Abstract

The purpose of  study  was  to find out the effect of different concentrations of  soya bean extractin sperm diluter, stored at 5°C toward sperm viability and necrosis. The semen was divided into four groups :  P0: 0.1 ml sapudi ram  + 1 ml of diluent yolk citrate, P1: sapudi ram  0.1 ml + 1 ml diluent  soybeans extract konsentration 5%,  P2: 0.1 ml sapudi ram  + 1 ml diluent soybeans extract konsentration 10%,  P3: 0.1 ml sheep semen + 1 ml diluent soybeans konsentration 15%. The result showed that the dose of  soya bean extract diluter konsentration of  15% could increase the viability percentage and could decrease the necrosis percentage of sperm.

Abstract

The purpose of this study was to prove that the addition of Vitamin B12 in egg yolk skim milk extender can maintain the motility and viability of post thawing Sapudi sheep spermatozoa. This study used sample of Sapudi sheep fresh semen that was collected with artificial vagina, and divided into 3 different treatments. The control treatment or P0, Sapudi sheep semen was diluted in the egg yolk skim milk extender + vitamin C 0,02 mg / ml, treatment group (P1 and P2), Sapudi sheep semen was diluted in the egg yolk skim milk extender+ vitamin B12 5 μg / ml and grouped in P1, Sapudi sheep semen was diluted in the egg yolk skim milk extender + Vitamin C 0.02 mg / ml + Vitamin B12 5μg / ml and grouped in P2. The data analysis was use One Way Analysis of Variant (ANOVA), then to identify the real difference among the treatments continued with Duncan distance test. The result of data analysis showed that P2 treatment was significantly different (p <0.05) with P1 treatment and P0 treatment or control. While P1 treatment was not significantly different (p> 0,05) with P0 treatment or control. The result showed that the addition of vitamin C 0.02 mg / ml with vitamin B12 5 μg / ml combination in egg yolk skim milk extender was the best addition to maintain the motility and viability of the post thawing Sapudi sheep spermatozoa.

Abstract

The purpose of this study was to determine the effect and the best concentrasion of green tea extract in skim milk diluent for motility, viability, and intact plasma membrane Sapudi sheep spermatozoa that was stored 50C. The semen was divided into four groups: skim milk  diluent; 0,05 mg green tea extract in skim milk diluent; 0,10 mg green tea in skim milk diluent and 0,15 mg green tea extract in skim milk diluent. Spermatozoa quality was observed first day until fifth day after diluent. The data obtained was analyzed with the analysis of variance one way ANOVA. The result showed that the percentage of motility, viability and intact plasma membrane spermatozoa at the 5th day after diluent showed a significant differece (p<0,05) between 0,05 mg green tea extract in skim milk diluent; 0,10 mg green tea in skim milk diluent and 0,15 mg green tea extract in skim milk diluent. Motility at the 3th day after diluent respectively 46,67b ± 2,582; 43,33bc ± 4,028; 40,00ab ± 3,162; and 37,50a ± 2,739. Viability  at the 3th day after diluent respectively are 79,50b ± 1,643; 77,67b ± 3,327; 77,00ab ± 3,230; and 73,83a ± 3,061. Intact plasma membrane at the 3th day after diluent respectively are 50,17b ± 2,483; 47,00ab ± 3,899; 46,83ab ± 2,858; and 46,00a ± 3,286. The conclusion of this study was the green tea extract of 0,05 mg in milk skim diluent could increase the percentage of sperm quality until 5th day after diluent.

Abstract

The purpose of this study was to determinated the effect and the best concentration of greentea extract in skim milk and egg yolk diluent for quality sapudi sheep spermatozoa were measured in motility, viability and intact plasma membrane that was stored on cold temperature. The semen was devided into four groups; skim milk and egg yolk diluent, 0,5% green tea extract in skim milk and egg yolk diluent, 0,1% green tea extract in skim milk and egg yolk diluent, and 0,15% green tea extract in skim milk and egg yolk diluent. Spermatozoa quality was observed day 1, day 2, day 3, day 4 and day 5 after being diluted. The data obtained was analyzed with the Analysis of Variant (ANOVA), followed by multiple range test duncan. The results showed that the highest percentage of motility, viability and intact plasma membrane derived from green tea extract 0,15% on skim milk and egg yolk diluent  for 1 day storage is 86.66b ± 2.58, 92.00b ± 1.78, and 74.16b ± 3.25. The lowest percentage of motility, viability and intact plasma membranes was obtained from the skim milk and egg yolk diluent for 5 days storage is 43.33a ± 6.05, 56.50a ± 3.08 and 28.33a ± 2.80. The conclusion of this study is the addition of 0.15% green tea extract in a skim milk and egg yolk diluent can maintain the quality of sperm for up to 5 days of storage at cold temperatures.

Abstract

The lowest precentage of motility, viability and intact plasma membrane were obtained from egg yolk citrate diluent for 5 day of storage which were 26.66 ± 6.05, 49.16 ± 6.43 and 26.66 ± 4.32. Conclution of this research was the addition of 0,05 % green tea extract can be used as a diluent could maintain the precentage of spermatozoa quality until 5 days after diluent.

Abstract

This study aimed to determine the influence of mixture diluter of chicken egg yolk and young coconut water on the integrity of plasma membrane and abnormalities of Sapudi sheep spermatozoa. The experimental design used in this study was a complete random method. The data was analysed using ANOVA, and if there is a significant difference then proceed with Duncan's Multiple Range Test ( DMRT ) at significance level of 5%. Based on the result of the research, the influence of mixture diluter of chicken egg yolk and young coconut water was not significantly different ( p > 0,05 ) on the intact plasma membrane of spermatozoa on the first day until the fourth day. Interaction between treatment and storage duration had no effect on spermatozoa abnormality on the first day, second day and the fourth day. It can be concluded that mixture diluter of chicken egg yolk and young coconut water had a good effect on the plasma membrane and spermatozoa abnormalities up to the third day.

Abstract

Semen storage in cold temperatures might cause an increase in reactive oxygen species (ROS) production. This condition resulted in spermatozoa damage and quality decrease. This study was conducted to investigate the effects of α-tocopherol supplementation in diluents on the motility, viability, and plasma membrane integrity of Simmental bull spermatozoa after cooling. Semen samples were diluted in skim milk egg yolk supplemented with 0, 0.5, 1.0, and 1.5 mM α-tocopherol respectively for control, Tl, T2, and T3. Spermatozoa were evaluated for their motility, viability, and membrane integrity in cooling temperature (5°C). The daily evaluation showed that 1.5 mM α-tocopherol was the best in maintaining motility, viability, and plasma membrane integrity, while 1.0 mM α-tocopherol was only good for maintaining viability. Therefore, it can be concluded that α-tocopherol at the concentration of 1.5 mM was an efficient antioxidant supplement for Simmental cattle semen in skim milk egg yolk diluent.

Abstract

Repeat breeding is a reproductive disorder that harms dairy farmers because of the unachieved target to obtain one calf per year and decreased milk production. The repeat breeding syndrome can be caused by malnutrition. This study aimed to determine the effect of feeding silage, complete feed, and growth promoters in repeat breeder cows to restore the services per conception (S/C) and daily milk production. Thirty repeat breeder Holstein Friesian cows were divided randomly into three groups. The control group (P0) was fed 30-40 kg forage and 3-4 kg concentrate/head/day. Group P1 was given 8-10 kg silage or complete feed alternately every day; whereas P2 was given the same as for the P1 group, added with 10 ml growth promoter/head/day. Milk production and S/C were measured after six months of treatment. The results showed that the P1 and P2 groups returned S/C to normal (1.40 ± 0.52 and 1.30 ± 0.48), while the S/C of P0 group was 3.20 ± 0.63 (p <0.05). Daily milk production in the P1 and P2 groups (14.20 ± 1.40 and 14.60 ± 1.43 l/d) was also higher (p <0.05) than the control group (8.30 ± 1.06 l/d). It could be concluded that feeding silage and complete feeds alternately daily, with or without the addition of growth promoter, improved the S/C to the normal range and increased milk production.

Abstract

The purpose of this study was to determine the reproductive efficiency of dairy cows in the presence of non-specific bacteria in the reproductive tract. This study was a descriptive study of 20 healthy and cycling dairy cows. The identification of non-specific bacteria in the reproductive tract was conducted in the previous study. Data were collected from interview, questionaire, and direct observation at the beginning of the study. Detection of estrus was performed visually, followed by artificial insemination 12-18 hours later. Non-return to estrus was checked after 21 days. Artificial insemination was repeated up to three times on cows returning to estrous. Pregnancy was checked through rectal palpation 60 days after the last insemination. Pregnant cows were followed until calving. The results showed that the estrus rate, non-return rate (NRR), conception rate (CR), calving rate (CvR), services per conception (S/C), and days open (DO), were respectively 100%, 85%, 85%, 85%, 1.3, and 93 days. It could be concluded that the presence of non-specific bacteria in the reproductive tract of dairy cows did not affect the reproductive efficiency so that pregnancy and calving could still occur.

Abstract

The purpose of this study was to identify the correlation among the percentage of feed concentrate, milk urea nitrogen (MUN), milk production, services per conception (S/C), calving interval (CI), and days open (DO) of Holstein Friesian cows on smallholder farms. Twenty four, 4-6 year old, non-pregnant, lactating cows, with body condition score (BCS) 5-7, having given birth 2-5 times were recorded their daily milk production, S/C, CI, DO, and the percentage of concentrate in feed given. Milk samples were collected to measure MUN. The correlation calculation among the six variables resulted 15 correlation values, ten of which were not significantly different (p >0.05), including the correlation between the percentage of concentrate and milk production on MUN. The other five correlation values were significantly different (p <0.05). The increase in concentrate intake was moderately correlated (r = 0.52), affecting 27.04% of the increase in milk production, and weakly correlated (r = 0.39), affecting 15.21% of the increase in S/C. The increase in milk production had a very strong correlation (r = 0.82), affecting 67.24% of the increase in S/C. The increase in S/C had a strong correlation (r = 0.61), affecting 37.21% of the increase in CI. The increase in CI was also affected 38.44% by the increase in milk production with a strong correlation (r = 0.62). It could be concluded that dairy cows with higher feed concentrate intake would be followed by an increase in milk production, but had a negative effect on reproduction in the form of increased S/C and CI.
