Abstract

Genetics characterization in livestock based on microsatellite has been widely implemented including for Bali cattle in three different breeding centres in Indonesia. This study aimed to determine diversity of Bali cattle microsatellites in three breeding centers namely BPTU Bali cattle in Bali, BPT-HMT Serading Sumbawa in West Nusa Tenggara, and Village Breeding Center in South Sulawesi. The number of animals used in this study was 95 head of cattle consisted of 32 heads from BPTU Bali Province, 32 heads from BPT-HMT Serading Sumbawa, and 31 heads from Village Barru Breeding Center Barru distric. Microsatellite loci used to determine the diversity was the locus SPS115, INRA037, MM12, and ETH185 based on flourescently labeled fragment method. Data analysis of microsatellite in Bali cattle at three different locations was performed by using POPGEN 1.2, Cervus, and POPTREE2 programs. The results showed that microsatellite diversity in Bali cattle detected 32 alleles from three different locations, and there were specific alleles at each location. Average values of observed heterozygosity (Ho) and expected heterozygosity (He) were 0.418 and 0.604 respectively, while the average value of polymorphism informative content (PIC) was 0.579. The Hardy-Weinberg equilibrium in general suggested that the loci used in the Bali cattle in three populations were in equilibrium, except INRA037 and ETH185. The genetic diversity between populations of Bali cattle was 0.033 (3%), while the inbreeding coefficient index in all populations was 0.296 (29.6 %). Bali cattle phylogeny tree with three populations showed that the populations of Bali cattle in BPTU Bali and VBC Barru had close genetic distance compared to the population of Bali cattle in BPT-HMT Serading Sumbawa. The results of this study provide information that the characteristics of Bali cattle breeding centers in three locations are different, so we need a directed breeding program in each population.Key words: Bali cattle, genetics diversity, microsatellite

Abstract

Calpastatin (CAST) gene is well known as an inhibitor of muscle protein degradation and relates to muscle growth and meat tenderness. The objective of this study was to determine the association of CAST gene with growth traits and carcass characteristics in Bali cattle. A number of data from 35 Bali bulls were collected from BPTU Bali Cattle to obtain growth traits, carcass characteristics, and blood samples. Polymorphism of CAST gene in Bali bulls was analyzed by using PCR-RFLP and DNA sequencing. The association of CAST gene with growth traits and carcass characteristics were analyzed by using General Linear Model (GLM). The result showed that there were two genotypes (GG and AG) of CAST gene with allele frequencies of 0.857 and 0.143, respectively, for G and A. Notably, mutation A to G occurred in 253 bp CAST fragment gene in Bali Cattle. Genotypes GG and AG of CAST gene significantly affected (P<0.05) the back-fat thickness and longissimus dorsi without a significant effect on the growth traits. It could be concluded that CAST gene had a potency as a marker gene for carcass quality in Bali cattle.  Key words: Bali cattle, CAST gene, PCR-RFLP, polymorphism

Abstract

This study aimed to determine the Pit-1|Hinf-1 gene polymorphism in Bali cattle (Bos javanicus) as Indonesian native cattle and besides Madura, Pesisir, Aceh, and Katingan cattle breeds as a comparison. DNA samples were extracted from 488 blood samples consisting of Bali (245 heads), Madura (68 heads), Aceh (25 heads), Pesisir (100 heads) and Katingan (50 heads) cattle. The diversity of the Pit-1|Hinf-1 gene wasere analyzed using PCR-RFLP. Whereas the nucleotide base mutations were identified by sequencing. Genotyping data were analyzed by calculating the allele frequency, observed heterozygosity (Ho) and expected heterozygosity (He) values as well as Hardry-Weinberg equilibrium test using POPGENE 1.31 program. Whereas, Tthe sequence data were analyzed by using MEGA6 program. The Pit-1|Hinf-1 gene fragment analysis showed that Bali, Madura, Pesisir, Aceh, and Katingan cattle had high BB genotype, resulting in B allele frequency of 0.982, 0.963, 0.925, 0.960, and 0.960, respectively. Ho and He values were 0.074-0.130 and 0.036-0.139, respectively. Hardy-Weinberg equilibrium test did not significant for all breed populations, except for Aceh cattle population (P<0.05). Mutation from guanine (G) to adenine (A) was found in Pit-1 gene fragment. Therefore, Pit-1|Hinf-1 gene fragment had low genetic diversity in Bali cattle and other breeds population.Key words: Bali cattle, Pit-1|Hinf-1 gene, polymorphism

Abstract

This research aimed to study the supplementation effects of mangosteen pericarp meal (MPM) and vitamin E (VE) in the diets on the egg quality and blood profile of laying hens. This research used 160 laying hens of Lohman strains 24 weeks of age. The observation was conducted for 11 weeks. A completely randomized design with four treatments and four replications (10 birds each) was used in this experiment. The treatments consisted of R0 (control diet), R1 (R0 + 1 g MPM/kg ration), R2 (R0 + 2 g MPM/kg ration) and R3 (R0 + 200 mg VE/kg ration). Variables measured were egg quality, yolk cholesterol, and blood profiles. The data were analyzed by using analysis of variance (ANOVA) and any significant difference between the treatment means were further tested by Duncan’s Multiple Range Test. The results showed that supplementation of 1 g MPM/kg ration in the diet significantly (P<0.05) decreased blood triglycerides compared with the control, laying hens fed with diet suplemented with of 2 g MPM/kg ration, and laying hens with diet suplemented with 200 mg VE/kg  ration. Supplementation of MPM and VE did not affect (P>0.05) egg quality (except shell thickness), blood cholesterol, and HDL, respectively. In conclusion, supplementation of 1 g MPM/kg in the diet of laying hens could decrease blood triglycerides.Key words: blood profile, laying hens, mangosteen pericarp meal, egg quality, vitamin E

Abstract

The existence of selection on the rabbits with potential for meat has only been seen from phenotypic aspects including performance and productivity, while the molecular genetic studies are still very rare. One of the candidate genes for meat production traits in rabbit is myostatin. Totally 50 blood samples of male rabbits from Rex, Satin, Reza (crossing from Rex and Satin), Flemish Giant and FZ3 (crossing from Flemish Giant and Reza) breed were used at Indonesian Research Institute for Animal Production (IRIAP). Genetic polymorphism by Polymerase Chain Reaction – Restriction Fragment Length Polymorphism (PCR-RFLP) method used FspBI restriction enzyme. PCR-RFLP data were analyzed by calculating allele and genotype frequencies. Sequencing was performed in rabbit with different genotypes which represents each of the samples. Genotype of AT had two cut points of the FspBI restriction enzyme at the base position of 508 bp and 444 bp. The cut point at the base position of 446 bp was site mutation base T became A. Genotype of TT had one cut point at the base position of 508 bp and no mutation site. Allele T had higher frequency than allele A and just Rex and Reza rabbit breeds had two alleles. The other rabbits (Satin, Flemish Giant and FZ3) only had one allele i.e., allele T. PCR - RFLP analysis of the MSTN|FspBI gene segments was polymorphic in Rex and Reza rabbit breeds. All of rabbit breeds in this study did not have AA genotype.Key words: myostatin gene, rabbit

Abstract

Myostatin (MSTN) gene plays a key role in skletal muscle homeostasis such as inducing muscle athrophy, poliferation of myoblast, increasing ubiquitin-proteasomal, downregulating IGF pathway, and glucolysis. Myostatin gene expression is controled by CpG island located in promoter region. The objectives of this research were to identify polymorphism of MSTN promoter gene and to associate the polymorphism of SNP with growth and muscling traits in Bali cattle. A total of 48 Bali cattle from BPTU-HMT Bali island was screened to identify genetic polymorphisms in MSTN promoter region using sequencing method. The growth and muscling traits were measured at 12 months of age. The muscling traits were evaluated using ultrasound console with linear transducer having frequency 6.5 Hz and scaning we conducted at 130 mm in deep. Analysis of polymorphism was conducted by using PopGen 1.32 software. The association of MSTN with growth and muscling traits were analyzed by using General Linear Model (GLM) procedure. This result showed that a total of 20 polymorphic SNPs (seven SNPs in CpG island) were detected in this region. Although, only 3 SNPs (g.-8078C>T, g.-7996G>C, and g.-7930A>G) had equilibrium condition in Hardy-Weinberg analysis. The association result showed that 2 SNPs (g.-7799T>C and g.-7941C>T) were significantly associated with intramuscular fat percentage (P≤0.05) in Bali cattle. Although the 2 SNPs were nominally significant at nominal P≤0.05 threshold, they were not significant after Bonferroni correction for multiple testing. It could be concluded that MSTN promoter gene was polymorphic in Bali cattle and there were 2 SNPs associated with carcass quality.

Abstract

The chicken growth hormone (GH) and its receptor (growth hormone receptor, GHR) play important roles in chicken performances due to their crucial functions in growth. The variations of GH and GHR genes were then thought to be associated with the variations of the performances. This experiment was designed to identify the g.2248G>A GH and the g.565G>A GHR loci polymorphisms and to evaluate their associations with carcass components in Kampung and broiler chicken cross. A total of 215 chickens including 4 chicken populations (Kampung, Cobb broiler, F1, and F2 Kampung x broiler chicken cross) were screened to identify polymorphism using PCR-RFLP technique with EcoRV and Eco72I restriction enzyme for GH and GHR loci, respectively. The carcass components were recorded at 26 weeks of age on F2 Kampung and broiler chicken cross (42 chickens) for association study. Both the g.2248G>A GH and the g.565G>A GHR loci were polymorphic with two alleles (G and A) and three genotypes (GG, AG, and AA). The GG genotype and the G allele of GH locus were predominant in all chicken populations. While in GHR locus, the AA genotype and the A allele were found to be higher in all chicken populations. The association study showed that the g.565G>A GHR locus polymorphism had significant effect on carcass components, including live weight, carcass weight, breast weight, thighs weight, breast muscle weight, and thighs muscle weight. There was no significant association was found between the g.2248G>A GH genotype and carcass components. It could be concluded that the g.2248G>A GH and the g.565G>A GHR loci were polymorphic in Kampung and broiler chicken cross and the g.565G>A GHR locus was accosiated with carcass components. This g.565G>A GHR SNP might be an important candidate marker for chicken growth and muscle mass improvement.

Abstract

The aim of this study was to identify the genetic polymorphisms of calpastatin (CAST), calpain 3 (CAPN3) and myostatin (MSTN) on local sheep at Jonggol Animal Science Teaching and Research Unit (JASTRU). A total number of 294 blood samples were collected from JASTRU. The identification of polymorhism in CAST and CAPN3 genes performed by using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) while MSTN gene by using PCR-SSCP methods. The results showed that CAST|MspI, CAST|NcoI and CAPN3|MaeII loci were polymorphic, whereas The MSTN locus was monomorphic for G (1.0). The frequency of allele M (0.87) on the locus (CAST|MspI) higher than the N allele (0.13). At locus CAST|NcoI, the frequency of allele M (0.96) higher than the N allele (0.04). At the CAPN3|MaeII, allele G (0.85) and allele T (0.15). Locus CAST|NcoI has higher observed heterozygosity (Ho = 0.92) compared to CAPN3|MaeII and CAST|MspI (Ho = 0.74-0.77), however has lower compared to CAPN3|MaeII and CAST|MspI in expected of heterozygosity (He = 0.08 vs 0.23-0.26) and in index fixation (Fis = -0.04 vs 0.03-0.12).
