Abstract

This study aimed to evaluate the protective effects of α-tocopherol and zinc sulfate on oxidative stress parameters in ovarian granulosa cells of female Wistar rats (Rattus norvegicus) exposed to lead acetate (Pb(C₂H₃O₂)₂). A total of 25 rats were randomly divided into five groups: Control (C), lead exposure only (T0), α-tocopherol + lead (T1), zinc sulfate + lead (T2), and α-tocopherol + zinc sulfate + lead (T3). Treatments were administered orally for 21 days. Superoxide dismutase (SOD) levels were measured spectrophotometrically, and p53 expression was analyzed using immunohistochemistry. The results showed that SOD levels significantly decreased in group T0 compared to the control (p <0.05), while treatment with either α-tocopherol (T1), zinc sulfate (T2), or their combination (T3) significantly improved SOD levels compared to T0. Moreover, p53 expression was markedly elevated in T0, indicating enhanced oxidative stress and potential apoptosis, whereas all antioxidant-treated groups showed reduced p53 expression, with T3 demonstrating values comparable to the control group. In conclusion, the combination of α-tocopherol and zinc sulfate provided a synergistic antioxidant effect, effectively enhancing SOD levels and suppressing p53 expression in granulosa cells of lead-exposed rats. This suggested a promising therapeutic potential of these compounds in mitigating lead-induced ovarian toxicity.

Abstract

Mineral and nutritive supplements improve animal reproductive performance, especially after Foot and Mouth Disease (FMD) infecteds, to establish sustainable food security solutions. This study aimed to investigate the effects of mineral addition, particularly through premix and concentrate booster supplementation, on the body weight gain and reproductive performance of Pote goats in Madura post-infected of foot and mouth disease (FMD). Thirty-six non-pregnant Pote goats that cured from foot and mouth lesion based on veterinarian examination, were divided into three groups and fed differently for 45 days. Group T0 received standard feed comprising 3-4 kg of forage and 300 g of concentrate with a crude protein content of 16–17%, along with ad libitum water. Groups T1 and T2 received standard feed supplemented with 15g/head/day of premix booster and immune booster, respectively. Results showed no significant increase in body weight with supplementation. However, higher blood urea nitrogen and albumin levels were observed in both T1 and T2-supplemented groups. The estrus rate was higher in goats given mineral supplements than with standard feed alone. Although the onset and duration of estrus did not differ significantly, ultrasound scans showed follicular development before estrus synchronization, indicating restoration of the estrus cycle. Overall, mineral addition in feeding post-FMD infected in Pote goats appears to enhance reproductive performance and overall health.

Abstract

Background : Chronic monosodium glutamate (MSG) exposure is linked to testicular toxicity, impacting spermatogenesis and oxidative stress markers. Objective : This study investigated the protective effects of Dayak onion (Eleutherine palmifolia) extract against MSG-induced testicular damage in mice. Methods : Twenty-five male mice (11 weeks, ~20 g) were divided into five groups: negative control (C-, 0.5% CMC-Na), positive control (C+, MSG 4 mg/g BW), and treatment groups (T1, T2, T3) receiving MSG plus Dayak onion extract (30, 60, 120 mg/kg BW, respectively) for 52 days. On day 53, testicular histology and serum malondialdehyde (MDA) and superoxide dismutase (SOD) levels were analyzed. One-way ANOVA and Duncan’s post hoc test assessed differences (p<0.05). Results : Group C+ showed significantly reduced spermatogenic cell counts, elevated MDA, and decreased SOD compared to C-. Group T1 exhibited no significant improvement in spermatogonia and spermatocytes but increased spermatids. Groups T2 and T3 significantly improved spermatogenic cell counts, with T3 showing the closest recovery to C-. MDA levels decreased, and SOD activity increased dose-dependently, with T3 nearing C- levels. Conclusion : Dayak onion extract, particularly at 120 mg/kg BW, mitigates MSG-induced testicular toxicity, preserving spermatogenesis and balancing oxidative stress markers.
