Abstract

Penelitian ini bertujuan mengetahui gambaran darah (hematokrit, eritrosit, leukosit, dan diferensial leukosit) tikus yang diinfeksi Trypanosoma evansi (T. evansi) dan diberi ekstrak kulit batang jaloh (Salix tetrasperma Roxb). Duapuluh lima ekor tikus jantan dibagi secara acak ke dalam 5 kelompok perlakuan yang masing-masing terdiri atas 5 ekor tikus. Kelompok 0 (K0) tanpa perlakuan, kelompok I (K1) hanya diinfeksikan dengan 103 T. evansi, kelompok II (K2) diinfeksikan dengan 103T. evansi dan diberikan ekstrak kulit batang jaloh 30 mg/kg bobot badan, kelompok III (K3) diinfeksikan 103 T. evansi dan diberikan ekstrak kulit batang jaloh 45 mg/kg bobot badan, dan kelompok IV (K4) diinfeksi dengan 103 T. evansi dan diberikan ekstrak kulit batang jaloh 60 mg/kg bobot badan. Infeksi T. evansi dilakukan secara intraperitoneal sedangkan ekstrak diberikan secara oral selama 3 hari berturut-turut. Hasil penelitian menunjukkan bahwa rata-rata ± SD nilai hematokrit dan eritrosit tikus dari K1, K2, K3 dan K4 lebih rendah dari K0. Sebaliknya, rata-rata ± SD jumlah leukosit (103/µl) lebih tinggi dari K0. Diferensial leukosit menunjukkan jumlah masing-masing sel leukosit semua tikus dalam kelompok perlakuan meningkat setelah pemberian ekstrak kulit batang jaloh kecuali eosinofil dan limfosit yang justru menurun. Dari penelitian dapat disimpulkan bahwa Infeksi T. evansi menurunkan kadar hematokrit dan eritrosit namun meningkatkan kadar leukosit tikus dan pemberian ekstrak kulit batang jaloh dosis rendah dalam waktu yang singkat mampu mengembalikan profil darah tikus mendekati nilai normal.

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak epididimis (EE) terhadap peningkatan kualitas spermatozoa kambing jantan  lokal. Dalam penelitian ini digunakan 12 ekor kambing jantan lokal, berumur 1,5 tahun dengan bobot badan 10-15 kg dan dibagi atas empat kelompok (K0, KP1, KP2, dan KP3). Kelompok K0, hanya diinjeksi dengan NaCl fisiologis sedangkan kelompok KP1, KP2, dan KP3 diinjeksi EE masing-masing 1, 2, dan 3 ml/ekor selama 13 hari berturut-turut. Pada hari ke-14, dilakukan pengambilan semen kambing dengan elektroejakulator dan selanjutnya dilakukan pemeriksaan kualitas spermatozoa. Hasil penelitian menunjukkan bahwa pemberian EE dengan dosis 1 dan 3 ml/ekor EE selama 13 hari berturut-turut menyebabkan peningkatan kualitas spermatozoa dibanding kelompok kontrol. Berdasarkan hal tersebut dapat disimpulkan bahwa EE berpotensi meningkatkan kualitas spermatozoa pada kambing jantan lokal.

Abstract

The aim of this research was to find out in vitro antiplasmodium activity of Malacca leaves (Phyllantus emblica) ethanolic extract against Plasmodium falciparum growth. In this study, Plasmodium culture contained 5% parasitemia in ring stage was cultured using candle jar method and antiplasmodial activity test was carried out using microculture. The treatments were divided into 7 groups with four repetitions. K1 as negative control group was given Roswell Park Memorial Institute (RPMI), while K2 as positive control group was given artesdiaquine. Groups K3, K4, K5, K6, and K7 group was added with 100 µg/mL, 75 µg/mL, 50 µg/mL, 25 µg/mL, and 5 µg/mL of Malacca leaves ethanolic extract, respectively. Antiplasmodial activity was determined by inhibition concentration of 50% parasite growth (IC50). The data were analyzed using ANOVA and followed by Duncan test. The average of parasitemia level in group K1, K2, K3, K4, K5, K6, and K7 were 55.25±15.62, 8.50±2.52, 8.50±3.00, 9.25±0.95, 9.00±2.70, 9.79±2.06, and 10.75±2.22, respectively. The average of inhibition percentage in group K1, K2; K3; K4; K5; K6; and K7 were 0.00±0.00%, 84.62±4.55%; 84.62±5.43%; 83.26±1.73%; 83.71±4,90%; 82.35±3,73%; and 80.54±6.83%, respectively (P0.01). The results showed that the administration of malacca leaves ethanolic extract significantly affect (P0.01) the inhibition of Plasmodium growth as compared to group K1 (negative control). Probit analysis reveals the IC50 value was 3.889 µg/mL. In conclusion, all doses of malacca leaves ethanolic extract used in this study was able to inhibit Plasmodium falciparum growth with IC50 value was 3.889 µg/mL.

Abstract

ABSTRACT The aim of this research was to asses the acute toxicity of ethanolic extract of malaka leaves using lethal dose 50 (LD50) on mice (Mus musculus). Twenty male mice weighing between 20-30 g were randomly divided into 4 groups (group K1-K4) of 5 mice each. All mice in group K1, K2, K3, and K4 were administered ethanolic extract of malaka leaves with the dose of 2, 4, 8, and 16 g/kg bw, respectively. Single dose of ethanolic extract of malaka leaves were given by oral gavage prior to clinical observation . The observation period was 14 days post administration, for sign of toxicity symptom, weight loss, and mortality. The result showed that no mortality was observed in the experimental animals during this study. Slight reduction of body weight was observed in group K2, K3, and K4, and no toxicity sign was found during fourteen days of observation. The LD50 of ethanolic extract of malaka leaves was higher than 16 g/kg body weight, thus, the substance was practically non toxic substance.

Abstract

Burns are caused by heat exposure, such as fire, radiation, electricity or chemicals that can damage the skin and affect the body's systems. The aim of this study was to find out the effect of the ethanol extract of Malacca leaves (Phyllanthus emblica) on the number of fibroblast cells in white rats (Rattus norvegicus) that have burned. This study used 24 white rats (Rattus norvegicus) were divided into 4 groups that smeared with aquadest as a negative control (P1), 5% ethanol extract gel of Malacca leaves (P2), 10% ethanol extract gel of Malacca leaves (P3), and positive control applied with bioplasenton® gel (P4). The IIA degree burn were created by placing a 2x2 cm hot iron plate on the back of the rat for 5 seconds. The euthanasia performed to all animal and the skin samples were collected after 15 days of treatment. Then histopathological preparations were made using HE staining. The number of fibroblast cells were analyzed by ANOVA test. The average number of white rats (Rattus norvegicus) fibroblast cells that suffered burns P1 (negative control) had a number of 7 ± 1.4 cells/visual. Whereas th e P2 group had a number of 4.2 ± 1.58 cells/visual. This value has a significant difference with the negative control. But the P2 and P3 values (3 ± 1.51 cells/visual) there is no significant different with the P4 value (positive control) with an average number of P4 fibroblast cells were 2 ± 0.4 cells/visual. The results of this study concluded that the ethanol extract of malacca leaves 5% and 10% had an effect againts accelerating burns healing in white rats ( Rattus norvegicus).

Abstract

Penelitian ini bertujuan mengetahui pengaruh anestesi per inhalasi dan anestesi per injeksi terhadap kadar hemoglobin anjing lokal. Hewan coba yang digunakan adalah 6 ekor anjing jantan lokal dengan umur 4-5 bulan dengan berat badan 5-6 kg. Anjing dibagi menjadi 2 kelompok, masing-masing kelompok terdiri atas 3 ekor anjing. Kelompok 1 (K1) diinduksi dengan halotan 3%, setelah teranestesi diturunkan menjadi 1% untuk maintenance sedangkan kelompok 2 (K2) diinjeksi dengan ketamin 10 mg/kg bobot badan dan xylazin 1 mg/kg bobot badan. Pengambilan darah dilakukan pada menit ke-0, 10, 20, 30, 40, dan 50 melalui vena cephalica dan dilakukan pemeriksaan kadar hemoglobin dengan metode Sahli. Data yang diperoleh dianalisis menggunakan analisis varian pola split plot. Hasil penelitian menunjukkan bahwa terjadi perbedaan yang sangat nyata (P0,01) terhadap kadar hemoglobin (g/dl) antara anestesi per inhalasi (14,28+1,71) dengan anestesi per injeksi (12,66+0,37), namun tidak terjadi perbedaan yang nyata (P0,05) pada masing-masing periode pengamatan. Dari hasil tersebut dapat disimpulkan bahwa pemakaian anestesi per inhalasi lebih aman dibandingkan anestesi per injeksi berdasarkan kadar hemoglobin.

Abstract

This study was done to investigate lipid profile in the blood of Sumatran elephants at the ECC of Saree. In this stud blood samples were drawn from the auricular vein of 8 sumatran elephants at the ECC. Reasurement of total cholesterol, low density lipoprotein (LDL), high density lipoprotein (HDL), and triglicerida (TG) was carried spectrophotometrically by the commercial service of research laboratory in Banda Aceh. The results showed that the average concentrations of total cholesterol, LDL, HDL, and TG in the blood of Sumatran elephants were 155,86±54,39 mg/dL, 40,13±3,40 mg/dL, 101,13±46,16 mg/dL, dan 84,89±32,86 mg/dL. It can be concluded that the average concentrations of the lipid profile in the blood of the sumatran elephants at the ECC in Aceh were higher than that of reported in other subspecies of Asian elephant

Abstract

Penelitian ini bertujuan mengetahui pengaruh peningkatan suhu terhadap jumlah eritrosit ikan nila. Sampel yang digunakan adalah darah ikan nila yang berasal dari 18 ekor ikan nila dengan bobot badan berkisar 40-50 g. Rancangan penelitian yang digunakan adalah rancangan acaklengkap (RAL) pola searah dengan tiga perlakuan dan enam kali ulangan. Perlakuan P1 sebagai kontrol merupakan perlakuan dengan suhu akuarium 29±1°C, P2 merupakan perlakuan dengan suhu akuarium 32±1°C, dan P3 merupakan perlakuan dengan suhu akuarium 35±1°C. Ukuran akuarium pada masing-masing kelompok perlakuan adalah 80 x 60 x 40 cm dengan ketinggian air 30 cm. Perlakuan dilakukan selama 15hari dan pada hari ke-16 dilakukan pengambilan sampel darah. Darah diambil melalui vena caudalis. Data dianalisis dengan analisis varian(ANAVA) dan dilanjutkan dengan uji Duncan. Rata-rata (±SD) jumlah eritrosit ikan nila (106/mm3) pada P1, P2, dan P3 masing-masing adalah 3,01x106/mm3; 2,63x106/mm3; dan 2,08x106/mm3. Hasil penelitian menunjukkan bahwa peningkatan suhu berpengaruh (P0,05) terhadap jumlaheritrosit ikan nila. Dapat disimpulkan bahwa peningkatan suhu air 32±1°C dan 35±1°C dalam akuarium dapat menurunkan jumlah eritrosit ikan nila.

Abstract

The aim of this research was to find out the effect of long bean (Vigna unguiculata) on level of blood glucose on mice (Mus musculus) induced by alloxan. Twelve mice with the weigh of 25-40 grams and clinically healthy were used in this research. All mice were randomly divided into 4 treatment groups, 3 mice each. K0 was negative control group, only given aquadest. K1, K2, and K3 groups were induced by alloxan 0.5 ml. K1 group was positive control. K2 group was fed with 100 grams long bean mixed with 50 ml distilled water. K3 group was fed with 100 grams long bean mixed with 100 ml distilled water. Long bean was fed orally 0.5 ml every morning and evening for 7 consecutive days. On day eight the examination of blood glucose level was performed. The average level of blood glucose were K0 (142.00±23.39), K1 (167.00±10.54), K2 (122.67±12.50), dan K3 (154.67±16.26) mg/dL. In conclusion, the administration of long bean for seven consecutive days does not decrease blood glucose level on mice induced by alloxan.Key words: blood glucose, alloxan, long bean

Abstract

The study aimed to isolate the Salmonella sp on quail eggs that failed to hatch in Garot village, Darul Imarah Subdistrict, Aceh Besar. A total of thirty quail eggs that failed to hatch were examined in microbiology laboratory of Veterinary Medicine Faculty, Syiah Kuala University. Modification of Carter method was used in this study to isolated samples. The eggs were opened for taking the embryo and then it was swabbed with sterile swabs.  The swab was put into Selenite Cystine Broth (SCB) and incubated at 37o C for 24 hours. The breed was then transferred onto Salmonella Shigella Agar (SSA). The growing Bacteria were observed for their morphology and Gram staining to make sure the bacteria belonged to the negative Gram. The result showed that 10 of the 30 of quail eggs that failed to hatch were positively infected with Salmonella sp. Based on this study, it can be concluded that the salmonella sp.can be isolated from quail eggs which were not hatched in the farm of Garot, Darul Imarah Subdistrict, Aceh Besar
