Abstract

Kanker tetap menjadi tantangan global karena heterogenitas tumor membuat respons terapi sulit diprediksi, sementara model pra-klinik konvensional kerap tidak mencerminkan fisiologi manusia. Studi ini mengevaluasi kemampuan organoids-on-chip memprediksi respons terapi kanker melalui tinjauan sistematis berpedoman PRISMA 2020. Berdasrkan hasil tinjauan yang telah dilakukan, ditemukan bahwa organoids-on-chip menyediakan perfusi mikro, gradien kimia, dan ko-kultur mikro-lingkungan tumor, sehingga uji berbasis patient-derived organoids dapat memetakan sensitivitas atau resistensi serta memantau toksisitas secara real-time. Mayoritas studi melaporkan peningkatan ketepatan prediksi untuk kemoterapi dan terapi target. Organoids-on-chip juga memfasilitasi penapisan regimen kombinasi dan identifikasi biomarker respons. Namun, variasi protokol, perbedaan matriks, dan parameter aliran membatasi generalisasi. Disimpulkan, organoids-on-chip menjanjikan sebagai dasar pengambilan keputusan dalam onkologi personal, dengan prasyarat standardisasi lintas laboratorium, validasi prospektif, pertimbangan biaya berkelanjutan, serta penetapan titik akhir klinis terstandardisasi dan ukuran efek yang konsisten.

Abstract

This Literature Study aims to analyze various research results on biotechnology and bioengineering that focus on the potential of chicken egg biomaterials as medical solutions. This study uses descriptive qualitative research based on literature studies by reviewing scientific articles from 2020–2025 from the PubMed, Scopus, Sciencedirect, and Google Scholar databases. The results of the study show that chicken egg biomaterials have significant potential in tissue engineering, drug delivery, and degenerative disease therapy. Eggshells are rich in calcium carbonate and hydroxyapatite, which are relevant for hard tissue regeneration. The shell membrane contains collagen and glycosaminoglycans with anti-inflammatory and regenerative activities. Egg whites contain bioactive proteins such as ovalbumin and ovotransferrin, which play a role in wound healing and anticancer activity, while egg yolks are rich in lipids and immunoglobulin Y, which support drug delivery and immunological therapy. Overall, chicken egg biomaterials show high potential as a sustainable, biocompatible, and economical alternative to synthetic materials for the development of drug delivery systems and future biomedical therapies. Abstract

Wastewater bioremediation demonstrates substantially improved performance when employing cross-kingdom microbial consortia, as bacteria, fungi, and microalgae complement one another in carrying out successive stages of organic and persistent pollutant degradation. This review summarizes current findings on the functional mechanisms of such consortia, including metabolic division of labor, metabolite exchange, enzymatic complementarity, and the role of biofilms in maintaining community stability. The literature indicates that cross-kingdom systems are highly effective in removing pollutants such as hydrocarbons, synthetic dyes, pharmaceuticals, nutrients, and microplastics. The integration of omics approaches provides a clearer understanding of the genes, metabolic pathways, and cellular activities involved in degradation processes. Synthetic ecology contributes to strengthening consortium design through spatial arrangement, directed functional roles, and controlled interspecies interactions. Overall, cross-kingdom microbial consortia supported by omics analyses and synthetic ecological concepts offer a solid foundation for developing more efficient and sustainable. wastewater bioremediation systems.

Abstract

Tanaman refugia merupakan salah satu metode agroekologis yang dapat digunakan untuk pengendalian hama secara alami melalui pelestarian musuh alami. Pengabdian kepada masyarakat ini bertujuan untuk meningkatkan pengetahuan dan keterampilan petani terkait manfaat tanaman refugia dalam sistem pertanian berkelanjutan. Kegiatan ini dilaksanakan di Desa Ka'ba, Kabupaten Pangkep, dengan mitra sasaran Kelompok Tani setempat. Metode pelaksanaan terdiri atas pemberian angket awal (pre-test), penyampaian materi melalui metode ceramah, pembagian bibit refugia, serta evaluasi melalui angket akhir (post-test). Hasil kegiatan pelatihan menunjukkan peningkatan pemahaman petani mengenai fungsi ekologis tanaman refugia, dengan 95% peserta menunjukkan peningkatan skor angket setelah sosialisasi. Selain itu, peserta menyatakan kepuasan terhadap materi, metode, dan manfaat kegiatan. Kegiatan ini menunjukkan bahwa pelatihan dan distribusi tanaman refugia efektif dalam membangun kesadaran petani terhadap pengendalian hama ramah lingkungan.

Abstract

This study aims to review various scientific publications related to the application of CRISPR-Cas9 in tomato genetic engineering to enhance food security. The research method used is a literature from reputable databases such as Scopus, ScienceDirect, and PubMed. The results show that CRISPR-Cas9 offers several advantages, including high specificity, multiplexing capability, and flexibility in genome editing. Its applications in tomatoes involve improving resistance to biotic stress (pathogens and diseases) and abiotic stress (drought, extreme temperatures), enhancing nutritional quality, extending fruit shelf life, and increasing productivity. These improvements are directly linked to food security by ensuring stable production, reducing postharvest losses, and increasing crop resilience to climate change. Therefore, CRISPR-Cas9 technology provides an innovative strategy for sustainable agricultural biotechnology development and global food security enhancement.

Abstract

HIV infection remains a global health challenge with high rates of new infections and low levels of viral suppression, especially in middle-income countries. Antiretroviral therapy (ART) has significantly reduced morbidity and mortality, but has not been able to eliminate latent reservoirs so lifelong therapy remains necessary. In the last decade, monoclonal antibodies, particularly broadly neutralizing antibodies (bNAbs), as well as CRISPR-Cas9 gene-editing technology have emerged as innovative therapeutic approaches with long-term remission potential. bNAbs is able to neutralize a broad spectrum of HIV strains and activate immune effector mechanisms, including antibody-dependent cellular cytotoxicity (ADCC) and phagocytosis, but cost limitations, viral resistance, and duration of effects limit its use as a substitute for ART. In parallel, CRISPR-Cas9 has demonstrated the ability to target HIV proviruses in the host cell genome and enable the engineering of immune cells to stably produce therapeutic antibodies. Pre-clinical studies show that B-cell engineering with CRISPR-Cas9 can maintain immune memory, perform isotype switching, and somatic hypermutation, thus creating "programmed immunity". The combination approach of bNAbs and CRISPR-Cas9 is considered promising because it is able to neutralize circulating viruses while removing latent proviruses, increasing the potential for functional cure. However, biological risks such as off-target effects, Cas9 immunogenicity, limitations of the delivery system, and the emergence of escape mutants remain challenges. This review shows that the integration of monoclonal antibodies with CRISPR-Cas9 opens up a new paradigm of HIV therapy, with the potential to improve efficacy, duration of protection, and decrease reliance on ART. Further research, including advanced phase clinical trials, is needed to ensure its safety, effectiveness, and feasibility of application in human populations.

Abstract

Limitations in accurate and efficient species identification techniques at various stages of development and sample conditions are major problems in the conservation of local plants. Conventional morphology-based identification approaches often encounter obstacles due to phenotypic variation and diagnostic character limitations. This study aims to examine the potential of DNA barcoding as a biotechnology innovation in supporting the identification and conservation of local plants. The research method used was a systematic literature study of national and international scientific articles published between 2020 and 2025. Data sources were obtained from the PubMed, Scopus, ScienceDirect, and Google Scholar databases, then analyzed thematically by grouping findings based on the type of genetic marker, identification procedure, and relevance to plant conservation. The results of the study show that DNA barcoding is a species identification technique carried out through the analysis of deoxyribonucleic acid sequences from specific standard marker genes. In plant taxonomy, chloroplast genes commonly used as genetic markers include ribulose bisphosphate carboxylase large chain, maturase K, and internal transcribed spacer. The genetic sequences obtained are then compared with a global database to accurately determine species suitability. This technique has been proven to provide fast and reliable species identification under various sample conditions. With these advantages, DNA barcoding has great potential as a key supporting tool in local plant conservation efforts. Keywords: DNA Barcode, Biotechnology, Plant Conservation

Abstract

Limbah minyak goreng bekas (WCO/FOG) menimbulkan penyumbatan, peningkatan COD/BOD, dan biaya pengelolaan tinggi, sementara melakukan bioremediasi kerap terhambat oleh matriks limbah yang kompleks dan kestabilan enzim yang rendah. Studi ini bertujuan mensintesis bukti tentang efektivitas konsorsium mikroba lipolitik lokal dalam mengoptimalkan aktivitas lipase dan meningkatkan kinerja bioremediasi WCO/FOG. Metode yang digunakan adalah tinjauan literatur sistematik (SLR) mengikuti PRISMA 2020 dengan penelusuran Scopus, Web of Science, PubMed, Dimensions, dan Google Scholar (2015-2025); skrining berpasangan; ekstraksi data terstruktur; penilaian kualitas (adaptasi JBI); serta sintesis naratif dan, bila homogenitas memadai, meta-analisis. Analisis juga memancarkan moderator: imobilisasi enzim, evolusi terarah , optimasi multivariabel , dan transfer massa (kLa/aerasi). Hasil menunjukkan sebagian besar penelitian melaporkan peningkatan aktivitas lipase dan penurunan FOG/COD yang substansial, dengan konsistensi kinerja lebih tinggi pada konsorsium lokal dibandingkan isolat tunggal. Imobilisasi meningkatkan stabilitas operasional dan siklus pemakaian, RSM memperjelas kondisi optimal dan meningkatkan, dan meningkatkan kLa kerusakan positif dengan laju degradasi. Keterbatasan utama meliputi heterogenitas satuan/pelaporan dan minimalnya analisis biaya/LCA, sementara bukti skala pilot mulai berkembang. Secara keseluruhan, strategi integrasi proses enzim konsorsium direkomendasikan untuk implementasi di IPAL/UMKM

Abstract

Antimicrobial resistance has become a major global health challenge that threatens the effectiveness of existing antibiotic therapies. The rapid emergence of multidrug-resistant bacteria highlights the urgent need for alternative antibacterial agents, including those derived from medicinal plants. This study aimed to evaluate the antibacterial potential of Phyllanthus niruri against Gram-positive bacteria and to identify the bioactive compounds and possible molecular mechanisms involved. A systematic literature review was conducted following the PRISMA 2020 guidelines. Articles were retrieved from the Scopus database using the keywords “Phyllanthus niruri” and “antibacterial” or “antimicrobial”. After applying predefined inclusion and exclusion criteria, 15 studies published between 2021 and early 2026 were included in the analysis. The results indicate that P. niruri consistently exhibits antibacterial activity against several Gram-positive bacteria, particularly Staphylococcus aureus. Antibacterial activity was commonly evaluated using inhibition zone assays, minimum inhibitory concentration, and minimum bactericidal concentration tests. Several bioactive compounds, including flavonoids, tannins, alkaloids, and lignans such as phyllanthin, were identified as contributors to antibacterial activity. These compounds may inhibit essential bacterial proteins such as DNA gyrase and penicillin-binding proteins and disrupt bacterial cell membranes. Overall, P. niruri shows promise as a natural source of antibacterial agents against Gram-positive bacterial infections.

Abstract

Biofortifikasi tanaman merupakan strategi penting untuk meningkatkan kandungan mikronutrien dan senyawa fungsional guna mendukung ketahanan pangan berkelanjutan. Pendekatan konvensional sering kali terbatas dalam mengatur jalur metabolisme yang kompleks dan melibatkan banyak gen. Artikel ini bertujuan untuk mengidentifikasi, menyeleksi, dan mensintesis bukti ilmiah terkini mengenai implementasi teknologi CRISPR/dCas9 (CRISPRa, CRISPRi, dan pendekatan epigenetik) dalam modifikasi jalur metabolisme untuk memperkaya nutrisi tanaman. Penelitian dilakukan menggunakan metode Systematic Literature Review (SLR) berbasis PRISMA terhadap artikel terindeks Scopus periode 2022–2026. Hasil kajian menunjukkan bahwa CRISPR/dCas9 mampu mengatur ekspresi gen secara presisi tanpa pemotongan DNA, baik melalui aktivasi, represi, maupun fine-tuning epigenetik. Strategi multipleks memungkinkan pengendalian beberapa gen dalam satu lintasan biosintesis untuk mengoptimalkan metabolic flux dan meningkatkan produksi metabolit bernilai nutrisi. Selain itu, pendekatan epigenetik berbasis dCas9 membuka peluang regulasi gen yang lebih fleksibel tanpa perubahan permanen pada sekuens genom. Meskipun demikian, tantangan seperti efek off-target, keterbatasan sistem pengiriman, dan stabilitas transgenerasional masih memerlukan optimalisasi lebih lanjut. Secara keseluruhan, CRISPR/dCas9 berpotensi menjadi teknologi biofortifikasi presisi yang mendukung pengembangan pertanian modern dan berkelanjutan.
