Abstract

Myostatin (MSTN) gene acts as a negative regulator of muscle growth. The aim of the present study was to identify polymorphism of g.-371T>A in promoter region of MSTN gene in Indonesian beef cattle. Blood samples were collected from 191 cattle, including Bali (BL) (42), Madura (MD) (20), Pesisir (PI) (17), Katingan (KT) (16), Ongole grade (PO) (22), Pasundan (PD) (14), Sumba ongole (SO) (10), Brahman (BH) (17), Simmental (SM) (15), and Limousin (LM) (18). Polymorphism of MSTN gene was analyzed using PCR-RFLP (DraI) and direct sequencing methods. Results of genotyping g-371T>A were polymorphic (TT, TA, and AA genotypes) in Simmental, SO, and Katingan. The frequencies of alleles T and A were 0.83; 0.90; 0.97 and 0.17; 0.10; 0.03 respectively. The values of Ho and He were 0.06–0.20 and 0.06–0.28, respectively. The results of this study are in Hardy-Weinberg equilibrium (P> 0.05). Whereas in PD, MD, PI, PO, BH, LM, and BL were monomorphic (TT genotype). The result of sequencing the promoter region of MSTN gene found that mutations transversion was occurred in T to A at g.-371. It was concluded that g.-371T>A of MSTN gene was polymorphic which was potential to be used as genetic markers of muscle growth in SM, SO, and KT cattle. Abstract

Etawah grade goat is a result of grading up line between Kacang and Etawah goats, which well adapted to Indonesia’s humid tropical climate. This goat is a dual-purpose goat breed for milk and meat production. The objectives of this study were to evaluate the polymorphism of fecundity genes through the investigation of four candidate genes for prolificacy, i.e., BMP15 (exon 1), BMPR1B (exon 1), KISS1 (exon 1), and KISS1 (intron 1) and their associations with the litter size of Etawah-Grade does. The diversity of BMP15 (exon 1), BMPR1B (exon 1), KISS1 (exon 1), and KISS1 (introns 1) genes were analyzed by the PCR-RFLP technique. A total of 106 of DNA samples were collected from Etawah-Grade does at BPTU-HPT Pelaihari in Pelaihari, South Kalimantan (51 heads) and Cordero Farm in Bogor, West Java (55 heads). Genetic polymorphisms between the two Etawah-Grade groups were calculated as well as their allele and genotype frequencies and the Polymorphic Informative Content (PIC). The Kruskal-Wallis one-factor ranked analysis of variance was used to determine whether there was any effect of genotype of the fecundity genes on litter size. The polymorphic information content (PIC) estimated at BPTU-HPT Pelaihari was 0.313, whereas at Cordero farm was 0.174, and overall PIC was 0.244. There was no association between genotypes of BMP15 (exon 1), BMPR1B (exon 1), KISS1 (exon 1), and KISS1 (intron 1) genes and litter size. Results indicated that BMP15 (exon 1) and KISS1 (exon 1) genes were monomorphic. However, the other two loci showed polymorphisms. The BMPR1B (exon 1) locus had two genotypes with the frequency for CC (96.23%) and for GG (3.77%), and KISS1 (intron 1) locus had three genotypes with the frequency for TT (80.19%), AT (17.92%), and AA (1.89%).

Abstract

Bali cattle (Bos javanicus) are native Indonesian cattle, domesticated from banteng (Bibos banteng). Genes that have an important role in meat quality are calcium-activated neutral protease genes, known as calpains (CAPN). The objective of this study was to evaluate the polymophisms of calpain gene SNP g.232 G>T by PCR-RFLP technique and its influence on growth trait and meat quality of Bali cattle detected by ultrasound imaging of longissimus dorsi thickness (LDT), back fat thickness (BFT), marbling score (MS), and intramuscular fat percentage (PIMF). The polymorphisms of CAPN1 gene were analyzed by PCR-RFLP using BglII restriction enzyme (n=52 cattle). The ultrasound images of longissimus dorsi muscle were carried out transversally and longitudinal between 12th -13th thoracic vertebrae then analyzed by Image-J NIH software. Result showed that  SNP g.232 G>T of CAPN1 gene was polymorphic in Bali cattle. SNP g.232 G>T of CAPN1 gene in Bali cattle has higher diversity which was showed of 0.48 heterozygosity value and was in Hardy-Weinberg equilibrium. The polymorphisms of SNP g.232 G>T was associated significantly (P<0.05) with bodyweight at 730 days, marbling score (MS), and intramuscular fat percentage (PIMF). It suggests that the CAPN1 gene in Bali cattle is a candidate for Marker Assisted Selection (MAS), which influences body weight at 730 days, marbling score, and percentage of intramuscular fat.

Abstract

Productivity data from 560 head of PO cattle have been collected for 9 years from 2004 until 2013 for evaluating heritability estimation and environmental factors affecting production traits of Indonesian Ongole cross from birth to yearling old. Heritability estimation were analyzed using varians analysis with data of paternal halfsib correlation. Varians component for to estimate of heritability value were analyzed using completely randomized design one - way classification. While a general linear model was used to analyze the environmental factors (sex, year, parity, season and type of births). The results showed that sex of calves and year of births had significant differences (P<0.05) for all production traits such as weight at birth, weaning and yearling, while the type of birth only presented a significant difference on birth weight. Parity, season and generation exhibited nonsignificantly effect on those production traits. Estimation of heritability on birth weight, weaning weight and yearling weight were 0.28+0.12; 0.47+0.15 and 0.63+0.17 respectively. The highest heritability values obtained in yearling weight, this means that yearling weight will be effective as selection criteria to improve the genetic of PO cattle.

Abstract

Bali cattle (Bos javanicus) are common species distributed throughout Indonesia to survive in tropical hot, and humid climates. Fluctuating asymmetry (FA) is a random deviation from perfect bilateral symmetry. FA is often used to measure developmental stability in individuals and can be used as a marker for the impact of environmental and genetic stress. This study aims to investigate the pattern of developmental instability in Bali cattle as caused by environmental stress at various altitudes using the FA index. FA indexes in this study were used to compare asymmetrical physical traits of Bali cattle in Sembalun high altitude and Serading low altitude. Sixty-five Bali cattle were used in this study reared at different altitudes, namely Serading, Sumbawa Island (50 m above sea level), and Sembalun, Lombok Island (1,186 m above sea level). The physical traits of Bali cattle measured were horn circumference (horn), a distance of hip to spine bone (pelvic), metatarsal circumference (metatarsal), and metacarpal circumference (metacarpal). The FA1 and FA5 indexes showed significant metacarpal differences between Bali cattle reared at Sembalun and Serading population (P<0.05). FA11 multiple trait index as a composite of all traits revealed a significant difference as well. (P<001). It can be concluded that various heat stress by altitude impacts the asymmetry of Bali cattle.

Abstract

The objective of this study was to evaluate effect of genetic and non-genetic factors on semen characteristics including ejaculate volume, sperm concentration, total sperm number and sperm moetility of Bali cattle. Semen data were collected from the National Centre of Artificial Insemination at Singosari, Malang, East Java, Indonesia. A total of 3,847 ejaculates of 17 Bali bulls from 2014 to 2016 were collected and evaluated. Data were analyzed by restricted maximum likelihood (REML) method using mixed models which the bull was a random effect, while age of bull, season of collection, frequency of ejaculation and collection intervals were the fixed effects. Results showed that age significantly affected all semen characteristics (P<0.01). Season affected only on sperm motility (P<0.01). Effect of frequency of ejaculation and collection intervals was significant on all studied variables (P<0.01), except sperm motility. Repeatability of ejaculate volume, sperm concentration, total sperm number and sperm motility was 0.43; 0.35; 0.32 and 0.31, respectively. It is concluded that age, frequency of ejaculation and collection intervals were the most factors affected semen characteristics of Bali cattle. Repeatability estimations of semen characteristics were moderate to high.

Abstract

The purpose of this study was to determine the diversity of qualitative traits in PO, Belgian Blue and PO-Belgian Blue crosses using principal component analysis. The total samples of cattle used were 36 consisting of 8 heads Belgian Blue (4 males and 4 females), 17 heads Belgian PO-Belgian cross-breed (11 males and 6 females) and 11 PO heads (all females)). The diversity of qualitative traits observed were 17 characteristics namely plain body-color (WTP), combination body-color (WTK), horned (MT), humped (MP), sagging (MG), black tail tip color (WUEH), white tail tip color (WUEH), black eyelash color (WBMH), white eyelash color (WBMP), circle hair head (PRK), circle hair back (PRP), circle hair bracket (PRB), black nail color (WKH), nail color white (WKP), double muscle (PG), black muzzle (WMH) and white muzzle (WMP). Qualitative data were analyzed descriptively by calculating the proportion value, while the determinant variables of each breed were analyzed using the principal component analysis method (AKU) with the Minitab version 7 program. The results showed that the characteristics of qualitative traits in PO, Belgian Blue, and PO-Belgian Blue cross-breed cattle have a high diversity. PO and Belgian Blue cattle breeds can be clearly distinguished based on the characteristics of qualitative traits, whereas PO-Belgian Blue crossbred cattle have clusters that are close to PO cattle clusters. Found character traits for each cattle breed in both PO, Belgian Blue, and PO-Belgian Blue cross-breed. There are three characteristics of qualitative traits possessed in each cow nation, namely the variable circle hair head (PRK), circle hair back (PRP) and horned characters (MT). Based on the results obtained that the qualitative traits in cattle can be used as a distinguishing character between PO, Belgian Blue, and PO-Blue Belgian cross cattle breeds.

Abstract

ABSTRAK Gen FTO berfungsi sebagai regulasi homeostasis, deposisi lemak dan pengaturan obesitas. Penelitian ini bertujuan untuk mengidentifikasi polimorfisme SNP g.125550A>T di ekson 3 gen FTO pada bangsa sapi potong Indonesia. Sampel darah diperoleh dari 209 ekor sapi, terdiri atas sapi bali (44), madura (20), Pesisir (20), katingan (20), Peranakan ongole (PO) (22), Pasundan (20), Sumba Ongole (SO) (11), brahman (20), simental (15), dan limousin (18). Polimorfisme gen FTO dianalisis menggunakan metode PCR-RFLP (HpyCH4III) dan direct sequencing. Hasil genotiping SNP g.125550A>T adalah polimorfik (genotipe AA, AT, dan TT) pada sapi madura, pesisir, katingan, PO, pasundan, SO, brahman, simental, dan limousin. Frekuensi alel A dan T masing-masing adalah 0,70, 0,68, 0,84, 0,89, 0,70, 0,86, 0,90, 0,73, 0,69 dan 0,30, 0,33, 0,16, 0,11, 0,30, 0,14, 0,10, 0,27, 0,31. Nilai Ho dan He masing-masing adalah 0,60-0,14 dan 0,44-0,18 serta dalam keseimbangan Hardy-Weinberg (P>0.05). Sementara pada sapi bali bersifat monomorfik hanya bergenotipe AA. Hasil sekuensing SNP g.125550A>T ditemukan mutasi tranvesi A menjadi T pada posisi nukleotida  g.125550. Berdasarkan hasil penelitian ini, dapat disimpulkan bahwa SNP 125550A>T gen FTO beragam dan berpotensi dijadikan marka genetik untuk kualitas daging pada bangsa sapi potong Indonesia.Kata Kunci: gen FTO, PCR-RFLP, Sapi, SNP g.125550A>TABSTRACTThe FTO gene functions as regulation of homeostasis, fat deposition and regulation of obesity. Results of sequencing SNP g.125550A>T of the FTO gene found a transverse mutation A to T at the nucleotide position g.125550. As a result of this study, it can be concluded that SNP 125550A>T of the FTO gene was diverse and potentially used as genetic markers for meat quality in Indonesian beef cattle.Keywords: cattle, FTO gene, PCR-RFLP, SNP g.125550A>T.

Abstract

Penelitian ini bertujuan untuk mengkaji pengaruh suplementasi tepung kulit manggis (TKM) dan Vitamin E (VE) di dalam ransum ayam ras petelur terhadap kualitas fisik telur selama penyimpanan pada waktu dan suhu yang berbeda. Ayam petelur diberi perlakuan pakan yaitu : R0 (ransum kontrol), R1 (R0 +1 g TKM/Kg ransum), R2 (R0 + 2 g TKM/Kg ransum) dan R3 (R0 + 200 mg VE/Kg ransum). Rancangan yang digunakan adalah rancangan acak lengkap pola faktorial 4 x 3 x 2 dengan 4 ulangan. Faktor pertama yaitu ransum, faktor kedua yaitu: waktu penyimpanan (1, 2 dan 3 minggu) dan faktor ketiga yaitu suhu penyimpanan pada refrigerator (4,13oC dan 22,50%) dan suhu ruang (27, 47oC dan 76,17%). Variabel yang diamati adalah: berat telur, persentase kerabang telur, persentase kuning telur, persentase putih telur, tebal kerabang, warna kuning telur dan Haught unit (HU). Hasil penelitian menunjukkan bahwa interaksi antara ransum, waktu dan suhu selama penyimpanan sangat nyata (P<0,01) menurunkan warna kuning telur. Waktu dan suhu penyimpanan sangat nyata (P<0,01) menurunkan HU dan meningkatkan persentase kuning telur. Interaksi waktu dan suhu penyimpanan nyata (P<0,05) menurunkan persentase putih telur. Tebal kerabang nyata (P<0,05) meningkat dipengaruhi oleh ransum dan sangat nyata (P<0,01) meningkat dipengaruhi oleh waktu penyimpanan. Berat telur dan persentase kerabang telur tidak dipengaruhi oleh ransum, waktu dan suhu penyimpanan. Kesimpulan penelitian adalah suplementasi tepung kulit manggis dan Vitamin E di dalam ransum ayam ras petelur strain Lohmann secara umum tidak mempengaruhi kualitas fisik telur (kecuali warna kuning telur dan tebal kerabang) yang disimpan pada waktu dan suhu yang berbeda. Kualitas fisik telur lebih utama dipengaruhi oleh waktu dan suhu penyimpanan yang berbeda. Suhu dan kelembaban terbaik untuk penyimpanan telur adalah 4,13oC dan 22,50% pada refrigerator, pada kondisi ini telur dapat disimpan selama 21 hari.

Abstract

The objective of this research was to characterized morphology and estimated genetic distance between intra population of Kuantan cattle. A Total of 213 cattle (44 male and 169 female with age ranging from 2-3 years) were used in this study and collected from extensive ranging systems in Three sub-population (Cerenti, Inuman, and Kuantan Hilir regions) Kuantan Singingi Regency, Riau Province. Five variables were measured that is Body Length (BL)(cm), Wither Height (WH)(cm), Hip Height (HH)(cm), Chest Girth (CG)(cm), and Chest Depth (CD)(cm). Data obtained were descriptive analysis, Principal Components Analysis (PCA) and Hierarchichal Clustering Analysis (HCA)  using XLSTAT program. All variables of body measurement in the Kuantan Hilir region were higher than Cerenti dan Inuman, Kuantan Singingi Regency. The first factor in PCA described body measurement contributed 32.77%, and the second factor described body shape contribute 25.83% of total variability. The dendrogram showed there is Three clusters of Kuantan Cattle based on this research.
