Abstract

Red betel plant (Piper crocatum) is a plant that has benefits in the treatment of various diseases. This study aimed to determine the antibacterial activity of red betel leaf extracted using n-hexane, ethyl acetate, and 96% ethanol as solvents against the growth of Escherichia coli in vitro. In this study, 5 concentrations of red betel extract namely 0%, 10%, 20%, 40%, and 60% were used. Chloramphenicol disc was used as a positive control. Antibacterial activity was examined using Kirby-Bauer method. The data were analyzed. The results showed that no inhibition zone was formed around the discs containing n-hexane and 96% ethanol extract, while the ethyl acetate extract at concentrations of 0%, 10%, 20%, 40%, and 60% showed inhibition zone with average diameter were 0 ± 0.8, 97 ± 0.40; 11.61 ± 0.47; 13.4 ± 0.98; 17.56 ± 0.62, respectively. The ethyl acetate extract is able to inhibit the growth of E. coli while the n-hexane and 96% ethanol extracts are unable to inhibit the growth of E. coli in vitro. Further research is needed to examine the effect of red betel ethyl acetate extract against E. coli in vivo.

Abstract

ABSTRAKTujuan penelitian ini untuk mengetahui pengaruh pemberian ekstrak jelly daun sikhoh-khoh dan lama waktu pengamatan terhadap penurunan luas luka pada proses penyembuhan luka terbuka pada tikus putih secara makroskopis. Penelitian ini merupakan penelitian eksperimental tipe The Postest Only Control Group. Sampel yang digunakan adalah tikus putih strain Wistar sebanyak 9 ekor. Dengan jenis kelamin jantan, umur 3 bulan, dan berat badan rata-rata 200 g. Sampel dibagi secara acak menjadi 3 perlakuan dan 3 kali ulangan, masing-masing 3 ekor yaitu perlakuan kontrol (P0) dengan perlakuan tidak diberi ekstrak jelly daun sikhoh-khoh, perlakuan 1 (P1) dengan perlakuan diberi ekstrak jelly daun sikhoh-khoh 10%, dan perlakuan 2 (P2) dengan perlakuan diberi ekstrak jelly daun sikhoh-khoh 30%. Pemberian ekstrak jelly daun sikhoh-khoh dilakukan sehari dua kali secara topikal di atas luka. Pengamatan kesembuhan luka dilakukan secara makroskopis. Pengukuran luas luka terbuka menggunakan jangka sorong untuk menghitung panjang dan lebar luka. Data yang diperoleh dianalisis dengan ANOVA split plot kemudian dilanjutkan uji Duncan dengan kesimpulan pemberian ekstrak jelly daun sikhoh-khoh perlakuan 10% dan 30%  dalam lama waktu pengamatan dapat menurunkan luas luka terbuka tikus putih sejak mulai hari ke 3, 7 dan 14. Penurunan luas luka terbuka pada kelompok pemberian ekstrak jelly daun sikhoh-khoh 10% lebih baik dibandingkan dengan jelly tanpa ekstrak daun sikhoh-khoh dan ekstrak jelly daun sikhoh-khoh 30%.Kata kunci: Daun sikhoh-khoh, tikus putih, luka terbuka ABSTRACTThe purpose of this study was to determine the effect of giving sikhoh-khoh leaf jelly extract and the length of time of observation on the decrease in wound area in the process of healing open wounds in white rats macroscopically. This research is an experimental research type The Posttest Only Control Group. The samples used were 9 white Wistar rats. Witgh male sex, age 3 months, and an average weight0f 200 g. Samples were randomly devided into 3 treatmens and 3 replicates, each 3 tails, namely control treatment (P0) with treatment not given sikhoh-khoh leaf jelly extract, treatment 1(P1) with treatment given sikhoh-khoh leaf jelly extact 10% and , treatment 2(P2) was treated given with 30% sikhoh-khoh leaf jelly extact. The administrationof sikhoh-khoh leaf jelly extract was applied topicaly twice a day over the wound. Wound healing was observed macroscopically. Measurement of the area of an open wound usinga caliper to calculate the length and width of the wound. The data abtained were analyzed by split plot ANOVA then followed by Duncan's test with the conclusion that giving sikhoh-khoh leaf jelly extract 10% and 30% treatment in the length of time of observation could reduce the area of open wounds in white rats from days 3, 7 and 14. Decreased open wound area in the group given 10% sikhoh-khoh leaf jelly extract was better than jelly without sikhoh-khoh leaf extract and 30% sikhoh-khoh leaf jelly extract.Keywords: Sikhoh-khoh leaves, white rat, open wound

Abstract

The research aims to find out the  incidence of gastrointestinal parasite in wild long tail macaque in wildlife  reserve  park, Weh  Island, Sabang. For  that purpose,  fecal  specimens  from 25 wild  long  tail macaques  in Weh  Island-  Sabang were  collected.  The  fresh  faeces  samples were  then  preserved  using AFA  (alcohol-formal-acetic)  liquid  prior  to  be  examined  in  Parasitology  Laboratorium,  Faculty  of Veterinary Medicine, Syiah Kuala University.   The  presence of gastrointestinal  parasite was  observed using centrifuge method as well as Boray  sedimentation method. The obtained data were tabulated and analyzed  descriptively.  The  result  revealed  that  two  parasite  species  were  found  in  gastrointestine; Eimeria  spp.    (12%)  and  Strongloides  spp.  (8%).  It  could  be  concluded  that  the  prevalence  of gastrointestinal  parasite  in wild  long  tail macaque  in wildlife  reserve  park, Weh  Island-  Sabang was considered mild.

Abstract

The  research  has  been  conducted  to  determine  hepatoprotective  effect  of  ethanolic  extract  of Acalypha  indica  L.  leaves  on  rats  (Rattus  novergicus)  induced  with  paracetamol.  The  study  was performed  following  a  completely  randomized  design, using  45 male  rats  (Rattus  novergicus)  divided into  5  groups.  Rats  in  group  1  (K1)  were  given  CMC  1  %  for  7  days  and  followed  by  aquadest administration 8 hours post given CMC on day 7. Group 2 was given CMC 1 % for 7 days and treated with paracetamol at dose 2,5 g/kg BW 8 hours post given CMC on day 7. Groups 3 (K3), 4 (K4), and 5 (K5) were pretreated with ethanolic extract of Acalypha indica L.  leaves at the dose of 50, 100, and 200 mg/kg BW for 7 days respectively, followed by paracetamol administration at the dose of 2.5 g/kg BW in the  next  8  hours  on  day  7. Paracetamol  hepatotoxicity  and  extracts  activity were measured  based  on SGPT and SGOT activity level on day 0, 8 hours after  last administration of  extract on day 7, and 24 hours post paracetamol administration. Data were analyzed using one way ANOVA. The result showed that  the  administration  of  ethanolic  extract  of Acalypha  indica  leaves  at  the  dose  of  50,  100,  and  200 mg/kg BW was  significantly  reduced SGPT  and SGOT  level  (P<0,05)  in  rats  that were  induced with paracetamol. Based on the results of SGPT and SGOT serum activity, it can be concluded that ethanolic extract of Acalypha indica L. potential as hepatoprotective agent.

Abstract

The objective of this study was to evaluate the effect of storage time of epididymis at 5 oC on spermatozoa quality of Aceh local goat. This research was conducted at Reproduction Laboratory, FKH Unsyiah from May to June 2009. Nine epididymis from local goat at the age of 1.5-2.0 years were used in this study. The epididymis were collected from abattoir in Banda Aceh. The experimental design used in this study was completely randomized design with 3 treatments and 3 replications. Spermatozoa quality which consist of sperm concentration, motility, amount of life sperm, and sperm abnormality were examined after the collection of epididymis on day 0 (H-0), day 1 (H-1), and day 3 (H-3) post storage at 5 oC. Data were analyzed using one-way analyses of variance (ANOVA). The result showed that no significant difference (P0.05) seen in the spermatozoa quality after different storage time. The average of spermatozoa concentration on H-0, H-1, and H-3 were 318x107/ml, 282x107/ml, and 241x107/ml respectively. On the average, the percentage of spermatozoa motility on H-0, H-1, and H-3 were 82,27±2,75; 80,25±2,83; and 78,07±0,92%, respectively. Life spermatozoa observed on H-0, H-1, and H-3 were 82,29±2,71; 80,63±1,87; and 80,09±3,31%, respectively. Observation on spermatozoa abnormality showed that the average of spermatozoa abnormality on H-0, H-1, and H-3 were 7,23±0,27; 8,21±0,55; and 10,75±3,14%, respectively. It could be concluded that the spermatozoa quality were not affected by the storage time at temperature of 5 oC.

Abstract

 An experiment was carried out to evaluate the effect of graded levels of cassava leaf meal (CLM) with varying cyanide content on growth performance of African catfish (Clariasgariepinus) fry. There were four groups of fish, each comprising 12 fish per group. One groupwas fed on a control diet (0 mg HCN/kg diet) with fish meal (FM) and soybean meal (SBM) asprotein source (30% CP). Groups two, three and four were fed graded levels of CLM to replace10, 20, and 30% of the total diet at the expense of SBM which contains 4.19, 7.47 and 11.96 mgHCN/ kg diet respectively. After 12 weeks, fish fed with increasing level of CLM showed asignificant growth depression and poor feed utilization compared to those fed the control diet.Apparent dry matter digestibility (ADMD) and apparent protein digestibility (APD) were alsosignificantly (P0.05) affected by cassava leaf meal inclusion, with the best values were found infish fed the control diet (52.26+0.24% and 83.11+1.17% for ADMD and APD respectively). Bodycomposition of fish fed higher CLM tended to have less protein and fat but more ash. The poormethionine availability as a result of low protein digestibility and the presence cyanide contentlimits the use of CLM in African catfish fry diet.Keywords: African catfish, cassava, growth, cyanide

Abstract

This study was conducted to determine the effect of water salinity changes on methionine content in tiger grouper juvenile (Epinephelus fuscoguttatus). A total of 2,560 tiger grouper juveniles were used and divided into five groups consist of 1 control group (without exposed to salinity changes) and 4 treatment groups. The salinity was changed every 2, 3, 4, and 6 hours in treatment A, B, C, and D, respectively. Salinity levels were changed during 24 hours by lowering salinity level from 32 psu to 22 psu. Twenty five of fish were collected from each treatment for methionine content analysis. Data were analysed using one way analysis of variance (ANOVA). The results showed that there was a decrease in methionine content in all treatments. A significant decrease (P0.05) of methionine content in treatment A, B, and C were observed after 20 hours (1.15%), 18 hours (1.27%), and 16 hours (1.24%), respectively. While at 0 hours (control), the methionine content was 2.02%. Methionine level in treatment D was not significantly different (P0.05) compared to control group. As conclusion, rearing the tiger grouper juvenile with salinity fluctuations every 6 hours did not lead to methionine deficiency.

Abstract

The purpose of this study was to evaluate the in vitro and in vivo antimicrobial activity of the commercial Lactobacillus acidophilus (L. acidophilus) cells and cell free extract against Aeromonas hydrophila (A. hydrophila). The in vitro method was carried out using well diffusion method. For in vivo evaluation, the effect of L. acidophilus on the survival rate of Pangasianodon hypophthalmus (P. hypophthalmus) infected with A. hydrophila was evaluated. The well diffusion method showed a significant inhibition ability of L. acidophilus cells against A. hydrophila compared to the cell free extract. The inhibition diameters obtained with cells and cell free extract were 17.23 mm and 15.17 mm, respectively. P. hypophthalmus injected with L. acidophilus cells and cell free extract following challenged with A. hydrophila cells showed survival rate of 70% and 60% respectively, at 2-week post challenged. The gas chromatography-mass spectrophotometry (GC-MS) result revealed that a diverse of compounds was detected in both the L. acidophilus cells and cell free extract, among them the most abundant component was pyrrolo[1,2-a]pyrazine-1,4-dione, hexahydro-3-(2-methylpropyl), which showed a promising anticancerous activity and might be played a significant role in the recovery of the infectious P. hypophthalmus. The current study revealed that both cells and cell free extract of L. acidophilus have antimicrobial activity against A. hydrophila.

Abstract

ABSTRACT The aim of this research was to asses the acute toxicity of ethanolic extract of malaka leaves using lethal dose 50 (LD50) on mice (Mus musculus). Twenty male mice weighing between 20-30 g were randomly divided into 4 groups (group K1-K4) of 5 mice each. All mice in group K1, K2, K3, and K4 were administered ethanolic extract of malaka leaves with the dose of 2, 4, 8, and 16 g/kg bw, respectively. Single dose of ethanolic extract of malaka leaves were given by oral gavage prior to clinical observation . The observation period was 14 days post administration, for sign of toxicity symptom, weight loss, and mortality. The result showed that no mortality was observed in the experimental animals during this study. Slight reduction of body weight was observed in group K2, K3, and K4, and no toxicity sign was found during fourteen days of observation. The LD50 of ethanolic extract of malaka leaves was higher than 16 g/kg body weight, thus, the substance was practically non toxic substance.

Abstract

This study has long-term objectives to obtain immunogenic prototype that can be used to induce multiple ovulation in goats. Working steps of this study were begun with the collection of ovarium from goats, collection of granulose cells, culture of granulose and characterization of molecular weight and isoelectric point (pI) of inhibin protein of granulose cells obtained from culture and non-culture of granulose cells, and followed by preparation of monoclonal antibody toward inhibin. The results showed that inhibin isolated either from culture or non-culture of granulose cells produced a 32 kDa band. Molecular weight of inhibin was measured by Western Blot. The 32 kDa band of SDS PAGE product appeared on Western Blot result was inhibin molecules produced by granulose cells collected fom culture and non-culture of granulose cells that can be identified by Mab-inhibin. Product of IEF gel electrophoresis suggested that inhibin molecule collected from culture of granulose cells has no charge at isoelectric points ranging from 5-6, depends on its total amino acid composition.
