixty days following AI, a pregnancy examination was conducted using USG. Estrus was detected in all treated cows, while the pregnancy rates varied. Compared with other groups, the groups given 100 and 150 IU PG-600 produced higher pregnancy rates. It could be concluded that the combination of 25 mg PGF2α, 150 IU PG-600, and 100 IU hCG was the most effective treatment to produce high pregnancy rates in cows with persistent corpus luteum.

Abstract

This study investigates the impact of mulberry leaf extract on the viability, plasma membrane integrity, and motility of spermatozoa from male white rats (Rattus norvegicus) exposed to e-cigarette smoke. A total of twenty-five male rats were divided into five groups: negative control (NC), positive control (PC), T1, T2, and T3. All groups, except for the NC group, were exposed to e-cigarette smoke. Rats in the T1, T2, and T3 groups received mulberry leaf extract in doses of 100, 200, and 400 mg/kg bw, respectively, while the NC and PC groups were given a placebo of 1% Na-CMC. Both the mulberry leaf extract and the placebo were administered daily, beginning three days prior to the start of e-cigarette smoke exposure, which lasted for 28 days. Results showed that spermatozoa motility, plasma membrane integrity, and viability in the experimental groups were significantly lower than those in the NC group (p <0.05). Conversely, rats in the T1, T2, and T3 groups that received mulberry leaf extract demonstrated significantly greater spermatozoa viability, plasma membrane integrity, and motility compared to the PC group (p <0.05). The T3 group exhibited the most pronounced improvements, with significantly enhanced spermatozoa viability, membrane integrity, and motility (p <0.05) relative to the PC group. These results indicate that mulberry (Morus alba L.) leaf extract enhanced spermatozoa viability, plasma membrane integrity, and motility in white rats (Rattus norvegicus) subjected to e-cigarette smoke.

Abstract

Pyometra is a reproductive disease that most often affects cats which is caused by bacterial infection of the uterus. This study aims to determine the risk factors that could influence the occurrence of pyometra in cats. The study was conducted at the Four Leg Pet Care, Asa Animal Clinic, and K-5 Animal Care in Kediri regency in December 2021 to February 2022. This retrospective cohort study took secondary data for the period 2020 to 2021 as research material and found 35 cats diagnosed with pyometra. The odds ratio was calculated to analyze the associations to some factors including age, body weight, use of hormonal contraception, frequency of administration of hormonal contraception, hormonal contraceptive drugs used, dose of hormonal contraception given.  Analysis of the relationship between risk factors for pyometra showed that the factors age, use of hormonal contraception, and frequency of giving hormonal contraceptives each have p values ​​of 2.17, 2.13, and 2.13, respectively. It could be concluded that age, use of hormonal contraception, and frequency of administration of hormonal contraception influenced or had an association with the risk of pyometra in cats.

Abstract

This study aimed to know the effect of cloves (Syzigium aromaticum) methanol extracts as an antioxidant on the germ cells of white rats (Rattus norvegicus) with induced cryptorchidism. The subjects of this study were 24 male white rats aged 21 days, who were divided into six groups. Rats were adapted for 5 days, after which cryptorchidism were induced. Cloves extract was given for 18 days and 36 days. On day-19 and day-37, rats were sacrificed and testicles were taken for Protein Gene Product (PGP) 9.5 detection by immunohistochemistry. These groups consisted of negative control group (given distilled water and sham surgery), positive control group (induction of cryptorchidism and given distilled water), and T treatment group (induction of cryptorchidism and given 70mg/kg bw cloves extract. Kruskal-Wallis test indicated a significant difference (p <0.05). The differences between treatments were shown based on the outcome of the Mann-Whitney test. Cloves acted as an antioxidant for cryptorchid testicular germ cells. It could be concluded that administering methanol extract of clove flowers could ameliorate the expression of PGP 9.5 in the testicular germ cells of white rats in a model of unilateral cryptorchidism.

Abstract

Livestock production requires Sapudi rams, a breed native to Indonesia, to meet meat demand and food security. In artificial high-quality frozen semen is needed to spread Sapudi rams. To maximize the survival of spermatozoa during cryopreservation, semen should be stored in an extender. Green tea leaf extract (GTLE) and skim milk-goat egg yolk (SM-GEY) may be a good cryoprotectants due to their antioxidant properties. This study aimed to determine the effect of adding GTLE to the SM-GEY extender on the quality of Sapudi ram spermatozoa stored at 5°C. The fresh semen sample was divided into four different GTLE treatment groups, which each contained a 0.1 mL semen sample and a 25-mL extender of SM-GEY. Group T0: no GTLE added to SM-GEY; Groups T1, T2, and T3: 0.1 mL semen diluted in 25 mL SM-GEY with 0.05, 0.10, and 0.15 mg GTLE. Extended semen was then stored at 5°C, and its quality was evaluated daily for five days. The variables observed included spermatozoa motility, viability, and membrane integrity. Data were analyzed using one-way analysis of variance followed by Duncan's test using Statistical Program and Service Solution version 23. The result of this study was that adding 0.05 mg GTLE to 25 mL of SM-GEY extender significantly maintained the spermatozoa motility, viability, and plasma membrane integrity of Sapudi ram spermatozoa for three days at 5°C (p <0.05). Therefore, it could be concluded that adding 0.05 mg of GTLE to the SM-GEY extender preserved Sapudi ram spermatozoa's motility, viability, and membrane integrity for three days at 5°C.

Abstract

This study aims to determine the motility and viability of kampung rooster (Gallus domesticus) spermatozoa in 5% Dextrose Ringer's solution as an extender with or without the addition of various egg yolk concentrations at 5ºC during storage. Semen was collected by massage in the morning. In this study, four types of extenders were used, i.e., 5% Dextrose Ringer's extender without egg yolk, 5% Dextrose Ringer's extender with 5, 7.5, and 10% egg yolk for T0, T1, T2, and T3, respectively. Then, 1000 IU Penicillin and 1 mg Streptomycin were added per mL extender. Fresh semen from each rooster was evenly divided into four volumes to be diluted 11 times in each group extender. Extended semen from all groups was stored at 5°C. Data were analyzed using ANOVA and Duncan's new multiple range test. The result showed a significant difference (p <0.05) between each treatment group for spermatozoa motility and viability. In conclusion, this study revealed that the addition of various concentrations of egg yolk into 5% Dextrose Ringer's solution as an extender maintained the motility and viability of kampung rooster spermatozoa.

Abstract

This study aims to determine the effect of adding melon (Cucumis melo L.) juice to Lactated Ringer's-egg yolk extender on the intact plasma membrane (IPM) and morphological abnormalities of native rooster spermatozoa. Five ejaculate each from two native roosters (Gallus gallus domesticus) were divided into four equal volumes. Ejaculate was extended 1:10 (v/v) in Lactated Ringer's-egg yolk without or with the addition of of 2, 4, and 8% (v/v) melon flesh juice, respectively for groups T0, T1, T2, and T3. Extended semen was stored in a refrigerator (5°C) and evaluated every two hours. The results showed that IPM of all groups decreased (p <0.05) consistently, lasting more than 40% for six hours in the T0 and T1 groups and four hours in the T2 and T3 groups. The percentage of spermatozoa morphological abnormalities in all groups stored for six hours was higher (p <0.05) than before storage. However, it was not significantly different (p >0.05) from that which had been stored for two and four hours. It was concluded that native rooster semen extended in Lactated Ringer's-egg yolk without (group T0) or with 2% melon flesh juice (group T1) and stored at 5°C retained spermatozoa plasma membrane integrity of more than 40% and morphological abnormalities of less than 20% for six hours. While the addition of 4 % (T2) and 8% (T3) melon flesh juice maintained the percentage of IPM and spermatozoa morphological abnormalities for only four hours.

Abstract

This study aims to determine the effect of the administration of white guava (Psidium guajava L.) fruit juice on spermatozoa plasma membrane integrity (PMI), morphological abnormality, viability, and motility of lead acetate induced rats (Rattus norvegicus). Twenty-five male rats were divided into five groups: NC (negative control) group, rats were administered with distilled water twice daily at four-hour intervals; T0 (positive control) group, rats were administered daily with lead acetate 50 mg/kg bw and distilled water four hours later; T1, T2, and T3 groups, rats were administered daily with lead acetate 50 mg/kg bw and 0.5 mL of 25, 50, and 100% white guava fruit juice four hours later. The treatment of the rats was conducted for 14 days, and on day 15, all rats were sacrificed to assess the spermatozoa quality. Data was analyzed using ANOVA followed by Duncan's multiple range test at a confidence level of 95%. The results showed that exposure to lead acetate (T0) caused lower spermatozoa PMI, viability, and motility as well as higher spermatozoa morphological abnormalities (p <0.05) compared to those of the T0 group. Administration of white guava fruit juice starting at a dose of 25% (T1) resulted in higher spermatozoa motility, viability, and PMI as well as lower spermatozoa morphological abnormalities (p <0.05) compared to rats in the T0 group. It could be concluded that white guava fruit juice maintained the spermatozoa quality of lead acetate induced rats.

Abstract

This research aims to determine reproductive data on Sumatran elephants (Elephas maximus sumatranus) in the ETC and ERU of Way Kambas National Park, Indonesia during 1988-2021. Data recorded from the elephant population at both locations (ETC and ERU) includes the number, gender of elephants, elephant calves, ages and birth dates. Calving intervals and service periods were calculated from calving records of cows with a minimum parity of two. Data collection produced data on 47 elephant calves from 13 female elephants with at least twice the parity and 12 primiparous cows. Elephant reproductive records at ETC and ERU were: age (37.44 ± 9.03 vs. 29.75 ± 3.30 years), parity (2.78 ± 1.09 vs. 2.50 ± 0.58), and age at first birth (18, 11 ± 3.92 vs. 17.75 ± 0.50 years). While the calving interval was 1857.56 ± 870.81 vs. 1833.00 ± 305.18 days, and service period respectively 1229.44 ± 846.18 vs. 1210.50 ± 283.59 days, respectively. It can be concluded that the calving interval and service period for captive elephants at ETC and ERU were not much different and were within the normal range. Young cows showed better reproductive efficiency than older cows. Monitoring calves is very important. Further assistance is needed to improve elephant mobile veterinary services, increase diagnostic laboratory capacity, and educate camp managers, veterinary assistants, and mahouts about elephant diseases, their monitoring and treatment. In addition, the use of reproductive technology such as monitoring ovulation using ultrasound and carrying out artificial insemination was expected to increase reproductive efficiency.
