Abstract

Penelitian ini bertujuan mengetahui motilitas dan kemampuan memfertilisasi sperma kucing yang berasal dari epididimis yang disimpanpada suhu 4 C. Epididimis disimpan dalam media phosphate buffer saline (PBS) pada suhu 4 C selama 1, 3, dan 6 hari. Viabilitas spermatozoa diamati dengan pewarnaan hoechst-propidium iodine. Fungsi biologis spermatozoa dievaluasi melalui teknik kultur in vitro dengan fertilisasi mikro dan perkembangan embrio di dalam media kultur CR1aa. Hasil penelitian menunjukkan persentase spermatozoa hidup pada hari ke-1, 3, dan 6 penyimpanan masing-masing adalah 81,0; 71,7; dan 70,7% (duktus deferens), 84,0; 81,2; dan 63,2% (kauda epididimis), 84,0%; 75,0; dan 74,7% (korpus epididimis). Persentase pronukleus (PN) yang didapat dengan teknik intra cytoplasmic sperm injection (ICSI) menggunakan spermatozoa epididimis pada hari ke-1, 3, dan 6 hari penyimpanan masing-masing adalah 8,0; 10,0; dan 5,9%. Preservasi epididimis pada suhu 4 C dalam PBS dapat digunakan untuk fertilisasi dan menghasilkan embrio kucing secara in vitro.

Abstract

Thorax ganglion was reported as a source of stimulating hormone for gonad development. The aim of this research was to study the development of mud crab (Scylla olivaceous) oocytes cultured in in vitro medium supplemented with thoracic ganglion extracts of Neopisesarma lafondi. Immature crabs with 110 mm carapace width were collected from the mouth of Bawana Marana Rivers, Maros Regency of South Sulawesi, Indonesia. Pieces of ovarian tissues from those crabs were incubated within 24 hours in culture medium (Medium-199 with 100 IU/ml Penicilin-G) supplemented with 2 mg/ml thoracicganglion extract. Abstract

In an attempt to produce the interspecies embryo transfer, this study was conducted to evaluate the efficacy of the production of reconstructed blastocyst by transferring the donor ICM into the recipient trophoblast. ICM cells were isolated from the donor blastocyst by immunosurgery method. Zona-free blastocysts were incubated in the medium (TCM-199) containing 20% of the heat-inactivated rabbit anti-bovine-serum. The embryo reconstruction was produced by three different methods. Recipient blastocyst was maintained on the holding pipette by gentle suction, with the ICM in a 9 o'clock position to have the possibility of developing incorporate ICMs (Method I), the ICM was in a 3 o'clock position to break the original ICM during injection (Method 2); cutting the original recipient ICM followed by insertion of the donor ICM (Method 3). Reconstructed blastocysts were then cultured overnight and examined morphologically according to the re-expansion of the reconstructed blastocyst with or without developed donor ICM. According to morphological observation in this study, 37.9% of the reconstructed blastocyst developed with the incorporation of two ICM originally from recipient and donor (Method I), 66.7% of the reconstructed blastocysts developed with a single ICM (Method 2), and 80.0% of the reconstructed blastocyst developed from the ICM originally from donor ICM (Method 3). These results showed that the reconstructed blastocyst is better produced by cutting the original recipient ICM followed by the insertion of the donor ICM (Method 3).Key words: embryo reconstruction, immunosurgery, ICM transfer, bovine.

Abstract

Knee joints are the primary sites affected by osteoarthritis (OA) in humans and animals. Although intra-articular human umbilical cord–derived mesenchymal stem cells (hUC-MSCs) show therapeutic promise, their combined use with hyaluronic acid (HA) remains underexplored in large animals. This study evaluated morphological and proteomic changes in a sheep osteoarthritis model treated with both agents. A total of twelve sheep were divided into four groups (n = 3 per group): control (normal and OA) and three treatment groups receiving hUC-MSCs, HA, or a combination of HA and hUC-MSCs (HA + hUC-MSCs). OA was induced by meniscectomy, and intra-articular injections were administered four weeks post-surgery. Macroscopic scoring was conducted according to the Osteoarthritis Research Society International system, while histological evaluation was performed using hematoxylin–eosin, Safranin O, and Masson’s trichrome staining. Proteomic analysis of synovial fluid was carried out using liquid chromatography–tandem mass spectrometry. The macroscopic OA score in the HA + hUC-MSC group was lower than in the other treatment groups and was not significantly different from the normal group (p > 0.05). Microscopic observations indicated that hUC-MSCs supported joint cartilage regeneration by preserving proteoglycan and collagen content. Proteomic profiling revealed that treatment with HA + hUC-MSCs modulated proteins associated with complement activation and coagulation pathways. The combination of HA and hUC-MSCs provided synergistic benefits for cartilage regeneration in a sheep OA model during a short treatment period. These findings suggest that this approach represents a promising regenerative therapy for OA in large animals within veterinary medicine and has translational relevance for human regenerative medicine.
