Abstract

This study aimed to investigate the genetic impact of single nucleotide polymorphisms (SNPs) of the sterol regulating element binding factor 1 (SREBF1) and scavenger receptor class B member 1 (SCARB1) genes on carcass and meat characteristics, as well as fatty acid composition, in the Bali cattle. The blood and beef samples used for DNA sequencing, physical assessment, and fatty acid analysis were collected from 95 male Bali cattle. The ultrasound images were analyzed using the Image-J NIH software. A total of 4 SNPs were identified in the SREBF1 gene and 5 SNPs in the SCARB1 gene. The results showed that the 4 SNPs in the SREBF1 gene, namely g.12629T>C, g.12731T>C, g.12881A>G, and g.12986C>T, were associated with heptadecanoic acid (C17:0) and cis-11-eicosanoic acid (C20:1). The SNPs g.12731T>C of the SREBF1 gene was associated with fat content, palmitoleic acid (C16:1), stearic acid (C18:0), cis-11-eicosanoic acid (C20:1), and total fatty acids. Furthermore, 4 SNPs in the SCARB1 gene, including g.72219C>T, g.72380C>A, g.72517G>A, and g.72607C>T correlated with longissimus dorsi thickness (LDT). All SNPs in the SCARB1 gene showed significant associations with cis-10 heptadecanoic acid (C17:1) and cis 8,11,14-eicosatrienoic acid (C20:3n6). The SNP g.72400A>G of the SCARB1 gene was related to caprylic acid (C8:0), lauric acid (C12:0), arachidonic acid (C20:4n6), monounsaturated fatty acids (MUFA), and unsaturated fatty acids (UFA). These results suggested that the identified polymorphisms in the SREBF1 and SCARB1 genes could serve as valuable references for investigating similar genes in other cattle breeds, particularly concerning fatty acids.

Abstract

This study aimed to investigate the genetic diversity of the Heat Shock Protein 70 (HSP70) gene in the coding sequence (CDS) region and its association with physiological responses in Bali cattle. The samples used in this study consisted of 62 Bali cattle from two different locations in Bali and Se-rading. Physiological data, including respiratory rate, heart rate, rectal temperature, and heat tolerance coefficient (HTC), were collected. The HSP70 gene was amplified using two pairs of primers to target the coding region. SNPs of the HSP70 gene were identified through sequencing. The diversity of SNPs in the coding sequence of the HSP70 gene was determined using the FinchTV 1.4.0 application and version X of the Molecular Evolutionary Genetic Analysis (MEGA) program. In contrast, the association of HSP70 gene SNPs with physiological responses in Bali cattle was evaluated using the GLM method in IBM SPSS Statistics 26. The research discovered nine SNPs within the CDS region of the HSP70 gene, comprising eight synonymous SNPs (c.24C>T, c.31C>T, c.117C>A, c.126G>A, c.324G>A, c.333C>T, c.573G>C, c.1074C>T) and one nonsynonymous SNP (c.1265C>T), which caused an amino acid substitution from threonine (T) to methionine (M). The SNP diversity in the coding sequence of the HSP70 gene showed no significant association (P > 0.05) with physiological responses, including respiratory rate, heart rate, rectal temperature, and heat tolerance coefficient (HTC). The SNPs discovered in the coding region of the HSP70 gene exhibited polymorphism in Bali cattle.

Abstract

The growing chicken population in Indonesia reflects the expanding poultry farming industry. To maintain chicken health and treat diseases, vitamins and antibiotics are commonly used, with guidelines for their use changing over time. To understand usage trends, data from PT Tekad Mandiri Citra (TMC) in Bandung is needed. This study evaluates the sales patterns of poultry antibiotics and vitamins in the veterinary drug industry between 2020 and 2022, with a focus on farm health and sustainability. Additionally, a SWOT analysis will be used to replace colistin antibiotics, based on changes to the Minister of Agriculture Regulation No. 14 of 2017. Data were processed using Microsoft Excel 2019 for descriptive analysis. The results show that 79% of broiler farms use antibiotics, and 81% use vitamins. In 2022, the most common antibiotics were from the fluoroquinolone group (34%), and the most used vitamins were from the miscellaneous group (35%). The SWOT analysis for 2021-2022 indicates that replacing colistin products has been successful.

Abstract

This study examines the Stearoyl-CoA desaturase (SCD) gene, which is essential for converting saturated fatty acids (SFA) into monounsaturated fatty acids (MUFA). The research aims to analyze the diversity of Single Nucleotide Polymorphism (SNP) g.10428C>T within the SCD gene using the PCR-RFLP method. The investigation involved 40 blood samples from male Bali cattle aged 1.5 to 3 years, sourced from the Banjarmasin Slaughterhouse (RPH) in South Kalimantan. The SNP g.10428C>T diversity in the SCD gene was analyzed using the PCR-RFLP technique with the HinfI restriction enzyme. Genotype frequencies, allele frequencies, heterozygosity values, and the Hardy-Weinberg equilibrium were calculated using Popgen32. The relationship between fatty acid composition and the genotype of the SCD gene, as well as the fatty acids in Bali beef, was analyzed using a t-test. SCD|HinfI was found to have homozygous CC (0.03) and TT (0.85) and heterozygous CT (0.13) genotypes with a C allele frequency of 0.09 and T of 0.91. SNP g.10428C>T in the SCD gene was not significantly associated with fatty acid composition.

Abstract

The aim of this study was to evaluate effect of genetic and non genetic factor on the gestation length (GL) and calf survival to weaning (CS) of Bali cattle. Data were analyzed using Generalized Linier Model (SAS) to observe non-genetic effect. To evaluate the genetic effect, the estimations of heritability were done using Mixed Models analysis with the dam and sire as random effect and parity, year of birth and type of mating as fixed effect in the model besides the residual. The results showed non genetic factors except type of mating influenced the variability of gestation length (GL) and calf survival to weaning (CS) in Bali cattle. The result showed that dam year of birth and parity were found significant source of variation in the gestation length, whereas calf survival to weaning were affected of age of dam and year of birth.  Estimated heritability of  GL and CS were 0.68 ± 0.07  and 0.28 ± 0.09, respectively. With regards to genetic effect, estimated heritability of GL was considered higher (0.68), which means that GL information would be more effective as a selection criteria. Selection for shorter GL might be reduce calving problems with less impact on calf survival to weaning.
