Abstract

Some Asteraceae plants have phytochemicals that are commonly used to treat some illness. However, research on the use of Asteraceae plants as tyrosinase inhibitor agents is still rare. Therefore, this study aims to select the most active fraction of Asteraceae species as tyrosinase inhibitors and suspect active compounds that play a role in it. About 12 species from Asteraceae are extracted using water. The resulting filtrate is partially fractionated using chloroform to obtain water extracts, water fractions, and chloroform fractions. Each extract and fraction of each plant determined the activity of its tyrosinase inhibitor using a microplate reader (ELISA) with L-DOPA as the substrate. Thitonia diversipholia water fraction provided the best activity of all the samples tested with the smallest IC50. The active compounds contained are analyzed using LC-MS / MS.  Compounds that act as tyrosinase inhibitors are alkaloid group that dominate the plant, namely trigonelline.

Abstract

Yogurt rosella susu kambing merupakan susu kambing fermentasi menggunakan bakteri stater “Lactobacillus bulgaricus dan Streptococcus thermophilus” serta ditambah ekstrak bunga rosella (Hibiscus sabdariffa L). Bakteri Lactobacillus plantarum IIA-1A5 dan Lactobacillus fermentum B111K digunakan sebagai probiotik pada pembuatan minuman yogurt rosella. Tujuan penelitian untuk mengevaluasi karakteristik dan kemampuan yogurt rosella probiotik dengan menggunakan L. plantarum IIA-1A5 dan L. fermentum B111K dalam mengasimilasi kolesterol secara in vitro dengan lama penyimpanan berbeda. Perlakuan yang dilakukan dalam penelitian ini adalah P1 : Yogurt dengan bakteri L. bulgaricus dan S. thermophilus, P2 : Yogurt rosella dengan L. bulgaricus dan S. thermophilus,  P3 : Yogurt rosella dengan bakteri L. bulgaricus, S. thermophilus dan L. plantarum IIA-1A5,  P4 : Yogurt rosella dengan bakteri L. bulgaricus, S. thermophilus dan L. fermentum B111K. Hasil penelitian menunjukkan bahwa bakteri L. plantarum IIA-1A5 dan L. fermentum memberikan pengaruh nyata (P<0.05) terhadap nilai viskositas, aktivitas air, total asam tertitrasi (TAT) selama penyimpanan 15 hari. Penggunaan bakteri L. plantarum IIA-1A5 dan L. fermentum B111K tidak memberikan pengaruh yang nyata (P>0.05) terhadap nilai total bakteri asam laktat (BAL) tetapi lama penyimpanan 15 hari memberikan pengaruh nyata (P<0.05). Karakteristik fisik, kimia dan mikrobiologi yogurt rosella probiotik Lactobacillus plantarum IIA-1A5 dan Lactobacillus fermentum B111K selama penyimpanan 15 hari masih baik dan layak untuk dikonsumsi. Selama penyimpanan 15 hari yogurt rosella probiotik Lactobacillus fermentum B111K (YRPF) memiliki potensi mengasimilasi kolesterol tertinggi dengan kolesterol terasimilasi sebesar 4.59 µg/ml dan persentase kolesterol terasimilasi sebesar 15.7%.Potency of Yogurt Roselle Probiotic Lactobacillus plantarum IIA-1A5 or Lactobacillus fermentum B111K in Assimilating CholesterolAbstractYogurt roselle milk goat is fermented milk goat using starter bacteria "Lactobacillus bulgaricus and Streptococcus thermophilus" probiotic bacteria added with rosella flower extract (Hibiscus sabdariffa L). The Lactobacillus plantarum IIA-1A5 and Lactobacillus fermentum B111K bacteria are used as probiotics in the manufacture of functional beverages of rosella yogurt. The objective of the study was to evaluate the characteristics and abilities of probiotic rosella yogurt by using L. plantarum IIA-1A5 and L. fermentum B111K in assimilating cholesterol by in vitro analysis with different storage times. Treatment conducted in this research was P1: yogurt with L. bulgaricus and S. thermophilus bacteria, P2: Yogurt with L. bulgaricus and S. thermophilus bacteria, P3: Yogurt with bacteria L. bulgaricus, S. thermophilus and L. plantarum IIA-1A5 bacteria, P4: rosella yogurt with stater bacteria and L. fermentum B111K. The results showed that L. plantarum IIA-1A5 and L. fermentum had significant effect (P <0.05) on viscosity, water activity, total titrated acids (TAT) during 15 days storage. The use of L. plantarum IIA-1A5 and L. fermentum B111K bacteria did not give significant effect (P> 0.05) to the total value of lactic acid bacteria (BAL) but 15 days storage time gave significant effect (P <0.05). Physical, chemical and microbiological characteristics of yogurt roselle probiotic L. plantarum IIA-1A5 and yogurt roselle probiotic L. fermentum B111K for 15 days storage are still good and feasible for consumption. During the 15 day storage of probiotic yogurt rosella L. fermentum B111K (YRPF) has the potential to assimilate the highest cholesterol with assimilated cholesterol by 4.59 μg/ml and the assimilated cholesterol percentage of 15.7%

Abstract

Marine-derived fungi are of great interest as new promising sources of bioactive secondary metabolites. The aim of this study was to determine the antioxidant activity, tyrosinase inhibitor and antiglycation of marine-derrived fungi collected from Kepulauan Seribu Marine National Park, Indonesia.  Antioxidant screening was determined using the 1,1-diphenyl-2-picryl hydrazyl (DPPH) method. The tyrosinase inhibitor was screened using L-tyrosine substrate, while the antiglycation test was determined by the ability to inhibit the formation of advanced glycation end products (AGEs).  A total of 28 marine fungi isolates had been screened for their activities.  Mycelium extract of MFP 271 had the best antioxidant activity with the IC50 of 287.25 ± 50 µg/mL. Mycelium extract of MFP 277 had the best tyrosinase inhibitory activity with the IC50 of 586.42 µg/mL. The MFP 274-broth extract had the highest antiglycation activity with the IC50 value of 298.57 µg/mL. Based on the screening, the crude extracts were considered weak as antioxidant, but tyrosinase inhibitor and antiglycation activity of MFP 277 and MFP 274 are needed to investigate in depth activities.

Abstract

Fingerprint analysis using high performance liquid chromatography (HPLC) has been developed for authentication of Kaempferia galanga from related species, such as Kaempferia pandurata and K. rotunda. By comparing the fingerprint chromatograms of K. galanga, K. pandurata and K. rotunda, we could identify K. galanga samples and detect adulteration of K. galanga from K. pandurata and K. rotunda by using their marker peaks. We also combined HPLC fingerprint with chemometrics for discrimination the three species and also for authentication of K. galanga. All the three species and K. galanga adulterated with K. pandurata and K. rotunda were discriminated successfully by using principal component analysis (PCA) and discriminant analysis (DA). This result indicates that HPLC fingerprint analysis in combination with PCA (PC1 = 30.06% and PC2 = 34.74%) and DA (DF1 = 94.59% and DF2 = 3.32%) could be used for authentication of K. galanga samples from the related species.

Abstract

Padina australis seaweed is known to contain active compounds that can be utilized in pharmaceutics and cosmetics. Ethanol concentration and maceration time are thought to affect the concentration of active compounds in the extract. This study aims to determine the effect of ethanol concentration and maceration time on yield, antioxidant activity, and the total phenol content (TPC).  Seaweed samples were taken from Binuangeun waters, Lebak-Banten. Extraction was conducted by maceration method using 0, 40, and 80 % (v/v) ethanol for 8, 16, and 24 hours. Antioxidant activity was carried out using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) test, while the TPC was determined by colorimetry method using FolinCiocalteu reagent. The results showed that the highest extract yield was obtained when using 40 % ethanol and 16 hours maceration time, the highest antioxidant activity was obtained when using  80 % ethanol and 24 hours maceration time, while the highest TPC was obtained when using 40 % ethanol and 8 hours maceration time. Based on this research, the best antioxidant activity of extract was obtained when maceration used 80 % ethanol with a 24-hour maceration time.

Abstract

Endophytic fungi play an important role on mangrove growth and development, however research on the endophytic fungi of the mangrove Rhizophora apiculata is limited. The endophytic fungi produce diverse bioactive compounds involved in the mangrove’s adaptation to varied biotic and abiotic stresses and could have applied uses in agriculture or medicine. The purpose of this research was to isolate endophytic fungi of Rhizophora apiculata mangrove and to study their activity as herbicides on the weed Gomphrena globosa. The fungi were isolated using the surface sterilization method and identified based on morphological characteristics and molecular characteristics using ITS regions of rDNA. Herbicidal activity of the fungal filtrates extracted by ethyl acetate were tested on seed germination and seedling growth of G. globosa. Five fungal isolates were obtained, namely Penicillium citrinum, Diaporthe eucalyptorum, Diaporthe musigena, Colletotrichum queenslandicum, and Diaporthe tectonae. All isolates were able to grow on PDA medium containing 0, 25, 50, 75, and 100 (% v/v) seawater concentrations, but the growth rate varied by species and seawater concentration. In general, all five isolates showed herbicidal activity by delaying seed germination and reducing shoot and root growth. P. citrinum showed the highest herbicidal activity compared to the other isolates. Analysis using Gas Chromatography-Mass Spectrometry of the crude extract of P. citrinum filtrate identified 7 main compounds: 3-Methoxy-2-methyl-cyclohex-2-enone, Cyclohexane-carbohexaldehyde, 6-methyl-3-(1-methylethyl)-2-oxo, Cyclopropane carboxylic acid, 1-(2-propenyl)-1,1-dimethylethyl ester, 2-Hydroxy-4-isopropyl-7-methoxytropone, Beta-Asarone, Oxane, 4-(2-amino-1-hydroxyethyl)-4-(3,4-dimethoxyphenyl)-, and Diisooctyl-phthalate. These compounds should be studied further to determine which ones are responsible for the herbicidal activity.

Abstract

Tujuan penelitian ini ialah mengetahui perbedaan kadar bahan aktif yaitu flavonoid daun tabat barito (Ficus deltoidea Jack.) pada dua tempat tumbuh yang berbeda. Daun tabat barito dikoleksi dari Taman Nasional Gunung Gede Pangrango (TNGGP) dan Taman Nasional Gunung Halimun Salak (TNGHS). Tabat barito di TNGGP yang digunakan dalam penelitian tumbuh sebagai efifit, sedangkan di TNGHS tumbuh di atas tanah. Daun tanaman diekstraksi dengan etil asetat lalu ditentukan kadar total flavonoidnya menggunakan spektrometri. Hasil penelitian menunjukkan kadar flavonoid daun tabat barito TNGHS lebih rendah dibandingkan daun tabat barito TNGGP. Perbedaan kadar flavonoid diduga karena perbedaan kadar kimia tanah pada kedua lokasi.

Abstract

Unit Konservasi Budidaya Biofarmaka (UKBB) and Kampoeng Djamoe Organik Martha Tilaar (KaDO) are two medicinal plantation that possibly as resources for stingless bees nectar and pollen. This study was aimed to describe the foraging behavior of Tetragonula laeviceps in medicinal plantations, determine the plant species which become the pollen resources, and the distance between the colony and medicinal plants. Observation on foraging behavior were divided into three activities, i.e. flying out from the nest, returning the nest with pollen, and returning the nest without pollen. We found a similar pattern of numbers of bees returning to the nest with pollen in both UKBB and KaDO that occurred in the morning with the highest between 9 and 10 am and gradually decreased in the afternoon. The bees in UKBB showed highly number flying out from the nest in around 3 pm which might be the activites of resin collection. We identified 11 and 16 pollen types at the UKBB and the KaDO, respectively. The pollen in both locations are from the families Amaryllidaceae, Anacardiaceae, Clusiaceae, Cucurbitaceae, Ebenaceae, and Sapindaceae. Our results revealed that the distance range of medicinal plants from the bees hives was 13.4-113,3 m. Thus, this data indicate that the hives should be located in the distance of the bee forage. The medicinal pollen identified in the T. laeviceps pollen pot confirmed the role of the bees as the ecosystem service in the medicinal plantations. Abstract

This research proposes new method to estimate spectral reflectances of Jati Belanda (Guazuma ulmifolia) leaf based on digital image color. Jati Belanda is a medicinal plant containing antioxidant compounds. Spectral reflectances can be used to estimate chemical compounds but the tools to obtain spectral reflectances is very expensive. To estimate the Jati Belanda leaf reflectance values based on digital image color, we used Fourier Transformation. The estimated spectral reflectances is evaluated using root mean square error (RMSE) and goodness-of-fit coefficient (GFC). According to the experiment, dataset used is 276 leaves of medicinal plants and the best polynomial transformation is 3rd order with RMSE value 4,83 and GFC value 0.96. The future work could estimate the amount of chemical compounds in Jati Belanda Leaf using estimated spectral reflectances. Estimated spectral reflectances is  an important part of estimated the amount of chemical compounds.

Abstract

Honey has natural bacteriostatic and bactericide activities; thus, it could be active compounds as an antibacterial against Propionibacterium acnes, the main bacteria in acne forming state. Our study aims to determine phenolic and flavonoid contents (qualitative and quantitative), antioxidants, along with antibacterial activity against P. acnes of selected Indonesian honey. Honey used in this study, including honeybees and stingless bees, were collected from Sumbawa, Riau, Belitung, Sukabumi, Banyuwangi, Bogor, and South Sulawesi. In the method, the quantitative content of phenols and flavonoid were measured using a spectrophotometer. Further, DPPH (2, 2’-diphenylpicryl hydrazyl) and ABTS (2’-azino-bis (3- ethylbenzothiazoline-6-sulfonic acid) assays were used to measure the antioxidant activities, and subsequently microdilution method was applied to determine antibacterial activity. The results showed that Indonesian honey contains phenolics and flavonoids in the range of 49.5-67.3 μg Gallic Acid Equivalent (GAE)/g and 766.5-1509.5 μg Quercetin Equivalent (QE)/g, respectively. All honey samples also show antioxidant and antibacterial activities. Of note, Apis cerana honey from Banyuwangi has the highest antioxidant and antibacterial activity compared to others. It has the Inhibition Concentration (IC50), Trolox Equivalent Antioxidant Capacity, Minimum Inhibitory Concentration (MIC) and Minimum Bactericide Concentration (MBC) values of 59.85 mg/L, 122 μg Trolox/g, 125 mg/mL, and 500 mg/mL respectively. Our study reveals that Indonesian honey can inhibit and kill P. acnes as well. Nonetheless, the phenolic content of honey is not always linear with antioxidant and antibacterial activities.
