Abstract

Penggunaan obat tradisional yang berasal dari tanaman dapat menjadi alternatif untuk mengatasi masalah kulit. Tanaman alpukat (Persea americana Mill.) diketahui memiliki banyak khasiat karena mengandung flavonoid, saponin, tannin, alkaloid, steroid, dan fenol yang merupakan antioksidan yang berperan dalam menangkal radikal bebas, sehingga dapat digunakan sebagai alternatif terapi pada beberapa kelainan kulit, serta menjaga kualitas dan penampilan kulit. Metode yang digunakan oleh penulis adalah kajian literatur dari jurnal nasional maupun internasional. Artikel dan jurnal yang didapat dibaca, ditelaah dan dianalisis untuk memberikan informasi baru yang relevan. Hasil penelusuran didapatkan bahwa tanaman alpukat, baik dari daging buah, daun, biji dan kulit memiliki khasiat dalam bidang dermatologi, diantaranya yaitu sebagai antibakteri pada antijerawat, antijamur, tabir surya, agen pemutih, pelembab kulit, dan antiaging. Dapat disimpulkan bahwa tanaman alpukat dapat digunakan sebagai alternatif untuk mengatasi berbagai masalah kulit, walaupun masih harus dilakukan penelitian lebih lanjut untuk menggali lebih banyak manfaat lain dari tanaman alpukat dalam bidang dermatologi. Abstract

African Swine Fever (ASF) is a disease affecting pigs, caused by a double-stranded DNA virus that is not transmissible to humans or other animals. It leads to significant economic losses due to high morbidity and mortality rates, particularly in densely populated pig regions like Bali Province. During the ASF outbreak from June to December 2023, this study conducted histopathological research on clinical samples. Biological materials from twelve pigs confirmed positive for ASF via qPCR examination were histopathologically analyzed. Tissue samples from various organs underwent processing and examination using hematoxylin and eosin staining. Immunohistochemistry (IHC) was employed to detect the ASF virus’s p54 protein. Clinical symptoms, anatomical pathology, and histological examination revealed characteristic acute ASF lesions. Immunohistochemistry consistently showed p54 viral antigen distribution in mononuclear cells/macrophages across various organs, with the spleen and lymph nodes being dominant sites in 12 pigs (100%). This comprehensive study demonstrates the effectiveness of IHC in detecting the ASF virus and characterizing its histopathology.Keywords: ASF; histopathology; immunohistochemistry

Abstract

Cardamom is one of the medicinal plants called perennial herbs from the Zingiberaceae family. Cardamom, with the scientific name Amomum compactum, contains various phytochemical compounds such as terpenoids, flavonoids, phenolics, tannins, sterols, and proteins that are pharmacologically useful as an anticancer, antioxidant, and have cytotoxic activity. Testing the total content of terpenoid compounds that affect the cytotoxic activity of A. compactum fruit using Box-Behnken Design with three independent variables (solvent ratio, ethanol concentration, and extraction time) has never been studied before. Thus, this study aimed to obtain the extraction optimization conditions on the total terpenoid content and cytotoxic activity using Artemia salina Leach larvae with the response surface method, namely Box-Behnken Design with Design Expert 13.0 application. Determination of the total terpenoid content of the A. compactum fruit extract was carried out based on the standard ursalic acid, and the cytotoxic activity was carried out using the Brine Shrimp Lethality Test (BSLT) method. The maximum total terpenoid obtained was 4.8970 mg UAE/g DW, and the minimum IC50 value was 38.8813 ppm. Optimal conditions for the extraction of A. compactum fruit obtained a combination of 1:8 mL/g solvent ratio, 50% ethanol concentration, and extraction time of 1 day with a desirability value of 0.772. Furthermore, the optimal extraction solution results from the Box-Behnken Design were verified and analyzed using a One-Sample T-Test. Using the Box-Behnken Design extraction optimization method, the content of terpenoid bioactive compounds with cytotoxic activity from A. compactum fruit can be optimally obtained.

Abstract

Artificial whitening agents available in the market can cause damage to melanocytes; therefore, a safe and naturally derived skin-whitening agent is needed. Avocado seeds (Persea americana Mill.) are often discarded as waste but possess numerous health benefits. One of the compounds in avocado seeds is catechin, a flavonoid metabolite. This compound has been reported to exhibit antioxidant activity and inhibit tyrosinase to prevent melanin formation, making it suitable for skin-whitening applications. This study aimed to conduct phytochemical screening, assess antioxidant activity using the DPPH method, analyze catechin content using HPLC, and perform cell viability tests using the MTT method from the ethyl acetate extract of avocado seeds. Phytochemical screening has revealed the presence of flavonoids, saponins, and steroids. The DPPH assay yielded an IC50 value of 89.47±0.73 ppm, indicating an intense antioxidant activity, and HPLC detected 0.09% catechin. The MTT test results yielded cell viability percentages ranging from 80-100% at test concentrations of 0.75-25 ppm, with an IC50 value of 61.7 ppm. In conclusion, our results indicate that the ethyl acetate extract from avocado seeds affects cell viability without toxicity, warranting further testing for tyrosinase inhibition in the mouse melanoma B16F10 cell line.

Abstract

BACKGROUND Open reduction and internal fixation are considered the gold standard for managing mandibular fractures, as they effectively restore anatomical structure and functionality. Three-dimensional (3D) miniplates were developed to obtain a geometrically stable structure. However, the rigid structure of 3D miniplate may cause screw holes to be located within the fracture lines or directly above important anatomical structures. Hence, we have developed customizable interlocking 3D miniplates that allow for modifications in the configuration of the horizontal miniplates and vertical cross-struts to meet specific requirements. METHODS This study included 24 goats divided into 2 groups: interlocking 3D miniplate and standard plate. Each group was further divided into subgroups sacrificed on Days 5, 28, and 56. Radiological assessments were performed to evaluate bone, muscle, peri-implant bone, and peri-implant muscle density. Histological assessments were performed using Masson's trichome (MT) for bone healing evaluation and hematoxylin and eosin (H&E) staining for biocompatibility. RESULTS Radiological assessment indicated an increase in density across all measured parameters from Day 5 to 56. Histological evaluation from MT staining showed a significantly higher fibrocartilage in the interlocking 3D miniplate group (6.109%, IQR 1.453–10.828) compared to standard plate (1.311%, IQR 0.636–2.553; p = 0.001). Furthermore, H&E staining showed significantly higher inflammatory cell counts in the standard group on Days 28 and 56 compared to the interlocking 3D miniplate group (p = 0.027). CONCLUSIONS This study concluded that the interlocking 3D miniplate showed better bone healing properties and biocompatibility than the standard plate.

Abstract

Bovine respiratory syncytial virus (BRSV) is highly prevalent in cattle. It is a major viral cause of bovine respiratory disease complex, which is associated with morbidity, mortality and substantial economic impact. Currently available treatments are only symptomatic, but no specific treatments are available for BRSV infection. This study aimed to identify new antiviral agents against BRSV, which could be used to control bovine respiratory disease complex in cattle with Indonesian herb extracts. Ethanol extracts prepared from Indonesian herbs including Andrographis paniculata, Phyllanthus niruri, Curcuma aeruginosa, and Curcuma xanthorrhiza were evaluated for anti-BRSV activity in A549 cells. The cytotoxicity of the herb extracts was evaluated using a CCK-8 cell viability assay. Antiviral activities of the herb extracts were examined using cell activity and cytopathic assays. The effect on virus production was evaluated by qRT-PCR and plaque-formation assays. Extracts of Curcuma xanthorrhiza (125 μg/ml), Andrographis paniculata (250 μg/ml), and Phyllanthus niruri (62.5 μg/ml) inhibited BRSV activity in A549 in pre-, simultaneously-, and post-infection treatment assays, respectively, as measured by the selective index. Reduction of BRSV activities by the herb extracts correlated with inhibition of viral gene expression and inhibition of plaque formation in a concentration- and time-dependent manner. Our findings suggest that these herb extracts have sufficient potency to be used not only as a therapeutic agent but also as a preventive agent to limit BRSV infection.

Abstract

Background: Zoledronate is an intravenous bisphosphonate that effectively treats osteoporosis by inhibiting osteoclast resorption, thereby reducing the risk of fractures. However, its use in post-extraction patients may increase the risk of bisphosphonate-related osteonecrosis of the jaws (BRONJ). Objective: This study aims to analyze the effects of zoledronate on alveolar bone structure in Sprague Dawley rats, focusing on changes in osteoblast, osteocyte, and osteoclast counts. Methods: This cross-sectional study employed an observational analytical approach. Samples were categorized based on zoledronate exposure. Osteoblast, osteocyte, and osteoclast counts were measured using ImageJ software at 40x magnification. Data were analyzed using SPSS, with one-way ANOVA employed for statistical analysis. Normality and homogeneity tests were conducted to ensure data validity. Results: Zoledronate exposure resulted in a decrease in osteoblast and osteoclast counts, while osteocyte counts increased. Statistical analysis showed no significant differences between the zoledronate-exposed group and the control group. Conclusion: Zoledronate reduces osteoclasts and increases osteocytes, potentially leading to cellular apoptosis in osteoporosis patients. The risk of BRONJ can be minimized by administering zoledronate at appropriate doses and maintaining optimal oral hygiene to prevent bacterial activity.

Abstract

Background: Black rice bran has the potential to be a functional food raw material due to its high content of bioactive compounds; however, it is susceptible to microbial contamination. This study aims to evaluate the effect of pasteurization on the microbiological quality of fermented rice bran tempeh using Rhizopus oligosporus and Rhizopus oryzae. Methodology: The study employed a comparative experimental approach, where fermented for 72 hours with R. oligosporus and R. oryzae, grown in PDA medium, with the addition of spore suspensions at 15% each (10⁶ spores/mL). After fermentation, the bran was divided into two groups: one group was steamed for 10 minutes, while the other group was not steamed. Both groups were freeze-dried for 24 hours and then stored at -18 °C for analysis of changes in the profiles of total microbes, total fungi, total enterobacteria, and total spores. The raw material of rice bran was also analyzed microbiologically as a control. Microbiological data were analyzed using the Generalized Linear Model (GLM) and further evaluated by Duncan Multiple Range Test (DMRT) at a 5% significance level, as determined by SPSS 22.0 software. Findings: The results showed the pasteurization on bran tempeh products was effective in reducing the total number of microbes, moulds, Enterobacteriaceae, and spores compared to those without pasteurization. However, bran tempeh with R. oligosporus culture between pasteurized and unpasteurized did not show a significant decrease (p>0.05), except for the results of the analysis of the total number of moulds which showed a significant decrease (p<0.05), which was <1 log CFU/g (pasteurization) and 3.52 log CFU/g (without pasteurization). It can be concluded that the 10 minutes pasteurization process is effective in reducing the number of Enterobacteriaceae, which are indicators of polluting and pathogenic bacteria. Contribution: this treatment has potential to be applied on a home industry and Micro, Small and Medium Enterprises (MSME) scale to improve safety and extend the shelf life of fermented food products based on bran.

Abstract

Sambiloto is one of the most diverse herbal plants. The active content found in sambiloto is believed to fight diseases, one of which is diarrhea. This study aimed to determine the content of secondary metabolite compounds with phytochemical tests, the antidiarrheal effectiveness of sambiloto leaf infusa using the intestinal protection method, and the effective concentration of antidiarrheal in mice. This study used 25 mice, which were divided into five groups: control (+), control (-), and three treatment groups of sambiloto leaf infusa with concentrations of 25%, 50%, and 100% given orally. The results of phytochemical tests of sambiloto that have the potential to be antidiarrheal compounds are alkaloids, flavonoids, steroids, terpenoids, saponins, and tannins. The parameters observed were the frequency of defecation and consistency of mice's feces every 30 minutes for 9 hours. The results of this study showed that bitter melon leaf infuse preparations at concentrations of 25%, 50%, and 100% had antidiarrheal activity, with the best antidiarrheal activity shown at a concentration of 25%.
