Abstract

The purpose of this research was to study the changes in uterine support capability power due to increase of litter size in Kacang goat. In this study, 9 pregnant female Kacang goats were divided into 4 different groups based on litter size, namely 1, 2, 3, and 4. At 7 weeks of pregnancy, the experimental Kacang goats were sacrificed to observe the macro and micro parameters of the uterus. The results showed that the litter size had a quadratic relationship with micro parameters of uterus but it had a linear relationship with macro parameters of the uterus. The variables of uterine glands area, glands lumen area, and cytoplasmic area reached maximum condition at the litter size of 2.5. Litter size had a linear relationship with the volume, weight, and dimension of the uterus. It can be concluded that the optimal number of litter size in Kacang goat was two offsprings which was proven by the optimal function of the uterus to support fetal development.

Abstract

Penelitian ini bertujuan mengetahui motilitas dan kemampuan memfertilisasi sperma kucing yang berasal dari epididimis yang disimpan pada suhu 4° C. Epididimis disimpan dalam media phosphate buffer saline (PBS) pada suhu 4 C selama 1, 3, dan 6 hari. Viabilitas spermatozoa diamati dengan pewarnaan hoechst-propidium iodine. Fungsi biologis spermatozoa dievaluasi melalui teknik kultur in vitro dengan fertilisasi mikro dan perkembangan embrio di dalam media kultur CR1aa. Hasil penelitian menunjukkan persentase spermatozoa hidup pada hari ke-1, 3, dan 6 penyimpanan masing-masing adalah 81,0; 71,7; dan 70,7% (duktus deferens), 84,0; 81,2; dan 63,2% (kauda epididimis), 84,0%; 75,0; dan 74,7% (korpus epididimis). Persentase pronukleus (PN) yang didapat dengan teknik intra cytoplasmic sperm injection (ICSI) menggunakan spermatozoa epididimis pada hari ke-1, 3, dan 6 hari penyimpanan masing-masing adalah 8,0; 10,0; dan 5,9%. Preservasi epididimis pada suhu 4 C dalam PBS dapat digunakan untuk fertilisasi dan menghasilkan embrio kucing secara in vitro.

Abstract

The aim of this study was to determine the best combination of cryoprotectant (Ethylene glycol, EG), dimethyl sulfoxide (DMSO) and propanediol (PrOH) and carrier system (hemistraw and cryotop) in improving ewe oocytes viability during cryopreservation. Oocytes with multi layers of compact cumulus cells were colleted from abbatoir and matured in TCM 199 medium supplemented with 10% fetal calf serum for 24-26 h at 38.5° C under 5% CO2 in the air. Matured oocyte was divided into six parts and vitrified in three different vitrification solutions; (i) 17% EG+17% DMSO with hemistraw as carrier system, (ii) 34% EG with hemistraw as carrier system, (iii) 17% EG+17% PrOH in hemistraw (iv), 17% EG+17% DMSO with cryotop as carrier system (v), 34% EG with cryotop as carrier system (vi), and 17% EG+17% PrOH in cryotop. Oocytes were cryopreserved for one week before revived and evaluated for viability. The result showed that oocytes vitrified in media containing EG and DMSO in cryotop had the highest viability (88.16%) compared to media containing EG only or EG and PrOH (70.95% and 68.76%, respectively) (P0.05). Moreover, oocytes viability that vitrified using cryotop and hemistraw as carrier system were not significantly different. The present results indicated that vitrification using combination of EG and DMSO as permeable cryoprotectant and cryotop as carrier system was the best system to maintain oocyte viability after vitrification-warming.

Abstract

Penelitian ini bertujuan mengetahui pengaruh penyuntikan pregnant mare serum gonadotropin (PMSG) terhadap peningkatan respons berahi kambing kacang. Sebanyak 20 ekor kambing kacang betina dewasa kelamin dibagi secara acak ke dalam 5 perlakuan dosis penyuntikan PMSG, yaitu 0,0 (K1); 1,0 (K2); 7,5 (K3); 15,0 (K4); dan 20,0 (K5) IU/kg bobot badan dan diulang 4 kali. Hasil penelitian menunjukkan bahwa pada K3, K4, dan K5 terjadi peningkatan jumlah folikel dominan, total diameter folikel dominan, total volume folikel dominan, dan konsentrasi estrogen dalam serum darah (P0,05), serta menunjukkan gejala berahi yang lebih jelas dibandingkan K1. Konsentrasi estrogen dalam serum darah memiliki korelasi positif dengan jumlah folikel dominan, total diameter folikel dominan, total volume folikel dominan, onset berahi, durasi berahi, perubahan vulva menjadi merah, kebengkakan vulva, peningkatan respons betina terhadap pejantan, dan pergerakan ekor pada saat berahiatau fleging berturut-turut sebesar 53,10; 82,60; 48,80; 53,0; 45,50; 39.40; 32,60; 32,60; dan 80,20% dengan pola kuadratik. Dari hasil penelitian disimpulkan bahwa penyuntikan PMSG pada dosis 7,5; 15,0; dan 20,0 IU/kg bobot badan mampu meningkatkan konsentrasi estrogen dan respons berahi kambing kacang.

Abstract

Penelitian ini bertujuan mengetahui pengaruh kombinasi intracytoplasmic sperm injection (ICSI) dan aktivasi dengan strontium untuk mengetahui perkembangan pronukleus dan perkembangan embrio sampai tahap blastosis. Kombinasi ICSI dan strontium 20 mM meningkatkan perkembangan pronukleus 2-PN mencapai 43,59%. Hasil perkembangan embrio pada perlakuan kombinasi ICSI dan strontium 20 mM mencapai tingkat perkembangan 2-4 sel (50,5%), 8-16 sel (43,73%), dan blastosis (15,63%). Kesimpulan dari hasil penelitian ini adalah kombinasi ICSI dengan aktivasi strontium 20 mM mampu menghasilkan perkembangan in vitro embrio sapi yang lebih baik.

Abstract

Penelitian ini bertujuan mengetahui efek asap rokok terhadap perubahan kualitas spermatozoa dan jumlah sel-sel spermatogenik. Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan perlakuan asap rokok 10 batang/ekor/hari selama 2,5 jam dalam smoking chamber terhadap 24 ekor tikus jantan yang dibagi menjadi 4 kelompok, masing-masing 6 ekor dalam tiap kelompok. Kelompok P1 adalah kelompok yang tidak diberi perlakuan (kontrol), P2 adalah kelompok yang dipapar asap rokok selama 20 hari, P3 adalah kelompok yang dipapar asap rokok selama 40 hari, P4 adalah kelompok yang dipapar asap rokok selama 60 hari. Parameter yang diamati adalah kualitas spermatozoa dan jumlah sel-sel spermatogenik. Hasil penelitian menunjukkan bahwa pemaparan asap rokok dapat menurunkan konsentrasi spermatozoa dan viabilitas spermatozoa serta meningkatkan abnormalitas spermatozoa. Disimpulkan bahwa pemaparan asap rokok 10 batang/ekor/hari menyebabkan penurunan kualitas spermatozoa dan jumlah sel-sel spermatogenik yang tidak kembali ke kondisi normal setelah proses penyembuhan.

Abstract

Cryopreservation is a technique for storing cells and tissues at very low temperatures for the possible usage of the stored cells and tissues throughout the year. Sperm cell cryopreservation in some species causes a decrease in sperm quality and DNA damage. Inappropriate cryopreservation protocols can cause changes in sperm physiology. Mitochondria are organelles that play a role in producing energy for sperm motility. Mitochondria have DNA molecules with small sizes compared to structures from nuclear DNA. This study analyzed the effect of cryopreservation on sperm motility and the Cytochrome Oxidase1 (COI) gene. The CO1 gene in mitochondrial DNA plays a role in energy production for spermatozoa motility. The cryopreservation was performed using skim milk and 10% methanol cryoprotectant, and the temperature in the equilibration process was 4-5°C for 10 minutes. Cryopreservation took place for 14 days in the freezer at -80°C. In addition, the thawing process was performed for 1-2 minutes at 40°C. This study found that the number of lesions per 10 kb in the CO1 gene in post-equilibration spermatozoa was (9.24±3.74), and post-thawing spermatozoa (was 10.26±7.54). Spermatozoa motility was obtained in fresh spermatozoa, i.e., 87±1.5%, post-equilibration spermatozoa 79±4.5%, and post-thawing spermatozoa 30±3.2%. This study concluded that cryopreservation of spermatozoa causes CO1 gene lesions and that in cryopreserved spermatozoa, there is a decrease in spermatozoa motility compared to fresh spermatozoa.

Abstract

Information regarding the domestic cat as a model animal in the biomedical research has not been widely carried out. This study was aimed to see the correlation between testicular weight, testicular volume and total sperm count between the left testicle, right testicle and both testes. In addition, this study was aimed to examine the effect of testicular weight and volume on quantitative histomorphometry of the testes in domestic cats in Indonesia. The results showed that there was no difference between the left and right testes. Testes weight has a positive and highly significant correlation with testes volume and total sperm count, both in the left, right, and both testes. The heavier the weight of the testes, the wider the diameter of the seminiferous tubules, the wider the lumen, and the taller the tubular epithelium. It can be concluded that the increase in testicular weight is in line with the increase in cells in the seminiferous tubules.

Abstract

Our study aims to scrutinize the hormonal and laboratory profile of women with PCOS visiting a fertility center in Indonesia. A computerized database of 120 PCOS women and 120 controls was retrospectively reviewed and analyzed. According to analysis, the concentration of AMH and basal LH were significantly more elevated in the PCOS group than in the control group (6.77 vs. 2.05, P<0.001; and 6.8 vs. 5.3, P<0.001, respectively). The level of basal FSH was significantly lower in the PCOS group (5.83 vs. 7.86; P<0.001). After an IVF cycle, PCOS women had significantly higher retrieved oocytes and mature oocytes compared to controls (18 vs. 8, P<0.001; 12 vs. 6, P<0.001). The number of fertilizations, cleavages, and top-quality cleavages were also higher in the PCOS group than in the control group (10 vs. 4, P<0.001; 10 vs. 4, P<0.001; 4 vs. 2, P<0.001). At the blastocyst stage, patients with PCOS had a higher number of blastocysts and top-quality blastocysts than control (9 vs. 4, P<0.001); 4 vs. 1, P<0.01). However, the clinical pregnancy was significantly higher in the control group than in the PCOS group (52.6% vs. 16.7%; P = 0.045).

Abstract

We aim to evaluate the efficacy and optimal dose of orally administered DHEA in the PCOS mice model by assessing their ovarian morphology and serum FSH, LH, and testosterone levels. Female C57BL/6 mice were divided into a control group (n=5, received daily injections of 0.2 ml sesame oil) and an experimental group, which was further classified into 1) D-45 group (n=5), 2) D-60 (n=5), and 3) D-90 group (n=5) that were treated with 45, 60, and 90 mg/Kg body weight of oral DHEA. After modelling, mice in the control group had a regular estrous cycle, while mice in all experimental groups had a disturbed estrous cycle. Ovarian histology showed several growing follicles and some corpora lutea (CL) in the control, D-60, and D-90 groups. However, some large antral follicles and decreased CL were observed in the D-45 group. Serum FSH was significantly lower in the D-45 group than in the control group (3.73 ± 0.12 vs. 5.28 ± 0.31 mIU/mL, P<0.01), but D-60 and D-90 groups had a similar FSH level to the control group (P>0.05). The serum level of LH and testosterone were significantly elevated in the D-45 group than in the control group (2.52 ± 0.43 vs. 1.30 ± 0.33 mIU/mL, P<0.01 and 1.80 ± 0.32 vs. 1.24 ± 0.23 ng/mL, P<0.01, respectively). Still, the level of LH and testosterone in the D-60 and the D-90 groups was comparable to the control group (P>0.05). Our study suggests that oral administration of DHEA is efficacious in establishing PCOS-like phenotype in mice with the suggested optimal dosage of 45 mg/Kg body weight.
