Abstract

Abstract:  Plants produce a variety of secondary metabolites, one of which is essential oils that contain a variety of volatile compounds and are useful for humans. Cymbopogon nardus contains volatile compounds that can inhibit the proliferation of cancer cells. This research aims to explore the antiproliferation activity of C.nardus’ essential oils with different concentrations against breast cancer Michigan Cancer Foundation-7 (MCF-7) cells. Antiproliferation test was carried out with total cells method using trypan blue and cells were calculated using a microscope. Gas Chromatography-Mass Spectrometer (GC-MS) was performed to identify the volatile compounds. The results showed that the Inhibitory Concentration 50 (IC50) value was 359.6 ppm with an inhibition percent of 44.49% at 170 ppm. Meanwhile, inhibition percent against Vero normal cell was 29.04%, compared to Doxorubicin 35.23%. The dominant volatile compound in C. nardus’ essential oil were Geraniol and Citronellol.Abstrak Tanaman menghasilkan berbagai macam metabolit sekunder, salah satunya dalam bentuk minyak atsiri yang mengandung berbagai macam senyawa volatil yang berguna bagi manusia. Cymbopogon nardus mengandung senyawa volatil yang dapat menghambat proliferasi sel kanker. Penelitian ini bertujuan untuk mengetahui aktifitas antiproliferasi minyak atsiri C. nardus dengan beberapa konsentrasi terhadap sel kanker payudara Michigan State Foundation-7 (MCF-7). Uji antiproliferasi dilakukan dengan metode total sel menggunakan trypan blue dan sel dihitung menggunakan mikroskop. Gas Chromatography-Mass Spectrometer (GC-MS) dilakukan untuk mengidentifikasi senyawa volatil. Hasil menunjukkan bahwa konsentrasi penghambatan 50 (IC50) sebesar 359.6 ppm dengan persen penghambatan sebesar 44.49% pada konsentrasi 170 ppm. Sementara itu, persen ihibisi terhadap sel normal Vero sebesar 29.04%, dibandingkan dengan Doksorubisin yang sebesar 35.23%. Senyawa volatil dominan yang terdapat pada C. nardus adalah Geraniol dan Citronellol.

Abstract

Background: Mucus is pre-epithelial gastric layer that may prevent damages due to direct contact between aspirin and gastric epithelial cells. The integrity of gastric mucosa and mucous cellular reaction may serve as primary and secondary prevention of extended aspirin-induced gastric mucosal damage. The aim of this study was to prove the function of mucus as defensive factor in rats. Method: The study was conducted in twenty white rats of the Sprague-Dawley strain at Department of Pathology and Clinical Reproduction, Bogor Agricultural University, between January and December 2008. The rat in the treatment group were given 400 mg aspirin diluted in aqua bidest through intra- gatric canules; while the control group received aqua bidest only once daily for 3 days. Necropsies, macroscopic and microscopic observation were performed by counting the number of Alcian blue- periodic acid Schiff-stained mucous cells at fundus/corpus and antrum/pylorus regions. Data analysis was performed using ANOVA and Duncan test. Results: The number of mucous cells with positive lesions in the treatment group was significantly different from the control group at both regions. There was no significant difference of negative lesions between treatment and control group at both regions. At antrum/pylorus region, there was no difference of negative lesions between treatment and control groups; however, both groups demonstrated significant difference of positive lesions in treatment group. Conclusion: In primary prevention for gastric mucosal lesions, there is no increasing number of mucous cells in normal mucosa. Increasing number of mucous cells is a secondary prevention against extended aspirin-induced gastric mucosal damage. Keywords: NSAIDs/ASA, mucus cells, gastric mucosal lesion, rat

Abstract

Background: Nonsteroidal anti-inflammatory drugs (NSAIDs) gastropathy is a common complication, which has characteristic symptoms of dyspepsia syndrome. Mostly, it includes epigastric discomfort with bloating and nausea. The aim of this study was to provide evidences that clinical symptoms of dyspepsia are related to macroscopic changes of gastric form in rats, which are expected to be applied in human. Method: The study was conducted in 20 white rats (Rattus norvegicus, Sprague-Dawley strain) at the Department of Pathology and Clinical Reproduction, Bogor Agricultural University between January and December 2008. The rats were divided to treatment group and control group and each group consisted of 10 rats. Acetyl salicylic acid (ASA/aspirin) was administered at 400 mg dose, diluted in distilled water and was given to the treatment group using gastric cannula, once daily for three days period; while the control group had received aquabidest only. Subsequently, necropsies were conducted for both groups, followed by macroscopic observation and measurement of sagittal and transversal diameter. Gastric incisions along the minor curvature were performed in both groups to recognize any macroscopic changes of gastric mucosa. ANOVA test was utilized for data analysis, which was followed by Duncan test when the results were significant. Results: Gastric diameters in treatment group with positive lesion were significantly different from the control group and the treatment group with negative lesion on anthrum/pylorus region, with p 0.05. Conclusion: Prominent gastric dilatation at anthrum/pylorus region found in the treatment group may become the initial cause and signs of dyspepsia in human. Abstract

Nitrit oksida (NO) adalah salah satu molekul terkecil yang termasuk radikal. Ketika nitrit saliva memasuki lambung terjadi kombinasi asiditas asam askorbat dari asam lambung yang merubah nitrit menjadi NO. Konsentrasi NO intraluminal maksimal pada gastroesophageal junction (GEJ) dan daerah kardia sehingga dapat mengakibatkan stress nitrosatif. Tujuan: mempelajari patofisiologi NO terhadap abnormalitas sfingter GEJ. Metode: menggunakan 48 ekor tikus putih jantan galur Wistar, dikelompokan menjadi 1 kelompok kontrol dan 3 kelompok perlakuan berdasarkan dosis natrium nitrit (NaNO3) dan waktu nepkrosi. Semua kelompok perlakuan diberikan 1 ml HCl dan 1 ml asam askorbat dengan sonde pada bagian bawah GEJ dan kelompok kontrol hanya diberikan 1 ml aquabidest steril. Kemudian dilakukan pemeriksaan NO luminal, NO jaringan, pemeriksaan morfologi untuk mengetahui gambaran makroskopis daerah GEJ, gambaran mikroskopis pada densitas pleksus Meissner dan pleksus Auerbach, menilai terjadinya perpanjangan lamina propria dan menganalisis serbukan sel radang limfosit. Hasil: didapatkan nepkrosi hari kedua terdapat korelasi antara NO jaringan dan perpanjangan lamina propria didapat nilai r=-0.630 dengan nilai p<0.05, serta korelasi antara perpanjangan lamina propria dengan limfosit nilai r = 0.590 dengan nilai p<0.05. Pada terminasi hari ke-8 terdapat korelasi antara NO luminal dan densitas sel saraf pleksus Meissner dan pleksus Auerbach dengan nilai r = 0.758 dan nilai p <0.05. Kesimpulan: Pemberian senyawa NO3 meningkatkan kadar NO luminal yang menyebabkan terjadinya perubahan morfologi makro-mikroskopis, inflamasi serta peningkatan densitas sel saraf pada pleksus Meissner dan pleksus Auerbach di daerah sfingter GEJ.Kata kunci: GEJ, GERD, NO

Abstract

Virus Bovine Viral Diarrhea (BVDV) secara luas diakui memiliki dampak ekonomi yang signifikan pada ternak yang terinfeksi. Kerugian penyakit ini meliputi, hewan Persistenly Infection (PI) yang kurang sehat, penyakit reproduksi, penurunan produksi, pertumbuhan yang buruk, dan berpengaruh immunosupressif terhadap hewan ternak. Penelitian ini bertujuan untuk melakukan suatu kajian pola distribusi penyebaran dengan karakterisasi gejala klinis serta mendeteksi adanya kaitan pemeliharaan kandang sebagai faktor risiko sumber penularan penyakit BVD pada sapi potong impor. Jumlah hewan yang diambil 100 ekor sapi yang didapatkan dengan metode penghitungan kajian lintas seksional dengan pertimbangan penyakit ini sudah ada di Indonesia. Persebaran penyakit pada setiap kandang kandang A sampai H dengan total 80 ekor sapi terlihat sampel positif merata sebanyak 21 sampel pada posisi nilai S-N sebesar 0,3-0,7 di pengambilan awal dan meningkat pada nilai S-N sebesar 0,9-1,5 sebanyak 53 sampel. Peningkatan penyebaran pada kandang diluar kandang karantina mencapai 2,5x lipat mengindikasikan penyebaran BVDV cukup tinggi. Pengelolaan limbah buruk akan berpeluang 2,483 kali lebih besar menimbulkan hasil ELISA antigen positif bila dibandingkan dengan peternakan yang memiliki pengelolaan limbah yang baik (OR=2,483; CI=1,066-5,783). Program biosekuriti buruk akan berpeluang 2,667 kali lebih besar menimbulkan hasil ELISA Ag BVD positif dibandingkan dengan peternakan yang memiliki program biosekuriti yang baik (OR=2,667; CI=1,145-6,210). Bangsa Brahman Cross berpeluang tiga kali lebih besar memiliki peluang hasil positif bila dibandingkan dengan sapi yang memiliki ras non-Brahman Cross (OR=3; CI=1,269-7,091). Sedangkan umur sapi > dua tahun akan berpeluang 3,241 dibandingkan dengan umur sapi yang < dua tahun (OR= 3,241; CI=1,411-8,912). Impak studi penelitian terhadap jurnal ini adalah memberikan informasi distribusi penyebaran penyakit BVD asal dari hewan sapi impor dari Australia yang berguna sebagai pertimbangan tindakan preventif dalam upaya pengendalian penyakit.

Abstract

Dalam penelitian ini isolasi dan karakterisasi hemaglutinin Staphylococcus aureus dilakukan dengan teknik afinitas kromatograf Karakterisasi hemaglutinin yang dihasilkan dilajutkan dengan teknik elektroforesis menggunakan metode sodium dodecyl sulphate gel electrophoresis (SDS-PAGE) dan dilanjutkan untuk melihat pengaruh suhu dan enzim terhadap aktivitas hemaglutinin. Hasil penelitia menunjukkan bahwa komponen hemaglutinin Staphylococcus aureus yang telah diisolasi memiliki berat molekul 46 kDa. Aktivit Staphylococcus aureus dalam meghemaglutinasi hilang pada pemanasan 60° C dan pengaruh enzim proteolitik. Hasil ini mengindikasika bahwa hemaglutinin Staphylococcus aureus adalah protein.

Abstract

This research was conducted in order to examine the goblet cells proliferation in duodenum, jejunum, and ileum of laying hens due to exposured with protein of excretory/secretory (ES) of Ascaridiagalli adult worm. Thirty heads of laying hens were divided in to two groups. The first group was treated with 4,000 infective larva (L2) of A. galli and the second group was immunized with 380µg of ES andfour hours later was challenged with 4000 L2. All treatments were given orally using stainless steelcanule directly to the oesophagus. Data was taken on the 3, 6, 9, 12, and 15 days post immunization(p.i.). The goblet cells were determined by Periodic Acid Schiff (PAS) staining. The result showed that immunization was able to increased goblet cells proliferation significantly at 12 and 15 day p.i. on theduodenum, and at 9, 12, and 15 day p.i. on the jejunum, but goblet cells proliferation did notsignificantly on the ileum. From this result we suggested that ES would beneficial in the strengthen thehost’s defence mechanisms in the intestinal mucosa.Keywords: Ascaridia galli, excretory/secretory, goblet cells

Abstract

Tujuan penelitian ini adalah mengetahui kemampuan hemaglutinin pada Staphylococcus aureus (S. aureus) dalam proses adhesi. Uji adhesi pada sel epitel ambing dilakukan dengan menggunakan metode Velentine-Weiggand. Hasil penelitian ini menunjukkan bahwa S. aureus yang memiliki hemaglutinin menunjukkan kemampuan adhesi yang jauh lebih baik (±380 bakteri/20 sel) dibandingkan dengan yang tidak memiliki hemaglutinin (±56 bakteri/20 sel).

Abstract

Javanese turmeric (Curcuma xanthorrhiza Roxb.) is important for study due to its increasing economic value in many aspects including marker-associated compounds for curcuminoids and xanthorrhizol. This systematic review aimed to summarize and find information about C. xanthorrhiza starting from its pharmacological activity and mechanism as an anticancer, anti-inflammatory, and antioxidant extraction process, and cultivation techniques. The data were obtained from relevant journals in national and international scientific databases PubMed, Scopus, Google Scholar, Web of Science, SINTA, ScienceDirect, and Wiley Online from 2000 to 2023. Results showed that from the perspective of anti-inflammatory pharmacological activities, xanthorrhizol inhibits pain response and inflammatory response. Its antioxidant activity showed a contribution to inhibiting oxidation which also successfully inhibits cancer cell proliferation. The extraction method of xanthorrhizol and curcuminoid compounds with Ultrasonic-Assisted Extraction (UAE) 20-2000 kHz performs better than other methods. C. xanthorrhiza cultivation techniques under shading produce better rhizomes. Based on research on cultivation techniques, extraction methods, and pharmacological activities, C. xanthorrhiza has an important role in the future.

Abstract

Breast cancer is a sickness caused by abnormal cell growth in the breast. Mahkota Dewa fruit or god’s crown fruit products (Phaleria macrocarpa) contain flavonoids, alkaloids, polyphenols, and tannins associated with active compounds. This work directs to influence the potency of P. macrocarpa fruit as an antiproliferative agent against breast cancer cells (MCF-7 cells). The antiproliferative potency of P. macrocarpa fruit was proved by extracting and fractionating P. macrocarpa fruit using maceration. The cytotoxicity of extracts and fractions was determined using Brine Shrimp Lethality Test (BSLT). The antiproliferative potency against MCF-7 cells was tested using the hemacytometer approach. This work demonstrates the crude ethanol extract, n-hexane fraction, ethyl acetate fraction, and water fraction. The LC50 values in crude ethanol extract, n-hexane fraction, ethyl acetate fraction, and water fraction were 13.72 ppm, 147.55 ppm, 405.81 ppm, and 149. 07 ppm severally. Phaleria macrocarpa fruit has shown antiproliferation potency against MCF-7 cells. The maximum part of crude ethanol extract antiproliferative potency (56 ppm) effectively suppressed MCF-7 cell growth by 70. 9% while doxorubicin (100 ppm) by 46. 92%. This work confirms that crude ethanol extract of P. macrocarpa fruit interacts synergistically as an antiproliferative compound against MCF-7 cells.
