Abstract

The religion generally creates religious society. The religious society principally could be seen from their attitude that follow religious norms, becouse religion it self guides the people not to do something bad. But in special case, religious Japanese people were founded something unusual and contradict to this ideal values. In the history, the concepts of Geisha, Jugan lanfu, and any sexual harassments had been founded in religious Japanese people. This contradiction may be caused by the change of their religion point of view.

Abstract

Fish was a highly perishable product. The deterioration of fish quality was followed by modification of biochemical components. These Biochemical components include a total volatile base, glycogen, cathepsin activity, and water holding capacity. The objective of this study was to determine the changes of biochemical components in fish deterioration quality. The highest TVB value showed on post rigor phase (38,40 mg N/100 g). The highest glycogen content showed on pre-rigor phase (765,1707 mg/ml). The decreasing of cathepsin activity showed on rigor mortis until post rigor phase, 0,0285 U/ml to 0,0095 U/ml. The ability fresh fish to retain water (76,39%) from total moisture content (76,89%).

Abstract

Keong mata lembu (T. setosus) memiliki potensi sebagai sumber protein dan mineral yang berfungsi sebagai aprodisiak seperti K, Mg, Ca, Fe, Zn, dan Cu. Penelitian bertujuan untuk mengetahui penanganan, pengolahan, dan komposisi kimia keong mata lembu. Cara panen dan penanganan keong ini masih sangat sederhana. Hasil analisis proksimat adalah 70,34% protein, 2,20% lemak, 6,87% abu, 10,06% karbohidrat, dan 10,15% air. Kandungan mineral makro tertinggi adalah kalium sebesar 8225,29 ppm sedangkan untuk mineral mikro tertinggi adalah sebesar 98,68 ppm. Kata Kunci: kandungan mineral, proksimat,Turbo setosus

Abstract

Fish Protein Hydrolysates (FPH) is the mixed products of polypeptide, dipeptides, and amino acid. It can be produced from materials that contained of protein by acid reaction, base reaction or enzymatic hydrolysis. The objectives of this study were to study the production of FPH from fish carp meat at post rigor phase and viscera by enzymatic hydrolysis, to determine the specific activity of papain enzyme, and to determine the solubility of FPH. Capacity of fish hydrolyzing can be identified by analyzing the content of dissolved total nitrogen (NTT) compared with nitrogen total ingredient (NTB) in order to get the value of total soluble nitrogen/total nitrogen material (NTT/NTB). The hydrolysis processes were carried out in 0,26% (w/v) papain, 60 ?C for 3 hours. The result showed that the specific activity of papain enzyme was about 3.28 U/mg. Solubility of FPH by comparing NTT/NTB was about 0.29% (fish meat) and 0.40% (fish viscera). Proximate test of protein content of fish meat was 18.34 ± 0.04 (g/100 g); while viscera was about 0.95±0.04 (g/100 g). The result indicated that product waste of fish carp had potential as a major of source of FPH.

Abstract

Collagenase was purified from intestines of milkfish (Chanos chanos, Forskal) by extraction,ammonium precipitation, ion exchange chromatography on DEAE Sephadex A-50 and gelfiltration on a Sephadex G-100 column. The purification and yields were 114.731 fold and1.26% when compared to those in the starting-crude extract. The molecular mass of milkfishserine collagenase was estimated to be 14.63 kDa and 27.46 kDa.

Abstract

Pathogenic bacteria produced protease that involved in molecular mechanism of foodborne disease. Produced protease involved in molecular mechanisms of foodborne diseases. The purpose of this research was to screen, identify and characterize the potential microorganisms associated with sponge as producer of protease inhibitor. Among 96 isolates examined, four isolates i.e 10A6, 6A3, 9A51, and 1A12 yielded protease inhibitors which were potential to inhibit protease substrates (40-90%). One of the most potential protease inhibitor producer, the bacteria isolate 6A3, was identified as Chromohalobacter sp. Chromohalobacter sp.6A3 produced protease inhibitor with optimum temperature and pH 300 C and 5, respectively. The inhibitor activity was stable when incubated at 400 C for ten minutes or at 300 C for 8 hours. Key words: Bacteria, Chromohalobacter sp., protease inhibitor, screen, sponge

Abstract

In the last decade, concern on protease as medical target for overcoming bacterial diseases and viral diseases has been rapidly increased because of the obvious involvement of this enzyme in the molecular of the diseases. The purpose of this research was to purify and characterize protease from pathogenic bacteria Pseudomonas aeruginosa. The bacteria were grown in media containing triptone 1%, NaCl 1% and Yeast extract 0.5%. protease of Pseudomonas aeruginosa was purified using column chromatography with Sephadex G-100 gel. There were three peaks of enzyme protein, which were detected on fractions 14,17 and 30. the optimum pH of the extracelluler protease from Pseudomonas aeruginosa was 8. The optimum temperature of Pseudomonas aeruginosa was 300C. Fe3+ (1dan 5 mM) was strong activator and Co 2+ was strong inhibitor. Study on the effect of metals ion and specific inhibitors indicated that protease from Pseudomonas aeruginosa was serin metaloprotease. The apparent moleculer weights, as determined by SDS-PAGE and zymogram technique, 36 kD and 42 kD. Key word : Protease, characterization, purification, pathogenic bacteria P. aeruginosa

Abstract

 Various sponges has been reported to produce protease inhibitor which could inhibit protease activity of pathogenic bacteria. The previous research showed that bacteria-associated with sponge could produce bioactive compound similar to their host, including protease inhibitor. The purposes of this research were to purify protease inhibitor from Chromohalobacter sp. 6A3 and to study the characteristics of the protease inhibitor. The result showed that the protein can be extracted by 30 % (v/v) acetone, purified by gel filtration (Sephadex G-75) and finally, purified by anion exchanger (Sephadex A-50). The molecular weight of the purified protease inhibitor after gel filtration was estimated as 21,31 kDa and 17,05 kDa, but anion exchanger gave protein with estimated molecular weight of 21,31 kDa The optimum temperature and pH were 30 oC and 5 respectively. The protease inhibitor could resist heating at 30 oC for 10 minutes. Incubation of the inhibitor at 30 oC, pH 5, still retained its activity until 3 hours. The purified enzyme inhibitor was also still active after incubated at pH from 5 to 6 for 1 hour. The most susceptible substrate (enzyme) for the inhibitor was protease from P. aeruginosa. The protease inhibitor was inhibited by metal ions except Na+ 1mM. Activity of the inhibitor increased twofold by SDS 5 mM. IC 50 of the protease inhibitor was 3.48 nM. The protease inhibitor inhibited the enzyme uncompetitively.

Abstract

Microalgae produce secondary metabolites with different characteristics for each genus, species or strain. A single species of microalgae can produce several bioactive compounds, including protease inhibitors which can prevent deterioration of fish. In this study, we observed the growth of Chaetoceros gracilis in the media NPSi and NPSi + NaHCO3 and determined the chemical content and the potency of protease inhibitor from Chaetoceros gracilis in both media. The culture was harvested at 8 and 15 days. Screening of protease inhibitor activity was performed by agar diffusion method. Protease inhibitor activity was tested on three pathogenic protease-producing bacteria, namely Staphylococcus aureus, Bacillus cereus and Escherichia coli. The pathogenic bacteria often contaminate foodstuffs. The results showed that media NPSi and NPSi + NaHCO3 affected protein and lipid content of C. gracilis, but the culture age did not affect them. The protein content of C. gracilis cultivated in NPSi media (34.75 and 32.94%) was higher than in NPSi + NaHCO3 media (28.13 and 27.13%), while the lipid content was 16.36 and 18.06, 23.86 and 25.40% respectively. Extracts of C. gracilis grown in NPSi and NPSi+NaHCO3 media had inhibitory activity against the test bacteria. Inhibitory activity against E. coli was greater than S. aureus and B. cereus.
