Abstract

Monosodium glutamate (MSG) enhances flavor during food processing by providing an umami taste, but its metabolic by-products can cause liver injury. The liver is crucial for the metabolism of xenobiotics including glutamate. Black garlic, derived from fresh garlic (Allium sativum L.) through chemical reactions and heat treatment, may have hepatoprotective properties; however, studies are limited. This study assessed the hepatoprotective effects of black garlic extract (BGE) on MSG-induced liver damage. Twenty-five Wistar rats (Rattus norvegicus) were divided into five groups (n = 5): P0 (negative control); P1 (MSG group), 8 mg/kg MSG for 21 days; and P2, P3, and P4, receiving the same MSG protocol as P1 plus BGE doses of 200, 400, and 600 mg/kg body weight, respectively, for 16 days. After the experiment, rats were euthanized for liver tissue extraction. Although liver weights in the BGE-treated groups were lower than those in the MSG group, the difference was not statistically significant. These results indicated that a BGE dose of 200 mg/kg body weight mitigated MSG-induced hepatic damage. Further biochemical and histological studies are required to elucidate the mechanism of BGE's hepatoprotective action of BGE against MSG-induced hepatotoxicity.

Abstract

Bulbus oculi prolapse is a condition characterized by the protrusion of the eyeball from the eye socket, which can be caused by various factors such as trauma, elevated intraocular pressure, and abnormalities in the eye muscles. This case study aimed to examine the treatment of bulbus oculi prolapse in domestic cats. A female calico cat named Mici presented with a chronic left eye injury that had become infected, leading to inflammation and persistent ocular discharge. The cats body weight was 0.7 kg, and its temperature was 38.2C. Surgical treatment was performed using an enucleation technique. The procedure involved an incision along the mid-anterior eyelid, extending to the lower eyelid. Dissection was carried out to expose the bulbus oculi, which was then ligated and excised. The eyelid margins were sutured using simple interrupted sutures. Postoperatively, the cat was administered antibiotics and antihistamines.

Abstract

The purpose of this case study is to determine the changes that exist in chickens infected with Chronic Respiratory disease (CRD). A broiler male chicken aged 40 days was found dead in a chicken basket at Ulee Kareng chicken slaughterhouse in a state of feather loss and feces attached to cloaca, on January 6th, 2023. The chicken was brought to pathology laboratory of Faculty of Veterinary Medicine, Universitas Syiah Kuala for necropsy and organ collection. Organs associated with CRD cases were trachea and lungs and histopathological preparations were carried out, stained with Haematoxylin Eosin, and observed under a microscope. The results of histopathological examination showed hemorrhage and hyperplasia of the tracheal epithelium. Hemorrhage was caused by edema which causes increased vascular permeability and epithelial cell hyperplasia occured due to the body's response to infection or inflammation. In the lungs there was bleeding due to damage to the blood vessel wall which caused increased vascular permeability, so that fluid and blood plasma come out of the blood vessels. Fibrinogen then underwent polymerization which resulted in fibrinous thrombi. Congestion happened as the result of fluid accumulation in a location that occured in a passive process so the fluid failed to escape. Because of the accumulation of fluid (congestion), the blood supply to the lung tissue is interrupted, resulting in several points of necrosis in the lungs.

Abstract

This study aimed to determine the effect of prostaglandin F2 (PGF2) administration on improving semen quality and testosterone levels on Bali cattle. This study used 3 Bali cattle aged 2, 3.5, and 5 years old. The sample used in this study was Bali cattle semen. In this study, the Latin Square Design was used with three different treatments were administered over three time periods. The treatments performed were P1 (5 mL physiological NaCl), P2 (25 mg prostaglandin), and P3 (37.5 mg prostaglandin) while treatment period was three weeks of treatment administration, which were 1st. 2nd, and 3rd week. Blood collected through coccygea vein 30 minutes after treatment to measure testosterone levels using enzyme-linked immunosorbent assay (ELISA) technique. Statistical analysis showed that PGF2 administration at a dose of 25 mg and PGF2 at a dose of 37.5 mg had no effect (P0.05) on volume, color, pH, consistency, concentration, and motility of Bali cattle spermatozoa. The volume of semen obtained from P1; P2; and P3 were 6.50.9; 6.30.6; and 6.21.2 mL, respectively. The color of the semen in P1, P2, and P3 were beige and milky white. The pH in groups P1; P2; and P3 were 6.70.6; 7.10.2; and 6.50.2, respectively. Spermatozoa concentration in P1; P2; and P3 were 1,32896.43 x 106 cells/mL; 1,354102.19 x 106 cells/mL; and 1,353.388.55 x 106 cells/mL, respectively. Spermatozoa motility in P1; P2; and P3 were 73.32.8%; 71.62.8%; and 73.32.8%, respectively. Testosterone levels in P1 and P3 were 5.050.22 and 6.741.38 ng/mL, respectively (P0.05). Based on the results of this study, it was concluded that the administration of PGF2 does not affect semen quality but can increase the level of testosterone on Bali cattle.

Abstract

The aim of this study was to further understand the histomorphometry of seminiferous tubules from local goat testicles that were fixed with different fixatives. Nine testicles were randomly chosen, each fixed with 10% formalin, 10% neutral buffered formalin (NBF), and 4% paraformaldehyde (PFA) solutions. The sample was then proceeded to histology preparation and stained with haematoxylin eosin (HE). Observation of the tissue was carried out using microscope and the data obtained was analyzed statistically. The results showed that there was a significant difference (P0.05) on each fixative in tubular diameter and membrane, and very significant difference (P0.01) on germinal cell and staining absorbability. Duncan test result on tubular diameter showed that the use of PFA fixative was significantly different (P0.05) compared to formalin fixative while the use of NBF fixative had no significant difference (P0.05) compared to PFA fixative. In addition, the measurement of interstitial membrane of seminiferous tubules showed no significant difference (P0.05) among three fixatives. To conclude, formalin, NBF, and PFA fixatives affect seminiferous tubules, basal membrane structure, and germinal cell appearance inside tubular lumen. Type of tissues and fixative must be considered in selecting the suitable fixative.

Abstract

This research was aimed to know histopathology change of white mouse ovary as result of EG intoxication. Eight white mice randomly divided into three treatment groups and one control group. Treatment of P1 group was given 10 ml EG/L drinking water, P2 was given 20 ml EG/L drinking water, and P3 was given 30 ml EG/L drinking water for two weeks. Control group was given aquadest. Then, white mice were euthanized and ovaries sample were taken for histology observation. The result for treatment groups (P1 and P2) had shown ovary hyperemic (congest). But, the change is worst in P2 rather than P1. Changes in P3 treatment group had shown ovary hyperemic (congest) and leukocyte infiltration in follicular antrum and follicular necrosis causing lost of ovum cell (cariolysis). Poisoned of EG in white mouse can cause inflammation and necrosis of ovary follicle thus the formed of ovum cell was failed (ovogenesis).

Abstract

The objective of this study was to identify the type of specific glycoconjugates and its distribution in testicular spermatogenic cells in muntjak (Muntiacus muntjak muntjak) based on lectins histochemistry. An adult male muntjak aged 4-5 years old in hard antler period was used in this study. Testicular tissue was fixed in Bouin solution and processed histologically. Histochemistry method was performed using six types biotinylated lectins such as peanut agglutinin (PNA), soybean agglutinin (SBA), wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin A (Con A), and ulex europaeus agglutinin I (UEA I) with 20 g/ml of concentration for PNA lectins and 15g/ml for other type of lectins. The results showed that glycoconjugates were detected by all type of lectins except UEA I in testicular spermatogenic cells with variation in distribution pattern and also the intensity of lectins binding. Glycoconjugates -galactose, -glucose, mannose, Nacetylgalactosamine, N-acetylglucosamine and sialic acid were stained intensely by lectins in golgy-cap phase and acrosomal phase of spermatids. Glycoconjugate N-acetylgalactosamine was the sugar residues which distributed abundantly that marked by positive reaction with PNA, SBA, and RCA lectins. In conclusion, glycoconjugates are detected in testicular spermatids cells of muntjak indicated that glycoconjugates have an important role in spermatogenesis particularly in spermiogenesis.Key words: glycoconjugates, lectins, spermatid, spermatozoa, muntjak
