Abstract

Abstrak. Bakteri penambat Nitrogen terdapat di rhizosfer tanaman. Salah satu tanaman yang dapat tumbuh dengan baik walaupun kondisi tanah kering dan tandus adalah Tanaman Kayu Jawa (Lannea coromandelica). Tanaman Kayu Jawa  memiliki empat senyawa kimia yang berkhasiat sebagai obat, yakni saponin, flavonoid, polifenol dan tanin. Tujuan penelitian ini adalah untuk mengetahui bakteri pelarut fosfat apa saja yang diisolasi dari rhizosfer tanaman Kayu Jawa. Penelitian dilakukan dengan  menginkubasi sampel tanah dari rhizosfer tanaman Kayu Jawa pada media Pikovskaya untuk menumbuhkan bakteri Pelarut Fosfat. Karakterisasi Morfologi koloni dalam hal bentuk, warna, margin dan tekstur. Isolat dikarakterisasi dengan berbagai metode biokimia yaitu nitrat, Uji Reduktase, Uji pemanfaatan sitrat, Uji metil merah, Uji VP, uji Katalase (metode slip penutup), dan Uji oksidase. Bentuk Sel dan sifat Gram bakteri Pelarut Fosfat juga dianalisis. Dari 7 koloni isolat, bentuk sel isolat RF3, RF4 dan RF7 adalah basil sedang isolat yang lain berbentuk coccus. Reaksi Gram isolat Bakteri Pelarut Fosfat yang diperoleh isolat RF3, adalah Gram Negatif, sedang 6 isolat yang lain adalah Gram positif. Hasil karakterisasi isolat bakteri Pelarut Fosfat menunjukkan bahwa isolat RF1, RF5 dan RF6 adalah Genus Micrococcus. Isolat RF2 adalah Genus Streptococcus. Isolat RF3 adalah Genus Pseudomonas. Isolat RF4 dan RF7 adalah Genus Actinomycetes. Kata Kunci: Isolasi dan Karakterisasi, Bakteri Rhizosfer Pelarut Fosfat, Tanaman Kayu Jawa (Lannea coromandelica)

Abstract

Program Pengabdian Masyarakat ini dilaksanakan pada bulan September 2024 di Laboratorium Sekolah SMA 1 Kabupaten Takalar, Sulawesi Selatan, dengan sasaran utama Guru MGMP Biologi dan IPA. Metode yang digunakan mencakup penyampaian teori, demonstrasi, latihan pembuatan media tumbuh, sterilisasi media, isolasi, serta diskusi dan sesi tanya jawab. Penyampaian teori bertujuan untuk memberikan pemahaman dasar mengenai praktikum Mikrobiologi. Demonstrasi dilakukan agar peserta dapat mengamati langsung prosedur dan tahapan sterilisasi serta pembuatan medium tumbuh menggunakan bahan dan alat yang murah dan mudah didapatkan disekitar sekolah. Praktik dimaksudkan agar peserta memperoleh keterampilan dalam melaksanakan proses kerja pembuatan media tumbuh mikroba. Diskusi dan sesi tanya jawab dirancang untuk mengatasi masalah yang muncul selama penyampaian teori dan kegiatan praktik. Berdasarkan kegiatan ini, dapat disimpulkan bahwa pelatihan ini berhasil meningkatkan pemahaman dan keterampilan guru dalam melaksanakan praktikum pembuatan media kultur mikroba menggunakan bahan dan peralatan sederhana dengan biaya rendah. Program ini membekali guru-guru dengan keterampilan untuk melaksanakan praktikum dengan mudah di laboratorium sekolah. Hasil pelatihan menunjukkan bahwa guru-guru peserta pelatihan mampu membuat media tumbuh mikroba serta memahami prinsip dasar pembuatan media dan isolasi mikroba. Kegiatan ini mampu meningkatkan minat dan pemahaman guru dalam merancang praktikum mikrobiologi menggunakan peralatan dan bahan yang ada di laboratorium sekolah dengan biaya yang sangat murah.

Abstract

The student's cognitive load is high, and the level of thinking and communication skills is low. So, the chunking technique combined with writing thinking is implemented. The purpose of this study was to determine the difference in communication skills of students of circulatory system material taught with chunking techniques combined with writing is thinking, chunking techniques, and lecture techniques in class XI students of SMA Negeri 20 Gowa in the 2024/2025 school year. This type of research includes experiments in the form of a pretest, posttest, and nonequivalent control group design. Sampling technique with purposive sampling with specific consideration in the form of almost the same initial ability of students from three treatment classes where class XI. 1 (experimental class), XI.2 (positive control class), and XI.3 (negative control class), each of which totaled 30 students. Data acquisition using non-test instruments to see students' communication skills and teaching module implementation sheet. Hypothesis testing uses the Anacova test because the data is normally distributed and homogeneous. The results of hypothesis testing showed that the significance value was <0.000. This shows the sig value < α (0.05), so it can be concluded that H0 is rejected and H1 is accepted. This means that there are differences in the communication skills of students who are taught with chunking techniques combined with writing thinking (experimental class), chunking techniques (positive control class), and lecture techniques (negative control class) on human circulatory system material.

Abstract

The aim of this research is to explore the development of gene therapy in the era of modern biotechnology and analyze its application in the treatment of genetic and non-genetic diseases. The research uses the literature review method, which involves collecting, analyzing, and synthesizing relevant scientific sources to answer research questions. The research results obtained based on a literature approach, namely gene therapy in the era of modern biotechnology, have shown significant progress in the treatment of various genetic and non-genetic diseases. Technologies such as CRISPR-Cas9 enable high-precision DNA editing, which has been used to correct genetic mutations, while for non-genetic diseases, gene therapy has provided innovative solutions, such as engineered CAR-T cells and RNA interference (RNAi). Gene therapy still faces various challenges, including the risk of off-target mutations, the body's immune response, and ethical issues and high costs. Gene therapy, as a result of technological developments and research, is expected to be an important solution for various diseases that have been difficult to treat.

Abstract

Aerobic gram-negative bacteria Pseudomonas putida is commonly found in soil and air. It is renowned for its extraordinary metabolic ability to break down various complex organic materials, such as aquaculture pollutants. This article discusses the role of P. putida in bioremediation and the ability of P. putida to break down organic compounds such as aromatic and aliphatic chemicals. This study identified key genetic traits and metabolic pathways that enhance the efficiency of bacterial bioremediation by synthesizing relevant literature using a descriptive qualitative approach. These results demonstrate the importance of utilizing the diverse characteristics of bacteria for innovative bioprocesses that can produce sustainably manufactured products, which in turn promotes biotechnology-based economic development and environmental sustainability.

Abstract

Wolbachia pipientis is an endosymbiotic bacterium commonly found in insects, arthropods, and certain nematodes, playing a pivotal role in host reproductive manipulation and pathogen resistance enhancement. This review aims to provide a comprehensive understanding of the characteristics, physiology, and ecological roles of Wolbachia pipientis, as well as its potential in vector control strategies to combat insect-borne diseases. A literature-based approach was employed, utilizing peer-reviewed scientific journals from reputable sources such as Scopus and Web of Science. The analysis revealed that Wolbachia pipientis is a Gram-negative bacterium with a small size (0.2 to 0.5 µm) that reproduces through binary fission within host cells and relies on the host for essential nutrients. It modulates host gene expression and enhances host immunity, thereby reducing the transmission of pathogens like dengue, Zika, and chikungunya through cytoplasmic incompatibility. Furthermore, Wolbachia pipientis has shown potential in managing filariasis by inhibiting nematode reproduction. The bacterium's ability to alter host reproductive dynamics through mechanisms like feminization and parthenogenesis makes it an effective tool in biocontrol programs. The findings underscore the importance of Wolbachia pipientis in improving public health and environmental sustainability by controlling disease vectors and supporting ecological balance. This review highlights the need for further research to explore genetic engineering approaches to enhance the effectiveness of Wolbachia pipientis in global health management.

Abstract

Polyethylene terephthalate (PET) is a type of plastic that is difficult to decompose, causing significant environmental impacts. PET biodegradation using microorganisms is a sustainable solution. One of the potential bacteria in this process is Ideonella sakaiensis, which is able to produce the PETase enzyme to decompose PET faster. This study aims to examine the potential of Ideonella sakaiensis in PET biodegradation through the enzymes it produces. The method used is a literature review from various scientific sources regarding the characteristics, degradation mechanisms, and the potential for genetic engineering to increase the efficiency of PET degradation. The results of the study showed that I. sakaiensis was able to accelerate PET degradation 5–120 times faster than natural methods. The potential use of genetic engineering by transforming the PETase gene into Escherichia coli also increases the efficiency of PET plastic degradation.

Abstract

Agrobacterium tumefaciens merupakan bakteri tanah Gram-negatif yang dikenal sebagai agen rekayasa genetika alami karena kemampuannya mentransfer DNA ke dalam sel tanaman. Kemampuan ini dimediasi oleh plasmid Ti yang memungkinkan integrasi DNA asing secara stabil ke dalam genom tanaman inang. Bakteri ini telah banyak dimanfaatkan dalam bioteknologi pertanian untuk menghasilkan tanaman transgenik dengan sifat unggul, seperti ketahanan terhadap hama, penyakit dan cekaman lingkungan. Keunggulan A. tumefaciens terletak pada efisiensi transformasi yang tinggi, spesifisitas terhadap tanaman tertentu serta stabilitas ekspresi gen yang baik. Meskipun demikian, terdapat beberapa tantangan dalam penggunaannya seperti keterbatasan jangkauan inang dan tingkat keberhasilan transformasi yang bervariasi antar spesies tanaman. Untuk mengatasi kendala tersebut, berbagai strategi telah dikembangkan termasuk penggunaan super-Agrobacterium, rekayasa plasmid Ti dan pendekatan biologi sintetis. Dengan perkembangan teknologi bioteknologi, pemanfaatan A. tumefaciens di masa depan diharapkan semakin luas dan berkontribusi dalam pengembangan varietas tanaman unggul yang lebih adaptif terhadap perubahan lingkungan serta mendukung ketahanan pangan global.

Abstract

Deinococcus radiodurans is an extremophilic bacterium with exceptional resistance to ionizing radiation, oxidative stress, and extreme environmental conditions. This resistance is supported by a unique multipartite genome structure, efficient DNA repair mechanisms, and complex global regulation. Key genes such as recA, ssb, pprA, and irrE play crucial roles in DNA repair and cellular protection. The bacterium can fully reconstruct its genome after severe radiation-induced damage, making it an ideal model for studying DNA repair and stress response mechanisms. Due to its remarkable resilience, D. radiodurans hold great potential for various applications, including radioactive waste bioremediation, radiation protection in medical fields, and advancements in biotechnology and genetic engineering. Further research is necessary to explore its molecular mechanisms and technological applications in industry, healthcare, and environmental sectors.

Abstract

Thermus aquaticus is a thermophilic bacterium known for its ability to produce thermostable enzymes, such as Taq polymerase, which plays an important role in the Polymerase Chain Reaction (PCR) technique. This study aims to examine the characteristics, ecology, and potential applications of thermostable enzymes produced by this bacterium in various industrial and biotechnological fields. The study was conducted through a literature review using a search method in scientific databases such as PubMed, Scopus, and Google Scholar. The results of the study showed that T. aquaticus has an optimal growth temperature between 70-75°C and is found in extreme environments such as hot springs and hydrothermal systems. Its characteristics, which include strong cell walls, unique membrane lipid content, and thermostable enzymes, allow this bacterium to survive in high temperature conditions. The Taq polymerase enzyme isolated from this bacterium has revolutionized the field of biotechnology, especially in PCR which is used for DNA amplification in genetics, medicine, and forensics research. In addition to PCR, thermostable enzymes from T. aquaticus have wide applications in the food, pharmaceutical, and bioremediation industries. Its existence in the ecosystem shows an important role in the biogeochemical cycle and its interaction with other microorganisms in extreme environments. This finding confirms that the exploration of extremophile microorganisms, including T. aquaticus, still has great potential in technological innovation and the development of biotechnology-based industries.
