Abstract

Rhizophora mucronata is the dominant species plant in Mangrove forests of Kwandang,North Gorontalo. The objectives of this study were to determine the characteristics of fruitand evaluate antioxidant activity including toxicity and bioactive compound of bakauextract . The morphometric value of the fruit has average length 66,75±3,64 cm and weight110,40±10,84 g. The content of ash, protein, and fat were 98±0,03%, 1,75±0,19 %, and1,69±0,36 %, respectively; and the amounts of soluble, insoluble, and total dietary fiberswere 6,75±1,08, 74,42±1,87, and 81,49±2,40 g/100 g, respectively. The boiling process ofbakau fruit until 30 minutes showed the best activity of toxicity and radical scavenging.The potency of bakau fruit as antioxidant related to the bioactive compounds of its such asflavonoid, hidroquinon, triterpenoid, tanin, and saponin.Keywords: Dietary fiber, Mangrove, morphometric, toxicity

Abstract

Fish skin can be used as raw material for producing collagen. The collagen can be extracted by chemicalor combination of chemical and enzymatic processes. Extraction of collagen chemically can do with theacid process that produces acid soluble collagen (ASC). This study aimed to determine the optimumconcentration and time of pretreatment and extraction, also to determine the characteristics of the acidsoluble collagen from the skin of yellow tail fish. Extraction of collagen done by pretreatment using NaOH atthe concentration of 0.05; 0.1; and 0.15 M and extraction using acetic acid at the concentration of 0.3; 0.5; and0.7 M. Pretreatment NaOH with concentration 0.05 M and soaking time of 8 hours is the best combinationfor eliminating non collagen protein. Combination treatment of acetic acid at the concentration of 0.3 Mfor 3 days obtained the best solubility. The yield of collagen ASC was 18.4±1.49% (db) and 5.79±0.47%(wb). Amino acid composition that is dominant in the ASC collagen was glycine (25.09±0.003%), alanine(13.71±0.075%), and proline (12.15±0.132%). Collagen from yellow tail fish skin has α1, α2, β and γprotein structure with the molecular weight of 125, 113, 170-181, and 208 KDa. The transition and meltingtemperatures of collagen were 67.69oC and 144.4oC. Abstract

Fillet of dory is very easy to be find in Indonesian market with various brand and produsen.Imported dory fillet is preferred by consumer so far because it has a white color compare than localfillets. Color is the important parameter that used by consumers to determine the quality of filet. Thisstudy was aimed to determine the quality of local and imported fillets, including protein profile usingSDS PAGE, color measurement, and myoglobin extractability. The results of water soluble protein profilesshowed dory fillet contained 13-15 bands. The redness value (a*) of local fillet (DN, DL, DM) was highercompared others. However, imported fillet (DI) had the highest if redness index (a/b). Imported fillet (DI)showed the lowest concentration of myoglobin compared other samples.

Abstract

Swim bladder is one of marine potential byproducts for alternative source of collagen. This study aimedto optimize hydro-extraction and characterized collagen. Extraction optimization of collagen was determinedusing Box-behnken design response surface method with three variables: CH3COOH concentration, soakingtime and extraction time to yield response. Hydro-extraction collagen was characterization based on theamino acid content, SDS-PAGE, FT-IR and DSC. Acording to the result, the concentration had significantlyinfluence yield. Optimum extraction conditions were variable combinations of acetic acid concentration of0.1 M, for 1 h and hydro-extraction time of 1 h to produce the highest yield 63.35%. collagen was classifiedas type I collagen by amino acid content, electrophoresis patterns and fourier transform infrared (FTIR)spectra. The glass transition of collagen was 67.23oC, showed high thermal stability that can be applied tothe cosmetics industry and nutraceutical.

Abstract

The coastal communities of Kendari believe that sea worm as traditional drug can cure a wound, because some chemical compounds that exist in sea worms are suspected to have the compound. The aims of this study were to determine chemical compound, amino acid, fatty acid content, and mineral of fleshsea worm (Siphonosoma australe-australe) from Toronipa Aquatic Province Sulawesi Southeast. The higest essential amino acid content on fresh sea worm was arginin  (3.04%), and freeze dry was 5.52%. The higest non-essential amino acid content on fresh sea worm and freeze dry were glutamic acid 6.53% and 8.53% for product resulted by freeze dry. The higest saturated attyacid (SF) content of fresh sea worm and freeze dry were palmatic acid (1.96% and 2.64%). The higest MUFA  fatty acid content of fresh sea worm and  freeze dry were plamitoleat acid 0.31% and 0.27%. The higest poly unsaturated fatty acid (PUFA) of fresh sea worm and freeze dry were were aracidonat 2.80% and oleat acid 1.97%. The higest mineral content of wet sea worm and frezee dry were Natrium 43,700 mg/kg and 127,334 mg/kg.

Abstract

Sardine is an economic fish industry product in Indonesia. Sardin fish oil of fish meal by-product can be processed into ethyl ester as a food grade product. The purpose of this study were to determine the chemical and physical the best ethyl ester of Semirefined and refined oil from sardine fish meal by-products. Results showed that heavy metals detected was cadmium (Cd) value, i.e. 0.02 ppm. SFA content of crude oil sardines was 29.39% with palmitic acid (16.24%) as the predominant fatty acids. The MUFA content amounted to 14.87% with palmitic acid as the predominant fatty acid (5.76%). The PUFA content were 35.47% with DHA (17.07%) as the predominant fatty acid, while EPA amounted to 13.82%. Semirefined oil  transformed into Semirefined ethyl ester oil was the best on oxidative and physical parameters. Oxidation process produced Semirefined ethyl ester with 1.50±0.00 mEq/kg peroxide value (PV), 0.90±0.15% fattyacids (% FFA), 5.46±0.32 mEq/kg Anisidin p-value (p-AV), 8.46±0.32 mEq/kg oxidation (TOTOKS), 62.15±0.27%T viscosity and and 5.65±0,26 cP clarity.

Abstract

Spoiled by catch fish is one of catch produce that have low economic value. Utilization and how to increase the selling value of byproducts by making peptone. Peptone fish is a derivative product or derivative of a water-soluble protein hydrolyzate and does not undergo any coagulation process in hot water. This research aim is to produce peptone with spoiled by catch fish as raw materials using microencapsulation technique and maltodextrin as coating ingredients. Microencapsulate of peptone has a chemical composition with moisture content 8.95%, ash content 5.26%, protein content 62.79%, Fat content 0.44% and carbohydrate content 21.48%. The chemical characteristic peptone by-catch rotten fish indicate that product has solubility 98.87%, nitrogen total 10.05%, α amino nitrogen 1.22%, AN/TN 12.14%, salt content 8.04% and pH  6.69. Microencapsulate peptone has a physical characteristic with lightness value 60.01, whiteness value 57.44 and dominated red color value 1.70 and yellow color value 10.33. The water activity of microencapsulating spoiled by catch fish peptone at room temperature of storage after 5 hours was lower than peptone spoiled by catch fish without micro encapsulation and product of commercial peptone. Amino acid analysis results indicate that microencapsulate spoiled by catch fish peptone arginine, serine, tyrosine,histidine and threonine amino acids higher than peptone spoiled by catch fish without microencapsulation and commercial product of peptone. Microorganism growth curve with Optical Density (OD). That indicatesmicroencapsulate spoiled by catch fish peptone has bacterial growth curve more good than a commercial product of peptone and peptone spoiled by catch fish without micro encapsulation.

Abstract

Bruguiera gymnorhiza fruit has potential as raw material for edible film production, because itsavailability in nature is quite abundant and has a quite high amylose and amylopectin content (31.56% and26.17%, respectively). The purpose of this study were to determined the physic and chemical charactersof B. gymnorrhiza fruit starch and to determined the physic characters of edible film added by glycerol asplasticizer and biopolymers (carrageenan and pectin). The edible film production was conducted by usingcommercial starch of B. gymnorrhiza fruit and B. gymnorrhiza fruit starch from North Halmahera. Thefirst stage of this study was characterized of lindur starch by microscopic and chemical composition. Thesecond stage is preparation of edible film contained by 4% starch, 0.4%, 0.5%, 0.6% carrageenan, 0.4%, 0.5%,0.6% pectin, 1% glycerol, subsequently the edible film was characterized by microscopic and mechanicproperties. The results showed that the thickness value of edible film ranged between 0.11–0.125 mm, thetensile strength value around 1.469-3.132 Kgf/cm2, percent of elongation around 8.38-19.6%, and watervapor transmission rate ranged around 0.0145–0.01775 g/s.m2.

Abstract

Formaldehyde found in the fish has been occurred naturally through the work of trimethylamine-N-Oxide demethylase (TMAO)ase enzyme that broke TMAO into formaldehyde and dimethylamine.Formaldehyde found in fish has been occurred during storage. This reserch was aimed to determine theTMAOase activity and its role in dimethylamine and formaldehyde formation in Greater lizardfish (Sauridatumbil), which stored at chilling temperature. Observations were made every 2 days for 16 days of storage.Analysis performed in formaldehyde content determination, dimethylamine content determination, andspecific activity of TMAOase enzyme. The result of formaldehyde test was increasing during storage, from0,22±0,00 ppm on day 0 to 7,45±0,46 ppm on day 16. The results of dimethylamine test increased duringstorage from 0,93±0,50 ppm at day 0 to 27,26±1,8 ppm on day 16. The results of TMAOase specific activityin Greater lizardfish increased during storage from 1,15 U/mg on day 0, to 16,47 U/mg on day 16.

Abstract

Transglutaminases have been found in various living organism, such as mammals, plants,microorganisms, and marine organisms including fishes. Transglutaminases have many various functionssuch as food or non-food application and in pharmaceticals industries. This research was aimed tocharacterize transglutaminase that obtained from fisheries byproducts such as catadromous fish the yellowpike conger’s liver.. The yield of yellow pike conger’s liver was 0.88±0.11%. Crude transglutaminase from liverof yellow pike conger had specific activity of 1.375 U/mg with optimum pH at 7.5 and optimum temperatureat 50°C. The activity of transglutaminase were increased on Mg2+ ion dependent. Crude transglutaminasehad molecular weight of 27.48; 37.00; 69.51; 78.89; 88.18; 108.45; 134.10; and 172.12 kDa.
