Abstract

The crude ethanol extracts of four Indonesian medicinal plants namely Curcuma xanthorrhiza Roxb.,Phyllanthus niruri Linn., Andrographis paniculata Ness., and Curcuma aeruginosa Roxb. wereexamined for their antioxidant (radical scavenging) activity using 2, 2-diphenyl-2-picrylhydrazyl(DPPH) free radical and cytotoxicity using brine shrimp lethality test (BSLT). The total phenoliccontent was used the Folin-Ciocalteu method. IC50 values for DPPH radical scavenging activityranged from 14.5 to 178.5 μg/ml, with P. niruri having the lowest value and therefore the mostpotent, and C. aeruginosa having the highest value. LC50 values for BSLT ranged from 210.3 to593.2 μg/ml, with C. xanthorrhiza and A. paniculata having the lowest and highest values,respectively. The total phenolic content of the Indonesian plants ranged from 133.0 ±3.7 to863.3±54.7 mg tannic acid equivalent per 1 g extract, with C. aeruginosa and P. niruri having thelowest and highest values, respectively. A positive correlation between free radical scavengingactivity and the content of phenolic compounds was found in the four of Indonesian medicinal plants.

Abstract

Green tea (Camellia sinensis) is one of tradisional plants that have potential as anticancer. Thesample used in this research was commercial green tea extract. The purpose of this study was to test theantiproliferation activity of green tea extract on breast cancer cell MCM-B2 in vitro. Green tea extract wasfractionated using three solvents, ie water, ethanol 70%, and n-hexane. Extract and water fraction of greentea have Lethality Concentration 50 (LC50) value of more than 1000 ppm. Fractions of ethanol 70% and nhexane had an LC50 value of 883.48 ppm and 600.56 ppm, respectively. The results of phytochemicalscreening of green tea extract are flavonoids, tannins, and saponins, while the phytochemical screeningresults of n-hexane fraction are flavonoids and tannins. Antiproliferation activity was tested on breast cancercells MCM-B2 and normal cells Vero by tripan blue staining method. The highest MCM-B2 cell inhibitoryactivity was achieved at a concentration of 13000 ppm green tea extract and 1000 ppm of n-hexane fraction,59% and 59%, respectively. The extract and n-hexane fraction of green tea are not toxic to normal Verocells characterized by not inhibiting normal cell proliferation.

Abstract

Primary lung cancers, or tumors originating in the lung, are relatively uncommon in dogs. The objectiveof this study was to describe the canine lung carcinoma that serially transplanted into severe combinedimmunodeficiency (SCID) mice, in order to established cell line from this tumor cell.  Morphology andcharacteristic of this canine lung carcinoma in SCID mice by histopathological and ultrastructuralexaminations with metastatic lesion in lung were also examined.  Histopathologically, the tumor masswere consisted of cuboidal to columnar cells with papillary pattern, uniform in size, the nuclei were oftenvariable in size, and some cells have vacuole on their cytoplasms.  Glandular forms were predominant withlobulated pattern, ductal pattern with papillary injected into tube-like structure were also encountered.Mitotic figures commonly found with inflammatory reaction were sometimes present in the interstitiumand lumen gland.  Ultrastructural analysis of the tumor cells showed round to oval cells with one or moreprominent nucleoli.  The cells possessed numerous mitochondria, smooth endoplasmic reticulum, andindividual cells which were interconnected via desmosomes.  Tonofilament characterize by long cytoplasmicmaterial was encountered.  Positive reaction of the round to oval tumor cells to anti keratin antibodyconfirmed that their epithelial cell nature.  Lung metastatic lesions were found in SCID mice aftertransplantation and this phenomenon indicated that canine lung carcinoma is tumorigenic to SCID mice.

Abstract

Penelitian ini bertujuan untuk mempelajari efektivitas ekstrak etanol daun keladi tikus (Typhonium flagelliforme) untuk terapi penyakit tumor. Penelitian ini menggunakan enam ekor mencit betina berumur lima minggu dengan bobot badan 20-25 g yang telah diinduksi karsinogen 7.12- dimetilbenz(?)anthracne (DMBA). Mencit dibagi menjadi dua kelompok perlakuan masing-masing kelompok terdiri dari tiga ekor mencit yaitu kelompok yang diberi plasebo (KK) dan kelompok yang diberi ekstrak etanol daun keladi tikus (KE). Kelompok KK diberi plasebo berupa aquabides. Kelompok KE diberi ekstrak etanol daun keladi tikus dengan dosis 120 mg/kg BB. Pengamatan dilakukan secara makroskopik dan mikroskopik. Massa tumor dari kedua kelompok mencit dikoleksi dan difiksasi dalam neutral buffered formalin (NBF) 10%. Kemudian dibuat preparat histopatologi dan diwarnai pewarna Hematoksilin-Eosin (HE). Hasil penelitian menunjukkan gambaran makroskopik permukaan kulit pada daerah tumbuhnya tumor mengalami alopesia, tumbuh benjolan berwarna kemerahan dan keropeng. Hasil pengukuran bobot badan mencit kelompok KK mengalami penurunan, sedangkan berat badan mencit pada kelompok KE mengalami kenaikan setiap minggunya. Diameter tumor kelompok KK mengalami peningkatan, sedangkan diameter tumor pada kelompok KE mengalami penurunan pada minggu ke empat pemberian bahan uji. Gambaran mikroskopik menunjukkan benjolan massa tumor yang terbentuk terdiri atas tiga jenis tumor yaitu anaplastic carcinoma (16,7%), fibrosarcoma (16,7%) dan squamous cell carcinoma (66,6%). Ketiga tumor digolongkan dalam diferensiasi baik 33,4%, diferensiasi sedang 33,4%, diferensiasi buruk 16,7%, dan tidak berdiferensiasi 16,.7%. Laju pertumbuhan tumor selaras dengan tingkat diferensiasi. Tumor yang bermetastasis sebanyak 50% dan tidak bermetastasis sebanyak 50%. Rataan indeks mitosis dan angiogenesis terjadi penurunan yang nyata (p<0.05) pada kelompok mencit yang diberi ekstrak etanol daun keladi tikus dibandingkan kelompok kontrol. Disimpulkan bahwa ekstrak etanol daun keladi tikus menurunkan pertumbuhan tumor dan dapat digunakan untuk terapi tumor.

Abstract

Study to determine the potential of tempe flour in improving bone quality of ovariectomized rats (ovx)based on calcium (Ca) and phosphorus(P) levels in the plasma and bones; osteoblast and osteoclastprofiles have been conducted for 4 months. A total of 20 female Sprague Dawley albino rats, age of 3months and average body weight of 200 g were used in this study. The rats were divided into 4 groups,i.e.normal rats + diets (K0); normal rats +tempe flour (K1); ovx + diets (OV0); and ovx + tempe flour (OV1),respectively. The Ca and P levels in plasma and bones were analyzed using Atomic AbsorptionSpectrophotometer (AAS) and spectrophotometric methods. In addition, the profiles of osteoblast andosteoclast were also observed. The results showed that Ca and P levels in plasma and bones of normal ratswere 11.08 mg/dL and 7.06 mg/dL; 30.88 mg/dL and 18.63 mg/dL, respectively. The highest Ca and P levelsin both plasma and bones were observed in normal rats which were given tempe flour (K1), 11.25 mg/dLand 7.42 mg/dL; and 31.60 mg/dL and 19.78 mg/dL, respectively. Similarly, Ca and P levels in plasma andbones of ovxwhich were given tempe flour were slightly higher (10.91 mg/dL and 6.96 mg/dL; and 29.89 mg/dL and 17.87 mg/dL) compared to ovx without tempe flour diets (9.38 mg/dL and 6.46mg/dL; and 26.58 mg/dL and 16.75 mg/dL). The numbers of osteblast was found slightly higher both in normal rats and ovxwhich were given tempe flour; whereas slightly decreased of osteoclast was observed only in ovx which weregiven tempe flour. The administration of tempe flour in ovx diets could improved bone quality as indicatedby the Ca and P levels in plasma and bones and decreased numbers of osteoclast.

Abstract

Isoflavone compounds have been known to have biological effects in improving health. The purpose ofthis study was to determine the bioavailability of isoflavone in plasma and its role as lowering bloodglucose levels in hyperglycemia rats. Bioavailability of isoflavones in plasma was performed in 18 ratsstrains Spraque Dawley age 3 months with 200 g of body weight on average and were given 300 ppm oralof isoflavone single dose. The analysis of isoflavon as done at 0, 1, 6, 12, 18 and 24 hours using HPLCmethod. Study effect on blood biochemical values used 25 male rats, which was divided into 5 treatmentgroups. One group as a negative control (K1) was given aquades; positive group hyperglycemia (K2), and 3other groups (K3, K4 and K5) were a positive groups of hyperglycemia and given isoflavone different doses,i.e. 1.0, 1.5 and 2.0 mg/200g bw per oral. Blood glucose were analyzed by using glucose oxidase biosensor,while cholesterol, HDL (Hight-density lipoprotein) and triglycerides were analyzed enzymatically using aspectrophotometer. The results showed the analyzes isoflavone genistein was more efficiently absorbedthan daidzein and glisitein. Isoflavone as found at highest concentration in plasma at 6 hours after oraladministration i.e: genistein of 11,17 ppm, daidzein of 2,86 ppm and glisitein of 2,47 ppm. Theadministration isoflavones at dose 2 mg/200g bw could decrease blood glucose levels in hyperglycemiarats, and also has hipocholesterolemia effect.

Abstract

This study aim at determining the activity of ethanol extract of Robusta green coffee beans as an antiproliferation agent in MCA-B1, MCM-B2, and Vero normal cell lines as a control. Ethanol extract of Robusta green coffee beans was obtained by maceration method, bioactive compounds in the extract were tested by phytochemical screening methods, testing levels of antioxidants were tested by 2,2-diphenyl-1-picrylhydrazyl (DPPH) method, cytotoxicity effects (LC50) were tested by the method Brine Shrimp Lethallity Test (BSLT) and antiproliferation activity were carried out by counting the number of cells after being tested with several extract concentrations namely 100, 250, 400, 550, 700 and 850 ppm and doxorubicin as a positive control with a concentration of 100 ppm. The results showed that the ethanol extract of Robusta green coffee beans contained flavonoids and tannins, had very strong antioxidant levels with IC50 values of 40.9923 ppm, cytotoxicity effects obtained LC50 values of 430.64 ppm, and the highest antiproliferation activity was achieved at a concentration of 850 ppm at 69.58 % in MCM-B2 tumor cells, 60.46% in MCA-B1 tumor cells, and 14.2% in Vero normal cells. Based on the results, the ethanol extract of Robusta Lampung green coffee beans has antiproliferation activity on the test tumor cells and is relatively non-toxic to normal cells.

Abstract

The experiment was designed to study the effects of sipatah-patah extract (ESP) on bone growth ingrowing female rats in order to prevent osteoporosis during postmenopausal period. Twenty growing femaleSprague Dawley rats in similar body weight were used with the average of age was 20 days old. Theexperimental rats were randomly divided into 5 (five) groups: control group (NOV-0, administered carboxylmethyl cellulose 1%) and groups administered ESP at the age of 30 days (NOV-1), 60 days (NOV-2), 90days (NOV-3), and 120 days (NOV-4) with dosage of 750 mg/kg body weight daily. Blood samples werecollected every 30 days to analyze serum calcium and phosphate concentrations. At the end of the treatment(180 day old), all of the animals were sacrificed to measure and analyze bone growth histologically. Ostibia-fibula slice was stained by using Hematoxylin Eosin (HE) method to observe osteoblast and osteoclastdensities, while Masson trichrome was used to observe trabeculae structure. The results showedadministration of ESP in rat during growing period improved bone growth. NOV-1 with longer duration(150 days) showed better growth rate with longer femur, optimum serum calcium, and phosphateconcentrations, and higher number of osteoblast with lower osteoclast densities compared to control rats(NOV-0). It was concluded that giving ESP at earlier age with longer duration during growth period couldimprove bone growth. This condition was expected could also improve the bone conditions during growingperiod.

Abstract

The purpose of this study is to examine the activity of Ambon banana extract in gel formulation on thewound healing process of mice skin (Mus musculus albinus) based on gross and microscopic observations.The wound healing process were observe grossly everyday while the microscopic lesion was observed on the3rd, 5th, 7th, 14th and 21st days after skin incisition. The wounded skin were sampled after the mice wereeuthanized for further microscopic observation. The gross parameters were the existence of blood coagulation,scab formation, wound covering and wound size. The microscopic parameters observed the percentation ofwound reepitelization and the the tickness of wound connective tissues (fibroblast). Gross lesion and thefibroblast thickness were presented descriptively as a qualitative data. Grossly, it was observed that thescab formation was faster in mice treated with the extract of ambon banana than that of negative control.Based on the research the ambon banana extrack in gel can be used in the acceleratiof wound healing.
