Abstract

Ubi jalar ungu (Ipomoea batatas var Ayumurasaki) dilaporkan dapat membantu diet harian bagi penderita diabetes melitus tipe 2 karena memiliki kadar glikemik yang rendah. Salah satu mekanisme menurunkan kadar gula darah pada penderita diabetes adalah dengan menghambat enzim α-glukosidase. Potensi fraksi alkaloid dari daun dan fraksi antosianin dari umbi ubi jalar ungu dievaluasi sebagai inhibitor enzim α-glukosidase. Ubi jalar ungu yang digunakan dari Ciampea, Bogor, Jawa Barat. Fraksi alkaloid daun ubi jalar ungu pada konsentrasi 2,00% aktif menghambat kerja enzim sebesar 61,88%, sedangkan fraksi antosianin umbi ubi jalar ungu tidak aktif (0,25%). Fraksi alkaloid daun ubi jalar ungu dipisahkan lebih lanjut menggunakan kromatografi kolom dengan fase diam silika gel dan kloroformsebagai eluen. Fraksi yang mengandung alkaloid dengan semprotan Dragendorf, yakni F10, kemudian dipisahkan kembali dengan kromatografi lapis tipis preparatif. Fraksi F10.15 positif mengandung alkaloid dan dapat menghambat enzim -glukosidase sebesar 18,07% pada konsentrasi 1,25%, yang berarti pemisahan menyebabkan penurunan aktivitas. Spektrum inframerah transformasi Fourier fraksi memperlihatkan gugus fungsi –NH−, –OH, dan –CH2– dan diduga mengandung alkaloid.

Abstract

ABSTRAKKamandrah (Croton tiglium L.) merupakan tanaman obat yang banyak terdapat di Kalimantan. Biji kamandrah banyak dimanfaatkan sebagai obat pencahar, racun ikan, dan pembunuh jentik nyamuk. Penelitian ini bertujuan mendapatkan formula minyak biji kamandrah untuk larvasida nabati yang efektif, aman dan mendapatkan minyak kamandrah terstandar sebagai bahan baku larvasida nabati. Analisis fisiko kimia minyak biji kamandrah hasil budidaya di Sukabumi memberikan hasil kadar air 0,33%, keasaman 0,09%, viskositas 4,1 cP, berat jenis 0,9425 g ml-1, indeks bias 1,4788 serta kadar asam lemak bebas 1,65%. Hasil uji ini lebih baik dibanding dengan tanpa budidaya dari Kalimantan dan Sukabumi. Uji menggunakan spektrofotometri menunjukkan kandungan piperine minyak biji kamandrah hasil budidaya Sukabumi sebesar 0,046%; tanpa budidaya dari Sukabumi dan Kalimantan masing-masing 0,043% dan 0,037%. Kandungan piperine berpengaruh terhadap hasil uji efikasi larva nyamuk Aedes aegypti instar III, nilai LC50 pengamatan 24 jam minyak kamandrah hasil budidaya Sukabumi sebesar 114,4 ppm, minyak kamandrah tanpa budidaya dari Sukabumi dan Kalimantan masing-masing 125,2 dan 212,9 ppm. Formulasi larvasida metode granulasi basah terhadap minyak biji kamandrah hasil budidaya Sukabumi menunjukkan, kandungan minyak kamandrah 15% dengan emulsifier gom arab memberikan hasil paling efektif dengan nilai LC50 24 jam sebesar 210,01 ppm. Uji stabilitas formula larvasida nabati minyak biji kamandrah yang disimpan pada temperatur 30, 40 dan 50oC selama 28 hari menunjukkan tidak ada perubahan fisik pada granul. Selama penyimpanan terjadi peningkatan kandungan piperine dalam formula larvasida antara 0,6-234%. Uji durabilitas formula larvasida terhadap larva A. aegypti menunjukkan penurunan potensi larvasida sampai di bawah 50% pada hari ke 12 setelah aplikasi.

Abstract

Xylocarpus granatum produces various bioactive compounds with diverse biological activities, one of them as an antioxidant. Endophytic actinobacteria can also produce bioactive compounds. This study aimed to explore the existence of the endophytic actinobacteria of X. Granatum and determine its antioxidant activity. The research method begins with the isolation of endophytic actinobacteria from fruits, seeds, and leaves using selective media Humic Acid (HV). Furthermore, characterizing the obtained isolates using four different media, namely YSA, ISP2, ISP3, and ISP4, measured antioxidant activity for actinobacterial supernatants and actinobacterial ethyl acetate extracts using the DPPH method. The results showed that from X. granatum, 15 isolates of endophytic actinobacteria were obtained from fruit, seeds, and leaf. Endophytic actinobacteria isolates from X. granatum showed different colors of aerial mycelium, substrate mycelium, and spore chain type. XGF11, XGF12, and XGF4 isolates produced high antioxidant activity of the supernatant; 57.30%, 49.77%, and 41.90% inhibition, respectively. The antioxidant capacity of XGF12 ethyl acetate extract was 501.60 μg AEAC/mg extract and 465.47 μg AEAC/mg extract for XGF11 ethyl acetate extract. DNA extraction and 16S rRNA amplification from endophytic actinobacteria X. granatum according to Presto Mini gDNA Bacteria Kit Protocol. Based on 16S rDNA gene analysis, isolates XGF12 and XGF11 showed 99.16% and 99.42% similarities with Streptomyces xylanilyticus, respectively. Meanwhile, XGF4 showed to have 99.82% similarities with Brevibacterium sediminis.

Abstract

Background : The treatment of some diseases caused by free radicals and pathogenic bacteria usually by using antioxidants and antibiotics. Due to excessive use of antibiotics and other environmental cues, some bacteria are now resistant to certain antibiotics or even to multiple antibiotics. Some Vibrio cholerae bacterial strains are multiresistant to many antibiotics. Objective : The antioxidant and antibacterial activities of Brugueira gymnorrhiza stem extracts against pathogenic bacteria V. cholerae. Method : The B. gymnorrhiza stem was extracted by gradient maceration method. The DPPH method was used to determine the antioxidant activity and the disc diffusion method was used to determine the antibacterial activities. The column chromatography method was used to fractionate the selective extract with the best activity. The LC-MS/MS method was used to identify the compound obtained from the fraction with the best antioxidant and antibacterial activity. Result : Ethyl acetate extract of B. gymnorrhiza stem had the best antibacterial activity with MIC and MBC values of 62.50 mg/L. Ethyl acetate extract also showed the best value of antioxidant activity as indicated by an IC50 value of 255.03 mg/L. The results of fractions test showed that fraction 3 had the best antibacterial and the best antioxidant activities with both the MIC and MBC values of 7.90 mg/L and IC50 value of 348.91 mg/L, respectively. Conclusion : Ethyl acetate extract of B. gymnorrhiza stem has good potential as antioxidant and antibacterial. The compound which is thought as antioxidant and antibacterial from Ethyl acetate extract is 2-Ethyl-4-methyl-1H-imidazole.

Abstract

Utilization of Kayu Bawang (Protium javanicum) plant is needed beside its utilization for building material, especially the utilization of the leaves which easily to get. The aim of this research was to describe the potency of Kayu Bawang leaves as tyrosinase inhibitor, an enzyme which responsible to synthesis melanin as natural color of the skin. The leaves of Kayu Bawang macerated with methanol. The methanol extract then partitioned with n-hexane, EtOAc, n-BuOH, and water. All fractions were concentrated and tested its ability to inhibit tyrosinase with two substrates namely L-DOPA (diphenolase reaction) and L-tyrosine (monophenolase reaction). The result showed that n-hexane fraction could inhibit diphenolase reaction with IC50 value of 114.2 ppm. EtOAc fraction could inhibit monophenolase reaction (IC50 834.0 ppm). Methanol extract, n-BuOH fraction, and water fraction could not inhibit tyrosinase activities. The conclusion is nonpolar and semi polar compounds in Kayu Bawang leaves could be utilized as whitening agent.

This food is popular as a functional food, which is not only contains nutrients, but also has a physiological benefits to prevent or cure diseases. One of the sources of functional food ingredients in Indonesia are spices and medicinal plants which are known containing various types of phytochemical compounds. Numerous reports showed a health benefits of phytochemical compounds derived from spices and medicinal plants such as ginger (Zingiber officinale), turmeric (Curcuma domestica), cardamom (Amomum cardamomum), legetan warak (Adenostemma lavenia), the god’s crown (Phaleria macrocarpa), and lambo guava (Psidium guajava). The phytochemical compounds from those plants are recognized to prevent or cure various diseases including high blood pressure, cancer, diabetes, flu, coughs, sore throat, etc. Functional food products based on spices and medicinal from these 6 plants are usually in the form of jamu, instant healthy drinks, tea, juices, sweets, syrups, etc. Functional food based on Indonesian spices and medicinal plants has the potential to be developed because of its high demand, adequate production and a lot of research that has been conducted. Although the Indonesian government has made the regulations on special nutritious food, these regulations are considered insufficient. Therefore, the government needs to make a specific and comprehensive regulations on functional food in Indonesia, specifically those based on spices and medicinal plants.

Abstract

The leaves of Syzygium polyanthum is widely used in Indonesia as a spice. It has been reported to have significant antioxidant activity. However, the reports on compounds responsible for the activity are very scarce. The purpose of this research was to characterize the chemical profile of S. polyanthumfractions which showed the highest antioxidant activity. The dried leaves of S. polyanthum were fractionated using the different combination of non-polar to polar solvent, those are n-hexane, acetone, water. Fourteen fractions were collected and analyzed for their DPPH antioxidant activity. The chemical profile of selected fractions was characterized using UHPLC-HRMS to identify compounds that might associate with the antioxidant activity of S. polyanthum. The results showed that at the dose of 500 μg/ml, four fractions showed the highest antioxidant activity with the following order: F9 (acetone:water=80:20), F10 (acetone:water=60:40), F1 (hexane 100%) andF3 (hexane:aceton=80:20). UHPLC-HRMSanalysis successfully identified several compounds in F9 and F10. Gallic acid, pyrogallol, and myricetin were identified in F9.  Aconitic acid was identified in F10. All of the identified compounds were previously reported to have antioxidant activity

Abstract

Yogurt rosella susu kambing merupakan susu kambing fermentasi menggunakan bakteri stater “Lactobacillus bulgaricus dan Streptococcus thermophilus” serta ditambah ekstrak bunga rosella (Hibiscus sabdariffa L). Bakteri Lactobacillus plantarum IIA-1A5 dan Lactobacillus fermentum B111K digunakan sebagai probiotik pada pembuatan minuman yogurt rosella. Tujuan penelitian untuk mengevaluasi karakteristik dan kemampuan yogurt rosella probiotik dengan menggunakan L. plantarum IIA-1A5 dan L. fermentum B111K dalam mengasimilasi kolesterol secara in vitro dengan lama penyimpanan berbeda. Perlakuan yang dilakukan dalam penelitian ini adalah P1 : Yogurt dengan bakteri L. bulgaricus dan S. thermophilus, P2 : Yogurt rosella dengan L. bulgaricus dan S. thermophilus,  P3 : Yogurt rosella dengan bakteri L. bulgaricus, S. thermophilus dan L. plantarum IIA-1A5,  P4 : Yogurt rosella dengan bakteri L. bulgaricus, S. thermophilus dan L. fermentum B111K. Hasil penelitian menunjukkan bahwa bakteri L. plantarum IIA-1A5 dan L. fermentum memberikan pengaruh nyata (P<0.05) terhadap nilai viskositas, aktivitas air, total asam tertitrasi (TAT) selama penyimpanan 15 hari. Penggunaan bakteri L. plantarum IIA-1A5 dan L. fermentum B111K tidak memberikan pengaruh yang nyata (P>0.05) terhadap nilai total bakteri asam laktat (BAL) tetapi lama penyimpanan 15 hari memberikan pengaruh nyata (P<0.05). Karakteristik fisik, kimia dan mikrobiologi yogurt rosella probiotik Lactobacillus plantarum IIA-1A5 dan Lactobacillus fermentum B111K selama penyimpanan 15 hari masih baik dan layak untuk dikonsumsi. Selama penyimpanan 15 hari yogurt rosella probiotik Lactobacillus fermentum B111K (YRPF) memiliki potensi mengasimilasi kolesterol tertinggi dengan kolesterol terasimilasi sebesar 4.59 µg/ml dan persentase kolesterol terasimilasi sebesar 15.7%.Potency of Yogurt Roselle Probiotic Lactobacillus plantarum IIA-1A5 or Lactobacillus fermentum B111K in Assimilating CholesterolAbstractYogurt roselle milk goat is fermented milk goat using starter bacteria "Lactobacillus bulgaricus and Streptococcus thermophilus" probiotic bacteria added with rosella flower extract (Hibiscus sabdariffa L). The Lactobacillus plantarum IIA-1A5 and Lactobacillus fermentum B111K bacteria are used as probiotics in the manufacture of functional beverages of rosella yogurt. The objective of the study was to evaluate the characteristics and abilities of probiotic rosella yogurt by using L. plantarum IIA-1A5 and L. fermentum B111K in assimilating cholesterol by in vitro analysis with different storage times. Treatment conducted in this research was P1: yogurt with L. bulgaricus and S. thermophilus bacteria, P2: Yogurt with L. bulgaricus and S. thermophilus bacteria, P3: Yogurt with bacteria L. bulgaricus, S. thermophilus and L. plantarum IIA-1A5 bacteria, P4: rosella yogurt with stater bacteria and L. fermentum B111K. The results showed that L. plantarum IIA-1A5 and L. fermentum had significant effect (P <0.05) on viscosity, water activity, total titrated acids (TAT) during 15 days storage. The use of L. plantarum IIA-1A5 and L. fermentum B111K bacteria did not give significant effect (P> 0.05) to the total value of lactic acid bacteria (BAL) but 15 days storage time gave significant effect (P <0.05). Physical, chemical and microbiological characteristics of yogurt roselle probiotic L. plantarum IIA-1A5 and yogurt roselle probiotic L. fermentum B111K for 15 days storage are still good and feasible for consumption. During the 15 day storage of probiotic yogurt rosella L. fermentum B111K (YRPF) has the potential to assimilate the highest cholesterol with assimilated cholesterol by 4.59 μg/ml and the assimilated cholesterol percentage of 15.7%

Active compounds utilization of its extractives for cosmetic products increase the value added of mangium tree. The purpose of this study was to determine the antioxidant and an inhibitor tirosinase activities of methanol extract of the various parts of mangium tree. Phytochemical properties of the best extract were also anlayzed. Extraction was conducted by soxhletation in methanol for 12 hours. The antioxidant and tyrosinase inhibitory activities of the extracts were tested in vitro to radical of 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging and inhibition to tyrosinase enzyme. The results showed that leave resulted in the highest methanol ectract followed succesively by bark, heartwood, and sapwood the value of 10.7; 4.4; 2.5; and 0.9%, respectively. The highest antioxidant activity was bark extract and followed by leave, heartwood, and sapwood extracts with EC50 values respectively of 8.3; 26.7; 66.9; and 137.9 ppm. Only bark extract which classified as an active tyrosinase inhibitor with IC50 value of 257.8 ppm in the difenolase reaction. The IC50 value of the positive control (kojic acid) was 116.7 ppm. The other extracts relatively inactive as a tyrosinase inhibitor because their IC50 values > 1000 ppm. The qualitative analysis detect the methanol extract of mangium bark as the best extract containing phenolic compounds (phenol hirokinon, flavonoids, and tannins) and alkaloids which were thought to contribute to the high antioxidant and tyrosinase inhibitor activities.Keywords: Acacia mangium, antioxidant, extracts, tyrosinase inhibitor
