Abstract

A total of 134 Pesisir cattle were genotyped for growth hormone (GH) gene by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The genotype and allele frequencies of the GH MspI and GH AluI Pesisir cattle were determined. The GH MspI gene frequencies for the C and T allele were 0.209 and 0.791 respectively, while GH AluI gene frequencies for the L and V allele were 0.992 and 0.008 respectively. The chi-square analysis indicated that this population is not in Hardy-Weinberg Equilibrium status. Expected heterozygosis value (He) for GH MspI and GH AluI were 0.3306±0.0266 and 0.0149±0.0073 respectively. The PCR-RFLP GH MspI marker has higher genetic variability compare to PCR-RFLP AluI marker. This finding showed that GH MspI T allele was favorable as a GH marker for Bos indicus breeds. Abstract

The aim of this study was to evaluate the effect of genetic and non-genetic factors on the growth traits including birth weight, weaning, and yearling weight of Bali cattle. Data were analyzed using generalized linear model (SAS) to observe non-genetic effect. To evaluate the genetic effect, the estimation of heritability were done using mixed models analysis with the dam and sire as random effect and sex, parity, year of birth, and season as fixed effect in the model besides the residual. The results showed that sex of calf had no significant influence on birth and weaning weight but had significant influence on yearling weight (P+0.07, 0.33+0.09, and 0.43+0.10 respectively. Heritability value of growth trait weaning and yearling weight in Bali cattle was quite moderate to high, so it was expected that selection in achieving increased gain on growth trait was effective.

Abstract

The aim of this study was to estimate genetic and phenotypic trends for growth traits including birth (BW), weaning (WW) and yearling weight (YW) in Bali cattle. The number of cattle used to determine growth traits of BW, WW, and YW were 235, 215, and 178 heads, respectively. Estimation of breeding value, phenotypic and genetic correlation were calculated by Restricted Maximum Likelihood and General Linier Model (GLM) procedures, respectively. Genetic trends analysis was performed using the regression mean breeding values on birth year. Phenotypic and genetic correlation among BW and WW were 0.10 and 0.08 respectively and 0.90 and 0.70 for WW and YW respectively. The phenotypic trends for traits of birth and weaning weight were constant, whereas yearling weight was fluctuating from 2000 to 2008. Likewise, in the case of genetic trends, the birth and weaning weight were constant from 2000 to 2008 except for WW in 2005, whereas the genetic trends for yearling weight showed a fluctuation of wide range. According to the breeding value estimated for all traits, the best was the sire No. 0565, whose breeding value for BW, WW and YW were +0.07, +2.79, and +10.25 kg, respectively higher than the mean value of the population. The genetic trends showed that there have been a significant and positive genetic improvement in all growth traits and indicate that selection would be effective. Genetic correlation between WW and YW was high (0.70) which indicates that the selection on weaning weight might also increase yearling weight in Bali cattle.

Abstract

Several bird species are sexually monomorphic. In this case, molecular approach is an efficient method for their sex determination. The sexes of monomorphic birds can be determined by PCR amplification of the CHD genes. CHD genes are preserved within avian Z and W sex chromosomes. The objective of this research was to determine sex of 21 individuals quails, pigeons, hill myna, salmon-crested cockatoos, and yellow-crested cockatoos based on CHD genes using two pairs of primers: 2550F-2718R and P2-P8. Samples from Kampong chickens and ducks were used as controls. PCR products were screened by agarose gel electrophoresis with ethidium bromide. Results showed that 2550F and 2718R primers could be used to identify sex of Kampong chickens, quails, ducks, pigeons, hill myna, salmon-crested cockatoos, and yellow-crested cockatoos. Individuals showed double (ZW) and single (ZZ) bands were identified as females and males, respectively in Kampong chickens, quails, hill myna, salmon-crested cockatoos and yellow-crested cockatoos. Males and females in ducks and pigeons showed single band in different length of base pairs. P2 and P8 primers could be used to identify the sex of pigeons, hill myna, salmon-crested cockatoos, and yellow-crested cockatoos. All of those samples showed bouble (ZW) bands for females and single (ZZ) band for males.

Abstract

Bacteriocins produced by Indonesian lactic acid bacteria Lactobacillus plantarum IIA-1A5, IIA-1B1, IIA-2B2 were purified and characterized. Plantaricin W gene had been successfully amplified from all strains. This amplicon showed the expected 200 bp size of plantaricin W gene. This bacteriocins purified from L. plantarum IIA-1A5, IIA-1B1, and IIA-2B2 were named plantaricin IIA-1A5, IIA-1B1, and IIA-2B2. Purification by cation exchange chromatography increased the purity (fold) and activity of plantaricins. Purity of plantaricin IIA-1A5 was increased by 3.13 fold with specific activity 13.40 AU/mg. Plantaricin IIA-1B1 had 2.98 fold purity with specific activity 5.12 AU/mg, while purity of plantaricin IIA-2B2 was 1.37 fold with specific activity 7.70 AU/mg. All plantaricins could inhibit the growth of pathogenic bacteria, such as Escherichia coli, Salmonella typhimurium, Bacillus cereus, and Staphylococcus aureus. Plantaricins could be digested by trypsin. Stability of plantaricins at 80 oC for 30 min and at 121 oC for 15 min were affected by type of plantaricin and species of pathogenic bacteria. Generally, plantaricin IIA-1A5 was better as antimicrobial agent than plantaricin IIA-1B1 and plantaricin IIA-2B2.

Abstract

The objective of this research was to identify genetic variation of mitochondria DNA especially in cytochrome oxidase subunit I (COI) among population of Indonesian buffaloes. Samples of swamp buffaloes were collected from Aceh (n= 3), North Sumatra (n= 3), Riau (n= 3), Banten (n= 3), Central Java (n= 3), West Nusa Tenggara (n= 3) and South Sulawesi (n= 3), and riverine buffalo from North Sumatra (n= 1) out of group for comparison. Sequence of COI was analyzed using MEGA 5.10 software with neighbor-joining method kimura 2-parameter model to reconstruct phylogeny tree. The result showed that three haplotypes for swamp buffalo and one haplotype for riverine buffalo in Indonesia resulted from 41 polymorphic sites. This finding showed that the COI gene could be considered as a marker to distinguish among swamp buffaloes in Indonesia.

Abstract

The aim of this study was to identify the genetic variability of PRL and STAT5A genes in Bali cattle from Bali, West Nusa Tenggara (NTB), and South Sulawesi (SS) using PCR-RFLP method. A total of 262 Bali cattle were identified using RsaI (PRL) and AvaI (STAT5A) restriction enzymes. PRL gene exon 3, PRL gene exon 4, and STAT5A gene exon 7 amplifications resulted fragments with the lengths of 156 bp, 294 bp, 215 bp, respectively. Genotyping of PRL gene both at exon 3 and 4 produced three genotypes in Bali population and two genotypes in NTB and SS population. For the PRL gene, frequencies of A allele (exon 3) and G allele (exon 4) were dominant to the B allele (exon 3) and A allele (exon 4) across all populations. The results showed that STAT5A|AvaI loci had monomorphic C allele.  Heterozygosity values were found low at both exons 3 and 4 of PRL gene in all population. Sequence analysis results of PRL gene both for exons 3 and 4 showed that there was a mutation between adenine (A) and guanine (G) bases in the RsaI recognized site, whereas in STAT5A gene we can confirm the existence of AvaI restriction site (C|CCGAG).Key words: bali cattle, genetic polymorphism, PCR-RFLP, prolactin gene, STAT5A gene

Abstract

Factor XI Deficiency (FXID) is caused by imperfect insertion of poly adenine which is resulted in introduction of premature stop codon in FXI gene.  Substitution of guanine into thymine in SLC35A3 gene caused Complex Vertebral Malformation (CVM). The research was aimed to detect the presence or absence of a genetic defect mainly CVM using SLC35A3 gene and FXID using FXI gene in Indonesian Bali cattle. The presence of this genetic defect may have a significant economic impact on the breeding program. The research of genetic defect was done mostly in dairy cattle, but there was no report for screening of genetic defect in Bali cattle. In this study, 303 fresh blood samples and 22 semen samples which were collected from Indonesian Bali cattle breeding center (BPTU HMT Denpasar, BPT HMT Serading West Nusa Tenggara and district Barru South Sulawesi) and artificial insemination centre (BBIB Singosari and BIBD Baturiti) were used for screening of FXID and CVM. The amplicons of FXI gene were obtained by using PCR and that for SLC35A3 gene were obtained by using PCR-RFLP method with PstI restriction enzyme. These PCR products were analyzed by using 2% agarose gels electrophoresis. All genotypes were confirmed by DNA sequencing to determine an allele mutant. The allele mutant was not found in all of the samples. The result of this study showed that CVM and FXID were not detected in Bali cattle from Indonesian Bali cattle breeding and artificial insemination centres.Key words: Bali cattle, genetic defect, SLC35S3 gene, factor XI gene

Abstract

Identification of genes in rabbits correlated to economic traits were intended to improve and develop their genetic quality. The objective of this research was to analyze the variability of growth hormone gene (GH) in three rabbit breeds, i.e. Rex, Satin, and Reza (Rex and Satin crosses) then was associated with ultrasound imaging of Longissimus dorsi muscle and perirenal fat thickness. Identification of the variability of growth hormone gene was analyzed using PCR RFLP technique from blood samples of 33 mature male rabbits in Indonesian Research Institute for Animal Production (IRIAP). Thickness of Longissimus dorsi muscle and perirenal fat were imaged and measured by using ultrasound unit at 2nd to 3rd lumbar vertebrae in the left body side. PCR product of GH gene fragment (231 base pair /bp) was digested with restriction enzyme Bsh1236I. PCR-RFLP patterns were allele T resulted in an undigested fragment of 231 bp;  allele C resulted in fragment of 169 bp and 62 bp. The result showed that Bsh1236I GH gene had three genotypes, i.e. CC, TT, and CT. There were signifficant association of Longissimus dorsi muscle thickness between rabbit breed (P<0.05). There was no significant association between GH Bsh1236I gene polymorphism and imaging ultrasound of Longissimus dorsi muscle and perirenal fat thickness. The association of characteristic genotype of GH|Bsh1236Igene with measurement phenotype was not significant, however it had potency as marker assisted selection (MAS). Key words: growth hormone gene, Longissimus dorsi, perirenal fat, rabbit, ultrasound

Abstract

The aim of this study was to estimate genetic and phenotypic parameters of growth traits consisting of birth weight (BW), weaning weight (WW), 6 mo body weight (6WM), 12 mo body weight (12WM), and 18 mo body weight (18WM) of Ettawa Grade goats. The number of goat used to determine growth traits of BW, WW, 6WM, 12WM, and 18WM were 316; 316; 259; 259 and 165 heads, respectively. Data were analyzed using General Linear Model (GLM) to identify non-genetic effect. Estimation of genetic and phenotypic parameters including heritability, repeatability, genetic and phenotypic correlation were calculated using Restricted Maximum Likelihood and GLM. Genetic trends were calculated using the regression of mean breeding values on birth year. The results showed that parity and type of birth had significant (P<0.05) influence on all growth traits. Estimated heritability of birth, weaning, 6WM, 12WM, and 18WM were 0.54±0.12; 0.35±0.07; 0.37±0.09; 0.68±0.16 and 0.63±0.19, respectively. Estimated repeatability of WW, 6WM, 12WM and 18WM, WW, 6WM, 12WM and 18WM were 0.98±0.01; 0.97±0.01; 0.94±0.03; 0.71±0.12 and 0.91±0.04, respectively. The genetic trends for traits of BW and 18MW were decreased fluctuatively. However, the WW, 6MW, 12MW were increased fluctuatively. The high and positive genetic correlations between all growth traits and 12WM traits in this study indicated that selection for high 12WM will improve genetic merit in Ettawa Grade goats. Key words: body weight, EBV, Ettawa Grade goat, genetic and phenotypic parameters
