Abstract

The objective of this study was to determine the effect of centella leaf extract administration on decreased of the molecule cAMP responsive element modulator (CREM) expression in the testicular seminiferous tubules of male rats (Rattus norvegicus). Eight rats, aged 3.5 months with 150-250 grams of body weight (BW) were used in this study. All rats were divided randomly into four groups as if K0 as a control group whereas K1, K2, and K3 were given the centella leaf extract with doses 125, 250, and 500 mg / kg body weight respectivelly that given once daily for 30 days. At the end of the treatment, rats were sacrificed and their testes were collected and subsequently fixed in buffered neutral formalin (BNF) 10% as fixative solution for histological preparation. The CREMs expressions were detected using immunohistochemical methods. The results showed that the number of CREM expression in the seminiferous tubules significantly differ (P 0.05) between K0 and the treatment group (K1, K2, and K3). Conclusion, the administration of centella leaf extract with of the dose 125, 250, and 500 mg/kg BW can decreased CREM expression spermatids of testicular seminiferous tubules in male rat.

Abstract

The objective of this study was to identified the development of the immunoreactive follicle stimulating hormone (ir-FSH) cells in pituitary glands of rats (Rattus norvegicus)in different age using immunohistochemical (IHC) methods. The pituitary organ that used in this study was collected from eight white female rats aged 2, 4, 6, and 12 months. Pituitary glands were processed histologically and stained with IHC methods to detect the ir-FSH cells. The result showed that the ir-FSH cells were observed in pars distalis (PD), pars tuberalis (PT), and pars intermedia (PI) of adenohypophysis with different number of cells. Furthermore, ir-FSH cells were found abundantly (+++) in the PT and a few (+) in PD of rats aged two months. In rats aged 4 months those cells were distributed in slight number (+) in PT and abundant (+++) in PD. In addition, the cells were also found in a great quantities in rats whose ages were 6 months, (+++) in both PT and PD, but these cells decreased (++) in rats aged 12 months in PD and absent (-) in PT. In conclusion, the devalopment and distribution of ir-FSH cells were found in pars tuberalis and pars distalis adenohypophysis in the pituitary glands of rats aged 2 up to 12 months with different number and distribution patterns.

Abstract

This study aimed to determine the effect of sperm washing by swim up and centrifugation in isotonic medium on sperm quality of aceh bull. In this study, fresh semen from healthy male aceh bull aged 3-4 months was collected using artificial vagina. Immediately after semen collection, fresh semen quality was examined macroscopically and microscopically. Subsequently, sperm washing was performed by centrifugation and swim up in sperm washing medium. Group 1 (P0) as control group, cement washed with isotonic solution (andromed medium: saline solution) with ratio of 1:8. 2. Group 2 (P1), cement was separated by centrifugation method, group 3 (P2), all cement was separated by swim up method then examined the sperm quality sperm washing results. Each treatment was repeated 5 times. Quality parameters measured were the percentage of spermatozoa motility, sperm viability, and plasma membrane integrity intact spermatozoa. Data were analyzed with analysis of variance one-way pattern, followed by Duncan's multiple test. The results showed the mean ± SD percentage of sperm motility of each treatment group (P0; P1; P2) respectively amounted to 72.00±3.74, 66.40±4.77, and 73.60±3.29%. The percentage of viability was 72.00 ±3.74%, 66.40±2.88%, 71.80±2.17%. The percentage of plasma membrane integrity is intact spermatozoa was 68.20±1.79%, 57.20±3.77%, 69.00±2.00%. Results of this study showed that the percentage of motility, live spermatozoa and plasma membrane integrity intact after separation by swim-up method were significantly different (P 0.05) compared with no separation.Key words: spermatozoa quality, aceh bulls, centrifugation, swim up

Abstract

This research aimed to observe and identify the development of estrogen receptor alpha (ERα) in the ovary and uterus of rats (Rattus norvegicus) at various age levels using immunohistochemistry (IHC). The ovary and uterus were collected from eight female rats aged 2, 4, 6, and 12 months and processed histologically and stained with IHC staining. This research showed, the intensity of ERα expression was found in high abundance (+++) in stroma and the corpus luteum and was weakly found (+) in the theca cells, blood vessels, oocytes and germinal epithelium of rats aged 2 months. Additionally, ERα expression in rats aged 4 months had similarities with rats aged 2 months, but the intensity of ERα had increased (++) in ovarian theca cells, blood vessels, and oocytes. In ovaries of rats aged 6 and 12 months, ERα expression had decreased (++) in the stroma and the corpus luteum and had a low expression (+) in the blood vessel and oocytes. The uterus of rats aged 2 months had low expression of ERα (+) in the stroma and luminal epithelium, moderate (++) in the myometrium, but was not found in the uterine glands. Furthermore, the expression in the uterus of rats at the age of 4 months had increased in the stroma and luminal epithelium (++) and uterine glands (+).  At the age of 6 months, it found excessively (+++) in the stroma, myometrium, and luminal epithelium, moderate expression (++) in the uterine glands.  The expression of ERα in uterus aged 12 months was almost equal to the uterus of rats aged 6 months, but the expression in the myometrium and epithelial luminal had decreased (++). It can be concluded that the development and distribution pattern of ERα in the ovaries and uterus of rats are different at each age levels, it may be related to the growth and development of reproductive organs and also closely related to the estrus cycles of rats.

Abstract

The objective of this study was to investigate the pattern of carbohydrate distribution of accressory sex glands in male muntjac (Muntiacus muntjak muntjak) in the velvet antler stage. In macroanatomy observation, muntjac accessory sex glands consist of the ampulla, vesicular gland, pars diseminate prostat, and bulbourethral gland. The observation of neutral carbohydrate distribution has done by periodic acid Schiff (PAS) staining. In addition, acid carbohydrate was observed by using alcian blue (AB) pH2,5 staining. Result of this research showed that neutral and acid carbohydrate scattering in each accessories sex gland with different pattern. Neutral carbohydrate with medium (++) and high (+++) intensity reaction was found in the ampulla, vesicularis gland, and bulbourethral gland and strong in prostat gland. The acid carbohydrate with low (+) and medium (++) intensity reaction was observed in the ampulla, low in vesicularis gland, and negative in prostat and bulbourethral glands. In conclusion, the neutral carbohydrate was found dominantly on surface of all accessory sex glands of male muntjac in velvet antler stage with highest concentration and intensity of colour in prostat gland.Key words: carbohydrate, accessory sex glands, muntjac (Muntiacus muntjak muntjak)

Abstract

This study aimed to determine the effect of Indigofera sp. leaf to increase protein content of rabbit meat (Origtolagus sp). This study used a complete randomized design, consisted of 3 treatments and 3 replications. The samples were nine local male rabbits aged two months. P0 was control group which was given commercial feed, P1 was given 80% commercial feed and 20% Indigofera sp. pellet leaf, and P2 was given 70% commercial feed and 30%  Indigofera sp. pellet leaf. The feeding and drinking was conducted at 07:00 to 08:00 am and at 04:00 to 05:00 pm ad libitum. Protein content was measured using Kjeldahl method after eight weeks of treatments. The data was analyzed using Analysis of Variance (ANOVA) with SPSS v16.0 for Windows. The results showed that mean value (±SD) of meat protein for P0, P1 and P2 were 18.79+0.81%, 19.40+0.33% and 20.88+0.74%, respectively. We can conclude that the leaves of Indigofera sp. was highly significant (P0,01) in increasing protein level of rabbit meat at a concentration of 30%.

Abstract

This study aims to determine the effectiveness of 10% Jatropha salve (Jatropha curcas L. )    to inflammation reaction cutaneous wound healing in mice (Mus musculus ) in the inflammatory phase. This experiment used 9 male mices, weighing 25-40 grams and age 2-3 months old, were divided into three treatment groups, each treatment consisting of three mices. The wound were made along 2 cms in the back area. Wound therapy was carried out  twice a day for 3 days.  Group KI was administered  and given by yellow vaseline, KII was given 10% Jatropha salve, and KIII was given gentamicin salve. The parameters measured are the increase in the number infiltrations of inflammatory cells,  neovascular, and re-epitelization. All quantitative data were measured using ANOVA and continued with Duncan Test, the qualitative data were presented descriptively by showing the results of observation under a microscope. The statistical test on the infiltrations of inflammatory cells and neovasculars parameter 10% Jatropha salve group significantly different (P0.05) compared to KI and KIII group. Histopatologycal observations indicated the 10% Jatropha salve could be reduce the infiltrations of inflammatory cells, increase neovasculars and re-epitelization percentage. According to the result can be concluded the 10% Jatropha salve can accelerate the inflammation phase cutaneous wound healing in mice.

Abstract

Penelitian ini bertujuan mengetahui efek pemberian ekstrak daun pegagan terhadap perkembangan sel spermatid tikus. Penelitian ini merupakan penelitian eksperimental dengan menggunakan rancangan acak lengkap (RAL) pola one way analysis of varian (ANOVA). Tikus yang digunakan adalah tikus jantan berumur 3,5 bulan dengan bobot badan 150-250 g sebanyak 12 ekor tikus. Tikus-tikus tersebut dikelompokkan menjadi empat kelompok perlakuan dengan masing-masing tiga ulangan. Kelompok pertama merupakan kelompok kontrol (K0) yang tidak diberi perlakuan, sedangkan kelompok K1, K2, dan K3 diberi perlakuan berupa ekstrak daun pegagan dengan dosis bertingkat, yaitu 125, 250, dan 500 mg/kg bobot badan selama 30 hari. Pada akhir perlakuan, tikus dikorbankan dengan diberi eter, lalu testis dinekropsi untuk selanjutnya dibuat preparat histologis.Preparat histologis diwarnai dengan pewarnaan periodic acid Schiff (PAS) untuk mengetahui perkembangan sel spermatid dengan cara mendeteksi karbohidrat netral pada tudung akrosom sel spermatid tersebut. Data dianalisis dengan ANOVA dan dilanjutkan dengan uji Tukey HSD. Hasil penelitian memperlihatkan bahwa rataan jumlah sel spermatid mengalami penurunan dibandingkan kontrol (K0). K0 mempunyai rataan jumlah sel spermatid tertinggi yaitu 308,00±56,33; diikuti oleh K1 yaitu 234,38±19,81; K2 yaitu 218,50±5,48; dan K3 yaitu 208,05±27,35. Hasil uji statistikmenunjukkan adanya pengaruh yang signifikan (P0,05) antara dosis dan rataan jumlah sel spermatid antara K0 dengan K2 dan K0 dengan K3. Kesimpulan dari penelitian ini adalah ekstrak daun pegagan dapat mengurangi laju perkembangan sel spermatid tikus.

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak daun pegagan terhadap konsentrasi testosteron pada tikus putih jantan.  Penelitian ini menggunakan rancangan acak lengkap (RAL) pola one way analysis of varian (ANOVA). Tikus yang digunakan adalah tikus jantan berumur 3-3,5 bulan dengan berat badan 150-250 g sebanyak 12 ekor. Tikus-tikus tersebut dikelompokkan menjadi empat kelompok perlakuan dengan masing-masing tiga ulangan. Kelompok pertama yang merupakan kelompok kontrol (K0) diberikan pakan komersil (T794P, Pokphand), kelompok K1, K2, dan K3 diberi pakan dan ekstrak daun pegagan dengan dosis bertingkat yaitu 125, 250, dan 500 mg/kg bobot badan selama 30 hari. Pada akhir perlakuan, diambil darah tikus sebanyak 1-2 ml pada vena orbitalis lalu disentrifus sehingga diperoleh plasma darah. Konsentrasi testosteron dalam plasma darah diperiksa dengan menggunakan metode enzim-linked immunosorbent assay (ELISA). Data dianalisis dengan ANOVA. Rataan konsentrasi testosteron pada K0; K1; K2; dan K3 masing-masing 4,66±1,93; 4,28±3,70; 4,01±4,93; dan 3,71±0,83.Dari hasil penelitian disimpulkan bahwa ekstrak daun pegagan tidak berpengaruh terhadap konsentrasi testosteron (P0,05).

Abstract

Preterm infants, born before the 37-week gestation period, have limited storage for nutrients at birth and are vulnerable to poor feeding, severe nutritional deficits and growth retardation. The immature gastrointestinal system leads preterm infants to experience a delay in initiating enteral nutrition. Inappropriate feeding can cause acute and long-term morbidity, prolonged hospitalization and increased treatment cost. Generally, preterm infants that are born after 32 weeks of gestation without severe comorbidities do not have dysphagia and should start oral feeding soon after birth. Preterm infants should have well-developed sucking-swallowing-breathing coordination by 32–34 weeks of gestational age. However, some infants take days or weeks to master the skill. The oral feeding development involves forkhead box protein 2 (FOXP2)-expressing neurons that are found in the deep layers of the cortex, basal ganglia, parts of the thalamus and Purkinje cells of the cerebellum. In mammals, these areas belong to the brain network circuits working for motor coordination in learning and acquiring sensorimotor skills. This review aimed to describe the role of FOXP2 in oral-motor skills in preterm infants, including oral feeding, sucking-swallowing-breathing coordination and language development. The oral-motor skills development could be an early predictor for language delay in premature infants, representing a vulnerable group susceptible to such delays.
