Abstract

In the last decade, a concern on protease as medicinal target for overcoming bacterial and viral diseases has been rapidly increased because of the obvios involvement of this enzyme in the molecular of the diseases mechanism. The porpuse of this research was to characterize proteases from fish pathogenic bacteria Aeromonas hydrophila. The bacteria were grown in media containing triptone 1%, NaCl 1% and yeast extract 0,5%. The optimum production time of A. hydrophila was 48 h, the optimum pH was 7,5, the optimum temperature was 50oC. Study on the effect of metals ion and spesific inhibitors indicated that protease from A. hydrophila was serin metaloprotease.Keywords: protease, characterization, pathogenic bacteria

Abstract

The process of deterioration of fresh  sh quality begins with an enzimatis damage, one of them is katepsin.To prevent deterioration of  sh, an inhibitor can be obtained naturally from the body of  sh. The purpose of this research was to use extract of cathepsin inhibitor from cat sh for inhibit deterioration of milk sh. This researchshowed that temperature of 80 ºC was the optimum temperature for extracting inhibitor complex with inhibitoryactivity of 92,88%. Cathepsin inhibitor gave the most effective to prevent deterioration (85,29% inhibitory activity) after diluted two fold on buffer solution. Soaking with inhibitors from cat sh katepsin capable to inhibit thedeterioration of  sh quality longer than 72 hours (3 days) compared with control  sh, milk sh (soaking in buffer).Keywords : catsh, cathepsin, inhibitor, milksh

Abstract

African catfish (Clarias gariepinus) is important freshwater fish commodity in Indonesia. African catfish also has high protein content, which is potential to be used as protein hydrolysate. Fish protein hydrolysate offered many advantages in food, feed, agriculture, microbiology and medicinal’s field. This research aimed to determine the optimal condition for hydrolysis of African catfish protein and to characterize protein hydrolysate from African catfish. The optimal concentration of papain used for hydrolysis of African catfish protein was 5% (w/v) for 6 hours hydrolysis time and degree of hydrolysis was 35.37%. Protein hydrolysate from African catfish revealed 21.16% of yield and water content of 5.46%; ash content of 5.71%; protein content of 53.29%; and fat content of 1.94%. Protein hydrolysate from African catfish contained 15 types of amino acids, with 9 types of essential amino acids and 6 types of non essential amino acids. The highest essential amino acid composition was lysin of 5.23%, while the highest non essential amino acid composition was glutamic acid of 7.77%. Protein hydrolysate from African catfish showed protein digestibility by in vitro enzymatic hydrolysis of 98.57%.Key words: African catfish, amino acid, fish protein hydrolysate, papain

Abstract

Decomposition of protein in the enzymatic process will lead to changes in odor, texture, and appearance of fish. The enzymes that play a role in the enzymatic process is primarily proteolytic enzymes. Cathepsin enzyme is one of the proteolytic enzymes found in animal tissue that hydrolyzes peptide bonds of proteins. Information on optimal conditions of cathepsin enzyme activity is useful in the process of good handling,  to avoid environmental conditions that can increase cathepsin enzymes activity, especially in milkfish. The purposes of this study were to partial purify the catepsin  enzyme of milkfish and characterize the enzymes. Crude extract of enzyme had specific activity 0.8598 U/mg and after presipitation with ammonium sulphate 70% obtained specific activity of 4.4643 U/mg and after 6 hours of dialysis obtained specific activity 14.4404 U/mg. The enzyme cathepsin worked optimally at 40 °C and pH 4, and 3%  substrate. Divalent metal ions inhibited the activity of the enzyme, higher compared to monovalent or trivalent metals. Cathepsin enzyme identified had molecular weight of 86.67 kDa.Key words: cathepsin, characterization, milkfish, purification

Abstract

AbstrakJeroan ikan tongkol memiliki kandungan protein yang hampir sama dengan daging ikan sehingga memilikipotensi yang besar untuk dijadikan pepton. Penelitian ini bertujuan memproduksi pepton dari jeroan ikantongkol menggunakan enzim papain dan mengujinya untuk pertumbuhan bakteri. Konsentrasi enzim optimalyang digunakan untuk hidrolisis protein jeroan ikan tongkol adalah 0,26% dengan waktu hidrolisis selama3 jam. Pepton jeroan ikan tongkol memiliki kadar air 7,66%; kadar abu 2,34%; kadar protein 50,18%; dankadar lemak 0,47%. Kadar asam amino tertinggi dan terendah masing-masing adalah asam glutamat (6,38%)dan sistein (0,68%). Pepton jeroan ikan tongkol memiliki kelarutan 98,20%; total nitrogen 8,03%; α-aminonitrogen bebas 1,12%; rasio asam amino bebas terhadap total nitrogen 13,93%; kadar garam 0,79%; rendemen3,92-5,54% dan pH 6,02. Pepton jeroan ikan tongkol dapat digunakan sebagai salah satu penyusun media luriabroth (LB) untuk pertumbuhan bakteri Escherichia coli dan Staphylococcus aureus. Kecepatan pertumbuhanspesifi k maksimum E. coli pada media yang menggunakan pepton jeroan tongkol lebih tinggi dibandingkandengan pepton komersial, sedangkan kecepatan pertumbuhan spesifi k maksimum S. aureus lebih rendah padamedia pertumbuhan LB yang mengandung pepton jeroan tongkol.Kata kunci: hidrolisis, jeroan ikan, papain, pepton

Abstract

Polifenoloksidase (PPO) merupakan enzim yang bertanggung jawab terhadap proses diskolorasi yang disebut melanosis. Blackspot atau melanosis merupakan proses penurunan mutu yang terjadi pada udang selama penanganan dan penyimpanan postmortem. Penelitian bertujuan untuk mengevaluasi aktivitas enzim PPO udang vaname dan mengkarakterisasi enzim yang meliputi suhu, pH, konsentrasi substrat optimum serta kinetika enzim. Enzim PPO telah berhasil diisolasi dari karapas udang vaname dengan ekstraksi buffer fosfat (1:3). Enzim ini dikarakterisasi menggunakan L-DOPA sebagai substrat spesifiknya. Ekstrak kasar PPO memiliki aktivitas optimum pada suhu 35°C, pH 6 dan konsentrasi L-DOPA sebesar 10 mM. Nilai Km dan Vmaks PPO untuk substrat L-DOPA adalah sebesar 1,17 mM dan 285,71 U.Kata kunci: L-Dihidroksifenilalanin, Litopenaeus vannamei, melanosis, polifenoloksidase

Abstract

Jeroan ikan adalah bahan baku dengan kualitas rendah atau limbah yang jika tidak dimanfaatkan dapat menimbulkan masalah lingkungan dan kesehatan. Limbah jeroan ikan kakap putih memiliki kadar protein yang tinggi sehingga dapat dimanfaatkan menjadi hidrolisat protein ikan. Penelitian ini bertujuan untuk menentukan kondisi terbaik pembuatan hidrolisat protein serta mengkarakterisasi produk hidrolisat protein yang dihasilkan. Tahap penelitian meliputi karakterisasi jeroan, pembuangan komponen lemak (defatting), penentuan kondisi optimum hidrolisis, dan analisis kimia hidrolisat jeroan ikan kakap putih. Jeroan ikan kakap putih memiliki kadar protein tinggi sebesar 31,20%±0,03 (bk) dan lemak 61,44%±1,22 (bk). Kandungan lemak dapat mempengaruhi proses hidrolisis sehingga membutuhkan proses pembuangan lemak (defatting). Defatting mampu menurunkan lemak sebesar 2,95% (bk) dari lemak awal yakni 61,44%±1,22 (bk) menjadi 58,71%±0,65 (bk). Proses hidrolisis jeroan ikan kakap putih dilakukan menggunakan enzim papain dengan aktivitas 30 Usp/mL dengan konsentrasi enzim 0,15% (b/v), suhu 55°C, pH 8 selama 4 jam. Karakteristik produk hidrolisat jeroan ikan kakap putih (Lates calcarifer) yakni kadar air (10,82±0,84%), kadar protein (62,85%±0,72), kadar lemak (0,84%±0,28), kadar abu (7,30%±0,03), karbohidrat (18,19%±1,32) dan daya cerna protein sebesar 87,03%. Hidrolisat protein jeroan ikan kakap putih memiliki kandungan 15 jenis asam amino. Asam amino tertinggi yakni asam glutamat (10,75%), sedangkan asam amino terendah yakni histidin (1,38%). Hidrolisat protein dapat diaplikasikan sebagai sumber protein dalam pakan ikan.Kata kunci: hidrolisat, jeroan, papain, pembuangan lemak

Abstract

Tuna is one of the mainstay fisheries commodities that involve many of the ordinary fishermen. The handling usually still not follows the principles of good handling that causing the poor quality as the result.The aims of this research were to analyze and to determine the quality of tuna meat (the abdominal wall, dorsal fin, and caudal fin). Raw material yellowfin tuna meat prepared for the light, the meat section of the abdominal wall, dorsal fin, and caudal fin. The result of organoleptic analysis showed that the tuna meat was above the minimum value for the criteria of fresh fish. The result of TVB analysis showed that the meat on the caudal fin had the highest value and significantly different with the other, but still exist on the safe limits and on the fresh category. For the result of analysis in histamine, it shown that at the fourth hour the meat at the dorsal fin had the histamine content which were above the minimum that have been recommended. For the result of analysis in peroxide number, it shown that the tuna meat did not sustain the fat damage until the fourth hour after the catching process. And for the result of analysis in TPC, it showed that at the fourth hour the tuna meat on the pectoral fin had the highest number of total colony and significantly different with the other.Keywords: handling, histamine, light flesh, microbiology, yellowfin

Abstract

Proteolytic enzyme is distributed acros all organism including fish. Cysteine proteases are the largest group of proteolytic enzyme. Lysosomal cathepsin, one of cysteine protease enzyme, cause softening and degradation of myofibril protein and it’s activity is regulated by endogenous inhibitors. The purposes of this study were to optimize the extraction cathepsin inhibitors from the skin, muscles, and viscera of fish, to partially purify the cathepsin inhibitors of selected sources, and to study the characteristics of the cathepsin inhibitor. The cathepsin inhibitor could be extracted from muscle fish and partially purified using ammonium sulfate of 70%. The purified cathepsin inhibitor had optimum temperature at 40°C and the optimum at pH 8. Metal ions decreased the activity of the protease inhibitor, except 1 mM of metal ion Mn2+ and Na+. Abstract

Sea urchin is highly valued seafood. Its bioactive compound also has a potential as natural antibacterial agent. The aim of this research was to screen the antibacterial activity using cup-plate method, toxicity potential using brine shrimp lethality test (BSLT) method, bioactive compound using phytochemical method, and proximate composition by AOAC. This research was devided into two phases, the preliminary research was to determine the best body part of sea urchin showing antibacterial activity. Sea urchins were collected from Pramuka Island and extracted by meseration method. For the preliminary research, gonad extract showed the high antibacterial activity against E. coli and S. aureus at 1.83 ± 0.74 mm and 1.5 mm, respectively. The main research includes the determination of proximate composition, toxicity, bioactive compound, and antibacterial activity from the best body part of sea urchin. The proximate composition from gonad of sea urchin showed that water content 64.97 ± 0.08%; ash 2.72 ± 0.13%; lipid 19.73 ± 0.04%; protein 12.26 ± 0.3%, and 0.33 ± 0.17%, respectively. The detected bioactive compounds from the three different solvents of gonads extracts were steroid, triterpenoid and saponin. The result of lethal toxicity (LC50) from the three gonads extract was 471.861 ppm (n-hexane), 563.226 ppm (ethyl acetate) and 577.531 ppm (methanolic), respectively. Gonads ethyl acetate extracts showed the highest antibacterial activity than n-hexane and methanolic extract, its inhibition zone was 2.71 mm against S. aureus and 4.13 against E. coli.
